Abstract

Background: The province of East Nusa Tenggara (NTT), Indonesia, boasts a significant expanse of marine territory, making its waters a potential treasure trove of aquatic resources. One such marine resource is the Stylotella SP sponge, which is found in this region. Sponges like Stylotella SP often have intricate relationships with microorganisms, including bacteria. Method: This research was to characterize and identify proteolytic bacteria and test bioactive compounds from isolates of proteolytic bacteria from the sponge Stylotella sp. Proteolytic bacterial strains were rejuvenated first in 2% TSA + Nacl medium and then characterized microscopically and biochemically using catalase and oxidase tests. The diffusion method used the antibacterial activity test against E.coli and S. aureus. Result: The results showed that proteolytic bacterial isolates had irregular and rounded colony shapes, wavy and intact colony edges, hilly and convex colony elevation, and white color, while biochemically, it was declared positive for the catalase and oxidase tests. Antibacterial activity tests on proteolytic bacterial strains showed the ability to inhibit the growth of Staphylococcus aureus with an inhibitory capacity of 9.5 mm and Escherichia coli with an inhibitory capacity of 7 mm.

Abstract

This study aimed to determine the effect of L-isoleucine supplementation in the diet on the growth performance of Kampong chickens at the starter phase (1-6 weeks). This study was conducted at Kefamenanu, East Nusa Tenggara Province, Indonesia, from May to June 2023. A total of two hundred and fifty-week-old Kampong chickens with an average initial weight of 59.03±0.52 g were used in the study. The chickens were divided into five treatments with five replications based on a completely randomized design. The diet treatments were: T0 (control feed); T1 (T0 + 0.20% L-isoleucine, NRC, 1994 for broilers); T2 (T0 + 0.30% L-isoleucine); T3 (T0 + 0.40% L-isoleucine); and T4 (T0 + 0.50% L-isoleucine). The parameters observed were body weight, weight gain, feed intake, feed conversion ratio, carcass weight, and carcass percentage. The data obtained were analyzed using variance analysis and the Duncan test. The study result showed that the groups T3 and T2 had the highest body weight (330.76±1.29; 334.48±1.47 g/bird, respectively). The highest weight gain was in group T2 (275.54±1.39 g/bird/5 weeks). Group T3 had the highest feed intake (613.15±5.79 g/bird/5 weeks) and the lowest feed conversion ratio (2.22±0.02). The highest carcass weight was in groups T2 and T3 (189.95±1.17 and 189.32±0.65 g/bird, respectively), while the highest carcass percentage was in group T2 (57.43±0.47 %/bird). The statistical analysis indicated that body weight, weight gain, feed conversion ratio, and carcass weight were affected by the level of L-isoleucine (p<0.05). In contrast, feed intake and carcass percentage were not significant. It was concluded that supplementation with 0.40% L-isoleucine to the diet of Kampong chicken resulted in optimal growth performance at the starter phase (1-6 weeks).

Abstract

Kelompok tani Perempuan Sion aktif dalam kegiatan bercocok tanam di lahan kering. Namun terdapat kesulitan dalam mengelola lahannya. Tujuan pengabdian ini adalah meningkatkan ketahanan pangan kelompok tani Perempuan Sion melalui penerapan pekarangan lestari. Pelaksanaan dilakukan melalui sistem learning by doing dengan sosialisasi dan demonstrasi yang melibatkan kelompok tani Perempuan Sion dan mahasiswa sebagai implementasi MBKM. Kegiatan berlangsung dengan durasi 3 (tiga) bulan mulai dari Juli – September 2022. Tahapan kegiatan dimulai dari persiapan lahan, pembuatan pupuk bokashi berbahan dasar limbah dan kotoran ayam, persemaian benih pakcoy, pemupukan, penanaman, perawatan tanaman, dan pemanenan. Berdasarkan hasil kegiatan pengabdian, kelompok tani Perempuan Sion mampu mengubah limbah menjadi bokashi, mampu mengelola lahan kering sebagai lahan pertanian yang lebih efektif dan efisien baik secara waktu dan tenaga, serta mampu meningkatkan ketahanan pangan.

Abstract

Indole Acetic Acid (IAA) merupakan hormon tumbuh yang memegang peranan penting pada semua tanaman dalam meningkatkan pertumbuhan dan perkembangan. Tujuan penelitian yaitu untuk mengkarakterisasi bakteri rhizosfer penghasil IAA dari tanaman kacang gude (Cajanus cajan L) sebagai penghasil IAA dan mengetahui pengaruh isolat bakteri tersebut terhadap perkecambahan benih padi (Oryza sativa L). Penelitian diawali dengan pengambilan sampel, isolasi bakteri menggunakan media Nutrient Agar (NA) dan seleksi isolat bakteri penghasil IAA. Seleksi isolate baketri penghasil IAA dilakukan dengan cara menumbuhkan isolat bakteri pada media Nutrient Broth (NB) + triptofan dan direaksikan dengan reagen Salkowski. Hasil isolasi diperoleh tujuh isolat dengan karakter morfologi koloni berukuran kecil dan sedang, warna koloni putih, bentuk koloni bundar, berserabut, tidak teratur, elevasi koloni datar, umbunate, pulvinate, dan cembung. Tepi koloni rata, bergelombang dan berlekuk. Hasil seleksi didapatkan tiga isolat yaitu RTG-01, RTG-02 dan RTG-03 yang menghasilkan IAA. Hasil identifikasi mikroskopis isolat RTG-01 dan RTG-02 bentuk sel bulat Gram negatif dan isolat RTG-03 bentuk sel batang Gram positif. Isolat bakteri penghasil IAA diaplikasikan pada benih padi dengan menggunakan Rancangan Acak Lengkap (RAL) non factorial. Hasil analisis ragam menunjukan bahwa perlakuan ketiga isolat bakteri penghasil IAA tidak berpengaruh nyata (p<0.05) terhadap potensi tumbuh maksimum, indeks vigor, dan berat kering kecambah normal padi, namun berpengaruh nyata (p>0.05) terhadap daya berkecambah.

Abstract

Enzim Endonuklease diisolasi dari beberapa jenis bakteri termasuk Escherichia coli. Tujuan penelitian ini adalah untuk mengetahui karakteristik enzim endonuklease restriksi kasar dari bakteri E. coli yang dipengaruhi kondisi suhu dan pH. E. coli diisolasi dari sampel air sumur dan ditumbuhkan pada media Eosin Methylene Blue Agar (EMBA). Isolat bakteri E coli digunakan dalam pengukuran kurva pertumbuhan untuk mengetahui fase pertumbuhan bakteri serta mendapatkan waktu yang optimum untuk memproduksi ekstrak kasar enzim endonuklease. Proses produksi enzim endonuklease dengan metode sentrifugasi pada kecepatan 10000 rpm selama 30 menit pada suhu 4°C dan diulang 4 kali. Hasil sentrifugasi dikarakterisasi dengan fraksinasi menggunakan ammonium sulfat dengan tingkat kejenuhannya berbeda-beda yaitu, (0-20%, 20-40%, 40-60%, 60-80%, 80-100%) dan diukur aktivitas enzimnya menggunakan spektrofotometer pada panjang gelombang 600 nm. Hasil fraksinasi yang aktivitasnya paling optimum pada fraksi 0-20%. Selanjutnya dilakukan pengujian aktivitas enzim dengan variasi suhu dan Ph menggunakan spektrofotometer dengan panjang gelombang 600 nm. Hasilnya pada variasi suhu aktivitas enzim paling optimum terjadi pada suhu 37°C, sedangkan pada variasi pH aktivitas optimum enzim terjadi pada pH 7.

Abstract

Tanaman kelor (Moringa oleifera Lam) marupakan salah satu tanaman yang dimanfaatkan sebagai tanaman obat di Nusa Tenggara Timur (NTT). Semua bagian tanaman kelor mengandung senyawa metabolit sekunder. Tujuan penelitian ini ialah mengetahui kandungan senyawa bioaktif dari ekstrak heksana bakteri endofit dari bunga dan biji kelor. Isolat bakteri endofit yang diperoleh dikarakterisasi morfologi koloni, mikroskopis, pengujian biokimia dan diidentifikasi molekuler. Kandungan kimia bakteri endofit dianalisis dengan metode standar untuk skrining senyawa kimia. Uji aktivitas biokontrol dilakukan terhadap penyebab penyakit layu Fusarium dengan difusi agar. Hasil isolasi menujukkan adanya 8 isolat, dan 2 isolat yaitu BiK.1 dan BiK.3 menghasilkan senyawa bioaktif yang mampu menghambat pertumbuhan jamur patogen Fusarium sp. Dua isolat BiK.1 dan BiK.3 memiliki golongan senyawa alkaloid, terpenoid dan saponin. Ekstrak kasar BiK.1 mempunyai zona hambat 3.1% terhadap Fusarium oxisporum TLPI, sedangkan isolat BiK.1 sebesar 1,2%.   Identifikasi secara molekuler berdasarkan gen 16S rRNA diketahui bahwa isolat BiK.1 memiliki kemiripan genetik dengan Bacillus sp. dan BiK.3 dengan Bacillus cereus dengan nilai query cover mencapai 100%.

Abstract

Mushrooms are eukaryotic organisms with spores, without chlorophyll that reproduce sexually and asexually, mushrooms can be used by humans as food ingredients and ingredients for traditional and modern medicine. The purpose of this study was to determine the type and habitat characteristic of macroscopic fungi conducted in the kateri procted forest area Malaka Regency. Using exploration and  identification methods where the plant identification process is a process of matching a plant according to a particular taxonomi  on each research transect line. There are 28 species of macroscopic fungi namely Dacrymyces stilatus, Auricularia auricula-judae, Xylaria hypoxylon, Daldinia concentrica, Microporus xanthopus, Earliella scabrosa, Pycnoporus sanguineus, Polyporus alveolaris, Lentinus squarrosulus, Hexagonia tenuis, Trametes pubescens, Tyromyces chioneus, Marasmius oreades, Marasmius haematocephalus, Baespora myosura, Marasmius rotula, Marasmius andorsaceus, Trogia infundibiliformis, Leucocoprinus brebissonii, Lepiota helveola. Agaricus micromegathus, Bolbitius vitelinus, Mycena albicocapilaris, Crepidotus variabilis, Parasola plicatilis, Coprinopsis fragilisimus, Lycoperdon pyriforme, Pleurotus djamor. Divided into 5 Orders namely Dacrymycetales, Auriculariales, Xylariales, Polyporales, Agaricales. And 12 family namely Dacrymycetaceae, Auriculariaceae, Xylariaceae, Polyporaceae, Marasmiaceae, Agaricaceae, Bolbitiaceae, Mycenaceae, Crepidotaceae, Psathyrellaceae, Schizophyllaceae, Pleurotaceae. The most abundant habitat Characteristics were on rotten wood branches as many 9 species, on dead  wood stem many as 8 species, in leaf little many as 4 species and in soil with hums many as 7 species.

Abstract

This Community Service Activity aims to increase the engagement and motivation of students at SMAN 1 Kefamenanu through training and mentoring using Kahoot as an interactive gamification-based learning medium. The activity was carried out in three main stages: training, mentoring, and evaluation, involving 50 teachers and 50 students. The training focused on improving teachers' abilities in designing and managing Kahoot quizzes, while mentoring was provided to ensure that teachers could implement the use of Kahoot in the classroom learning process. Evaluation was carried out through observation, questionnaires, interviews, and analysis of quiz results to obtain quantitative data. The results of the activity showed a very high level of participant enthusiasm with an attendance rate of 97%. 93% of teachers were able to create and manage quizzes well and 86% were able to implement them in the classroom learning process. From a student perspective, 95% said Kahoot could increase their engagement in learning and 96% experienced increased learning motivation and 82% were able to answer quiz questions correctly. The level of participant satisfaction reached 85% and 78% of teachers expressed a commitment to using Kahoot continuously. Based on these results, it can be concluded that training and mentoring on the use of Kahoot are effective in improving teacher competence, student engagement, and learning quality, and have good potential for sustainability if supported by continued mentoring and school policies that support the use of learning technology.Keyowords: Student Engagement, Motivation, Kahoot, Gamification, Training, Mentoring

Abstract

This study aimed to isolate and identify bacteria from the rumen fluid of Bali cattle and to evaluate their effect as microbial inoculants on the in vitro digestibility of rice straw. The experiment was conducted using a Completely Randomized Design (CRD) consisting of four treatments and four replications. The treatments included rice straw silage with the addition of 0%, 5%, 10%, and 15% of local microorganisms (MOL) derived from cattle rumen fluid. The observed variables included the isolation and identification of bacteria from the rumen fluid, and the assessment of in vitro digestibility, including Dry Matter Digestibility (DMD), Organic Matter Digestibility (OMD), Volatile Fatty Acid (VFA) concentration, and ammonia (NH₃) production. The results showed that the addition of rumen fluid MOL significantly (P<0.05) improved the digestibility parameters. The highest DMD (52.44%) and OMD (47.11%) were obtained from the treatment with 15% MOL. VFA and NH₃ concentrations also increased, indicating enhanced microbial activity and fermentation efficiency. The study concludes that MOL derived from cattle rumen fluid, particularly those containing Bacillus sp., is effective in improving the nutritional quality and digestibility of rice straw
