Abstract

Penelitian ini bertujuan mengetahui pengaruh suhu dan waktu penyimpanan ovarium terhadap tingkat fertilisasi oosit secara in vitro pada domba. Ovarium dibawa dari rumah potong hewan (RPH) dalam medium NaCl fisiologis pada suhu yang berbeda yaitu 27-28 C, 36-37 C, dan 4 C. Oosit kemudian dikoleksi dari setiap kelompok berdasarkan waktu penyimpanan yang berbeda yaitu 2-4, 5-7, dan 8-10 jam setelah domba dipotong. Oosit dikoleksi dan dimaturasi secara in vitro dalam inkubator 5% CO2, 38,5C selama 28 jam. Oosit kemudian difertilisasi ke dalam drop spermatozoa selama 14 jam dalam inkubator CO2 5%, 38,5 C. Tingkat fertilisasi dievaluasi berdasarkan jumlah pronukleus yang terbentuk. Tingkat fertilisasi oosit yang dikoleksi dari ovarium yang disimpan pada suhu 27-28 C tidak berbeda dengan tingkat fertilisasi oosit yang disimpan pada suhu 36-37 C dan pada suhu 4 C, 2-4 jam setelah kematian hewan (masing-masing 53; 66,66; dan 63%) (P0,05). Tingkat fertilisasi oosit mulai mengalami penurunan pada tiga kelompok perlakuan setelah ovarium disimpan selama 8-10 jam, tingkat fertilisasi oosit yang disimpan pada suhu 27-28 C; 36-37 C; dan suhu 4 C masing-masing berturut-turut sebesar 9,8; 22,22; dan 12,24%. Dari hasil penelitian disimpulkan bahwa penyimpanan ovarium pada suhu 27-28 C dan 36-37 C selama 5-7 jam dapat mempertahankan kompetensi oosit dibandingkan dengan penyimpanan pada suhu 4 C.

Abstract

Penelitian ini bertujuan mengetahui kemampuan perkembangan awal embrio sapi in vitro menggunakan media tunggal untuk maturasi, fertilisasi, dan kultur berbahan dasar tissue culture medium (TCM) 199. Oosit sapi dikumpulkan dari rumah potong hewan dengan teknik aspirasi dan diklasifikasikan berdasarkan kekompakan sel kumulus dan sitoplasma yang homogen. Oosit dimaturasi pada medium TCM 199 yang disuplementasi dengan 10 IU/ml pregnant mares serum gonadotropin (PMSG), 10 IU/ml human chorionic gonadotropin (hCG), dan 10% fetal bovine serum (FBS), dilakukan selama 24 jam pada inkubator 5% CO2, 39 C. Fertilisasi dilakukan pada dua media yang berbeda yaitu media rutin fertilisasi dan media berbahan dasar TCM 199 dengan suplemen bovine serum albumin (BSA) dan heparin. Setelah fertilisasi, kumulus sel dihilangkan (denudasi), kemudian dikultur pada media TCM 199 yang disuplementasi dengan asam amino esensial dan non-esensial serta 10% FBS selama 3 hari. Hasil penelitian menunjukkan tingkat maturasi oosit pada sistem yang digunakan mampu mendukung 81,5% oosit mencapai tahap metafase II (M-II). Tingkat pembelahan embrio lebih tinggi pada media rutin dibandingkan dengan media TCM 199 yakni masing-masing 44,4 dan 23,2%. Jumlah embrio tahap 4-8 sel pada kedua perlakuan tidak berbeda nyata. Dapat disimpulkan media tunggal berbasis TCM dapat digunakan untuk produksi embrio in vitro.

Abstract

Penelitian ini bertujuan mengetahui profil biokimia darah pada sapi perah yang mengalami kawin berulang. Sapi yang digunakan dalampenelitian ini adalah dua puluh ekor sapi perah peranakan Friesian Holstein, berumur 3-8 tahun sudah pernah beranak minimal satu kali, mempunyai siklus reproduksi normal, dan kondisi tubuh sehat. Seluruh sapi dibagi ke dalam dua kelompok yang masing-masing terdiri atas 10 ekor sapi. Kelompok I adalah sapi fertil sedangkan kelompok II adalah sapi yang telah diinseminasi lebih dari tiga kali tetapi belum atau tidak bunting. Profil biokimia darah yang diukur adalah kadar total protein, total kolesterol, glukosa, dan kalsium. Data dianalisis menggunakan t-test. Hasil penelitian menunjukkan bahwa total protein; total kolesterol; glukosa; dan kalsium pada kelompok I vs kelompok II masing-masing adalah 6,770,38 vs 6,820,821 g/dl (P0,05); 166,0837,06 vs 125,9538,108 mg/dl (P0,05); 68,409,60 vs 48,586,675 mg/dl (P0,01); dan 9,901,43 vs 9,230,94 mg/dl (P0,05). Disimpulkan bahwa sapi-sapi perah yang mengalami kawin berulang mempunyai total kolesterol dan kadar glukosa yang lebih rendah dibanding sapi-sapi perah yang fertil.

Abstract

One factor that affects frozen semen characteristics is bull age. Increasing age induces changes in sperm that reduce frozen semen quality. This study aimed to evaluate the frozen semen characteristics of Bali bulls aged 3, 8, and 13 years. Frozen semen was derived from fresh semen with sperm motility > 70%. The frozen semen characteristics tested included motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Sperm motility was analyzed by computer-assisted sperm analysis (CASA). Viability was assessed using eosin-nigrosin staining; plasma membrane integrity was assessed using the hypoosmotic swelling test (HOST); acrosome integrity was assessed using Giemsa staining; and sperm abnormalities were assessed using Williams staining. The plasma membrane integrity of the 13-year-old group was significantly lower (P < 0.05) than that of the younger age groups. However, there were no significant differences (P > 0.05) in the other parameters. We conclude that age affects the integrity of the sperm plasma membrane in frozen semen from Bali bulls.

Abstract

Lumba-lumba hidung botol Indo-Pasifik merupakan model potensial untuk pengembangan preservasi semen pada mamalia laut. Pengencer komersial berbasis lesitin digunakan untuk preservasi semen cair lumba-lumba hidung botol indo-pasifik. Penelitian ini bertujuan untuk mengeavaluasi karakteristik semen cair lumba-lumba hidung botol pasca penyimpanan pada suhu 5 °C dalam pengencer komersial berbasis lesitin. Semen segar dari dua lumba-lumba hidung botol indo-pasifik berusia 11,5 dan 18 tahun dikoleksi satu minggu sekali sebanyak 3 kali ulangan. Semen segar dievaluasi secara makroskopis dan mikroskopis. Semen dengan motilitas sperma lebih dari 70% dibagi menjadi empat tabung, masing-masing dengan dan tanpa perlakuan sentrifugasi. Semen dibagi menjadi konsentrasi 100×106 ml-1 dan 200×106 ml-1, kemudian disimpan pada suhu 5 °C selama 4 hari. Evaluasi dilakukan setiap 24 jam meliputi motilitas, viabilitas, dan membran plasma utuh (MPU) spermatozoa. Hasil penelitian menunjukkan adanya penurunan yang signifikan (P < 0,05) pada kualitas sperma setiap 24 jam pemeriksaan pada semua perlakuan. Kualitas semen cair dipertahankan dengan motilitas ≥40% hingga hari ke-2 setelah penampungan semen. Preservasi semen cair sperma lumba-lumba hidung botol indo-pasifik dapat dilakukan dalam jangka waktu penyimpanan yang singkat yaitu 48 jam pada semua perlakuan dengan pengencer komersial berbasis lesitin.

Operational implementation, such as laboratory staff and equipment, differ for each frozen semen producer, resulting in variability in the quality assessment of the frozen semen produced. Conventional assessments subject to subjectivity, human error, and high variability. Computer-assisted semen analysis (CASA) is considered more objective. This study aimed was to conduct a comparative analysis of motility and concentration assessment of frozen semen sperm using conventional and the portable CASA AndroScope. Twenty-one laboratory assistants from 21 national and regional frozen semen producers participated in this study. Sperm motility and concentration were assessed conventionally (using a Neubauer chamber) and AndroScope was used with five replicates. The results of the comparison of sperm motility scores revealed significant differences (p<0.05) between conventional examination and using AndroScope. The results of the comparison of the sperm concentration calculations revealed no significant difference (p>0.05) between the calculations performed using the Neubauer chamber and AndroScope. Assessment with the AndroScope is considered reliable and can replace conventional assessment in assessing motility and sperm concentration, thereby improving and standardizing the assessment performed by frozen semen producers.

Abstract

This study evaluated the effectiveness of animal, plant, and synthetic lecithin-based extenders in preserving local Indonesian ram semen during cryopreservation. Post-thaw sperm quality was assessed using Computer-Assisted Sperm Analysis (CASA), focusing on motility, viability, membrane integrity, and kinematics. Data were analyzed using ANOVA and Duncan’s Multiple Range Test (DMRT) at a 95% significance level. Results showed that sperm motility, viability, and membrane integrity were significantly higher (P<0.05) in the animal lecithin-based extender (53.66±1.33%, 66.45±2.50%, and 67.68±2.33%, respectively) compared to plant (51.84±0.48%, 62.98±1.94%, and 64.27±1.51%) and synthetic (50.14±0.22%, 60.72±0.81%, 62.44±1.37%) extenders. While kinematic parameters were not significantly different among groups, synthetic lecithin showed slightly higher values in velocity-related measures, whereas animal lecithin resulted in the most consistent improvement in overall sperm quality. All lecithin-based extenders met the Indonesian National Standard (SNI) for semen quality, and sperm kinematics remained within optimal fertilization values after cryopreservation.
