CHARACTERIZATION AND IDENTIFICATION OF SM4 BACTERIAL ISOLATE FROM Stylissa massa SPONSE AS PRODUCING ANTIMICROBIAL COMPOUNDS AGAINST PATHOGENIC BACTERIA Lukas Pardosi

Abstract

East Nusa Tenggara (NTT) is one of the archipelagic provinces in Indonesia with a sea area of 200,000 km² and contains potential marine resources including the Stylissa massa sponge. Bacteria have been known to associate with sponges and have the ability to produce antimicrobial compounds. This study aims to characterize, identify the 16S rRNA gene, and test the bioactive compounds of SM4 bacterial isolates from the Stylissa massa sponge. The bacterial isolates of SM4 were previously rejuvenated on NA medium, then characterized by macroscopic morphology, and biochemistry by citrate test and TSIA test. The culture was extracted using petroleum ether solvent and the antibacterial activity was tested against E. coli and S. aureus by the well method. The isolates were then identified based on the 16S rRNA gene by PCR method. The results showed that, SM4 isolates had small colony size, yellow color, round colony shape, flat elevation, flat margins, while biochemically they had positive results with Citrate and TSIA tests. The antagonist test of the SM4 bacterial isolate was found to have the ability to inhibit the growth of S. aureus and  E. coli, while the antibacterial activity test of the bioactive compound bacterial isolate SM4 was known to have the ability to inhibit the growth of S .aureus with an inhibition zone of 1.99 mm and E. colli 1.56 mm. The results of the molecular identification of the bacterial isolate SM4 had 99% similarity with the Bacillus cereus strain Nr_074540.1

Abstract

Kabupaten Timur Tengah Utara (TTU) merupakan salah satu Kabupaten di Provinsi Nusa Tenggara Timur (NTT) yang memiliki luas wilayahnya ± 2,669,70 km². Kabupaten TTU memiliki lahan pertanian yang luas dan berpotensi untuk budidaya tanaman cabai merah (Capsicum annuum L.). Penelitian ini bertujuan untuk mengisolasi, mengkarakterisasi dan menyeleksi potensi bakteri pelarut fosfat (BPF) yang berada disekitar perakaran tanaman cabai merah. Isolasi BPF menggunakan metode gores dan metode sebar. Hasil isolasi diperoleh 20 isolat. Hasil karakterisasi ke 20 isolat memiliki bentuk koloni bulat, rhizoid, panjang, ukuran kecil, sedang, besar, elevase rata, warnah putih, bening. Hasil seleksi BPF diketahui 5 isolat berpotensi sebagai pelarut fosfat dan yang memiliki indeks pelarutan fosfat tertinggi yaitu isolat CMB04 sebesar 2,10 mm.

Abstract

Research on the isolation, toxicity test, antimalarial test, and identification of the active compound from the ethyl acetate fraction of Stylissa massa sponge from Oenggae waters, Rote Island, has been conducted. This study aimed to investigate the antimalarial activity of the ethyl acetate fraction of the Stylissa massa sponge. Isolation was carried out by the extraction method using a mixed solvent of methanol: dichloromethane of 3: 2 (v/v), then the extract was partitioned in a solvent mixture of ethyl acetate: water of 1: 2 (v/v). The ethyl acetate extract obtained was separated by column chromatography using the gradient polarity system method. The toxicity test of each fraction was carried out by the Brine Shrimp Lethality Test (BSLT) method, and the antimalarial test was carried out by the haematin polymerization inhibition method. Identification of compounds from the active fraction in the antimalarial test was carried out using Liquid Chromatography-Mass Spectrometry (LC-MS). The extraction yield was 1.14 g (0.23%) of the ethyl acetate extract in the form of a dark brownish-yellow oily solid. Separation by column chromatography resulted in 15 fractions. Toxicity test results showed the four most active fractions with LC50 values, which are very promising for new drug discovery. The IC50 value in the antimalarial activity test of the four fractions indicated that the Stylissa massa sponge ethyl acetate extract was more active than the standard chloroquine compound (115 μg/mL). The LC-MS analysis indicates that fraction 11 contains two compounds that have been reported, and 1 compound is unknown. In contrast, fraction 14 indicates that it contains three compounds that have been reported and one unknown compound.

Abstract

Soil is a habitat for small animals and microorganisms. The most abundant microorganisms in soil are bacteria. The soil used in the study was soil obtained from paddy fields in Naen, North Central Timor Regency.The study aimed to characterize the bacterial isolates obtained from the paddy fields which are known to produce antibiotics. Rice soil samples were taken at a depth of about 15 cm from the soil surface and then cultured in Nutrient Agar media with graded dilutions. Characterization was carried out by macroscopic observation, namely direct observation, then a challenge test was carried out against S. aureus bacteria. The results showed that there were 6 bacterial isolates with macroscopic results mostly irregular in shape and 1 isolate fibrous. There were 4 bacterial isolates namely STS1, STS3, STS4, and STS6 which had inhibitory power against the growth of S. aureus.

Abstract

The training on tempeh fermentation from pigeon pea was carried out by a lecturer team from Timor University to the people of Tublopo Village (Sehati Farmer Group). Tublopo Village has one of the natural resources that potentially to be developed, namely Pigeon pea. The lack of knowledge and skills of the local community in processing the abundant pigeon pea is the reason for conducting this activity. Training on pigeon pea processing into tempeh can improve the selling value of the product and produce products with new innovations that have better quality. The used method in this service activity is in the form of counseling and the practice of making pigeon pea tempeh. At the counseling stage, the service team described the employed materials, tools and the steps for tempeh fermentation, then continued with a demonstration of making tempe according to the described stages. The main product from this service is pigeon pea tempeh. In addition to tempe products, members of the Sehati Farmer Group have been equipped with the knowledge and skills to produce tempe independently.

Abstract

The people of Dusun Banopo mostly work as farmers who produce a variety of agricultural products. One of the products produced is mung beans. The yield of bean sprouts is very good and plentiful, however the utilization of this product is still very low, especially in supporting family health and increasing family income. Most of the produce is sold immediately and a quarter of the harvested are usually left at home to be cooked with corn. Mung bean seeds can be germinated into bean sprouts which have a higher nutritional content and can be used as marketed products. This service activity was well received by the community, especially the women from the Women Farmers Group. This group gains knowledge and skills in making bean sprouts with media that can be taken from the surrounding environment. Besides being able to be used as a nutritious vegetable in improving family health, it can also marketed to increase family income.

Abstract

The aim of the study was to explore, identify and describe the types of Lichenes in the foothills of Mount Mutis in Eban Village. This research was conducted in April-July 2022 using a qualitative descriptive approach with the reconassance method. Sampling used a purposive sampling technique by taking samples, collecting and collecting them to assist in the laboratory. The results of research that have been carried out by lichens found at the foot of Mount Mutis, Eban Village, West Miomaffo District, amount to 21 types of lichen species with descriptions: Leptogium azureum, Pertusaria sp, Lepraria Loficans, Lepraria incana, Parmelia saxatilis, Parmotrema tinctorum, Cetrelia olivetorum, Heterodermia japonica, Usnea hirta, Parmotrema perlatum, Graphis scripta, Graphis sp 1, Graphis sp 2, Haematomma accolens, Cryptothecia striata, Bacidia schweinitzii, Bacidia sp, Dirinaria applanate, Opegrapha gyrocarpa, Lecidella elaeochroma, Caloplaca ferruginea. The most common lichen groups were the 12 families Parmeliaceae most commonly found in research lines with 5 species, Graphidaceae 4 species, Bacidia 2 species, Stereocaulaceae 2 species, and Collemataceaee, Pertusariaceae, Haematommataceae, Artho-niaceae, Physciaceae, Opegraphaceae, Lecidellaleceae, Teloschistaceae 1 species each. Lichen substrate found living on tree trunks, tree stalks and rocks. The form of lichen thallus that was most commonly found was Crustose which was 62%, then Foliose thallus was 33% and Fruticose was 5%. The diversity of mosses in the study area is strongly influenced by both abiotic and biotic environmental factors. The type of Licen found in Eban Village is a Lichen which is able to adapt to an environment which incidentally is dominated by dry land.

Abstract

This community service was held because of the low awareness of sellers in Pasar Baru to wear masks and the lack of knowledge of sellers about how to wear masks correctly. The socialization activity for the prevention of the spread of Covid-19 to sellers at Pasar Baru Kefamenanu was held for one day. Pasar Baru is located on Jl. Sisingamangaraja, Benpasi, Kefamenanu City, North Central Timor, East Nusa Tenggara, Indonesia. The main target of this activity is the sellers operating in the new market area of ​​Kefamenanu city. The activity began with socializing the prevention of the spread of Covid-19 and closed with the distribution of masks for free to sellers at Pasar Baru Kefamenanu and how to use masks correctly.

Abstract

Utilization of information and communication technology media has penetrated into all areas of life. One of the uses of information and communication technology media in the field of education is e-learning. E-learning (electronic learning) is a learning process that utilizes information technology in this case utilizing online media such as the internet as a method of delivery, interaction and facilitation. The use of e-learning platforms can make it easier for teachers and students in the learning process. The objectives of this activity are: First, to provide concepts and to disseminate the use of Edmodo for the learning process. Second, providing knowledge about how to use and apply the features contained in the Edmodo platform to facilitate the learning process. The targets for this community service are teachers at SMP Negeri 1 Ndona, Ende. This community service is expected to be able to provide additional skills for teachers how to use the e-learning platform (edmodo) for free to support conventional learning during this covid-19 pandemic.

Abstract

Abstract One of the e-learning platforms that can be used during the Covid-19 pandemic is Edmodo. Edmodo learning media which is very functional, besides being able to be used for students and lecturers, can also be used by parents. Assignments are given through the assignment and quiz features that have a time limit, so they can practice discipline in doing assignments. The purposes of implementing this activity are; First, to provide concepts and socialization of the use of Edmodo for the learning process. Second, to provide knowledge about how to use and apply the features contained in the Edmodo platform to facilitate the learning process. The targets for this service are teachers at SMA Negeri 1 Demon Pagong, East Flores. This service is expected to be able to provide additional skills for teachers how to use the e-learning platform (edmodo) for free to support conventional learning during this covid-19 pandemic. This activity lasted for one day by using Zoom.
