Abstract

FTA Cards are an easy way to collect blood samples, deliveries, and stores. The totalvolume of blood samples is less is needed when compared to the blood samples taken through a vein(venipuncture). The purpose of this research is a method that can be applied to isolate DNA samplesfrom FTA Cards using the Wizard® Genomic DNA Purifi cation Kit (Promega), PureLink® GenomicDNA Kits, and Chelex-100. This research was a type of experimental research, and data analysis wasshowed in qualitative and quantitative descriptively. The samples used were 16 samples of human bloodtaken using a lancet and then placed on the FTA Card, sampling was done by accidental samplingtechnique. The variations of the PureLink® Genomic DNA kits method with the addition of SDS 10%showed thin and smear DNA bands on agarose gels. Furthermore, the Chelex-100 method showed aclear DNA band. The purity data obtained varied between 1 to 1.1 and concentrations between 190 to950 ng/μl. The conclusion of this study is DNA kits method with the addition of 10% SDS and theChelex-100 method still have impurities, with the purity value of DNA was less than 1.8 and a lowconcentration to be used as a DNA template for further molecular analysis.

Abstract

One of the important DNA repair pathways is base excision repair (BER), which plays in the human body to maintain DNA integrity, prevent cancer and DNA damage. X-ray repair cross-complementing group 1 (XRCC1) is the most important DNA repair protein in base excision repair (BER) pathways and has been reported to have a relationship with the risk of developing various cancers. The study was purposed to assess the genetic polymorphism of XRCC1 exon 6 and 10 as a risk factor for thyroid cancer. A total of 90 participants were enrolled in this study, consisted of 30 thyroid cancer patients as a case group and 60 non-cancer patients as a control group. Examination of XRCC1 genotypes was carried out by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method and statistical analysis using a Chi-square test. In this study, we reported the frequency of XRCC1 genetic polymorphism was not significantly different between cancer patients and control groups both in exon 6 and 10, and we predict that these polymorphisms were not a risk factor for thyroid cancer (P-value>0.05). In conclusion, both mutant variants of XRCC1 exon 6 and 10 are not risk factors for thyroid cancer. In further studies, it is necessary to assess genetic polymorphisms in populations with controlled non-genetic factors, such as diet, lifestyle, and environmental factors.

Abstract

Objective: High number of antibiotic resistance cases in livestock will impact on increasing cases of microbe’s infection. On the other hand, the use of synthetic antibiotics is currently limited. A possible alternative can be used is the utilization of foods that possess medicinal effects (nutraceuticals). The use of animal feed sourced from Diptera larvae has been increasing widely, due to its high protein content. In addition, it is comprised of nutraceuticals as antimicrobial peptides like lucifensin. This study aimed at determining the potential of lucifensin as a nutraceutical product for livestock based on bioinformatics study.Methods: This study is descriptive research. The lucifensin protein data was obtained from the Protein Data Bank (PDB). The protein relationship analysis was performed using Blast-NCBI and lipinski Rules of 5 (Ro5) analysis to determine its potential as an oral drug (bioavailability). The molecular docking was used Swiss Docking online with lucifensin as target protein and Lipoteichoic Acid (LTAs) of Staphyllococus aureus as ligand.Results: The relationship analysis results showed that the lucifensin protein was related to two proteins of the other orders of diptera such as phormicin and sapecin. This protein belongs to the defensin protein family in which its coding genes are conserved until the order level. The Ro5 results showed that the mass of lucifensin protein was 312 daltons, its high lippopolycity was -0.5, it has five hydrogen bond donors, six hydrogen bond acceptors, and its molar refractivity was 77.14. Toxicity test also showed that this protein is classified as a safe component, and the docking results illustrate a strong energy bond that plays an important role in antimicrobial activity.Conclusions: In conclusion, lucifensin protein in the Lucilia sp. larvae is potential to be used as a livestock nutraceutical source that acts as an antimicrobial peptide.

Abstract

An autoimmune disorder is an abnormality that causes a disease. It is caused by a weakened immune system. One of the autoimmune diseases is Sjogren’s syndrome, which affects the salivary and lacrimal glands and causes dry mouth, dry eyes, and dry skin. Sjogren’s syndrome influences humans of every age, with the symptoms occurring at the age of 45–55 years and rarely in children. One of the factors causing Sjogren’s syndrome is genetic disorders. To identify genes that can influence Sjogren’s syndrome in this study, we used several databases, including GWAS Catalog, HaploReg Version 4.1, GTEX portal, and Ensembl, particularly to identify the gene expression profiles of TNIP1, TNFAIP3, and IRF5 and the quantitative properties of locus’ expression. This research showed that the missense variants and splice donor rs2233290, rs2230926, and rs2004640 influenced the susceptibility of autoimmune diseases, especially Sjogren’s syndrome, in the fibroblast tissue, sigmoid tissue, sigmoid colon, skin, esophagus, and adrenal glands. The allele frequency of each variant was then assessed in African, American, European, and Asian populations. Our data showed that TNIP1, TNFAIP3, and IRF5 genes in African and American populations had higher frequencies than in the Asian population. This implies that the last of the aforementioned populations might be relatively susceptible to the autoimmune disease Sjogren’s syndrome.

Abstract

Association studies of SNPs have become very important in determining how genetic variants are linked to complex diseases, quantitative traits, and physiological responses. Genetic polymorphisms in the GLP-1R gene have potentially decreased protein stability and are associated with many diseases, especially diabetes and obesity. This study aimed to screen the expression and investigate the genetic polymorphism of the GLP-1R by using several beneficial bioinformatics tools. We observed database Ensembl, GTEx portal, and KEGG to identify the structure, expression, and molecular pathway of GLP-1R. In silico computational methods (SIFT, PolyPhen v2, PROVEAN, and PhD-SNP) were used to identify nsSNPs of the GLP-1R that potentially influence protein structure and function. I-Mutant was used to investigate possibly damaging nsSNPs, while GeneMANIA was used to investigate GLP-1R gene-gene interactions. GLP-1R is localized on chromosome 6p21, contains 13 exons, and has the regulation variant (CTCF, promotor, enhancer, and promotor flank region). GLP-1R is highly expressed in the pancreas to stimulate glucose-dependent insulin secretion and suppress glucagon secretion. Seven nsSNPs of the GLP-1R gene were found to be potentially deleterious: rs10305421, rs201672448, rs10305492, rs2295006, rs6923761, rs1042044, rs140642887, and rs10305510. I-Mutant server showed that nsSNPs rs140642887 was unstable, decreased GLP-1R protein stability, and impaired other genes' interaction and function (SP1, SP3, GNAS, and GCG). This study is the first in silico analysis of the polymorphic GLP-1R gene, and will serve as a great resource for developing precision medications to treat diseases associated with these polymorphisms.

Abstract

The Coronavirus Disease 2019 (COVID-19) pandemic has not yet been fully under public health control, which is still currently impacting a large number of people worldwide in 2023. Since the pandemic emerged, the growing number of publications related to TMPRSS2 as a SARS-CoV-2 receptor worldwide has increased rapidly with various findings and qualities. It is important to determine the trend of TMPRSS2 publication as no such studies currently exist that represent the publication trend related to this critical field of study. Here, we employed a bibliometric-based approach to evaluate the research trends of TMPRSS2 mechanistically as the SARS-CoV-2 receptor. We identified 1012 research documents published between 2020 and 2022 for this study. The most common document category was "Research Article" (646 articles, 63.84%) followed by "Review Article" (261 articles, 25.79%), and letters to editors (57 articles, 5.63%). Germany was the most cited country with a total of citations (9400 citations), followed by the USA (6409 citations) and China (1788 citations), respectively. In conclusion, given the impact of COVID-19, this study indicated TMPRSS2 as a SARS-CoV-2 receptor as a timely and highly relevant research topic.

Abstract

Kain ecoprint pada saat sekarang mengalami masa populer dan memiliki harga ekonomis yang tinggi, bahkan sampai menembus pasar internasional. Pembuatan kain ecoprint dengan mengembangkan keterampilan kain kreatif yang melibatkan pewarna alami dari alam seperti daun, ranting dan bunga merupakan salah satu produk yang mencerminkan karya yang kreatif dan berkarakter cinta lingkungan (ecofriendly). Kegiatan pengabdian ini bertujuan untuk meningkatkan keterampilan dan wawasan Ibu-Ibu PKK kampung wisata Bukit Nobita, Kelurahan Kampung Jua, Padang, dalam memanfaatkan potensi lingkungan yang ada disekitarnya untuk menghasilkan suatu produk/barang yang memiliki potensi ekonomis dan dijadikan salah satu produk kreatif dari kampung wisata bukit nobita nantinya. Metode pelaksanaan pengabdian meliputi beberapa tahap yaitu tahap observasi dan koordinasi, tahap persiapan, tahap pelaksanaan yang terdiri dari empat kegiatan dan tahap evaluasi. Hasil yang didapatkan dari pelatihan, Mitra telah bisa membuat kain batik ecoprint dan sudah memahami tahapan-tahapan pembuatan kain ecoprint. Kegiatan pengabdian ini meningkatkan keberdayaan mitra pada pengetahuan dan keterampilan, kreatifitas yang nantinya bisa digunakan untuk menjadi salah satu usaha atau kegiatan Ibu-Ibu PKK di kampung wisata Bukit Nobita.

Abstract

ABSTRAK Diabetes Melitus (DM) merupakan penyakit yang ditandai dengan kadar gula darah yang tinggi (hiperglikemia) akibat gangguan kerja insulin, gangguan insulin atau kombinasi keduanya. Studi Genomik DM di Indonesia khususnya di daerah pedalaman masih jarang dilaporkan karena kesulitan pengambilan sampel whole blood pasien DM. Saat ini, kartu FTA adalah alat yang sangat mudah untuk mengumpulkan, mengirim dan menyimpan sampel biologis. Penelitian ini bertujuan untuk mengetahui kualitas DNA yang diisolasi dari kartu FTA menggunakan metode Chelex-Saponin kemudian dilakukan analisis amplifikasi gen target yang berperan dalam DM menggunakan metode PCR. Sepuluh sampel kartu FTA pasien DM tipe 2 digunakan. Amplifikasi PCR menggunakan primer spesifik ABCC8, KCNJ11 dan TCF7L2. Hasil penelitian menunjukkan DNA genom berhasil diekstraksi dari kartu FTA untuk pasien DM tipe 2. Selanjutnya DNA dapat digunakan sebagai template untuk amplifikasi gen ABCC8 (100%), KCNJ11 (60%), dan TCF7L2 (40%). Penelitian ini dapat disimpulkan bahwa metode Chelex-saponin dapat diterapkan sebagai metode ekstraksi DNA sampel darah pada FTA cards pasien DM dan dilanjutkan dengan analisis genomik. ABSTRACT Diabetes Mellitus (DM) is a disease characterized by high blood sugar levels (hyperglycemia) due to insulin work disorders, insulin disorders or a combination of both. Genomic Study on DM in Indonesia, especially in the rural areas, is still rarely reported because of difficulties in whole blood patient DM sample collection. Currently, the FTA card is a very convenient tool for collecting, shipment and storage of biological samples. This study aims to determine the quality of DNA isolated from FTA cards using the Chelex-Saponin method and then amplification analysis of target genes that play a role in DM using the PCR method. Ten samples of FTA cards of DM type 2 patient were used. PCR amplification was used with specific primers ABCC8, KCNJ11 and TCF7L2. Results of this stud showed DNA genome was successfully extracted from FTA cards for patient DM type 2. Furthermore, DNA can be used as a template for amplification of genes ABCC8 (100%), KCNJ11 (60%), and TCF7L2 (40%). This study concludes that the Chelex-saponin method can be applied as a method for DNA extraction of blood samples on FTA cards of DM patients and continued to genomic analysis. Abstract

SKBR3 cancer cells are cell cultures isolated from pleural effusion of a 43-year-old Caucasian woman with metastatic breast cancer. Many plant species are used in alternative medicine which has increased in recent decades for purposes related to breast cancer. The purpose of this systematic review is to make it easier to study the potential of bioactive compounds of medicinal plants and to analyze the effects of medicinal plants on cancer cells. This study uses a search strategy to retrieve articles, including the following items: related scientific questions, database to be used, selection of keywords, inclusion and exclusion criteria, definition of results and conclusions regarding the selected articles. Eight articles that met the inclusion criteria and had compounds derived from plants were tested in vitro on the SKBR3 cell line to see their toxicity. These plant bioactive compounds play an important role in the proliferative phase, cell cycle, fatty acid synthesis and apoptosis of cancer cells.

Abstract

Menjaga kesehatan serta kebugaran tubuh merupakan hal yang sangat penting. Memiliki tubuh yang sehat dan bugar dapat mencegah tubuh terserang penyakit. Kegiatan pengabdian kepada masyarakat (PKM) ini bertujuan untuk meningkatkan pengetahuan dan wawasan Ibu-Ibu PKK kampung wisata Bukit Nobita terhadap kesehatan kulit maupun penyakit metabolisme yang dilaksanakan di Keluarahan Kampung, Padang Sumatra Barat. Metode yang digunakan pada kegiatan ini adalah observasi langsung dengan memberikan penyuluhan dan pemeriksaan kesehatan pada 11 orang mitra. Hasil dari kegiatan ini bahwa terjadi peningkatan pengetahuan dan wawasan mitra terhadap kesehatan baik penyakit kulit dan penyakit metabolisme seperti diabetes mellitus dengan menjaga pola makan, kebersihan dan olah raga.  Kegiatan PKM ini diharapkan dapat mewujudkan masyarakat yang sehat melalui peningkatkan pengetahuan, sikap, dan kesadaran untuk bisa melakukan pencegahan dan pengendalian terhadap penyakit.
