Abstract

Non-alcoholic fatty liver disease (NAFLD) is a condition marked by excessive fat accumulation in the liver and poses a significant health challenge. The leaves of Blumea balsamifera and Sargassum aquifolium have been reported to have anti-atherogenic effects. This study aims to determine the effectiveness of B. balsamifera extract (BBLE) and S. aquifolium extract (SAE) in preventing and treating liver fat accumulation in Wistar rats induced by a high-cholesterol diet through the expression of the AMP-activated protein kinase (AMPK)/ Sirtuin 1 (SIRT1)/peroxisome proliferator-activated receptor γ (PPARγ) pathway, and the leptin receptor. The experimental design of this study is laboratory-based, involving, 20 Wistar rats were fed a high-cholesterol diet over a period of 21 days. The rats were divided into four groups for the evaluation of BBLE and SAE effect: negative control (P0): induced with a high-cholesterol diet + distilled water, positive control (P1): induced with a high-cholesterol diet + simvastatin, P2: induced with a high-cholesterol diet + 4 mg/kg/bw BBLE, and P3: induced with a high-cholesterol diet + 4 mg/kg/bw BBLE and 4 mg/kg/bw SAE. The treatment duration extended over three months. Immunohistochemical analyses were performed on liver tissues to measure AMPK, SIRT1, PPARγ, and leptin receptor expression. The results indicated that leptin expression was lower in the BBLE+SAE group compared to the simvastatin group, and differences were significant between the BBLE and BBLE+SAE groups. No significant differences were noted in AMPK, SIRT1, and PPARγ expression between the simvastatin and BBLE+SAE groups (p≥0.05). In conclusion, BBLE and SAE effectively reduce liver lipid accumulation and enhance fat metabolism in hypercholesterolemic rats.

Abstract

Brown algae are considered to be a source of antioxidants that can contribute to improving health. The purpose of this study was to determine the best combination of brown algae and solvent types based on the parameters of antioxidant capacity, toxicity to Vero cells, and bacterial contamination that affects brown algae simplicia during 30 d of storage. Brown algae, Sargassum aquifolium and Padina australis, were collected from Sanur Beach at the lowest point in the intertidal zone. Extraction was performed using three solvents: ethanol, methanol, and distilled water. The antioxidant capacity of the brown algae extracts was measured using the DPPH method. The brown algae extract, which showed antioxidant capacity and IC50 in the strong category, was used for cytotoxicity testing. Bacterial contamination testing was performed based on the simple storage time of brown algae for 30 d. The results showed that the combination of S. aquifolium and P. australis ethanol extract (ratio 1:10) had a yield percentage of 37.2% with an antioxidant capacity of 0.935±0.003 mg/L GAEAC and an IC50 value of 89.03 μg/mL (strong category). The combination of S. aquifolium and P. australis ethanol extracts had an IC50 value of 382.30 μg/mL (the weak category). Total plate count (TPC), Enterobacteriaceae, and coliforms increased with the storage time of simplicia for 30 days. These findings indicate that the combination of ethanol extracts of S. aquifolium and P. australis is a potential source of natural antioxidants.

Abstract

Infectious disease is a major challenge in fish and shrimp farming systems caused by microorganisms including bacteria, viruses and parasites. Physical detection has limitations in detecting diseases in cultivated animals earlier, because of the varied clinical symptoms. The aims of this study was to identify the physical and molecular presence of infectious pathogens in fish and shrimp cultivated in several ponds in West Java, Banten and Jakarta. Monitoring was carried out in two stages, namely the period March–April 2021 and September 2021 by taking samples from several ponds in Bogor, Tangerang, Depok and Jakarta. The sample criteria used were fish and shrimp showing normal and clinical symptoms of being attacked by a disease which were collected by purposive sampling. The collected samples were examined in two stages, namely physical examination of clinical symptoms and molecular examination using PCR. The results showed that of the 24 species samples collected, 17 species showed normal clinical symptoms and seven species showed clinical symptoms. Of the 24 fish and shrimp samples, four samples were infected (2 samples from normal species and 2 samples with clinical symptoms). The infected normal samples were Osphronemus goramy and Litopenaeus vannamei. The infected samples with clinical symptoms were Carassius auratus by Aeromonas salmonicida and red zebra cichlid (Metriaclima estherae) by Red Sea Bream Iridoviral Disease (RSBIV). Physical testing supported by molecular detection of aquatic animals can be an effort to manage aquaculture systems in Indonesia. Keyword: aquaculture, emerging disease, fisheries products, molecular assay ABSTRAK Penyakit infeksi adalah tantangan utama pada sistem budidaya ikan dan udang yang disebabkan oleh mikroorganisme termasuk bakteri, virus, dan parasit. Deteksi secara fisik memiliki keterbatasan dalam mengetahui lebih dini penyakit pada hewan budidaya, karena gejala klinis yang bervariasi. Tujuan dari penelitian ini adalah untuk melakukan identifikasi fisik dan molekuler keberadaan patogen infeksi pada ikan dan udang yang dibudidayakan di beberapa pertambakan di Jawa Barat, Banten, dan Jakarta. Pemantauan dilakukan dalam dua tahap yaitu periode Maret – April 2021 dan September 2021 dengan mengambil sampel dari beberapa pertambakan di Bogor, Tangerang, Depok, dan Jakarta. Kriteria sampel yang digunakan adalah ikan dan udang yang menunjukkan gejala klinis terserang oleh penyakit yang dikumpulkan secara purposive sampling. Sampel yang terkumpul diperiksa dalam dua tahap yaitu pemeriksaan fisik gejala klinis dan pemeriksaan molekuler menggunakan PCR. Hasil menunjukkan dari 24 sampel spesies yang terkumpul, sebanyak 17 spesies menunjukkan gejala klinis normal dan tujuh spesies menunjukkan gejala klinis. Dari 24 sampel ikan dan udang, terdapat empat sampel yang terinfeksi (2 sampel dari spesies normal dan 2 sampel dengan gejala klinis). Sampel normal yang terinfeksi adalah Osphronemus goramy dan Litopenaeus vannamei. Sampel yang terinfeksi dengan gejala klinis adalah Carassius auratus oleh Aeromonas salmonicida dan red zebra cichlid (Metriaclima estherae) oleh Red Sea Bream Iridoviral Disease (RSBIV). Pengujian fisik yang didukung dengan deteksi molekuler pada hewan budidaya dapat menjadi upaya manajemen sistem akuakultur di Indonesia. Abstract

Bovine respiratory syncytial virus (BRSV) is highly prevalent in cattle. It is a major viral cause of bovine respiratory disease complex, which is associated with morbidity, mortality and substantial economic impact. Currently available treatments are only symptomatic, but no specific treatments are available for BRSV infection. This study aimed to identify new antiviral agents against BRSV, which could be used to control bovine respiratory disease complex in cattle with Indonesian herb extracts. Ethanol extracts prepared from Indonesian herbs including Andrographis paniculata, Phyllanthus niruri, Curcuma aeruginosa, and Curcuma xanthorrhiza were evaluated for anti-BRSV activity in A549 cells. The cytotoxicity of the herb extracts was evaluated using a CCK-8 cell viability assay. Antiviral activities of the herb extracts were examined using cell activity and cytopathic assays. The effect on virus production was evaluated by qRT-PCR and plaque-formation assays. Extracts of Curcuma xanthorrhiza (125 μg/ml), Andrographis paniculata (250 μg/ml), and Phyllanthus niruri (62.5 μg/ml) inhibited BRSV activity in A549 in pre-, simultaneously-, and post-infection treatment assays, respectively, as measured by the selective index. Reduction of BRSV activities by the herb extracts correlated with inhibition of viral gene expression and inhibition of plaque formation in a concentration- and time-dependent manner. Our findings suggest that these herb extracts have sufficient potency to be used not only as a therapeutic agent but also as a preventive agent to limit BRSV infection.

Abstract

Potensi Desa Luwus sebagai desa pendukung pariwisata karena dilewati jalur utama penghubung antar kota kabupaten serta beberapa tujuan destinasi pariwisata di Bali menjadi faktor pendukung utama mendukung pengembangan produk-produk agraris di desa tersebut. Salah satu produk budidaya lokal yang saat ini berkembang di desa Luwus adalah buah lemon. Tanaman lemon mulai dikembangkan masyarakat setempat sejak pandemi covid-19 mewabah di Indonesia, karena melihat kebutuhan masyarakat terhadap bahan alam yang mampu meningkatkan kekebalan tubuh. Namun dalam perkembangannya saat ini para petani lemon menemui kendala dalam hal pengembangan diversifikasi produk olahan buah lemon, pemasaran dan penurunan jumlah produksi buah lemon segar akibat serangan hama lalat buah. PKM ini memberikan solusi untuk mengatasi permasalahan tersebut. Sehingga ke depannya diharapkan setelah dilakukan pelatihan diperoleh luaran untuk keberdayaan mitra: 1) Peningkatan 100% keterampilan pengolahan buah lemon menjadi minuman kesehatan yang dapat dikomersilkan, 2) Peningkatan 80% kemampuan memasarkan buah lemon dan produk minuman sari buah lemon dengan memanfaatkan media sosial dan e-commerce 3), Peningkatan 100% keterampilan petani untuk menanggulangi hama lalat buah berbahan dasar atraktan alami ramah lingkungan. Hasil yang diperoleh dari kegiatan PKM ini meliputi 1)Aspek produksi yaitu terjadi peningkatan pemahaman terkait manfaat air lemon untuk kesehatan dan peningkatan keterampilan mitra dalam membuat dan mengemas minuman sari buah lemon; 2)Aspek teknologi informasi dan pemasaran yaitu usaha mitra memiliki akun media sosial (facebook, Instagram) dan situs e-commerse berbasis web dan keterampilan mitra dalam membuat dan mengelola akun media sosial dan situs e-commerse meningkat setelah pelatihan dan pendampingan; 3)Aspek solusi hama lalat buah yang menyerang tanaman atau buah lemon yaitu .terjadi peningkatan pengetahuan ekologi dan siklus hidup hama lalat buah dan peningkatan keterampilan mitra dalam membuat perangkap dan menanggulangi hama lalat buah.

Abstract

DNA barcoding is an effective tool for identifying species, with additional uses for measuring molecular diversity, authentication of illegal products, and early detection of disease types. In this review, we discussed the use of DNA barcoding for Avifauna research, that is known to have an important impact on environmental health. The existence of Avifauna is threatened with extinction due to human anthropogenic activities. This review starts by providing a general overview of DNA barcoding, focusing on its application in Avifauna research, metabarcoding, as a tool in detecting food samples from the Avifauna, tracking enandgered and protected exotic birds, and tracking avifauna diseases. This review concludes with definitive statements and challenges regarding the use of DNA barcoding, especially in regions with high levels of avifauna diversity. This review can contribute to a better understanding of DNA barcoding and its potential in managing Avifauna genetic resources in the wild and conservation institutions.

Abstract

Batu Lumbang Mangrove Ecotourism, located in Pemogan Village, Denpasar City, is one of the natural destinations located in a conservation area. Problems still faced by partners include not understanding the use of non-timber forest products (NTFPs) from mangroves as raw materials for food and traditional medicine and partners still need assistance to develop diversified tea bag products from mangrove leaves. Mentoring activities were carried out for 3 months at the Segara Guna Batu Lumbang Mangrove. The aim of this program is to provide education and training in making tea bags made from jeruju leaves (Acanthus ilicifolius) that are hygienic, in the right dosage, and increase partners' understanding of NTFPs through a pocket book. The method used in this community partnership program is a qualitative approach and observation. The target and output of this program is to increase partners' knowledge and skills in processing herbal drinks made from mangrove leaves and increase partners' understanding of the use of mangrove NTFPs. The results of the activity showed that there was an increase in partners' skills and knowledge of 88.87% after the program took place in processing and making herbal tea bags and utilizing mangrove pocket books.

Abstract

Ikan koi (Cyprinus carpio L.) merupakan salah satu komoditas ikan hias air tawar introduksi, yang sampai saat ini masih menjadi primadona di pasar internasional dan termasuk jenis ikan hias yang bernilai jual tinggi dengan harga yang relatif stabil.  Penelitian ini bertujuan untuk menyelidiki pengaruh pemberian hidrogen peroksida dan waktu pengangkutan terhadap respon fisiologis ikan koi serta interaksi antara hidrogen peroksida dan waktu pengangkutan. Metode yang digunakan Rancangan Acak Lengkap Faktorial dua faktor dan tiga kali ulangan. Ikan koi (panjang 10,70+0,97 cm dan berat 14,45+0,21 gram, n = 540 ekor) digunakan dalam penelitian ini. Faktor pertama (A) yaitu konsentrasi H2O2 sebanyak enam taraf (A1) 0,15 ml/l, (A2) 0,35 ml/l, (A3) 0,55 ml/l, (A4) 0,75 ml/l, (A5) 0,95 ml/l, dan (A6) kontrol.  Sedangkan faktor kedua (B) waktu pengangkutan sebanyak tiga taraf yaitu (B1) 10 jam, (B2) 15 jam, (B3) 20 jam. Parameter yang diukur adalah sintasan, kadar kortisol, glukosa, histologi hati, penilaian pasca pengangkutan, dan kualitas air.  Hasil penelitian menunjukan bahwa interaksi konsentrasi hidrogen peroksida dan waktu pengangkutan berpengaruh nyata terhadap sintasan, kortisol, dan glukosa.  Berdasarkan hasil pengujian, perlakuan terbaik diperoleh pada perlakuan A1B1 dengan konsentrasi hidrogen peroksida sebesar 0,15 ml/l dan waktu pengangkutan 10 jam, yang menghasilkan sintasan sebesar 100 %, kadar kortisol sebesar 0,45 ng/ml, dan kadar glukosa sebesar 56,80 mg/dL, tidak terdapat kerusakan pada jaringan.

Abstract

In aquaculture, using sustainable feed, especially feed produced from fishery waste, is an effective strategy for improving fish production performance and reducing disease susceptibility. Protein sourced from yellowfin tuna offal, bones, and by-products can be processed into fish protein hydrolysate in the form of fish meal. This raw material can then be used to produce alternative feed products. These products have economic value and can reduce the production costs of feed for white snapper fish farming among partner communities. Based on observations, the priority problems faced by the KUB Segara Guna Batu Lumbang fishermen group in their white snapper cultivation are: the suboptimal availability of independent feed, a lack of periodic water quality measurement, and low white snapper production performance. This community service program (PKM) aims to improve the knowledge and skills of the fishermen group in white snapper cultivation management through training in feed production, water quality monitoring, and production performance analysis. The PKM activities will be carried out from July to August 2025 at the Batu Lumbang Mangrove Ecotourism in Denpasar. Education will be provided through lectures and workshops, and partners' understanding will be measured through pre-tests and post-tests. The success of the program will be evaluated to determine if the activities of making alternative feed based on fishery waste, water quality management, and production performance analysis can significantly improve the partners' knowledge and skills (p≤0.05). This PKM program can contribute to achieving the Sustainable Development Goals (SDGs), specifically Goal 2 (Zero Hunger), Goal 12 (Responsible Consumption and Production), and Goal 14 (Life Below Water). Furthermore, this program aligns with the Government Program's Asta Cita Point 2, which aims to encourage national independence through food self-sufficiency sourced from the fisheries sector and to support a sustainable blue economy.

Abstract

Problems related to landfills of plastic packaging waste are a source of environmental pollution. This research provides innovation in utilizing biodegradable plastic from cassava and leri peel starch with the addition of chitosan. This study aimed to analyze the results of the synthesis and characterization of chitosan membranes with a combination of cassava and leri peel starch and determine the biodegradation time of biodegradable plastic in EM-4 bioactivation. This study characterized biodegradable plastic membranes using test parameters, namely the SEM, FTIR, Swelling, and tensile strength tests. The biodegradation test was carried out by immersing biodegradable plastic membranes in an EM-4 bioactator for 7 days. Based on the results obtained from the characterization of biodegradable plastic membranes, namely the swelling test with concentrations of 5:3, 7:3, and 9:3, with yield values ​​of 62.9921%, 69.8902%, and 71.4286%. The SEM test obtained the best results at a concentration of 7:3. FTIR test on biodegradable plastic membranes containing hydroxyl (O-H) and carbonyl (C-O) groups. The tensile test obtained optimum results at the concentration of biodegradable plastic membranes, namely 9:3. As for the biodegradation test that has been carried out, the start time for degradation is 3 days.
