Abstract

Malaria is an endemic disease that is still a problem globally, especially in countries with tropical and subtropical climates. People generally use mosquito repellents from synthetic materials, but they still cause effects and toxicity. Using active compounds from plants is an alternative to developing mosquito repellents. The 30 active compounds from the GC-MS analysis of methanol extract of Arabica coffee skin fruit and we selected the highest percentage compound, namely n-Hexadecanoic acid, Caffeine, Hexadecanoic acid methyl ester, 3-O-Methyl-d-glucose and desulphosinigrin. The highest compounds were carried out by molecular docking with Odorant binding protein 1 (OBP1) as the protein target and N, N-Diethyl-3-methylbenzamide (DEET) as the native ligand. Desulphosinigrin has the highest binding affinity, which is -6.2 Kcal/mol, close to the native ligand DEET. It can be concluded that the active compound desulphosinigrin has the potential as a repellent. This study concludes that active compounds from the methanol extract of Arabica coffee skin have the potential as a repellent.

Abstract

Kirsten rat sarcoma viral oncogene (KRAS) gene mutations lead to abnormal activation of the RAS signalling pathway and have been associated with poor prognosis and resistance to some therapeutics. This study aimed to identify mutation characteristics of the KRAS genes codon 12 and 13 in colorectal cancer patients in West Sumatra. KRAS mutations were analyzed in 20 DNA of colorectal cancer patients' tissue samples by using polymerase chain reaction (PCR) with specific primer and direct sequencing analysis. Our findings showed five samples (25%) with mutated KRAS at codons 12 and 13 (including three samples with GGT→GAT, one sample with GGT→GTT in codon 12, and one sample with GGC→GAC in codon 13). In conclusion, we found two variations of amino acid changes at codon 12 (G12D and G12V) and one at codon 13 (G13D). More research with many samples is required to obtain conclusive data on the relationship between these gene mutations and colorectal cancer response to therapy and prognosis.

Abstract

Recent scientific developments in the stem cell field have made it possible to create complex organoids, or structures that resemble complete organs, in vitro. In the majority of these methods, stem cells produced from stem cells or tissue progenitors are allowed to self-organize into three-dimensional (3D) structures using culture systems in three dimensions. The purpose of this study is to ascertain the use of organoid culture in the area of health. This kind of study employs the literature review methodology. The Preferred Reporting Items for Systematic Review and Meta-Analysis (PRISMA) criteria are referenced throughout the stages of the literature review performed in this study. The four processes that make up this level are identification, screening, eligibility, and acceptance. These discoveries showing that human stem cells and patient-derived pluripotent stem cells can be used to generate organoids open up a wide range of opportunities for modeling and disease development, toxicological research and medication discovery, and the field of regenerative medicine. Here, we discuss some of the most significant recent advancements in 3D human organoid production as well as the field's historical advancements. Finally, we discuss present restrictions and provide illustrations of how organoid technology may be used in the healthcare industry.

Abstract

Liver cancer is a global health challenge, with more than one million cases expected by 2025. Hepatocellular carcinoma (HCC) is the most common form of liver cancer and accounts for approximately 90% of cases. Nature provides a large source of natural medicines and compounds, one of which is plants. So, this article is made to contain information on the potential of several plants as antitumor agents. The method of preparing articles through literature review studies that collect articles from various sources such as ScienceDirect, google scholar, PubMed and others in the range of 2013 to 2023. There are ten plant bioactive compounds that have been tested in vitro on Hep3B cell lines to see their toxicity. These plant bioactive compounds play an important role in the proliferation phase, cell cycle, and cell apoptosis in liver cancer and have potential as antitumor.

Abstract

Colorectal cancer is one of the major health problems in Indonesia and the third most common cause of death worldwide. Colorectal cancer is a cancer of the colon and the rectum. There is considerable research evidence demonstrating the important role of complex gut microbiota communities in developing intestinal tumorigenesis. This study aims to isolate and identify gram-negative bacteria found in the colonic lumen fluid of colorectal cancer patients using the gram-staining method and biochemical test. From microscopic identification results, macroscopic gram staining, motility test, Simon citrates agar (SCA), and triple sugar iron agar (TSIA) tests against H1 and H2 isolates described gram-negative stem bacteria included in the Enterobacteriaceae family belonging to the species Shigella sp.

Abstract

Corn silk (Zea mays L.) is an agricultural by-product that is discarded as agricultural waste. Corn silk has secondary metabolite compounds that have the potential to become useful bioactive compounds. The purpose of this study is to recognize the bioactive potential of corn hair through compound identification and biological activity. The method used is preparation and extraction by maceration method followed by identification of compounds through LC-MS analysis method and Phytochemical test, bioinformatic PASS test based on data through PubChem, ChemSpider, and way2drug web. The results obtained from corn silk have a group of active compounds, including alkaloids, flavonoids, saponins, steroids and triterpenoids. Analysis of potential bioactivity received Acaciin compounds have high antioxidant bioactivity with a Pa value of 0.815, 2-arachidonoylglycerol has high potential antiinflammatory bioactivity with a Pa value of 0.736 and 1,2-di-O-methyl-4-[(2R)-2,4-dihydro butyramide shows moderate possible antibacterial activity with a Pa value of 0.643. This study concludes that corn silk has high bioactive potential as an antioxidant, anti-inflammatory and antibacterial.

Abstract

Cancer is a non-communicable disease and one of the highest causes of human death. It has been well documented that active substances have been shown in vitro to have anticancer properties. The purpose of this article is to examine the therapeutic implications of active substances involved in colorectal anticancer activity. We looked through the databases of PubMed, Science Direct, and Google Scholar in our systematic review to find papers released between 2013 and 2023. Ten studies were selected and published between 2013 and 2023. From the results obtained on plant data used as colorectal anticancer, it was reported that plants contain active phenolic compounds, flavonoids, alkaloids, gallic acid, saponins, triterpenoid/steroid, hydroquinones, triterpenoids, phenols, glycosides, quercetin, kaempferol, artemisinin, and tannins that can act as colorectal anticancer agents and are proven to be an inhibitor of cell line growth in colorectal cancer.

Abstract

Malaria is one of the main public health problems in Indonesia. Anopheles sp. mosquito larvae is vector of malaria that must be treated before it develops into a mosquito. Control of larvae by using larvicides containing hazardous chemicals causes side effects on the environment and resistance. This study aimed to determine the effect of giving the Arabika Coffee (Coffea arabica L.) pulp extracts on the mortality of Anopheles sp. larvae. Arabika coffee pulp extracts was fractionated with polar, semi-polar and non-polar. This study used ten larvae of Anopheles sp. Observations of larval mortality were carried 24 hours with five concentration series (0, 125, 250, 500, and 1000 ppm). The results of the mortality test showed that non-polar fraction of Arabika coffee pulp extracts had the highest larvicidal activity with values of LC50 and LC90, 278 ppm and 22950 ppm. In semi-polar and polar fractions, the LC50 and LC90, 1718 and 92683 ppm, 34085 and 40862 ppm were obtained. The results of compound analysis obtained 16 active compounds from non-polar extract of arabika coffee pulp with the five highest compounds, namely Hexadecanoic acid, 9,12-Octadecadienoic 15-Octadecenoic acid, Octadecanoic acid and 3-Furanacetic acid. Non-polar fraction of arabika coffee pulp extracts had larvicidal activity against Anopheles sp.

Abstract

This study aims to identify the types of methods and effects of human sperm cryopreservation. This review was written using a literature review method from international databases such as Scopus and PubMed. The results of the research show that by choosing a cryopreservation method, choosing a cryoprotectant agent such as TAT-PRDX2, canthaxanthin, or Holotheria parva coleomic extract, can increase sperm motility, viability and integrity after cryopreservation, the right cryoprotectant agent can defend cells from oxidative stress and fragmentation DNA.
