Abstract

The response of plants to environmental stress depends on the type of plant, the level, and the duration of stress. Drought stress affects growth, development, and the content of secondary metabolites produced by the plant. Kumis kucing (Orthosiphon aristatus (Blume) Miq.) is one example of a tolerant plant to drought stress. A study was conducted to compare the effect of several drought stress levels on plant growth and its sinensetin content. For this purpose, the experiment was carried out in a greenhouse with a one-factor complete randomized design, namely the level of water availability. The treatment tested consisted of: 100; 50; 37.5; 25; 12.5 percent of field capacity. Data were analyzed using analysis of variance and further tested with the Duncan Multiple Range Test level of 5% on significant results. The High-Performance Liquid Chromatography method was employed to test the sinensetin content (in percentage). The results showed that low water availability (50-12.5% field capacity) harmed plant growth and negatively affected leaf, stem, and root dry weight. The reduction in growth was highly significant in plants with water availability of 12.5% of field capacity. In low water availability, the plant that experience drought stress produced greater sinensetin content (0.0133%). Increased sinensetin in drought stress condition is a biochemical and physiological response of plants to unfavourable conditions.

Abstract

Sweet corn (Zea mays saccharata L.) is an important food commodity with high economic value. Sweet corn has a high business opportunity because of its fast production age and high selling value. The experiment was conducted to study the effect of the interaction between the composition of liquid organic fertilizer and plant spacing on the growth and yield of sweet corn. The experiment has designed in a factorial randomized block design (RAK) with two factors and three repetitions. The first factor is the composition of liquid organic fertilizer consists of 4 levels, namely: cow urine 42%, coffee skin 58% (P0); cow urine 35%, coffee skin 65% (P1); cow urine 72%. coffee skin 28% (P2); cow urine 21%, coffee skin 79% (P3). The second factor is the spacing, which consists of 3 levels, namely: 20x20 cm (J1); 30x30cm (J2); 40x40 cm (J3). Data analysis used variance and continued with the smallest significant difference test (BNT) at 5% level if the results were significantly different. The combination of liquid organic fertilizer composition and plant spacing affects the number of leaves and the length of the cob. Sweet corn with POC (72% cow urine, 28% coffee husk) and 30x30 cm spacing was able to produce 12,933 leaves. Combination of POC ingredients (21% cow urine and 79% coffee skin); 40x40 cm resulted in the length of the cob an increase of 1.5%. There was an interaction between the composition of liquid organic fertilizer and plant spacing on plant height, but the results were not significantly different. Sweet corn plants using 21% POC from cow urine and 79% coffee husk with a spacing of 40x40 cm produced cobs weight per plot of 240.27 grams and 208.22 grams, respectively. Keywords: sweet corn, spacing, material composition, liquid organic fertilizer

Abstract

Pesticides are one of the components of agri input needed to protect plants from plant pests. However, massive use of pesticides can cause various negative impacts on plants, the environment, and human health. Excessive use of pesticides in rice cultivation is still a problem that is often found in Sukorame Village, Trenggalek. Seeing this condition, there needs to be more intensive education efforts for farmers. Seeing this problem, community service activities were carried out by the Faculty of Agriculture team at Kadiri University. Community service activities were carried out with the aim of providing education to farmers regarding more environmentally friendly and sustainable pest management. This program is also expected to reduce dependence on chemical pesticides and introduce Integrated Pest Management (IPM) techniques that can help farmers increase productivity without damaging the environment. Community service activities were carried out using the Participatory Aural Appraisal (PRA) approach. This method emphasizes community participation and involvement starting from the preparation, education, training and mentoring stages. Overall, the main objectives of the program to increase farmer awareness of the dangers of pesticides and introduce the IPM method have been achieved. The community began to understand the importance of environmentally friendly pest management. After the program was implemented, farmers began to provide follow-up. It is seen that some farmers have started to adopt the IPM method as a sustainable agricultural system.

Abstract

Kepel (Stelechocarpus burahol), designated as the floral identity of the Special Region of Yogyakarta, Indonesia, faces challenges from plant pests that threaten its sustainability. Understanding the morphology and impact of pests such as planthoppers on kepel is crucial for developing effective pest management strategies. Despite its significance, there is a gap in research regarding the specific pests affecting kepel and their impact. This study aims to identify the morphological characteristics of planthopper pests and examine their infestation levels on kepel plants. Using a qualitative method with purposive sampling, samples were collected from kepel plants identified as infected by tree planthoppers. Morphological observations identified the pest species as Sanurus indecora from the Flatidae family. Sampling from two infected plants resulted in 81 nymphs and 16 adult S. indecora. Infestation analysis showed that kepel plant XII.G.D had a 7.4% infestation rate, while XIV.G.II had a 23% infestation rate. These findings highlight the presence and impact of S. indecora on kepel plants, providing critical information for the bioecology of this pest. This study not only fills a research gap by documenting the pests affecting kepel but also contributes to the broader knowledge required for developing targeted pest management strategies, ensuring the conservation and health of kepel plants in Yogyakarta, Indonesia.

Abstract

Ananas comosus L. belongs to the Bromeliaceae family and has around eight subfamilies. Accessions of local pineapple have spread widely in several areas, one of which is in Kediri. However, some accessions and closely related species (Bromeliaceae) likely remain poorly understood relationships. The highly diverse morphological characters and low levels of sequence divergence in Bromeliaceae have been problematic in resolving the reconstruction of phylogenetic relationships. This study aimed to evaluate the efficiency of the matK gene as a DNA barcode to determine genetic diversity and reconstruct a phylogenetic tree in local pineapple and closely related species (Bromeliaceae). A total of 15 specimens were used in this research, of which 7 were local pineapple accessions from Kediri and eight were closely related species from the Bromeliaceae family. Whole genome DNA was isolated using a Tiangen kit and amplified using the polymerase chain reaction (PCR) technique using a specific primer. The results showed that matK was easily amplified with a DNA fragment length of 750-800 bp. The average composition value of each base was 36.9% T (U) bases, 30.1% in A bases, 15.9% in G bases, and 17.1% in C. BLAST analysis of the sequences showed that all samples were confirmed and identified according to the species names spread across local pineapple production areas (with query cover of 96-98%). However, the species names registered in the PBG database were confirmed to be different species but still in the same genus, i.e., samples B9-B14. The phylogenetic relationship analysis distinguished each accession into two large clades according to sub-families, i.e., Bromelioideae and Pitcairnioideae. This research shows that the matK gene is suitable and recommended as a DNA barcode for local pineapple and closely related species.

Abstract

Fruit maturing enhances maturity and quality, making fruits more attractive. This process involves physiological and biochemical changes, including increased respiration, ethylene production, texture alteration, and heat improvement. This study examines the effects of different ethylene sources: calcium carbide (CaC2), 500 mg vitamin C, and potassium permanganate (PK), on the maturing of Burahol fruit (Stelechocarpus burahol). A Completely Randomized Factiorial Design (CRFD) with five replications was employed. Treatments included E1 (control, no ethylene), E2 (0.5% CaC2 of fruit weight wrapped in cloth), E3 (500 mg vitamin C/kg fruit), and E4 (PK crystals in a sealed chamber). Data collected on day three evaluated skin color (RHS charts), texture, and mucus presence. Results showed CaC2 and PK induced optimal maturing, yielding uniformly brownish-yellow fruits (20A/Brilliant Yellow), firm texture, and no mucus. Vitamin C fails to prevent decay due to mechanical damage from forced harvesting. This study highlights CaC2 and PK as effective maturing agents, providing practical solutions for Burahol fruit post-harvest management.
