Abstract

Kulit dapat mengalami penuaan atau aging yang didefinisikan sebagai hilangnya keseimbangan homeostatik suatu organisme secara progresif. Indikator aging diantaranya keberadaan hyperpigmentasi yang disebabkan oleh enzim tyrosinase. Banyak Cara yang dapat dilakukan untuk  mencegah aging adalah dengan melakukan perawatan kulit menggunakan serum antiaging. Senyawa bioaktif dari tanaman berkembang dan populer untuk digunakan sebagai bahan kosmetik dalam formulasi karena banyak dilaporkan mengandung vitamin, antioksidan, minyak essensial, protein, senyawa fenolik, dan zat aktif lainnya. Buah Ceremai (Phyllantus acidus) diidentifikasi mengandung senyawa asam glikolat dan asam sitrat, sedangkan kulit buah semangka (Citrullus lanatus) mengandung saponin, tanin, alkaloid dan flavonoid. Tujuan penelitian ini adalah untuk membuat formulasi serum  antiaging  dari kombinasi buah ceremai (Phyllantus acidus) dan ekstrak kulit buah semangka (Citrullus lanatus) sebagai inhibisi enzim tirosinase. Penelitian ini  meliputi pengujian aktivitas ekstrak dan sediaan serum terhadap inhibisi enzim tirosinase dengan metode in vitro menggunakan ELISA. Hasil penelitian aktivitas inhibisi enzim tirosinase ekstrak buah ceremai menunjukan nilai IC50 sebesar 9.551 µg/mL dan ekstrak kulit buah semangka 3.304 µg/mL. Inhibisi enzim tirosinase serum F1 mendapatkan nilai IC50  1.137 µg/mL sedangkan F2 sebesar 1.025 µg/mL.  Hasil yang dapat disimpulkan bahwa ekstrak kulit buah semangka lebih aktif sebagai inhibisi enzim tirosinase daripada ekstrak buah ceremai tetapi hasil sediaan serum menunjukan hasil yang memiliki potensi sebagai inhibisi enzim tyrosinase.

Abstract

Mahogany (Swietenia macrophylla King.) is one of the plants used as an antidiabetic agent. 96% ethanol extract of S. macrophylla stem bark can inhibit the α-glucosidase enzyme. Methanol and aqueous extracts of S. macrophylla stem bark can reduce rat blood glucose levels induced by streptozotocin. Molecular docking is one of the most widely used structural-based drug design strategies due to its wide application in the analysis of binding energy and molecular interactions between drug compounds and receptors. This study aims to identify the compounds in the stem bark of S. macrophylla and analyze the molecular anchoring of the compounds produced by the aid. The research was carried out by means of the water fraction of the stem bark of S. macrophylla and was purified by column chromatography. Fraction 2 was identified by LCMS-MS. Molecular docking using AutoDock 4.2.6. The results of Fraction 2 showed the presence of catechins, evodionol, swietemacrophyllanin, swietenitin K and β-sitosterol. The results of an in-silico study showed that swietemacrophyllanin provided a better affinity than acarbose with a binding energy value of -8.57 kcal/mol and an inhibition constant of 0.52 µM. Forms hydrogen bonds with catalytic residues (Glu277 and Asp352) from the active site of the α-glucosidase enzyme. Swietemacrophyllanin has potential as an antidiabetic agent

Abstract

Soybeans contain proteins that have the potential to produce anticancer bioactive peptides. This study aims to determine the anticancer activity of soy protein hydrolysate against MCF-7 breast cancer cells. Soybean protein hydrolyzed by bromelain enzyme 0.5% (w/v) at the optimum temperature and pH for protein hydrolysis using the Bergmeyer and Grassl method. The degree of hydrolysis value of protein hydrolysate was determined by the Alder-Niesen method and the protein profile was analyzed by SDS-PAGE. The hydrolysate with the best degree of hydrolysis value was analyzed for anticancer activity against MCF-7 breast cancer cells by the Presto Blue assay method, and fractionation of protein hydrolysates by gel filtration chromatography (Sephadex G-15). The molecular weight of the peptide was characterized by LCMS/MS. Soy protein hydrolysis using 0.5% (w/v) bromelain enzyme was optimum at 65 ºC and pH 7.0 for 4 hours, with a hydrolysis degree value of 20.57%. The SDS-PAGE analysis showed that the protein hydrolysates had quite thick protein bands in the range of <35 kDa with an IC50 value of 70.37 mg/mL. Based on the LCMS/MS results, the peptide from fractionation has a molecular weight of 5.133 kDa.

Abstract

Alzheimer's is a chronic neurodegenerative disease characterized by low levels of acetylcholine and the accumulation of abnormal neuritic plaques, leading to rapid memory decline and cognitive impairment. Compounds found in the kedondong plant (Spondias dulcis) have been reported to exhibit in vitro activity as acetylcholinesterase inhibitors. This study examines the potential of active compounds in Spondias dulcis in their interaction with acetylcholinesterase, an enzyme implicated in the pathogenesis of Alzheimer's disease. The enzyme was obtained from the Protein Data Bank (PDB ID: 4EY7). The test ligands were screened based on Lipinski's rule and docked with the receptor. The results of molecular docking which yielded the five best affinity energy values were followed by ADMET testing (absorption, distribution, metabolism, excretion, and toxicity). The test ligand ellagic acid deoxyhexoside showed binding energy at -11.213 kcal/mol. Molecular dynamics simulations were performed using YASARA with AMBER14 force fields for 50 ns. The test ligand ellagic acid eoxyhexoside showed an MM-PBSA value of -51.277 kcal/mol and exhibited good complex stability with an average total RMSD value of 2 Å and low inter-residue fluctuation values. These findings are consistent with the results obtained from the comparator ligand, donepezil. Therefore, compounds in Spondias dulcis have the potential to act as acetylcholinesterase inhibitors and can be considered for the development of therapies for Alzheimer's disease.

Abstract

Analysis of the halalness of food products based on animal origin needs to be carried out, especially for critical food products such as processed meat products (sausages). Some processed meat products are found that they are contaminated by pork in the processing. This research was conducted to determine the validity of the analysis method of pork contamination in one of the processed products (beef, pork, and goat sausages) based on the characteristics of the protein profile. Beef, pork, and goat sausages that have been purchased from supermarkets in Jakarta and South Tangerang are used as the sample.The praparation of sample was preceded by extraction of the sausage protein in PBS buffer pH 7.2. Then, the dissolved protein content was tested, and the protein extract was separated by SDS-PAGE (sodium dodecyl sulphate polyacrylamide gel electrophoresis). The protein profile result from SDS-PAGE separation was followed by densitometric analysis (ImageJ) to obtain biomarker protein candidates. The validation of the analytical method includes the precision (repeatability), test for accuracy, linearity, and robustness of the method by varying the extraction pH. The results show that the protein extract has the differences in protein content and composition for each sample. The SDS-PAGE analysis results show that biomarker protein candidates appeared below 50 kDa which were thought to be the protein fraction of actin. The precision and accuracy test results obtained for each sample have met the required standards, namely with a KV value <5% and a percent recovery value> 95%. The results of the linearity test and the toughness of the method also show that the test method is quite effective in testing the halalness of animal food products, especially in sausage products which are contaminated by pork.

Abstract

The community service activities carried out in Sukalangu Village, Pandeglang Regency, had the main objective of increasing the utilization of local potential through the development of useful and economic herbal products. The potential of herbal plants in this village is quite abundant, but their utilization is still limited to traditional use for household needs. Therefore, a systematic effort is needed to enable the community to process them into products with added value. The implementation method of the activities included counseling on the benefits of herbal plants for health, technical demonstrations of making simple herbal products such as health drinks and traditional potions, and participatory assistance in the production process. The results of the activities showed a significant increase in community knowledge, with a 40% increase in understanding of the types, benefits, and processing methods of herbs. In addition, technical skills in mixing and processing herbs increased by 50% based on pre- and post-training evaluations. Furthermore, community interest and motivation to develop herbal-based businesses also grew, as demonstrated by the formation of small groups of residents who took the initiative to produce herbal products as household businesses. Other impacts that emerged were increased individual capacity, collective awareness to protect and utilize natural potential, and the opening of sustainable local economic opportunities. Overall, this activity makes a real contribution to supporting the independence of village communities through the utilization of local resources based on traditional wisdom.
