Abstract

Vanilla is a plant that has many benefits, high selling value and potential to be developed. Conventional propagation of vanilla such as through cuttings has not been effective, so a more effective method of propagation is needed, one of which is propagation through tissue culture. In-vitro propagation of vanilla can be through shoot multiplication to produce fast plantlets. One of the successes of shoot multiplication is influenced by the hormone given. This study aims to determine the effect of giving a combination of 2,4-D and BAP on the multiplication of vanilla shoots. The study design used CRD with a combination factor of 2,4-D (concentration 0.5 mg/L, 1.0 mg/L, and 1.5 mg/L) and BAP (concentration 0.25 mg/L, 0.5 mg/L, and 0.75 mg/L). The explants used were vanilla stem segments. Based on the research results, the combined treatment with concentrations of 2,4-D 1 mg/L and BAP 0.5 mg/L had a very significant effect on the multiplication of vanilla shoots with the fastest emergence of shoots with an average of 6.6 wap, the highest number of shoots was 7 shoots, and the survival percentage was 100%.

Abstract

Stunting is a major health problem in Indonesia. Sukogidri Village is one of the villages that contribute to stunting in Jember Regency. There are 18 stunting children under five from 353 total children under five in the village. The effort to solve the stunting problem should involve community participation through empowerment. This community service aimed to increase community empowerment to solve the stunting by forming the nutritionally-aware family (Keluarga Sadar Gizi = KADARZI) model and the stunting-alert cadres. KADARZI model was chosen in selected families and trained on the definition of KADARZI, the efforts and steps undertaken as a model for KADARZI in selected families. The stunting-alert cadres were formed from health cadres who were provided with intensive education and training materials on the stunting, its dangers and how to prevent it. After going through serial training, 10 KADARZI models and 10 stunting-alert cadres were formed in Sukogidri Village, Jember. It is hoped that the KADARZI model and stunting alert cadres will play an active role in overcoming stunting in the village.

Abstract

Anther culture is an effective biotechnological approach to accelerate the production of homozygous lines in tobacco breeding. This study evaluated the interaction between culture media (Murashige and Skoog/MS and Chu’s N6) and silver nanoparticle (AgNP) concentrations on in vitro anther regeneration of tobacco (Nicotiana tabacum L.). The experiment was arranged in a completely randomized design with ten treatment combinations consisting of two basal media and five AgNP concentrations (0, 2.5, 5, 7.5, and 10 ppm), each replicated three times. Quantitative parameters included callus formation, shoot regeneration, rooting response, regeneration percentage, and contamination rate. MS medium supplemented with 2.5 ppm AgNPs (K2) produced the optimal response, showing 100% callus formation and the highest shoot regeneration percentage (89%). Root formation was observed only in treatments K2 and K4. In contrast, higher AgNP concentrations (≥7.5 ppm), particularly in Chu’s N6 medium, significantly reduced regeneration percentages (ANOVA, p<0.05). The promotive effect of low AgNP concentration is associated with suppression of ethylene activity, antimicrobial action, and regulation of reactive oxygen species (ROS), which collectively enhance cell division and organogenesis. These findings indicate that MS medium supplemented with 2.5 ppm AgNPs is optimal for tobacco anther regeneration and suitable for doubled-haploid production.

Abstract

Increasing crop productivity is a top priority in supporting global food security amid increasingly complex environmental challenges. Sustainable agricultural technology emerges as a promising approach to enhance agricultural yields without harming the environment. This study aims to explore the application of sustainable agricultural technology in enhancing crop productivity. Through a comprehensive literature review, we identify various sustainable agricultural technologies that can be implemented, such as the use of organic fertilizers, water-efficient irrigation, environmentally friendly pest management, and soil conservation practices. We evaluate the positive impact of each of these technologies on major crop productivity such as wheat, rice, corn, and soybeans. The results of the analysis indicate that the implementation of sustainable agricultural technology effectively enhances crop yields by maintaining ecological balance and reducing negative environmental impacts. The practical implications of this research are that the integration of sustainable agricultural technology into agricultural practices can be an effective solution for improving crop productivity sustainably, while supporting environmental and social sustainability in the long term

Abstract

Anggrek Dendrobium sp. merupakan tanaman hias bernilai ekonomi tinggi yang memerlukan teknik perbanyakan in vitro yang efisien. Penelitian ini bertujuan untuk mengevaluasi respon pertumbuhan Dendrobium sp. terhadap perbedaan konsentrasi BAP (1; 1,5; dan 2 ppm) pada media cair dan media padat secara in vitro. Penelitian dilakukan di UPA PL2T Universitas Jember pada Januari–April 2025 menggunakan rancangan faktorial. Parameter pertumbuhan dianalisis menggunakan ANOVA dan uji DMRT 5%. Hasil menunjukkan bahwa konsentrasi BAP tidak berpengaruh nyata terhadap seluruh parameter, sedangkan bentuk media berpengaruh sangat nyata terhadap jumlah tunas, jumlah daun, panjang akar, dan luas daun. Media padat menghasilkan pertumbuhan vegetatif lebih optimal dan direkomendasikan untuk perbanyakan Dendrobium sp. secara in vitro.

Abstract

Desa Purwoasri kecamatan Gumukmas kabupaten Jember memiliki luas wilayah sebesar 3092,34 km2. Sebagian besar penduduk memiliki mata pencaharian sebagai petani dan peternak. Pemenuhan kebutuhan pupuk bagi petani desa Purwoasri Gumukmas sering terkendala oleh langkanya ketersediaan pupuk pada waktu diperlukan. Di sisi lain, limbah kotoran sapi menumpuk karena tidak termanfaatkan dan dapat menjadi penyebab utama polusi lingkungan. Dengan demikian upaya pemahaman dan peningkatan kemampuan petani dalam memanfaatkan limbah kotoran sapi menjadi pupuk organik melalui penyuluhan dan pelatihan merupakan satu upaya untuk menyelesaikan permasalahan yang ada di desa Purwoasri. Kegiatan pengabdian pada masyarakat ini dilakukan melalui kerja sama antara peneliti, mahasiswa KKN dan PPL kecamatan Gumukmas. Pembuatan pupuk organic cair (POC) dilakukan dengan bahan dasar kotoran sapi yang dicampur dengan bahan lainnya. Hasil pengabdian menunjukkan bahwa petani desa Wonoasri sangat antusias untuk membuat POC dari limbah kotoran sapi yang selama menumpuk di halaman. Warga selanjutnya juga menginginkan desa Purwoasri sebagai desa binaan Universitas Jember

Abstract

Lettuce (Lactuca sativa L.) is a leaf vegetable that has a high level of consumer demand and commercial value and can be cultivated using a combination of hydroponic substrate and drip irrigation methods. This research aims to determine the effect of the time interval for providing nutrients on the growth and harvest results of three lettuce plant varieties. Plants were cultivated on cocopeat and husk charcoal media (ratio 1:3) and arranged using a Randomized Block Design (RAK) consisting of two treatments. The first treatment is the nutritional interval which consists of an interval of 2 hours with a discharge of 17 ml (I1), 3 hours with a discharge of 25 ml (I2), and 4 hours using a discharge of 34 ml (I3). The second treatment is the plant variety which consists of the Grand Rapids variety (V1), the Green Coral variety (V2), and the Red Coral variety (V3). The data is analyzed using analysis of variance and if the results obtained are significantly different then a DMRT test will be carried out. The research results showed that the Green Coral lettuce variety was better than the Grand Rapids and Red Coral varieties. This is shown by the results with the highest and best values for the observation variables of number of leaves, plant fresh weight, and chlorophyll content. The nutritional interval which consists of an interval of 2, 4, and 6 hours gave results that were inversely proportional to the variety treatment, that is, they were not significantly different in all observed variables. Keywords: lettuce plants, plant varieties, nutritional interval, substrate hydroponics

Abstract

Genetic transformation in plants involves the insertion of isolated foreign genes into the plant genome. Sucrose Phosphate Synthase (SPS) is a key enzyme in sucrose biosynthesis, the primary sugar serving as an energy source and carbon transport molecule in most plants. Dendrobium orchids, which utilize Crassulacean Acid Metabolism (CAM), often exhibit a slow growth rate, thereby limiting their commercial potential. The insertion of the SPS gene into the orchid genome is expected to optimize the sucrose metabolic pathway, potentially influencing the rate of photosynthesis, carbon allocation, and growth characteristics of the orchid. Optimizing the optical density (OD) of the Agrobacterium suspension and the dosage of kanamycin is crucial for enhancing the success rate of the gene transformation process. This study aimed to determine the optimal concentrations of OD and kanamycin to achieve efficient SoSPS1 gene transformation in orchids. Three-month-old Protocorm-Like Bodies (PLBs) of Dendrobium Glory White were used as the explant source. The transformed explants were subsequently evaluated for the effectiveness of the kanamycin dosage, the percentage of explant shoots, the number of shoots per explant, the transformation efficiency, and phenotypic changes in the transformed plants. The results showed that a transformation efficiency of up to 40% was achieved using an Agrobacterium suspension at an OD600nm of 0.4. A lethal dose (LD50) of 100 mgL-1 kanamycin resulted in 34% of plantlets surviving post-selection. Transformation with the SoSPS1 gene in Dendrobium sp. induced distinct phenotypic plants including split leaves, swollen stems (pseudobulb), and increased shoot multiplication.

Abstract

The majority of residents in Suling Wetan Village work as farmers and rely heavily on self-produced seeds due to the village’s remote location. One high-potential crop for marginal lands in the area is patchouli (Pogostemon cablin), a plant valued for its essential oil. However, the propagation of patchouli remains a significant challenge for local farmers due to limited knowledge and skills. This community engagement program aimed to enhance farmers’ capacities in patchouli propagation through a combination of awareness campaigns, technical workshops, hands-on training, direct field mentoring, and participatory monitoring and evaluation. Evaluation was conducted through direct observation and participant surveys. The results showed that 96% of participants gained a clear understanding of patchouli propagation techniques, and all participants acknowledged the importance of the program and expressed hope for its continuation. The activity successfully contributed to increasing farmers’ independence in sustainable patchouli seedling production.
