Abstract

Hepatotoxicity is a condition characterized by liver cell damage caused by toxic chemicals. This article aims to explore the effects of compounds that contribute to mitigating hepatotoxicity, focusing on in vitro studies, particularly primary cell culture. In this systematic review, we conducted searches in the PubMed, Science Direct, and Google Scholar databases to find articles published between 2008 and 2022. Several active compounds were identified, including N-Benzylpiperazine (BZP), 1-(3-trifluoromethylphenyl) piperazine (TFMPP), antimycin A, coenzyme Q (CoQ) substrates, the antioxidant Vitamin C, L-glutamine (Gln), Nimesulide, Luteolin, glutathione, angelica sinensis polysaccharide (ASP) from Angelica sinensis, ammonium glycyrrhizin (CAG) from Glycyrrhiza glabra, L-arginine, silymarin from Silybum marianum, and glucurolactone (GA). However, only six compounds were shown to have remedial and treatment effects on hepatotoxicity and utilized primary cell culture with MTT Assay. These six compounds are Luteolin, Glutathione (GSH), Angelica sinensis polysaccharide (ASP) from Angelica sinensis, Ammonium glycyrrhizin (CAG) from Glycyrrhiza glabra, Antimycin A (AA), and Glutamine (Gln). The mechanism of action of these compounds involves preventing further damage to liver cells and repairing cells that have already been damaged. In summary, these compounds play a significant role in addressing hepatotoxicity caused by toxic substances and drugs.

Abstract

The continuous cell line known as T47D cells originates from the isolation of epithelial cells from the ductal breast tissue of a 54-year-old woman with metastatic breast cancer. Numerous plant species have been employed in alternative anti-breast cancer therapy, showing advancements in recent years. The aim of this systematic review is to assess the potential bioactive compounds from herbal plant extracts and analyze the effects of herbal plants on T47D breast cancer cells. This research employs a systematic approach to gather articles, encompassing related research questions, chosen databases, keyword selection, inclusion and exclusion criteria, as well as outcomes and conclusions regarding the collected articles. From the reviewed articles, several plants exhibit anti-breast cancer activity on T47D cell lines, namely Calamintha incana, Sterculia quadrifida, Psidium guajava, Uncaria nervosa, Syzygium aqueum, Capsicum annum, Bauhinia scandens, Lansium domesticum, and Garcinia cowa due to their content of bioactive compounds such as triterpenoids, alkaloids, phenolics, flavonoids, saponins, and tannins. These bioactive compounds from the plants exert significant influence on cell cycle, proliferation, fatty acid synthesis, and apoptosis of cancer cells.

Abstract

Jamur endofitik yang berasosiasi dengan mangrove Sonneratia sp. diketahui menghasilkan metabolit sekunder yang berpotensi sebagai antioksidan. Kawasan pesisir Mandeh merupakan habitat alami yang kaya akan Sonneratia sp., sehingga menjadi lokasi strategis untuk eksplorasi jamur endofitik.Tujuan dari penelitian ini adalah untuk mengeksplorasi jamur endofitik yang berasosiasi dengan mangrove Sonneratia sp., melakukan skrining aktivitas antioksidan secara in vitro, serta mengidentifikasi isolat potensial berdasarkan karakteristik makroskopis dan mikroskopis.Penelitian ini menggunakan metode eksperimen dengan tahapan sebagai berikut: isolasi jamur dilakukan melalui teknik direct planting, skrining aktivitas antioksidan dilakukan secara in vitro menggunakan metode DPPH, dan identifikasi isolat dilakukan berdasarkan karakteristik morfologi makroskopis dan mikroskopis. Hasil penelitian menunjukkan bahwa sebanyak 20 isolat jamur endofitik berhasil diperoleh. Berdasarkan hasil skrining aktivitas antioksidan, ditemukan empat isolat yang menunjukkan aktivitas antioksidan dengan kategori kuat, sedang, dan lemah. Isolat dengan kode T4L1C4 dan T3L2C3 menunjukkan aktivitas antioksidan kuat, sedangkan T3L2C2 menunjukkan aktivitas sedang. Berdasarkan karakteristik mikroskopis, ketiga isolat tersebut teridentifikasi sebagai anggota genus Fusarium sp. Kata kunci: Antioksidan; Endofitik; Jamur; Mandeh; Skrining

Abstract

Coffee fruit skin contains antioxidant compounds that can repair damaged tissues, especially those of reproductive or-gans. This study was designed to assess to what extent the metha-nolic extract of coffee fruit skin affected sperm characteristics and testicular histology in mice receiving 15% ethanol for 15 days. It used 25 male mice aged four months and employed a completely randomized design with two controls (K) and three treatments (P), namely K1 (without ethanol), K2 (15% ethanol), P1 (15% ethanol and the methanolic extract of coffee fruit skin, or MECS, at 125 mg/ kg BW), P2 (15% ethanol and MECS 250 mg/kg BW), and P3 (15% ethanol and MECS 500 mg/kg BW). These treatments were administered orally with a 1 mL disposable syringe for 15 days. The parameters observed were sperm characteristics (viability, morphology, and sperm count) and the microscopic structure of the testicles (lu-men area, diameter, and area of seminiferous tubules). These data were then analyzed using the one-way ANOVA, continued with the LSD and Duncan’s tests (P <0.05). The results showed that P2 (250 mg/kg BW) substantially improved sperm count (86.5x105±1.73 cells/ mL), sperm viability (87.26±0.05%), and the proportion of normal sperm morphology (93.33±0.004%) of mice with 15% ethanol-in-duced sperm damages. Testicular histology also confirmed improve-ments in spermatogenic cells, as evident from the lumen, diameter, and area seminiferous tubules after receiving P1 and P2. In conclu-sion, the optimum dose of the coffee skin extract for improving sperm quality and microscopic structures of mice testicles is 250 mg/kg BW.

Abstract

Tumbuhan mangrove Ceriops tagal diketahui sebagai salah satu sumber jamur endofitik yang mampu menghasilkan senyawa metabolit bioaktif dengan berbagai aktivitas biologis, termasuk potensi sebagai antibiotik untuk mengatasi masalah resistensi antimikroba yang terus meningkat secara global. Penelitian ini bertujuan untuk mengisolasi jamur endofitik Aspergillus spp. dari daun, akar, dan ranting C. tagal, serta mengevaluasi potensinya sebagai penghasil senyawa antibiotik terhadap Staphylococcus aureus, Escherichia coli, dan Candida albicans. Isolat dikultivasi pada suhu ruang selama 7 hari dalam kondisi statis, kemudian metabolit sekundernya diuji menggunakan metode difusi cakram (Kirby-Bauer). Sebanyak delapan isolat Aspergillus spp. berhasil diperoleh, yaitu AS7, AS8, AS12, AS26, AS47, AS51, AS53, dan AS85, yang menunjukkan variasi morfologi koloni serta potensi antibakteri. Hasil uji menunjukkan sebagian besar isolat mampu menghambat pertumbuhan E. coli dan S. aureus, tetapi tidak menunjukkan aktivitas terhadap C. albicans. Aktivitas terhadap E. coli umumnya lemah, kecuali AS8 (10,45 mm) yang tergolong kuat. Aktivitas terhadap S. aureus lebih tinggi, dengan tiga isolat AS7 (11,89 mm), AS12 (11,45 mm), dan AS51 (10,92 mm) masuk dalam kategori kuat. Dari delapan isolat yang diperoleh, AS7, AS8, AS12, dan AS51 menunjukkan aktivitas kuat terhadap Staphylococcus aureus dan Escherichia coli, namun tidak menunjukkan aktivitas terhadap Candida albicans. Temuan ini menegaskan potensi jamur endofitik mangrove sebagai sumber kandidat antibiotik baru.

Abstract

Ekowisata Sungkai Green Park (ESGP) terletak di kelurahan Lambung Bukit Kecamatan Pauh Kota Padang. Luas kawasan 4 ha dengan topografi berbukit, dan kekhasan konservasi habitat pohon Sungkai. Bentuk kegiatan ekowisata yang sudah terlaksana yaitu agrowisata dan perkemahan. Jumlah kunjungan masih didominasi oleh peserta kemah, sedangkan destinasi agrowisata belum menjadi daya tarik. Berdasarkan potensi kelurahan, dilakukan pengembangan ESGP dengan ikan hias. Tujuan kegiatan ini yaitu memberikan pengalaman dan keterampilan kepada anggota P4S Sungkai Permai dalam mengembangkan ESGP dengan kolam ikan hias.  Kegiatan pengembangan dilaksanakan dengan metode Participatory Action Research, melibatkan partisipasi aktif dan kolaboratif mitra P4S Sungkai Permai. Pada kawasan ESGP telah dibuat lima kolam yang diisi dengan 24 jenis ikan hias. Berdasarkan evaluasi kelayakan terdapat 12 jenis ikan hias yang layak dikembangkan pada kolam terbuka kawasan ESGP, yaitu Glofish Tetra, Glofish Tiger Barb, Dwarf Gourame, Angle Fish, Guppy, Platy Molly varian Golden Black, Glofih Danio, Komet Slayer, Red Jewel, Blue Jewel, dan Zebra Tilapia.

Abstract

Hypoxia, a condition characterized by insufficient oxygen supply, can cause tissue damage in vital organs, including the liver. The liver is particularly vulnerable to hypoxic injury, which may result in impaired function and hepatocyte death. Purslane (Portulaca oleracea L.) is known for its antioxidant properties, largely attributed to its flavonoid content, which can inhibit the production of reactive oxygen species (ROS). This experimental study aimed to evaluate the effects of acetone extracts of purslane herb on hepatic histopathological changes in hypoxia-induced rats. Five experimental groups were established, each consisting of six rats (n = 6): normal control (N), hypoxia (H), hypoxia treated with dexamethasone (H+DEXA), hypoxia treated with purslane extract at 150 mg/kg body weight (HP1), and hypoxia treated with purslane extract at 300 mg/kg body weight (HP2). Purslane extract was prepared using the acetone maceration method. Hypoxia was induced by exposing rats to a gas mixture of 10% O₂ and 90% N₂ for 10 consecutive days. Liver tissues were processed using paraffin embedding and stained with hematoxylin–eosin for histopathological evaluation, focusing on hepatocyte necrosis, inflammatory cell infiltration, and hemorrhage. Statistical analysis was performed using one-way analysis of variance (ANOVA) followed by Duncan’s post hoc test. The results showed that the 300 mg/kg dose of purslane extract significantly reduced hepatocyte necrosis (35.33 ± 2.52 cells), inflammatory cell infiltration area (2488.51 ± 112.82 µm²), and hemorrhage area (1031.10 ± 17.38 µm²) compared with the hypoxia group (p < 0.05). In conclusion, acetone extracts of purslane demonstrated significant hepatoprotective effects in hypoxia-induced rats, suggesting their potential as a natural therapeutic agent for the management of hypoxia-related liver injury.
