Abstract

The development of novel chemical entities, novel pharmaceuticals, and novel drug leads frequently involves the use of natural sources. One of the most well-known plants, Pandanus odoratissimus (Pandanaceae), is native to Malaysia and has long been used in Ayurvedic medicine to treat a variety of illnesses. The purpose of this study is to determine P odoratissimus's antioxidant and anticancer properties against MCF-7 human breast cancer cell lines. The 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity was used to assess antioxidant activity, and among the other extracts, core exhibited the highest antioxidant value. An MTT assay was used to assess the effects of plant components on MCF-7 cell lines (3-(4, 5-dimethylthiazolyl)-2, 5-diphenyl-tetrazolium bromide). The concentration of samples was shown to considerably boost the antioxidant activities in P. odoratissimus extracts. Our findings demonstrated that, after receiving treatment for 72 hours, sample treated cancer cells experienced a decline in cell viability. The IC50 value of 88.00 mg/ml indicates that the extracts have a poor cytotoxic impact and have no promise as anticancer agents against MCF-7, which is why it’s killing efficiency in cancer cells is regarded as lower.

Abstract

Pemerintah Indonesia perlu mendorong penggunaan teknologi environmental DNA (eDNA) sebagai metode utama dalam pemantauan biodiversitas di Kawasan Konservasi Laut (KKL) guna mengatasi tantangan logistik di wilayah yang luas dan beragam. Diperlukan investasi dalam pengembangan database eDNA nasional yang mencakup sekuens referensi spesies laut Indonesia dan pelatihan serta sertifikasi bagi para pemantau eDNA untuk memastikan kualitas data yang konsisten. Monitoring dengan eDNA dapat dilakukan secara rutin dan berkelanjutan di seluruh KKL untuk mengumpulkan data yang akurat dan mendukung pengamatan tren jangka panjang dalam keanekaragaman hayati laut. Selain itu, dukungan untuk penelitian lanjutan dalam identifikasi spesies baru dan pemahaman yang lebih dalam tentang ekologi laut Indonesia melalui teknologi eDNA juga perlu diberikan. Dengan mengimplementasikan rekomendasi ini, Indonesia dapat memperkuat upaya pelestarian keanekaragaman hayati lautnya secara efektif dan efisien, menjaga keberlanjutan ekosistem laut yang kaya dan unik di negara ini.

Abstract

Over the past five decades, there has been a growing trend in the capture of kawakawa (Euthynnus affinis), which, alongside its potential to support small-scale commercial fisheries, highlights its significance.However, there is inadequate data on this species for suitable management and conservation status. This study aimed to investigate the genetic diversity, population structure, and connectivity of the kawakawa in the three major fishing port in Sumatra, namely Belawan , Padang, and Lampung by using DNA mitochondria control region (d-loop) Sequence. In total, 78 sequences of kawakawa with an average size of 303 bp, we found 36 polymorphic sites and 56 haplotypes from all population were identified with DNA compatibility values of 97-100%. We found the genetic diversity values in Sumatera Island were high (h = 0.950; π = 0.034), with the highest genetic diversities value in Padang (h = 0.990; π = 0.082) and the lowest in Belawan (h = 0.929; π = 0.082). AMOVA and Fst analyses revealed no differentiation in each population (Fst = 0.005). The haplotype distribution and connectivity analyses showed genetic mixing among the three populations. This study showed a single stock at the study sites and suggests management measures at a regional level to maintain the population.

Abstract

Walking shark is one of elasmobranch that has been protected by the government of the Republic of Indonesia and can be found in the eastern seawaters of Indonesia. There are six species of walking shark, one of it is Triton Bay Epaulette Shark (Hemiscyllium henryi) which is endemic to the coastal waters of Kaimana. There are still lack of information regarding biology abundance and population of H. henryi, therefore it is necessary to do a study related to this species. This study aims to determine individual information and abundance of H. Henryi in Kaimana coastal waters during the observation in 2022 and 2023 based on density value and potential area. The method used is a census based on the Global Positioning System, through snorkeling and walking along the coastline, as well as observations of total length, wet weight, and sex in H. henryi which encountered during census. The total census track is 5.783 meters with a sweep of 2,46 Ha monitoring area. The density or abundance of H. henryi in 2022 is 1,2 ind/Ha and in 2023 is 2,4 ind/Ha. Total length of H. henryi ranged between 55–88 cm (Mean 70,6 cm ± SD 12,29). The length and weight relationship obtained W=0,0004L3,4343 with R2 value of 0,9526. The abundance trend of H. henryi at the coastal waters of Kaimana in the last two years has multipled by 2 times based on density value determining after being designated as a fully protected fish species.

Abstract

Kegiatan pengenalan dan pelatihan pembuatan eco enzyme pada peternak ayam dan ikan di Kabupaten Tapanuli Selatan, Sumatera Utara, bertujuan untuk meningkatkan pemahaman dan kemampuan para peternak dalam memanfaatkan limbah organik. Eco enzyme, yang dihasilkan dari fermentasi limbah organik seperti sisa buah dan sayuran, digunakan sebagai pupuk organik yang ramah lingkungan, desinfektan rumah tangga, maupun solusi sanitasi untuk peternakan. Program ini melibatkan penyuluhan dan pelatihan langsung kepada peternak ayam dan petani ikan, yang terbukti meningkatkan pengetahuan peserta tentang pembuatan dan aplikasi eco enzyme berdasarkan hasil pengetahuan pre test dan post test. Hasil dari kegiatan ini diharapkan dapat mendukung praktik peternakan yang berkelanjutan dan meningkatkan kesejahteraan ekonomi para peternak.

Abstract

The Halmahera Walking Shark (Hemiscyllium halmahera) is an endemic fish in the Halmahera Sea. This species is distributed and found in the Kao Bay sea. The International Union for Conservation of Nature (IUCN) data shows that the Halmahera Walking Shark is in the Near Threatened status. Protection is carried out to protect the population, therefore scientific data and information are needed. This study focuses on the biological structure of the Halmahera Walking Shark population in Kao Bay sea. Data collection was carried out exploratively in coral reef, seagrass, mangrove ecosystems and by-catch. Samples were taken for morphometric data, weight and sex. Morphometric data were analyzed to obtain cohorts, separation index, class intervals, length-weight relationships and condition factors of walking shark. The result were found to be 33 individuals (13 male and 20 female). Morphological characteristics based on morphometric variables found no significant differences. Analysis of the cohort of the Halmahera Walking Shark (H.halmahera) found 3 groups namely the juvenile, mature and old age phases. The separation index shows that individuals come from different populations based on length. Growth pattern of the Halmahera Walking Shark (H.halmahera) is negative allometric. The histogram of the frequency distribution between classes found small to large size structures with different relative frequency percentages. The condition factor found that the body proportions of the Halmahera Walking Shark (H.halmahera) were in the plump category. Abstract

Sponges (Phylum Porifera) are a group of highly diverse, sessile, and filter-feeding basal metazoans, except spongillids. The majority of sponges are marine species that play an important role in benthic habitat by maintaining the stability of marine ecosystems through nutrient cycling, habitat provision, and bioerosion. However, marine sponges are not easily identifiable because of their lack of species-level distinctive morphological features, which limits efforts to monitor actual species biodiversity. Indonesia is home to approximately 850 identified species of marine sponges, and Raja Ampat archipelago of West Papua province is known for its exceptional marine biodiversity. Despite the species abundance, the exact number of sponges in the region is not well-documented due to the lack of specific studies providing comprehensive data on their diversity. Therefore, this study aimed to conduct a DNA barcoding analysis using the mitochondrial cytochrome oxidase I (COI) gene as a marker, combined with morphological analyses of 22 individuals collected in the waters of Waigeo, Mansuar, and Gam Islands in Raja Ampat. The results showed that 3 samples were identified at the species level (Halichondria sp. and Stylissa carteri) with good query cover and percent identity. This showed the possible presence of undescribed or cryptic species, suggesting a severe lack of reference data for both morphology and molecular analyses of marine sponges in the region. Consequently, the analysis showed the presence of a significant gap in the understanding of sponge biodiversity in Raja Ampat's waters.

Abstract

The marine sponge Aaptos suberitoides has been shown to have anticancer properties, with clear evidence of its capacity to suppress the growth of cancer cells. However, the pharmaceutical exploration of chemicals from marine organisms causes significant environmental concerns. Aspergillus unguis, a fungal symbiont of the marine sponge A. suberitoides, has been isolated for its potential in sustainable natural products resources. This study assesses the antioxidant activity of ethanol extract and the toxicity of four levels fractions from ethanol extracts of A. unguis mycelium and these results were reported for the first time in this study. The antioxidant of ethanol extract was determined using the 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method. The fractions were examined for toxicity using the Brine Shrimp Lethality Test (BSLT). The ethanol extract of A. unguis shows very strong antioxidant bioactivity (IC50 = 42.84 mg/L). The LC50 values for hexane, chloroform, ethyl acetate, and butanol fractions were determined to be 74.11 µg/mL, 93.84 µg/mL, 59.37 µg/mL, and 142.79 µg/mL, respectively. It indicates significant toxicity. These preliminary results are important knowledge for further research into the bioactivity potential of the metabolites as candidate anticancer compounds, aligning with marine pharmaceutical drug development.

Abstract

The South Halmahera Islands feature a comprehensive marine coastal ecology. The ecosystems found were seagrass, mangrove and coral reef ecosystems. The components that sustain the existence of this ecosystem are employed equitably because of its geographical location on the equator. Mangrove ecosystems that provide benefits directly and indirectly to the community. The purpose of this study was to determine the condition of the mangrove ecosystem based on the hemispherical photography method and the type of zonation. Tawabi Island, Kayoa, South Halmahera Regency served as the site of the equator research. The research station is determined as many as 3 stations. Mangrove percent cover canopy using the hemispherical photography and transect quadrant for density. Field data gathering procedures with transects drawn perpendicular from the sea to the land. Zonation characteristics and thickness using zone tracking. Density mangrove using ecology analysis and zonation using canvas 9.0. The results of the research founded were Rhizophora apiculata, Rhizophora mucronata and Sonneratia alba. Mangrove percent cover canopy were good category (75-81%), where station I (75%), station II (81%) and station III (77%). Station I having 600 trees per ha, station II having 500 trees per ha, and station III having 400 trees per ha (low density category). Mangroves were found in 3 zones Sonneratia alba, Rhizophora apiculata and Rhizophora mucronata.
