Abstract

The aim this study was to determine the effect of Monostearin and Miglyol 808 lipid ratio in NLC system formulation resulting in physicochemical characteristics, release rate, and penetration rate. The NLC making was done by using emulsification method. In the formulation of NLC meloxicam, 3 different lipid ratios were used, including ratios of 6:4, 7:3, 8:2. Meloxicam served as active ingredient, monostearin served as solid lipid, miglyol 808 served as a liquid lipid, and tween 80 was surfactant. NLC meloxicam physicochemical characteristics include tests of organoleptic, pH, viscosity, particle size, particle morphology and entrapment efficiency. NLC meloxicam belongs to semisolid preparations with pH value range of 5,72-5,87. Increasing viscosity of NLC system are cause by increase of solid lipid. The measurement results of particle size of three different lipid formulas indicated that the lipid particle size was <1000 nm. Test of NLC particle morphology by using Transmission Electron Microscopy (TEM) indicated the spherical particle shape (round). Entrapment efficiency test of all NLC-lipid compositions revealed quite high result (> 80%). The determination of release rate (flux) and penetration rate (flux) was conducted by using Franz diffusion cells with a cellophane membrane for the release and Wistar rat’s skin membrane for the penetration. The release rate values of three NLC meloxicam formulas showed p value (sig) 0,005, while the penetration rate obtained p value (sig) 0,091. Abstract

The purpose of this research was to design an implant for a ciprofloxacin-based drug delivery system by combining bovine hydroxyapatite and collagen with genipin as the crosslinking agent. The production of ciprofloxacin implants using bovine hydroxyapatite:collagen blend (70:30). In addition, this synthetic preparation was made using three various concentrations of genipin (0.6, 0.8, and 1.0%). The pellets were created by compressing the implants. The tablets are cylindrical with a diameter of 4.0 mm and a weight of 100.0 mg. Ciprofloxacin cultures were characterized for swelling rate, porosity, density, compressive strength, morphology (SEM), dose, and drug release in vitro. The addition of genipin as a crosslinking agent may maintain ciprofloxacin release consistent with in vitro therapeutic levels of ciprofloxacin. These results are supported by compressive strength data, where the addition of genipin concentrations induces higher implant stiffness and scanning electron microscopy photomicrographs reveal small pore sizes and BHA adhere to collagen fibers so that ciprofloxacin is completely dispersed in the implant after cross-linking with genipin. As a drug delivery system for osteomyelitis, it can be concluded that the use of genipin as a cross-linking agent can sustain ciprofloxacin release commensurate with in vitro therapeutic levels of ciprofloxacin for 30 days.

Abstract

Nanostructured Lipid Carrier (NLC) appears as the next generation of lipid nanoparticles which overcomes the weaknesses of Solid Lipid Nanoparticles (SLN). NLC is a delivery system that offers several advantages, such as significantly improved entrapment efficiency, reduced drug expulsion during long-term storage, and increased contact between the drug and the stratum corneum, thereby enhancing drug penetration. The methods used to manufacture NLC have high energy-required such as high-pressure homogenization and high shear homogenization, low energy-required such as microemulsification and phase inversion, and very low or no energy-required such as solvent evaporation and solvent injection. The basic components of an NLC are solid lipids, liquid lipids, and surfactants or mixtures of surfactants. This narrative review refers to several previously published databases on the types, preparation methods, and physical properties of NLC. This review focuses on descriptions related to the NLC as a topical drug delivery system.
