Abstract

Herba ketumpang air (Peperomia pellucida L) contains bioactive compounds such as alkaloids, saponins, flavonoids, and steroids which are thought to be efficacious as antihypercholesterolemia. The purpose of this study was to determine the effectiveness of herba P. pellucida extracts as antihypercholesterolemia in vitro and characterize the active fraction. Extraction using ultrasonication methods with stratified organic solvents, such us n-hexane, ethyl acetate, and methanol. The fraction of n-hexane, ethyl acetate, and methanol were tested for cholesterol reduction activity in vitro by photometry method, cholesterol was reacted with Lieberman-Burchard reagents then analyzed using a UV-Vis spectrophotometer. The fraction that has the best activity is characterized by using a Liquid Chromatographic Mass Spectrometer (LCMS). Ethyl acetate fraction had the best activity to reduce cholesterol with a value EC50 of 196.43 ppm. Based on phytochemical tests the active fraction of ethyl acetate contains alkaloids, flavonoids, saponins, and steroids. Results of LCMS analysis of ethyl acetate fraction at a retention time of 4.97; 7.82; and 10.80 are thought to contain active compounds of the flavonoid group namely velutin, casticin, and dihydrochalcone.

Abstract

Desa wisata merupakan sebuah kawasan pedesaan yang memiliki keunikan dan karakteristik khusus untuk menjadi destinasi wisata. Masyarakat disekitar desa wisata akan melakukan persiapan untuk memperbaiki kondisi desanya bersama-sama dengan aparat desa dan mempersiapkan diri menghadapi kedatangan wisatawan yang akan datang ke desa mereka. Ramea merupakan desa wisata yang dikembangkan di Kabupaten Pandeglang. Tujuan pengabdian ini adalah mengembangkan desa wisata Ramea melalui pengembangan ekonomi kreatif masyarakat dengan menggali potensi desa untuk menjadi desa wisata yang unggul. Pemberdayaan desa wisata Ramea dilakukan dengan pemberdayaan masyarakat melalui peningkatan usaha kecil mikro, pengembangan fasilitas infrastruktur penunjang wisata dan pengembangan aktivitas masyarakat dalam pengembangan pengelolaan desa wisata berkesanambungan. Proses dilakukan secara bertahap dan berkelanjutan. Hasil yang dicapai pemberdayaan yang dilakukan diikuti oleh sebagian masyarakat menjadikan harapan kesejahteraan dan akan timbul suasana yang baru dan meraka siap menerima wisatawan yang akan dating ke desa mereka. Analisis SWOT memperlihatkan bagaiamana desa Ramea dapat mengembangkan desa nya menjadi desa wisata yang mendatangkan kesejahteraan bagi masyarakat. A tourist village is a rural area that has uniqueness and special characteristics to become a tourist destination. The people around the tourist village will make preparations to improve the condition of their village together with village officials and prepare themselves for the arrival of tourists who will come to their village. Ramea is a tourist village developed in Pandeglang Regency. The purpose of community service is to develop the Ramea tourism village through the development of the community's creative economy by exploring the potential of the village to become a superior tourist village. The empowerment of Ramea tourism village is carried out by empowering the community through the improvement of micro-small businesses, the development of infrastructure facilities to support tourism and the development of community activities in the development of tourism village management. The process is carried out gradually and continuously. The results achieved by the empowerment carried out were followed by some communities making hopes of well-being and a new atmosphere will arise and ready to accept tourists who will come to their village The SWOT analysis shows how Ramea village can develop its village into a tourist village that brings prosperity to the community.

Abstract

Gandaria (Bouea macrophylla) plants contain antioxidant compounds that can reduce free radicals. Free radicals are one of the causes of various degenerative diseases. The purpose of this study was to determine the fraction that had the highest antioxidant activity, and to determine the secondary metabolites contained in the most active fraction of B. macrophylla seeds based on LCMS/MS analysis. Maceration of B. macrophylla seeds with ethanol solvent. Ethanol maserate was concentrated and fractionated by liquid-liquid extraction method using n-hexane, ethyl acetate and ethanol as solvents. Antioxidant activity was measured by the free radical scavenger method with 1,1-diphenyl-2-picrylhidrazil as a source of radicals and characterization of the secondary metabolites contained in the most active fraction using a Liquid Chromatography-Mass Spectrometer. B. macrophylla seed ethanol extract has a % yield of 31.75%. The ethyl acetate fraction of B. macrophylla seeds has the highest mass compared to other fractions and has strong antioxidant activity with an IC50 value of 2.95 ppm. The ethyl acetate fraction of B. macrophylla seeds contains tannins, phenolic, flavonoid dan alkaloid compounds. Based on the results of the identification of secondary metabolites by LC-MS/MS, the ethyl acetate fractions of B. macrophylla seeds contained phenylalalnine compounds.

Abstract

Soybean is a source of vegetable protein that is rich in nutrients and a source of producing anticancer bioactive peptides. This study aims to determine the optimum conditions for hydrolysis of soybean protein with papain enzyme and their cytotoxicity against the MCF-7 cells. Soybean protein was isolated using an acid precipitation technique. Then, the protein isolate was hydrolyzed using papain enzyme with variations of papain concentration 0,5; 1; and 5% (v/v) and variations of incubation time 0, 1, 2, 3, and 4 hours at 50 °C. The hydrolysates were tested for their degree of hydrolysis (%DH), molecular weight profile using SDS-PAGE, and cytotoxicity against the MCF-7 cells through an in-vitro assay. The most active hydrolysate was fractionated using Sephadex G-15 and characterized by the molecular weight by LCMS/MS. The result showed that the optimum condition for hydrolysis was 1% (v/v) of enzyme concentration and 3 hours of incubation time with a %DH value of 3.01%. Based on the SDS-PAGE result, the hydrolysate had protein bands in a lower range (<25 kDa). That hydrolysate has cytotoxicity with an IC50 value of 1.87 mg/mL, and the molecular weight of its bioactive peptide is 7.70 kDa.

Abstract

Degenerative diseases can be caused by free radicals, whereas antioxidants can overcome free radicals. Kedondong leaves (Spondias dulcis Parkinson) and soursop leaves (Annona muricata Linn.) are potential sources of suitable antioxidant compounds. This study aims to determine the antioxidant activity and the relationship between antioxidants and total phenolics from the combination of S. dulcis and A. muricata leaf extracts. S. dulcis leaves, and A. muricata leaves were macerated with 96% ethanol and then formulated with a combination of extracts (1:1, 2:1, and 1:2). Each was tested for its antioxidant activity by the DPPH method. The ethanol extract of S. dulcis leaves had an antioxidant activity value of 36.72±4.01 ppm with a total phenol of 76.84±0.50 mgGAE/g extract, an ethanol extract of A. muricata leaves of 81.45±8.19 ppm with a total phenol of 104.48±7.55 mg GAE/g extract and a combination of extracts with a ratio of ethanol extract of S. dulcis leaves and ethanol extract of A. muricata leaves (2:1) of 29.98 ± 0.37 ppm with a total phenol of 36.99 ± 0.98 mgGAE/g extract. The results showed that the phenolic content of the ethanol extract of A. muricata leaves was more significant than the combination (2:1), which had the most excellent antioxidant activity among the other comparisons. Total phenolic is inversely related to antioxidant activity. Antioxidant activity is not only played by phenolic compounds.

Abstract

Swamedikasi (pengobatan sendiri) meningkat dikalangan masyarakat, tidak terkecuali masyarakat di Desa Kutakarang, Cibitung Kabupaten Pandeglang. Hal ini perlu diimbangi dengan pengetahuan cara mengelola obat dengan baik dan benar agarr obat dapat memberikan efek terapeutik yang diharapkan dan menghindari kesalahan dalam penggunaan dan pengelolaan obat. DAGUSIBU adalah dapat, guna, simpan dan buang obat dengan benar merupakan salah satu upaya peningkatan penggunaan obat bagi masyarakat yang diselenggarakan melalui kegiatan pelayanan kesehatan oleh tenaga kefarmasian. Pelayanan kesehatan yang dapat diberikan tenaga kefarmasian kepada masyarakat yakni kegiatan pemberian informasi tentang penggunaan dan penyimpanan sediaan farmasi dan alat kesehatan. Tujuan pengabdian ini adalah peningkatan pengetahuan masyarakat dalam memahami cara mendapatkan, menggunakan, menyimpan dan membuang obat, serta memahami cara mengkonsumsi obat dengan baik dan benar. Metode yang digunakan adalah presentasi dan penyulihan. Peserta kegiatan sosialisasi tentang DAGUSIBU berjumlah 54 peserta. Para peserta yang hadir sangat antusias dalam mendengarkan penjelasan dan aktif bertanya terkait penggunaan obat dan penanganannya. Harapan setelah Pengabdian kepada Masyarakat ini, yaitu peserta dapat lebih memahami dalam mengkonsumsi dan mengelola obat dengan baik dan benar. Self-medication is increasing among the community, including the people in Kutakarang Village, Cibitung, Pandeglang Regency. This needs to be balanced with knowledge of how to administer drugs properly and correctly so that drugs can provide the expected therapeutic effect and avoid errors in drug use and management. DAGUSIBU is getting, using, storing and disposing of drugs properly is one of the efforts to increase the use of drugs for the community which is organized through health service activities by pharmacists. Health services that can be provided by pharmacists to the community are the activities of providing information about the use and storage of pharmaceutical preparations and medical devices. The purpose of this service was to increase public knowledge in understanding how to obtain, use, store and dispose of drugs, and understand how to take drugs properly and correctly. The method used is presentation and lecture. Socialization activities about DAGUSIBU went well. There were 54 participants in this activity. The participants who attended were very enthusiastic in listening to explanations and actively asked questions regarding the use of drugs and their treatment. The hope after this Community Service is that participants can have a better understanding of consuming and managing drugs properly and correctly.

Abstract

Oxidative stress is a condition in which the amount of free radicals and antioxidants in the body is not balanced, causing various diseases. One of them is a degenerative disease. Oxidative stress can be reduced by antioxidant compounds. Anacardium occidentale L. is a type of plant from the Anacardiaceae family. This plant is found in many trophic countries included Indonesia, and is believed to ward off degenerative diseases caused by oxidative stress. The purpose of this study was to analyze the activity of A. occidentale stem bark extract against DPPH radical inhibition (1,1-Diphenyl-2-Picrilhidrazil) and the profiling of secondary metabolites in their active fraction. The bark of A. occidentale was extracted by a gradient maceration method. The solvents used were n-hexane (nonpolar), ethyl acetate (semi-polar), and methanol (polar). The macerate was concentrated using a rotary vacuum evaporator, followed by DPPH radical inhibition analysis using a spectrophotometer. The most active extract was analyzed for chemical content by LC-MS/MS. The results showed that the extract of A. occidentale stem bark had antioxidant activity. Methanol extract obtained very strong and best antioxidant activity with an IC50 value of 47.13 ppm compared to ethyl acetate extract and n-hexane extract, which had IC50 values of 163.92 ppm and 501.72 ppm, respectively. Secondary metabolites that are thought to have antioxidant activity in the methanol extract of A. occidentale stem bark were epigallocatechin (EC) and epigallocatechin gallate (EGCG) compounds.

Abstract

Quercetin is a drug candidate with bioavailability but currently limited, necessitating the use of a delivery matrix, such as Santa Barbara Amorphous-15 (SBA-15). Rice husk ash, containing a significant amount of silica, served as a precursor in the synthesis of SBA-15, which can be functionalized with nanoparticles, including zinc, using sonochemical methods. Therefore, this study aimed to synthesize Zn/SBA-15 using sonochemistry and evaluate the potential as a drug delivery matrix for quercetin. SBA-15 was formed by sonication of a sodium silicate from rice husk and pore director Pluronic P-123, followed by impregnating Zn at 2%, 4%, and 6% w/w. The characterization of X-ray diffraction (XRD) and fluorescence (XRF) showed a broad peak at 2theta 18-30◦ with Zn levels of 1.89%, 3.69%, and 5.06%. Furthermore, the analysis of transmission electron microscopy (TEM) showed the presence of Zn nanoparticles with a range size of 4-14 nm. In vitro drug delivery experiments were carried out using Zn/SBA-15 at concentrations of 1:1 and 1:2 to quercetin. The in vitro test results for a drug loading of quercetin with Zn/SBA-15 6% 1:1 was 28.30 mg/g. The encapsulation efficiency was 10.96% and the drug release reached 33.64% after 240 minutes.

Abstract

Melanin is the main pigment thfunctions to protect the skin from exposure to ultraviolet light, excessive melanin synthesis can cause skin hyperpigmentation. A. bilimbi and B. macrophylla fruits have bioactive compounds such as phenolics, flavonoids that function as antioxidants and tyrosinase inhibitors. This study aims to formulate a gel preparation of a combination of fruit extracts of A. bilimbi and B. macrophylla, test antioxidant activity using the DPPH method, and test tyrosinase inhibition enzymes. Three formulas were made, namely F0 (Basis), FI (50x extract), and FII (100x extract). Physical evaluation includes organoleptic, homogeneity, pH, adhesion, spreadability, viscosity, viscosity and irritation. Test of antioxidant activity and tyrosinase enzyme inhibition. Then the data were analyzed descriptively and statistically. The results of the evaluation of homogeneous preparations, dispersion, pH, viscosity, mandatory, meet the standards. Adhesion below standard, irritation test (mild irritation). The results of IC50 antioxidants were F0 (2,352 ppm), FI (1,448 ppm), FII (419,74 ppm) and gel brand x (77,429 ppm). The results of the tyrosinase inhibition test were the highest concentration of 10% F0 (7.828%), a concentration of 10% FI (18.245%), a concentration of 10% FII (25.814) kojic acid gel (brand x) as a comparison with a concentration of 10% (51.211%). Abstract

Ambarella (Spondias dulcis) is a tropical plant which is known to have antioxidant activity. This study aims to determine the antioxidant activity of the n-hexane, ethyl acetate and methanol extracts of S. dulcis leaves through superoxide radical scavenging and to characterize the phytochemical compounds in the extract using LCMS/MS. S. dulcis leaf powder was macerated with n-hexane, ethyl acetate and methanol. Antioxidant activity was tested through superoxide radical scavengers. The n-hexane extract of S. dulcis leaves had the best antioxidant activity with an IC50 value of 2080 µg/mL. Based on LCMS/MS, 6 compounds were detected in the n-hexane extract at retention time (Rt) 3.23-9.89 minutes. Compounds Ajmalin, Scopoletin, scoparone, 3-methyl-6-[[2-(2-methylbut-3-en-2-yl)-1H-indole-3-yl] methyldene]piperazine-2,5-dione, [3-hydroxy-3-methylbut-1-enyl]-5,7-dimethoxy-2-phenyl-2,3-dihydrochromene-4-one, and 4-hydroxy-3-(3-methylbut-2-enyl)phenyl ]ethanone.
