Abstract

Soybeans contain proteins that have the potential to produce anticancer bioactive peptides. This study aims to determine the anticancer activity of soy protein hydrolysate against MCF-7 breast cancer cells. Soybean protein hydrolyzed by bromelain enzyme 0.5% (w/v) at the optimum temperature and pH for protein hydrolysis using the Bergmeyer and Grassl method. The degree of hydrolysis value of protein hydrolysate was determined by the Alder-Niesen method and the protein profile was analyzed by SDS-PAGE. The hydrolysate with the best degree of hydrolysis value was analyzed for anticancer activity against MCF-7 breast cancer cells by the Presto Blue assay method, and fractionation of protein hydrolysates by gel filtration chromatography (Sephadex G-15). The molecular weight of the peptide was characterized by LCMS/MS. Soy protein hydrolysis using 0.5% (w/v) bromelain enzyme was optimum at 65 ºC and pH 7.0 for 4 hours, with a hydrolysis degree value of 20.57%. The SDS-PAGE analysis showed that the protein hydrolysates had quite thick protein bands in the range of <35 kDa with an IC50 value of 70.37 mg/mL. Based on the LCMS/MS results, the peptide from fractionation has a molecular weight of 5.133 kDa.

Abstract

The impact of extraction methods on the potency of plant extracts is crucial. Bouea macrophylla Griff., a tropical fruit plant rich in phenolic and flavonoid compounds, exhibits strong antioxidant activity. This study aimed to evaluate the effect of maceration and soxhletation extraction techniques on B. macrophylla plants on antioxidant activity in vitro using the method of inhibiting DPPH radical compounds. Ethyl acetate and ethanol solvents were utilized to macerate and soxhlet all parts of the plant. Subsequently, each extract was subjected to quantitative analysis for antioxidant activity, total phenolic, and flavonoid content through UV-Vis spectrophotometry. Quantitative analysis revealed substantial antioxidant activity and notable levels of total phenolic and flavonoid content across all plant parts. The result indicated that although both maceration and soxhletation showed a significant impact on yield (p=0.009) and total flavonoids (p=0.038), significant differences in antioxidant activity (p=0.409) or total phenolics (p=0.874) within the plant extracts of B. macrophylla were not observed at a significance level of 5%.

Abstract

Mahogany (Swietenia macrophylla King.) is one of the plants used as an antidiabetic agent. 96% ethanol extract of S. macrophylla stem bark can inhibit the α-glucosidase enzyme. Methanol and aqueous extracts of S. macrophylla stem bark can reduce rat blood glucose levels induced by streptozotocin. Molecular docking is one of the most widely used structural-based drug design strategies due to its wide application in the analysis of binding energy and molecular interactions between drug compounds and receptors. This study aims to identify the compounds in the stem bark of S. macrophylla and analyze the molecular anchoring of the compounds produced by the aid. The research was carried out by means of the water fraction of the stem bark of S. macrophylla and was purified by column chromatography. Fraction 2 was identified by LCMS-MS. Molecular docking using AutoDock 4.2.6. The results of Fraction 2 showed the presence of catechins, evodionol, swietemacrophyllanin, swietenitin K and β-sitosterol. The results of an in-silico study showed that swietemacrophyllanin provided a better affinity than acarbose with a binding energy value of -8.57 kcal/mol and an inhibition constant of 0.52 µM. Forms hydrogen bonds with catalytic residues (Glu277 and Asp352) from the active site of the α-glucosidase enzyme. Swietemacrophyllanin has potential as an antidiabetic agent

Abstract

Isolation and identification of secondary metabolite compounds from stem bark of Bintangur (Calophyllum bicolor) have been reported. Isolation was carried out using the maceration method with 70% ethanol solvent and fractionation of the ethanol extract by column chromatography using n-hexane solvent. The n-hexane fraction resulting from column chromatography was subjected to gravity chromatography with the mobile phase n-hexane: ethyl acetate (8:2) producing isolate 1. The results of purification of isolate 1 using fast column chromatography with the mobile phase dichloromethane: methanol (1:1) and a UV light detector at ƛ 254 nm, compound 1 was obtained. Meanwhile, identification of compound 1 was carried out through UV-Vis spectroscopy, FTIR and LCMS analysis. The identification results of compound 1 using UV-Vis spectroscopy showed the presence of an aliphatic ring double bond chromophore (λmax 233.5; 295 nm). FTIR results indicated the presence of vibrations of the OH group (3417 cm-1), CH alkanes (2933; 2864; 1456; 545 cm-1), and C=C alkenes (1618 cm-1). Analysis results using LCMS showed a molecular ion peak at [M]+ m/z = 386.96 at a retention time of 11.6 minutes, indicating a molecular weight of 386 and a molecular formula (C27H46O) that corresponds to a terpenoid compound. Keywords: isolation, identification, terpenoid, bintangur (Calophyllum bicolor) AbstrakIsolasi dan identifikasi senyawa metabolit sekunder dari kulit batang bintangur   (Calophyllum   bicolor)   telah   dilaporkan.  Isolasi dilakukan dengan metode maserasi dengan pelarut etanol 70% dan fraksinasi ekstrak etanol dengan kromatografi kolom menggunakan pelarut n-heksana. Fraksi n-heksana hasil kromatografi kolom dikromatografi grafitasi dengan fase gerak n- heksana : etil asetat (8:2) menghasilkan isolat 1. Hasil pemurnian isolat 1 menggunakan kromatografi kolom cepat dengan fase gerak diklorometana : metanol (1:1) dan  detektor sinar UV pada ƛ 254 nm diperoleh senyawa    1. Sedangkan identifikasi  senyawa 1 dilakukan melalui analisis spektroskopi UV-Vis, FTIR, dan LCMS. Hasil identifikasi senyawa 1 menggunakan spektroskopi UV-Vis  menunjukkan adanya kromofor ikatan rangkap cincin alifatik (λmaks 233,5 nm; 295 nm). Hasil FTIR mengindikasikan adanya vibrasi gugus OH (3417     cm-1), CH alkana (2933; 2864; 1456; 545 cm-1), dan C=C alkena (1618 cm-1). Hasil Analisa menggunskan LCMS menujukkan puncak ion molekul pada [M]+ m/z = 386,96 pada waktu retensi 11,6 menit, menunjukkan berat molekul 386 dan rumus molekul (C27H46O) yang sesuai dengan senyawa terpenoid. Kata kunci: isolasi, elusidasi, terpenoid, bintangur (Calophyllum bicolor)

Abstract

Soybean is a source of vegetable protein that is rich in nutrients and a source of producing anticancer bioactive peptides. This study aims to determine the optimum conditions for hydrolysis of soybean protein with papain enzyme and their cytotoxicity against the MCF-7 cells. Soybean protein was isolated using an acid precipitation technique. Then, the protein isolate was hydrolyzed using papain enzyme with variations of papain concentration 0,5; 1; and 5% (v/v) and variations of incubation time 0, 1, 2, 3, and 4 hours at 50 °C. The hydrolysates were tested for their degree of hydrolysis (%DH), molecular weight profile using SDS-PAGE, and cytotoxicity against the MCF-7 cells through an in-vitro assay. The most active hydrolysate was fractionated using Sephadex G-15 and characterized by the molecular weight by LCMS/MS. The result showed that the optimum condition for hydrolysis was 1% (v/v) of enzyme concentration and 3 hours of incubation time with a %DH value of 3.01%. Based on the SDS-PAGE result, the hydrolysate had protein bands in a lower range (<25 kDa). That hydrolysate has cytotoxicity with an IC50 value of 1.87 mg/mL, and the molecular weight of its bioactive peptide is 7.70 kDa.

Abstract

Stunting merupakan permasalahan kesehatan yang disebabkan oleh kekurangan gizi kronis, terutama pada anak-anak pada masa awal pertumbuhannya. Kegiatan pengabdian masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan warga Desa Pasanggrahan dalam mencegah stunting melalui pemanfaatan bahan pangan lokal, yaitu daun kelor dan gula merah. Kegiatan ini mencakup penyuluhan mengenai stunting dan gizi seimbang serta pelatihan pembuatan makanan tambahan bergizi kue talam daun kelor dan gula merah. Melalui program ini, peserta yang terdiri dari ibu hamil, ibu menyusui, kader kesehatan, dan masyarakat umum dibekali dengan pengetahuan serta keterampilan untuk mengolah bahan lokal yang kaya nutrisi menjadi menu makanan tambahan yang mudah dibuat dan disukai. Hasil kegiatan menunjukkan adanya peningkatan pemahaman peserta mengenai pentingnya gizi seimbang untuk pencegahan stunting, serta kemampuan mereka dalam mempraktikkan pembuatan menu tersebut. Program ini diharapkan dapat menjadi solusi berkelanjutan dalam upaya penanggulangan stunting di desa-desa melalui pemberdayaan dan pemanfaatan potensi lokal.

Abstract

Analysis of the halalness of food products based on animal origin needs to be carried out, especially for critical food products such as processed meat products (sausages). Some processed meat products are found that they are contaminated by pork in the processing. This research was conducted to determine the validity of the analysis method of pork contamination in one of the processed products (beef, pork, and goat sausages) based on the characteristics of the protein profile. Beef, pork, and goat sausages that have been purchased from supermarkets in Jakarta and South Tangerang are used as the sample.The praparation of sample was preceded by extraction of the sausage protein in PBS buffer pH 7.2. Then, the dissolved protein content was tested, and the protein extract was separated by SDS-PAGE (sodium dodecyl sulphate polyacrylamide gel electrophoresis). The protein profile result from SDS-PAGE separation was followed by densitometric analysis (ImageJ) to obtain biomarker protein candidates. The validation of the analytical method includes the precision (repeatability), test for accuracy, linearity, and robustness of the method by varying the extraction pH. The results show that the protein extract has the differences in protein content and composition for each sample. The SDS-PAGE analysis results show that biomarker protein candidates appeared below 50 kDa which were thought to be the protein fraction of actin. The precision and accuracy test results obtained for each sample have met the required standards, namely with a KV value <5% and a percent recovery value> 95%. The results of the linearity test and the toughness of the method also show that the test method is quite effective in testing the halalness of animal food products, especially in sausage products which are contaminated by pork.

Abstract

Koperasi Anugerah Aren Banten Nusantara merupakan koperasi yang menghasilkan produk gula aren dan turunannya termasuk gula aren cair. Produksi gula aren cair selama ini masih menggunakan teknik konvensional yang memerlukan durasi waktu yang lama dan menghasilakn kualitas produk yang kurang baik. Pada kegiatan pengabdian ini dilakukan pendampingan dan transfer teknologi alat produksi gula aren cair dengan metode vakum evaporasi tekanan rendah. Pelaksanaan kegiatan dilakukan dari bulan September -November 2024, melalui sosialisasi, pelatihan dan pendampingan. Kegitan melibatkan bagian produksi dan managemen Koperasi Aren Banten Nusantara. Sosialisasi dilaksanan dengan metode ceramah menggunakan alat bantu leafleat. Pelatihan dan pendampingan penggunaan alat vakum evaporasi tekanan rendah dilaksanakan di area produksi Koperasi Aren Banten Nusantara. Penggunaan alat produksi gula aren cair metode vakum evaporasi dapat mempercepat produksi gula aren cair. Berdasarkan hasil produski gula aren cair didapatkan nilai brix sebesar 60, 50 dan 50 dengan total rendemen 10 kg dari nira sebanyak 100 L. Pelatihan terhadap divisi produksi gula aren efektif dilaksanakan dengan perolehan peningkatan pengetahuan di atas 50% dari nilai sebelum dilakukan pelatihan. The production of liquid palm sugar has so far used conventional techniques that require a long duration and produce poor product quality. In this community service activity, assistance and technology transfer were carried out for liquid palm sugar production equipment using the low-pressure vacuum evaporation method. The implementation of the activity was carried out from September to November 2024, through socialization, training and assistance. The activity involved the production and management divisions of the Banten Nusantara Aren Cooperative. Socialization was carried out using the lecture method using leafleat aids. Training and assistance in the use of low-pressure vacuum evaporation equipment were carried out in the production area of ​​the Banten Nusantara Aren Cooperative. The use of liquid palm sugar production equipment using the vacuum evaporation method can accelerate the production of liquid palm sugar. Based on the results of liquid palm sugar production, the brix values ​​obtained were 60, 50 and 50 with a total yield of 10 kg from 100 L of sap. Training for the palm sugar production division was effectively implemented with an increase in knowledge of more than 50% from the value before the training.

Abstract

Program KKN (Kuliah Kerja Nyata) dengan tema restorasi kampung berkualitas di Desa Sindanghayu Saketi Pandeglang Banten untuk penanggulangan gizi buruk dan stunting bertujuan meningkatkan kesejahteraan masyarakat, khususnya anak-anak dan ibu hamil, melalui intervensi gizi dan kesehatan yang terpadu. Program ini melibatkan mahasiswa dalam pemberdayaan masyarakat melalui edukasi gizi, penyuluhan kesehatan dan peningkatan akses terhadap layanan kesehatan. Kegiatan utama meliputi penyuluhan tentang pentingnya asupan gizi seimbang, pemberian makanan tambahan (PMT), dan pelatihan pengelolaan kebun gizi berbasis bahan lokal untuk memastikan ketersediaan pangan bergizi di tingkat keluarga. Kader Posyandu dan tim PKK berperan aktif dalam program ini, termasuk dalam deteksi dini stunting dan gizi buruk serta pemantauan kesehatan anak-anak. Restorasi ini juga menekankan pada peningkatan sanitasi dan kebersihan lingkungan untuk mencegah penyakit yang dapat mempengaruhi status gizi. Hasil dari program ini diharapkan mampu menurunkan prevalensi stunting, meningkatkan kualitas gizi masyarakat, serta memberdayakan masyarakat dalam menjaga kesehatan keluarga. Dengan kolaborasi antara mahasiswa, pemerintah, dan masyarakat, program ini menciptakan kampung yang lebih sehat, berkualitas, dan berkelanjutan.

Abstract

Pemberdayaan perempuan menjadi kunci strategis dalam mendorong kemandirian ekonomi dan pembangunan sosial di tingkat lokal. Program pemberdayaan berbasis komunitas yang dilakukan oleh PT. Permodalan Nasional Madani (PNM) di Kabupaten Pandeglang bertujuan meningkatkan kapasitas perempuan melalui pelatihan kewirausahaan, pendampingan usaha, dan akses permodalan mikro. Pengabdian ini menggunakan pemberdayaan ekonomi untuk menganalisis dampak program yang diluncurkan PT. PNM terhadap kelompok perempuan di beberapa kecamatan di Kabupaten Pandeglang. Hasil menunjukkan bahwa program yang diluncurkan oleh PT. PNM dalam bidang pemberdayaan ekonomi ini tidak hanya meningkatkan keterampilan dan kepercayaan diri perempuan dalam mengelola usaha, tetapi juga memperkuat solidaritas sosial dan partisipasi perempuan dalam pengambilan keputusan di komunitas. Hasil penilaian kepuasan terhadap pemberdayaan ekonomi memiliki tingkat kepuasan t mencapai lebih dari 95%. Faktor pendukung keberhasilan program meliputi pendekatan partisipatif, keberlanjutan pendampingan, serta dukungan lokal dari perangkat desa dan kecamatan. Sementara itu, tantangan utama masih ditemukan pada keterbatasan akses informasi dan kesiapan teknologi. Secara keseluruhan, inisiatif pemberdayaan ini berkontribusi nyata terhadap terwujudnya Pandeglang yang berdaya dan mandiri, terutama melalui penguatan peran perempuan sebagai agen perubahan dalam pembangunan desa. Women's empowerment is a strategic key in encouraging economic independence and social development at the local level. The community-based empowerment program carried out by Permodalan Nasional Madani (PNM) in Pandeglang Regency aims to increase women's capacity through entrepreneurship training, business mentoring, and access to micro-capital. This service uses economic empowerment to analyze the impact of the program launched by PNM on women's groups in several sub-districts in Pandeglang Regency. The results show that the program launched by PNM in the field of economic empowerment not only improves women's skills and confidence in managing businesses but also strengthens social solidarity and women's participation in decision-making in the community. The results of the satisfaction assessment of economic empowerment have a satisfaction level of more than 95%. Supporting factors for the success of the program include a participatory approach, sustainability of mentoring, and local support from village and sub-district officials. Meanwhile, the main challenges are still found in limited access to information and technological readiness. Overall, this empowerment initiative has made a real contribution to the realization of an empowered and independent Pandeglang, especially through strengthening the role of women as agents of change in village development.
