Abstract

Black cumin (Nigella sativa) is a herbal plant that has been cultivated locally in Indonesia and is traditionally used for various diseases. Thymoquinone, one of the main components, is rich in biological activity. In several countries, topical application of its oil on human skin with vitiligo can stimulate skin repigmentation with minimal side effects. This study aims to determine the viability of B16F10 melanoma mouse cells against ethanol and n-hexane extracts of black cumin seeds through the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The ethanol extract (EE) yield was 14.306%, and the n-hexane extract (NHE) was 7.442%. Phytochemical screening of EE detected flavonoids, alkaloids, saponins, and steroids, and High-performance liquid chromatography (HPLC) detected 0.040% thymoquinone. The MTT test showed cell viability was >100% from EE at all treatment concentrations, namely 0.75-100 ppm and only 0.75-6.25 ppm for NHE. In conclusion, this study indicates that 96% EE of Nigella sativa is less toxic than NHE on B16F10 mouse melanoma cells and has potential as an alternative treatment for vitiligo, which needs to be proven in further research.

Abstract

This research investigates the impact of orange juice consumption on the histological structure of the duodenum, a critical site for nutrient absorption and digestion. Focusing on pediatric animals, orange juice (with a pH of 3) was administered to post-weaning rats aged 4 weeks to 6 weeks at dosages ranging from 0g/kg BW to 20g/kg BW over two weeks. Duodenal histopathology was assessed, focusing on parameters such as the quantity of crypts of Lieberkuhn, as well as the length, height, width, and depth of villi (measured in μm). Anatomical pathology examination included measurements of the length and width of the duodenum (also in μm), as well as the color mean (OD). The research findings indicate no significant changes in histo-pathology or anatomy pathology of the duodenum. This suggests that young individuals can adapt to acidic challenges without compromising gastrointestinal health. These findings offer reassurance regarding the short-term consumption of acidic orange juice (with a pH of 3) with minimal risks to intestinal integrity, both during growth and development.

Abstract

This research aims to determine the role of the TPPK (Violence Prevention and Control Team) SRA (Child-Friendly School) in the implementation of early childhood education at the PAUD unit level in Tandes District, Surabaya City. This research is descriptive qualitative. The data obtained in this work was carried out with the help of a literature review and field research, using several data collection methods, namely. observation, interviews, and documentation. Data analysis was carried out descriptively to determine the role of TPPK in the child-friendly school model at the PAUD level in Surabaya City, Tandes District. The results of the data analysis show that the role of TPPK in the child-friendly school model for early childhood in Tandes District, Surabaya City is quite good. In practice, the role of TPPK residents in educational activities according to the principles of Child-Friendly Schools is almost close to the existing theory. This is proven by the results of observations and documents carried out at all levels of PAUD units in Tandes District, Surabaya. The researcher's observations and documentation focused on the completeness of TPPK data, action programs and facilities and infrastructure used in early childhood education units, implementation of teaching methods, attitudes towards students and environmental health in early childhood education units in Tandes sub-district, Surabaya city which are organized in this way in such a way that the environment generally supports children's physical, spiritual and motoric activities. The application of learning methods designed and prepared by TPPK alone, especially teachers as learning leaders, cannot be implemented optimally for several reasons. Apart from that, teachers are quite varied in using teaching methods that are adapted to the material and supported by a game environment in delivering educational material, as well as active communication between teachers and students.

Abstract

Primary objective of this study was to ascertain the presence of Coxiella burnetii (C. burnetii) as the causative of Q fever in cattle within Bandung Regency. Tissue samples were procured from 240 slaughtered cattles in abattoirs across Bandung Regency. Each breed of cattle consists of Brahman cross (BX), Simmental, Limousin, Friesien Holstein crossbreed, and Friesien Holstein (FH) with total of 192, 3, 13, 29, and 3, respectively. Detection method employed was nested-Polymerase Chain Reaction (PCR) to identify the presence of C. burnetii Deoxyribonucleic Acid (DNA) within tissue samples. Positive samples underwent further examination employing hematoxylin-eosin (HE) staining to identify lesions attributed to C. burnetii. Five BX cattle were identified as positive for C. burnetii in lungs and heart. However, histopathological analysis of both organs did not reveal any notable changes. These findings suggest the occurrence of C. burnetii infection in cattle within Bandung Regency, without specific histopathological changes indicative of Q fever lesions. In conclusion, presence of asymptomatic C. burnetii infections should be regarded as a significant threat to human health.
