Abstract

Exploration of bioactive compound were continuously done as more new diseases appearing, from infection, cancer, and other degenerative diseases. The invention of new antibiotic compound and antioxidant compound is very needed to solve this problem. Endophytic microbes are microbes which lives in plant’s culture and produce the active secondary metabolite. In previous research, 2 compounds from endofitic microbes Penicillium sp in kunyit putih had been successfully isolated and identified as di-(2-ethylhexyl)phthalate   and  pyranon derivated as 5 (4’-ethoxy-2’-hydroxy-5’-methyl-2’,3’-dihydrofuran-3’-il (hydroxy) methyl-4-isopropyl-3-methyl-2-pyran-2-on). The goal of this research is to test antibacterial and antioxidant activitiy from derivate pyranon, which is resulting from endofitic microbes Penicillium sp in kunyit putih (Curcuma zeodaria). The method used to test the antibacterial activity is dic diffusion, using E. Coli, S. Dysenteriae, S. Aureus, and B. Subtilis. with variation of concentrations 250, 500, 1000, dan 2000μg/mL, and to test the antioxidant activity used 1,1-diphenyl-2- picrylhydrazyl ( DPPH)  method with variation of concentrations 1000, 500, 250, 125, 62.5, 31.25, 15.625, and 7.8125 µg/mL. The research         shown that  the antibacterial activity is strongest at S. aureus with 11mm-blocked-zone at 250µg/mL  concentration. Pyranon derivate also has active antioxidant manner with IC5016.05µg/mL.

Abstract

Telah dilakukan isolasi senyawa fenolat dari fraksi etil asetat buah tumbuhan mempelas (Tetraceraindica Merr.). Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut metanol dan pemisahansenyawa menggunakan teknik kromatografi. Hasil isolasi diperoleh kristal berbentuk jarum berwarna kuning seberat 4,01 mg dengan titik leleh 297-298◦C. Munculnya puncak serapan spektrum UV pada 277 nmmengindikasikan bahwa pada senyawa hasil isolasi memiliki ikatan rangkap terkonjugasi yang lazimnya merupakan cincin aromatis. Spektrum IR memberikan puncak absorbsi pada bilangan gelombang 3424, 3103-3010,2947, 1715, 1438 -1608, 1386-1354, 1182 cm−1yang masing-masing menunjukkan adanya gugus O-H bebas,C-H aromatik, regang C-H alifatik, C=O keton, C=C aromatik, lentur C-H alifatik, C-O-C. Berdasarkanspektrum LC-MS menunjukkan berat molekul senyawa isolasi adalah 300.

Abstract

In Indonesia, Scurrula atropurpurea is a medicinal plant known as benalu. Triacylglycerols can be produced by plants, animals, algae, and microorganisms such as endophytic fungi. Triacylglycerols can be used in cosmetics, food, and medicine because they have biological activities such as antitumor, antibacterial, and cytotoxic. Besides, the compound can be used as a biodiesel substitute for triacylglycerols sourced from oil palm. This study aims to isolate and characterize triacylglycerol from biomass of endophytic fungal N. surinamensis from the S. atropurpurea  leaves. The compound was isolated and purified by the column chromatography method. The structure of the compound was determined by spectroscopic data ( FTIR, 1H-NMR, and 13C-NMR). Analysis of the spectrum and compared with the literature, the isolated compound is a triacylglycerol.

Abstract

A tetraoxygenated xanthone, 1,7-dihidroxy-3-methoxy-4-(3methylbut-2-enyl),6’,6’ –dimethylpyrano (2’ , 3’ : 5, 6) xanthone had been isolated from the ethyl acetate extract of the stem bark of Garcinia nigrolineata Planch Ex T. Anders. The structure of these compounds were determined on the basis of spectroscopic NMR 1D and 2D.

Abstract

Ogan ethnic community in South Sumatera has been used a decoction of the stem bark of Syzygium samarangense L (locally named “jambu air” in Indonesia) for hypertension medicine. Hypertension is a degenerative disease caused by free radical activity in the body. This study aims to scientifically prove the use of a decoction of the stem bark of S. samarangense so that its use by the Ogan ethnic community can be justified. The research began with fractionation of the stem bark by gradient solvents, and each fraction was tested for antioxidant activity. Isolation of antioxidant compounds from active fractions was carried out by chromatographic techniques. Antioxidant tests on fractions and pure compounds were carried out by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Based on spectroscopic data analysis and by comparing with literature, the pure compounds were identified as quercetin and gallic acid. Both of these compounds have strong antioxidant activity with IC50 quercetin 11.16 and gallic acid 11.43 µg/mL. This research proves that the use of the stem bark of S. samarangense for a degenerative disease by the Ogan ethnic community contains the truth.

Abstract

Garcinia sizygiifolia  is  a native plant to the South Sulawesi region popularly known as sula and has been cultivated in several regions in Indonesia.  The plant by local community use as a food and source of wood, but has not found information on chemical content and biological activity.  Therefore, this study was carried out to Isolation and evaluation of the antioxidant activity of the phenolic constituent of the G. sizygiifolia.  A 30.0 g portion of ethyl acetate extract of the stembark G. sizygiifolia were separated by column chromatography method using silica G 60 F254 (230-400 mesh), eluted gradient polarity mixtures of n-hexane-ethyl acetate were collected and sorted into fractions. Fraction  F 1 (5.2 g) were separated and purified again by chromatography method until pure compound obtained. The structure of the isolated compound was determined using UV, IR, and NMR spectroscopy. The antioxidant activity was tested by DPPH method. The isolated  pure compound was a yellow solid with melting point 148-149 oC. Base on spectroscopy data and by comparison with data from the literature, isolated compound is a known compound 2,4-dihydroxyphenylethanone. The compound exhibited antioxidant activity with IC50 96 µg/mL against DPPH.

Abstract

Leaves of jambu air (Syzygium samarangense L.) has been used by local residents as medicine for various diseases caused by free radical agents in human’s body. This study aims to find a new source of antioxidants from medicinal plants and their endophytic fungi Syzygium samarangense leaves was fractionated by maceration method using gradient solvent i.e.  n-hexane, ethyl acetate, and methanol. The antioxidant activity of the leaf fractions was determined using 1,1diphenyl-2-picryl hydrazyl (DPPH) method. The chemical compound was isolated from active fraction by chromatographic techniques and their chemical structure was identified using spectroscopy techniques.  The endophytic fungi was isolated from S. samarangense leaves and continued to cultivation in potato-dextrose broth (PDB) medium for four weeks. The medium was extracted using ethyl acetate and determined its antioxidant activity. Fungi’s isolate with highest activity was analyzed to identify its molecular. Ethyl acetate fraction of S. samarangense leaves showed highest antioxidant activity. Spectroscopy analysis result concluded the isolated compound is 5,7-dihydroxy-6,8-dimethyl flavanone. Four endophytic fungi had been isolated form S. samarangense leaves. Antioxidant activity test showed that ethyl acetate extract of endophytic fungi BJA-1 has the highest value. Molecular identification of BJA-1 shows high homology with Lasiodiplodia venezuelensis strain CBS 129753.

Abstract

Tetracera indica Merr. in Musi Banyuasin, is one of the traditional medicine used by the community for the treatment of kidney stone disease and gout, but this claim is not recorded in the treatment of kidney stones and gout in Indonesia.  In this study, isolation of antioxidant compound from ethyl acetate extracts of supit (Tetracera indica) was done. The isolation was carried out through step gradient polarity extraction, and separated and purified by chromatography technique. The determination of the structure of the isolated compound was performed by spectroscopy method including UV, IR, and NMR 1D and 2D, and antoxidant activity was determined  by DPPH method.  An  active antioxidant compound was isolated from ethyl acetate extract in form of yellow solid (15 mg).  Based on spectroscopic analysis the isolated compound was 5,8-dihydroxy-7-methoxyflavone.  The compound showed strong antioxidant activity (IC50 8.25 μg/mL) higher than standard ascorbic acid (IC50 11.3 μg/mL).  This data concluded that efficacy of supit (Tetracera indica) for the treatment related to antioxidant activity (uric acid) is proven by the identification of one antioxidant compound of this plant.

Abstract

Flacourtia  rukam is a plant popular among people to treat hypertension, especially the Musi Banyuasin of south Sumatera, Indonesia. Isolation and identification of chemical compounds from F. rukam leaves and evaluation of their effects on antihypertensive activity have been conducted. Isolation of chemical compounds using chromatographic methods and identification using spectroscopic methods were compared with the reported data. The drug’s effects on antihypertension were determined using the  angiotensin converting enzyme (ACE) inhibitory method. Two compounds were first reported and isolated from the leaves of F. rukam and identified as apigenin (1) and lupeol (2). These compounds were demonstrated to be effective in treating antihypertension with IC50 656.51 ± 1.55 µg/mL for apigenin and 15.12 ± 0.72 µg/mL for lupeol. It can be concluded that  F. rukam leaves is a potential ACE inhibitor can be explored further as an effective antihypertensive agent.

Abstract

Telah dilakukan isolasi satu senyawa metabolit sekunder dari ekstrak n-heksana batang Tinospora crispa. Ekstraksi dilakukan dengan metode maserasi. Pemisahan dan pemurnian dilakukan dengan teknik kromatografi. Terhadap senyawa hasil isolasi dilakukan uji aktivitas antioksidan menggunakan metode DPPH (1,1 - difenil-2-pikrilhidrazil) dengan variasi konsentrasi 1000, 500, 250, 125, 62,5, 31,25, 15,625 dan 7,8125 μg/mL. Senyawa hasil isolasi diperoleh dalam bentuk minyak berwarna kuning. Berdasarkan data spektroskopi NMR 1D dan dengan membandingkan data senyawa yang telah dilaporkan dari jamur endofitik batang brotowali pada literatur disimpulkan bahwa senyawa hasil isolasiadalah bis-(2-etilheksil)ftalat dengan rumus molekul C24O4H38. Uji aktivitas antioksidan dari senyawa hasil isolasi memberikan nilai IC50 sebesar 232,9 μg/mL dan dikategorikan tidak aktif antioksidan.
