Abstract

The objective of this research was to examine the correlation between estrogen level and estrous cycle of rat fed with soybean flour and tempeh flour. A completely randomized design was applied in this study with 6 treatment groups and 5 replications. Rats in group 1, 2, and 3 were non-ovariectomized rats which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/ 100 g body weight/day, respectively. The rats in group 4, 5, qnd 6 were ovariectomized rat which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/100 g body weight/day, respectively. The rats were after fed with pellet, soybean and tempeh flour for 4 weeks, the estrous cycle phases of rats were examined based on the presence of vaginal epithelial cells and the number of qualitative vaginal epithelial cells, while the estrogen concentrations in serum were measured by radioimmunoassay method. The data was analyzed using analysis of variance and followed by Duncan multiple range test at 95% confidence. Non-ovariectomized rats given tempeh flour have longer estrus cycle than the non-ovariectomized rats which given soybean flour. The isoflavon contained in tempeh flour and soybean flour induced proliferation and cornification of vaginal epithelial cell. Feeding the soybean flour and tempeh flour to ovariectomized rats could optimize the estrogen hormone in initiating the estrus phase, in which of the tempeh flour addition had better effect than soybean flour. Correlation between the level of estrogen to the length of estrous cycle in both nonovariectomized rats and ovariectomized rats was positive, in which the higher the levels of estrogen the longer the length of estrous cycle.

Abstract

The study aimed to identify genetic variation of cDNA sequence from low density lipoprotein receptor (LDLR) gene of hyporesponder cynomolgus macaques. The animal used in this study was one hyporesponder cynomolgus macaque obtained from selection result in Primate Research Center-Bogor Institute of Agriculture (PSSP IPB). Amplification of cDNA from LDLR gene was performed using polymerase chain reaction (PCR) method with 4 pairs of walking primer. Alignment of amplification result sequence from 4 pairs of walking primer generated target sequence of 2353 bp which located on position 188-2540. Analysis of target sequence alignment on reference sequence in GenBank found 8 single nucleotide polymorphism (SNP), namely c408CT; c.1200CT; c.1497CT; c.1644TC; 1791TC; 1804AG; 2088CT; and 2377GA. Of 8 SNPs, c.1804AG and 2377GA. Two SNP (c.1804AG; dan 2377GA) caused changing of amino acids composition namely p.K602E (lysine glutamate) and p.V793I (valine isoleucine). This result proved the potential use of genetic variation of cDNA sequence from LDLR gene as genetic marker for selection of hyporesponder cynomolgus macaques.

Abstract

Background: The Banteng population in Ujung Kulon National Park (TNUK) is less than 500 individuals. The habitat of Java Banteng in conservation areas has largely decreased. One approach to assessing the current status of biodiversity at all levels, especially in endangered species, is to use geospatial technology such as remote sensing and geographic information systems combined with spatial data science. This study aims to create a spatial model of the suitability of the Javan Banteng habitat in the TNUK area and identify the use of the Java Banteng habitat and environmental variables that affect the presence of Javan Banteng. Methods: This research data was collected through coordinate data for stool sampling and data from BTNUK using a method called maximum entropy (maximum). The analysis used the Relative Use Index, Maximum Entropy modeling, and Relative Abundance Index. Results: Based on research, the use of habitat by Java Banteng with the value of making a spatial distribution model can be analyzed by analyzing the contribution of environmental variables based on the level of contribution in percent and the results of the jackknife test, namely the percentage of contribution of environmental variables in this study showed that environmental parameters, slope (37.6%) were the highest parameters, followed elevation (25.8%), land cover (25.3%), and NDVI (6%),  rivers (5.3%). The analysis of five environmental variables used in making the Javan Banteng distribution model showed that at an altitude of 45 meters above sea level, Java Banteng preferred to show 95%. The graph decreased at an altitude above 45 mdpl, and Java Banteng at 200 meters above sea level looked at 0%. Conclusions: Javan Banteng do not like or do not choose places with altitudes ranging from 200 - 625 meters above sea level.

Abstract

This research aims to analyse the variation on morphometric and cytochrome b (cyt b) DNA mitocondria gene of Ompok hypopthalmusin Batang Hari river. Cyt b DNA mitocondria gene was amplified by polymerase chain reactionand targeted1104bp nucleotids. The results show there is no significant differentiation on morphometric structure of the samples. This result is consistent for both of morphometric (p=0.96) and index (p=0.99). Amplification of cyt b DNA mitocondria gene generates927 bp nucleotids with cytosin as the majour component and guanin as the lowest. Only one nucleotide(0.11%) is vary among all the sequences i.e.883thof the partial cyt b gene. The 883th nucleotidein O. hypopthalmus Simpang and Sungai Bengkal is guanin, while inMandiangin dan Pelayangan is adenin. This subtitution occured on the first codon and changed the 295th putative amino acid ofcyt b gene. The putative amino acid in O. hypopthalmus Simpang-Sungai Bengkal is alanina (GCC), while inMandiangin-Pelayangan is treonina (ACC).

Abstract

Gambier is a dry extract obtained from both the leaves and twigs of the Uncaria gambir. It is an agricultural product produced by the local community through traditionally unique methods. The main objective of this study was to evaluate the antidiabetic potency of gambier. The study used two types of gambier, namely traditional aqueous gambier (TAG) from traditional extraction and maceration ethanol gambier (MEG) from maceration extraction. The phytochemical compound, alpha-glucosidase inhibition, and antioxidant activity were analyzed. Moreover, in vivo hypoglycemia activity study was carried out. The results showed that both TAG and MEG contained flavonoids, tannin, and triterpenoids, while saponins were detected in TAG and steroids in MEG only. Both TAG and MEG also contained quercetin and catechin which were higher in the TAG compared to the MEG. The DPPH scavenging activities of the TAG and MEG, measured in IC50 values, were 15.40 ± 0.21 μg/ml and 12.25 ± 0.04 μg/ml, respectively. The IC50 values of the alpha-glucosidase inhibition of TAG and MEG were 35.84 ± 1.75 μg/ml and 83.14 ± 1.26 μg/ml, respectively. The best hypoglycemic activity was observed in the TAG group. The group given TAG and high glucose had lower blood glucose (127 mg/dl) compared to the group only given high glucose (178 mg/dl). These results indicated that TAG had a better potential for antidiabetic activities compared to MEG, and it can be proposed for further investigations as a potential antidiabetic agent.
