Abstract

Tembakau (Nicotiana tabacum L.) merupakan salah satu tanaman komoditas perkebunan yang memiliki nilai ekonomi cukup tinggi. Curah hujan tinggi dapat menyebabkan terjadinya genangan dan merupakan kendala utama dalam budidaya tanaman tembakau. Banyak studi telah menunjukkan bahwa genangan mempengaruhi respon morfologi, fisiologi dan anatomi pada tanaman. Kalsium berperan dalam membantu proses metabolisme tanaman saat mengalami cekaman abiotik. Pemberian kalsium pada tanaman dapat meningkatkan adaptasinya terhadap genangan. Penelitian ini bertujuan untuk mengetahui toleransi tanaman tembakau terhadap cekaman genangan pada berbagai taraf aplikasi kalsium berdasarkan respon fisiologi, morfologi serta anatominya. Penelitian ini dilaksanakan pada bulan November 2021 hingga Januari 2022 di Kebun percobaan Universal PT. Tempu Rejo menggunakan rancangan acak lengkap (RAL) faktorial. Faktor pertama adalah penggenangan tanaman tembakau pada usia yang berbeda (tanpa penggenangan/kontrol; penggenangan pada 21 hari setelah tanam (HST), 28 HST, dan 35 HST). Faktor kedua adalah aplikasi kalsium dengan dosis (0, 8.5, 10, dan 11.5 g per tanaman). Hasil penelitian menunjukkan bahwa kalsium 11.5 g per tanaman yang diaplikasikan efektif meningkatkan toleransi tanaman terhadap cekaman genangan berdasarkan karakteristik morfologinya. Hasil penelitian juga menunjukkan bahwa penambahan kalsium dapat menekan keberadaan H2O2 sebagai ROS (reactive oxygen species) serta dapat mempertahankan struktur anatomi akar dan anatomi stomata di bawah cekaman genangan. Abstract

The growth and development of plants are determined by photosynthesis, which ultimately results in sucrose. Sucrose is synthesized in the source then translocated to all parts of the plant (sink). The translocation process of sucrose from source to sink is controlled by sucrose protein called sucrose transporter. SoSUT1 is a gene that encodes a sucrose transporter 1 (SUT1) protein in sugarcane. Rice transformation with the SoSUT1 causes overexpression SUT, which is expected to increase the translocation of sucrose into the seed of rice plants. This research was conducted by introducing SoSUT1 in rice plants Inpari 14 SS. Transformation using Agrobacterium tumefaciens vector in apical bud explant Indica rice cv. Inpari 14 SS results in 26 events positive rice contains genes SoSUT1. This study aims to elucidate the inheritance of transgene in the next generation and to characterize its effect on the morphology and the yields of a subsequent generation. The study is conducted by planting the seeds of T1 and T2 plants in media containing Hygromycin and using PCR analysis for further analysis. As the results, from 26 events on the T1 plant, only 3 events of T3 plants were confirmed on the T3 plant. The overexpression of the SoSUT1 gene could increase the number of tillers, the number of productive tillers, panicle length, and panicle exit length, also increasing the number of spherical grains, reducing the number of empty grains and increasing the weight of 1000 grains.

Abstract

Callus formed in somatic embryogenesis is divided into two types, embryogenic and non-embryogenic callus. Embryogenic callus can be distinguished by the expression of genes as markers related to somatic embryogenesis which are expected to be used as markers to detect callus that has embryogenic capabilities in sugarcane. The aim of this study was to determine the expressions of somatic embryogenesis-related genes in sugarcane. The genes analysis was carried out using somatic embryogenesis callus and using a kit for RNA analysis. Results showed that there were expressions of Baby Boom (BBM), Leafy Cotyledon (LEC), dan Somatic Embryogenesis Receptor Like-Kinase (SERK) gene with specific primer of those three genes, which were collected from embryogenic callus in mass, globular, scutelar, and coleoptilar pre-embryo stages, meanwhile there were no expressions of those genes collected from non-embryogenic callus. Amplification result from PCR product of cDNA using three gene primers detected were in 500 bp for BBM, 400 bp for LEC, and 700 bp for SERK gene.

Abstract

Vanilla is a plant that has many benefits, high selling value and potential to be developed. Conventional propagation of vanilla such as through cuttings has not been effective, so a more effective method of propagation is needed, one of which is propagation through tissue culture. In-vitro propagation of vanilla can be through shoot multiplication to produce fast plantlets. One of the successes of shoot multiplication is influenced by the hormone given. This study aims to determine the effect of giving a combination of 2,4-D and BAP on the multiplication of vanilla shoots. The study design used CRD with a combination factor of 2,4-D (concentration 0.5 mg/L, 1.0 mg/L, and 1.5 mg/L) and BAP (concentration 0.25 mg/L, 0.5 mg/L, and 0.75 mg/L). The explants used were vanilla stem segments. Based on the research results, the combined treatment with concentrations of 2,4-D 1 mg/L and BAP 0.5 mg/L had a very significant effect on the multiplication of vanilla shoots with the fastest emergence of shoots with an average of 6.6 wap, the highest number of shoots was 7 shoots, and the survival percentage was 100%.

Abstract

Stunting is a major health problem in Indonesia. Sukogidri Village is one of the villages that contribute to stunting in Jember Regency. There are 18 stunting children under five from 353 total children under five in the village. The effort to solve the stunting problem should involve community participation through empowerment. This community service aimed to increase community empowerment to solve the stunting by forming the nutritionally-aware family (Keluarga Sadar Gizi = KADARZI) model and the stunting-alert cadres. KADARZI model was chosen in selected families and trained on the definition of KADARZI, the efforts and steps undertaken as a model for KADARZI in selected families. The stunting-alert cadres were formed from health cadres who were provided with intensive education and training materials on the stunting, its dangers and how to prevent it. After going through serial training, 10 KADARZI models and 10 stunting-alert cadres were formed in Sukogidri Village, Jember. It is hoped that the KADARZI model and stunting alert cadres will play an active role in overcoming stunting in the village.

Abstract

Tanaman tembakau (Nicotiana tabacum L.) merupakan tanaman musiman bernilai ekonomi tinggi, namun produksinya di Indonesia terus mengalami penurunan dalam beberapa tahun terakhir. Penggunaan varietas hibrida dapat menjadi solusi karena memiliki produktivitas dan ketahanan penyakit yang lebih baik, tetapi pembentukannya secara konvensional memerlukan waktu lama. Teknik kultur antera dapat digunakan untuk mempercepat pembentukan galur murni karena menghasilkan tanaman homozigot hanya dalam satu generasi. Penelitian ini bertujuan untuk menganalisis interaksi antara jenis media kultur (MS dan Chu’s N6) dengan penambahan beberapa konsentrasi Nanopartikel silver (AgNPs) yaitu 0,0; 2,5; 5,0; 7,5; dan 10,0 mg L⁻¹ terhadap pertumbuhan antera tembakau secara in vitro. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 10 perlakuan dan 3 ulangan, kemudian data dianalisis menggunakan ANOVA dan diuji lanjut DMRT dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa media MS merupakan media terbaik dalam mempercepat dan memperbanyak induksi swelling, kalus, tunas, dan akar, sedangkan konsentrasi AgNPs terbaik diperoleh pada 2,5 mg L⁻¹ yang mampu meningkatkan respons pertumbuhan serta sedikit mengurangi tingkat kontaminasi media. Hasil penelitian ini menunjukkan bahwa kombinasi media MS dan AgNPs dosis rendah mampu meningkatkan efisiensi regenerasi antera tembakau dan berpotensi digunakan sebagai protokol standar dalam produksi galur murni secara cepat, efisien, dan steril di laboratorium kultur jaringan. ABSTRACT Tobacco (Nicotiana tabacum L.) is a seasonal crop with high economic value, but its production in Indonesia has continued to decline in recent years. The use of hybrid varieties can be a solution because they have better productivity and disease resistance, but their conventional formation requires a long time. Anther culture techniques can be used to accelerate the formation of pure lines because they produce homozygous plants in just one generation. This study aims to determine the interaction between the types of culture media (MS and Chu's N6) with the addition of several concentrations of silver nanoparticles (AgNPs), namely 0, 2.5, 5, 7.5, and 10 mg L⁻¹ on the growth of tobacco anthers in vitro. The study used a Completely Randomized Design (CRD) with 10 treatments and 3 replications, then the data were analyzed using ANOVA and further tested with DMRT at the 95% level. The results showed that MS medium was the best medium in accelerating and increasing the induction of swelling, callus, shoots, and roots, while the best concentration of AgNPs was obtained at 2.5 mg L⁻¹ which was able to increase the growth response and slightly reduce the level of media contamination. The findings of this study indicate that the combination of MS medium and low-dose AgNPs can increase the efficiency of tobacco anther regeneration and has the potential to be used as a standard protocol in the production of pure lines quickly, efficiently, and sterilely in tissue culture laboratories.

Abstract

Anther culture is an effective biotechnological approach to accelerate the production of homozygous lines in tobacco breeding. This study evaluated the interaction between culture media (Murashige and Skoog/MS and Chu’s N6) and silver nanoparticle (AgNP) concentrations on in vitro anther regeneration of tobacco (Nicotiana tabacum L.). The experiment was arranged in a completely randomized design with ten treatment combinations consisting of two basal media and five AgNP concentrations (0, 2.5, 5, 7.5, and 10 ppm), each replicated three times. Quantitative parameters included callus formation, shoot regeneration, rooting response, regeneration percentage, and contamination rate. MS medium supplemented with 2.5 ppm AgNPs (K2) produced the optimal response, showing 100% callus formation and the highest shoot regeneration percentage (89%). Root formation was observed only in treatments K2 and K4. In contrast, higher AgNP concentrations (≥7.5 ppm), particularly in Chu’s N6 medium, significantly reduced regeneration percentages (ANOVA, p<0.05). The promotive effect of low AgNP concentration is associated with suppression of ethylene activity, antimicrobial action, and regulation of reactive oxygen species (ROS), which collectively enhance cell division and organogenesis. These findings indicate that MS medium supplemented with 2.5 ppm AgNPs is optimal for tobacco anther regeneration and suitable for doubled-haploid production.

Abstract

Increasing crop productivity is a top priority in supporting global food security amid increasingly complex environmental challenges. Sustainable agricultural technology emerges as a promising approach to enhance agricultural yields without harming the environment. This study aims to explore the application of sustainable agricultural technology in enhancing crop productivity. Through a comprehensive literature review, we identify various sustainable agricultural technologies that can be implemented, such as the use of organic fertilizers, water-efficient irrigation, environmentally friendly pest management, and soil conservation practices. We evaluate the positive impact of each of these technologies on major crop productivity such as wheat, rice, corn, and soybeans. The results of the analysis indicate that the implementation of sustainable agricultural technology effectively enhances crop yields by maintaining ecological balance and reducing negative environmental impacts. The practical implications of this research are that the integration of sustainable agricultural technology into agricultural practices can be an effective solution for improving crop productivity sustainably, while supporting environmental and social sustainability in the long term

Abstract

Anggrek Dendrobium sp. merupakan tanaman hias bernilai ekonomi tinggi yang memerlukan teknik perbanyakan in vitro yang efisien. Penelitian ini bertujuan untuk mengevaluasi respon pertumbuhan Dendrobium sp. terhadap perbedaan konsentrasi BAP (1; 1,5; dan 2 ppm) pada media cair dan media padat secara in vitro. Penelitian dilakukan di UPA PL2T Universitas Jember pada Januari–April 2025 menggunakan rancangan faktorial. Parameter pertumbuhan dianalisis menggunakan ANOVA dan uji DMRT 5%. Hasil menunjukkan bahwa konsentrasi BAP tidak berpengaruh nyata terhadap seluruh parameter, sedangkan bentuk media berpengaruh sangat nyata terhadap jumlah tunas, jumlah daun, panjang akar, dan luas daun. Media padat menghasilkan pertumbuhan vegetatif lebih optimal dan direkomendasikan untuk perbanyakan Dendrobium sp. secara in vitro.
