Abstract

The Halmahera Walking Shark (Hemiscyllium halmahera) is an endemic marine biota in the Halmahera Sea. This species has a local migration pattern and is territorial. The distribution of this species is in the intertidal area to a depth of 10-15 meters. Previous studies have shown that this species is spread across several islands in the North Maluku Islands. Maitara Island is a small island located between the North Maluku archipelago. The Halmahera Walking Shark species is also found on this island. Information on the Halmahera Walking Shark species is not fully documented. The purpose of the study is related to biological information and emergence points (emergence points) in the Maitara Island Sea. Data collection was carried out on coral reefs, seagrass and mangroves. Samples were captured and morphometric measurements, weight and sex observations were taken. The sea area where the species was found was used to obtain coordinates for the emergence point. An analysis of length-weight relationship data was carried out to see growth patterns. The study found that morphometric measurements varied among individuals, although there were general similarities. Sexes were found in pairs (Male and Female), indicating that proportions and mating were still normal in nature. The habitat of this species is found in the mangrove area, coral reefs, and seagrass, thus describing that there is an association in nature. The emergence point shows that the Halmahera Walking Shark is found along the coast of Maitara Island although the frequency of the number is different. The length-weight relationship found a negative allometric growth pattern. Information on biological aspects and emergence points is important, to be used as a reference for migration areas and habitats for the interests of species, waters, and ecosystem conservation.  Hiu Berjalan Halmahera (Hemiscyllium halmahera) merupakan biota laut endemik di laut Halmahera. Spesies ini memiliki pola migrasi lokal dan bersifat teritorial. Penyebaran spesies ini di wilayah intertidal hingga kedalaman 10-15 meter. Penelitian terdahulu menunjukan bahwa spesies ini tersebar di beberapa pulau di Kepulauan Maluku Utara. Pulau Maitara merupakan pulau kecil yang terletak diantara gugusan kepulauan Maluku Utara. Pulau ini juga ditemukan speises Hiu Berjalan Halmahera. Informasi belum terekam lengkap terkait spesies Hiu Berjalan Halmahera. Tujuan penelitian terkait informasi biologi dan emegence point (titik kemunculan) di Laut Pulau Maitara. Pengambilan data dilakukan di terumbu karang, lamun dan mangrove. Sampel ditangkap dan diambil ukuran morfometrik, berat dan pengamatan kelamin. Area laut ditemukannya spesies diambil koordinat untuk di jadikan sebagai titik kemunculan.  Analisis data hubungan panjang-berat dilakukan untuk melihat pola pertumbuhan. Hasil penelitian ditemukan ukuran morfometrik bervariasi pada setiap individu, meskipun terdapat kemiripan secara umum. Kelamin ditemukan sepasang (Jantan dan Betina), dengan demikian menunjukan proporsi dan perkawinan masih normal di alam. Habitat tertangkap spesies ini di area mangrove, terumbu karang dan lamun, sehingga mendeskripsikan terdapat asosiasi di alam. Titik kemunculan menunjukan bahwa, Hiu Berjalan Halmahera ditemukan di sepanjang pesisir laut Pulau Maitara meskipun frekuensi jumlah berbeda. Hubungan panjang berat menemukan pola pertumbuhan allometrik negatif. Informasi aspek biologi dan emergence point penting, untuk dapat dijadikan rujukan wilayah migrasi dan habitat untuk kepentingan konservasi spesies, perairan dan ekosistem.

Abstract

ABSTRAKPenelitian ini bertujuan menganalisis perkembangan, fokus tematik, dan arah riset terkait penggunaan environmental DNA (eDNA) metabarcoding untuk mendeteksi dan mengidentifikasi spons laut (Porifera). Pendekatan yang digunakan adalah systematic literature review (SLR) berbasis PRISMA dan analisis bibliometrik terhadap artikel terindeks Scopus tahun 2009–2025. Dari 116 publikasi awal, 42 artikel memenuhi kriteria inklusi dan dianalisis lebih lanjut menggunakan VOSviewer untuk mengidentifikasi tren publikasi, pola kolaborasi penulis-negara, serta struktur klaster kata kunci. Hasil menunjukkan peningkatan signifikan publikasi sejak 2017, dengan kontribusi terbesar berasal dari Britania Raya, Spanyol, dan Amerika Serikat. Analisis kata kunci menghasilkan lima klaster utama yang mencerminkan arah riset global, yaitu (1) genetika dan biodiversitas, (2) mikrobioma dan metagenomik, (3) ekologi terumbu karang, (4) invertebrata laut, serta (5) lingkungan ekstrem. Meskipun eDNA metabarcoding terbukti sensitif untuk mendeteksi komunitas spons, tantangan utama yang ditemukan mencakup keterbatasan marker gen yang belum universal serta kelengkapan basis data taksonomi yang masih rendah. Novelty penelitian ini terletak pada penggabungan SLR dan bibliometrik secara khusus pada spons laut, sehingga mampu mengungkap kesenjangan geografis, keterbatasan metodologis, serta menawarkan rekomendasi strategis berupa standardisasi protokol, penguatan basis data referensi lokal, dan perluasan kolaborasi internasional, terutama di wilayah megabiodiversitas seperti Indonesia.Kata Kunci: eDNA, metabarcoding, Porifera, bibliometric, keanekaragaman hayati.ABSTRACTThis study analyzes the development, thematic focus, and research direction of environmental DNA (eDNA) metabarcoding applied to the detection and identification of marine sponges (Porifera). A systematic literature review (SLR) following PRISMA and a bibliometric analysis of Scopus-indexed publications from 2009–2025 were conducted. From an initial 116 records, 42 articles met the inclusion criteria and were analyzed using VOSviewer to identify publication trends, author–country collaborations, and keyword cluster structures. Results show a substantial increase in publications since 2017, with major contributions from the United Kingdom, Spain, and the United States. Keyword analysis revealed five major thematic clusters: (1) genetics and biodiversity, (2) microbiome and metagenomics, (3) coral reef ecology, (4) marine invertebrates, and (5) extreme environments. Although eDNA metabarcoding demonstrates high sensitivity for sponge detection, key challenges include non-universal genetic markers and incomplete taxonomic reference databases. The novelty of this study lies in integrating SLR and bibliometric approaches specifically for marine sponges, allowing the identification of geographic gaps, methodological limitations, and providing strategic recommendations for protocol standardization, the development of local reference databases, and enhanced international collaboration, particularly in megabiodiverse regions such as Indonesia.Keywords: eDNA, metabarcoding, Porifera, bibliometric analysis, biodiversity.

Abstract

Terumbu karang merupakan ekosistem penting yang mendukung keanekaragaman hayati laut, dengan ikan karang memainkan peran kunci dalam menjaga keseimbangan ekosistem tersebut. Penelitian ini bertujuan untuk menganalisis hubungan antara kelimpahan ikan karang dan struktur komunitas terumbu karang secara spasial dan temporal di Kawasan Konservasi Pulau Pieh, Sumatera Barat. Penelitian menggunakan data primer yang didapatkan dari 16 stasiun pengamatan yang tersebar di berbagai zonasi konservasi (zona inti, pemanfaatan, dan rehabilitasi) pada kedalaman 5–10 meter, menggunakan metode Underwater Visual Census (UVC) untuk ikan karang dan Underwater Photo Transect (UPT) untuk terumbu karang. Data sekunder didapatkan dari hasil monitoring pengelola kawasan konservasi selama tahun 2021-2024 pada periode yang sama setiap tahun. Hasil menunjukkan bahwa keanekaragaman ikan karang berada pada kategori sedang (1 ≤ H’ ≤ 3), keseragaman komunitas sedang hingga tinggi (E > 0,5), dan dominansi rendah (C < 0,5), yang mengindikasikan kondisi ekosistem yang sehat. Hasil Component Analysis (CA) kelimpahan ikan karang dan tutupan karang menunjukan adanya keterkaitan antara stasiun pengamatan, tutupan karang dan kelimpahan ikan. Penurunan tutupan karang dan kelimpahan ikan pada tahun 2024 menandakan adanya pengaruh dari tekanan ekologis maupun aktivitas antropogenik sehingga pengelolaan yang optimal dan pengetahuan masyarakat perlu ditingkatkan. Hasil penelitian ini dapat menjadi dasar ilmiah dalam pengelolaan kawasan konservasi secara efektif dan berkelanjutan.Coral reefs are vital ecosystems that support marine biodiversity, with reef fishes playing a key role in maintaining the balance of these ecosystems. This study aims to analyze the spatial and temporal relationship between reef fish abundance and coral reef community structure in the Pieh Island Marine Conservation Area, West Sumatra. The research used primary data conducted at 16 observation stations across different conservation zones (core, utilization, and rehabilitation zones) at depths of 5–10 meters, using the Underwater Visual Census (UVC) method for reef fish and the Underwater Photo Transect (UPT) method for coral reefs. Secondary data was obtained from monitoring conducted by conservation area managers during the same period each year from 2021 to 2024. The results showed that reef fish diversity was in the moderate category (1 ≤ H’ ≤ 3), community evenness was stable in moderate to high (E > 0.5), and dominance was low (C < 0.5), indicating a healthy ecosystem condition. The Component Analysis (CA) of reef fish abundance and coral cover revealed a correlation between observation stations, coral cover, and fish abundance. A decline in coral cover and fish abundance in 2024 indicates the influence of ecological stress and anthropogenic activities, highlighting the need for improved management and increased community awareness. The results of this study can serve as a scientific basis for effective and sustainable marine conservation area management.

Abstract

Ecological conditions are shifting due to increased anthropogenic activities along coastal areas. These activities are affecting the life cycle of aquatic organisms, especially phytoplankton. This study, conducted in the unique ecosystem of Hurun Bay Lampung, aims to analyze the composition and structure of phytoplankton communities in eutrophic waters and water quality conditions. Sampling was conducted in two different months, August and October, by vertical hauling from a depth of 5 meters to the surface. The results revealed a rich diversity of phytoplankton, with 72 genera, 48 families, and 6 classes identified in August, and 55 genera, 42 families, and 6 classes identified in October. The diatom group was the most prevalent, accounting for over 90% of the total, with the highest abundance found in Skeletonema and Chaetoceros. The diversity, uniformity, and dominance indices were all in the low to medium category, indicating a balanced ecosystem.

Abstract

Turbidity is a parameter of the marine environment that greatly affects the condition of seagrass whose habitat is an intertidal zone in shallow sea waters. Seagrass is an important type of ecosystem that can be found in several coastal areas of Ternate Island. This study aims to analyze the turbidity conditions of seagrass habitat waters and apply a remote sensing algorithm using Sentinel 2B images. The turbidity research method was carried out by field measurements. The turbidity algorithm model used refers to references with mathematical equations (Rrs665-0.014)/0.013, and the development of a new algorithm as a comparison algorithm. Both algorithms were validated with field data to determine the level of accuracy using the Normalized Mean Absolute Error (NMAE) and determination coefficient (R2). The results were obtained from turbidity data with values ranging from 0.3 NTU to 1.5 NTU with an average value of 0.87 0.45 NTU. The Sentinel 2B image in this study was restored with geometric corrections, atmosphere, radiometric digital values, land masking, and sun glint. The turbidity algorithm model used obtained good accuracy in mapping and monitoring the turbidity of seagrass habitat waters on Ternate Island. The application of the turbidity algorithm used as a reference in this study has an NMAE value of 50.44 and R2 of 0.8822, while the newly discovered turbidity algorithm has an NMAE value of 29.38 and R2 of 0.8827.Keywords:remote sensingecosystemscoastalsedimentationNorth Maluku

Abstract

The Halmahera Walking Shark (Hemiscyllium halmahera) is an endemic species in the Halmahera Sea. Information related to the environment in general has not been described in a complex manner. The environment is the main factor to support the life of biota. Environmental characteristics are a determining factor in the sustainability of biota. The study aims to provide information related to the physical environment of the habitat of the H. halmahera species. Primary data collection was carried out directly (in situ). Current data was taken using the Lagrange method using a currents drogue. Tides using tidal palms and observed for 24 hours. Secondary data was taken from Marine Copernicus and the Geospatial Information Agency (BIG). All data were tabulated and analyzed by Ocean Data View (ODV) software. The results of the study found that the surface current velocity was found to be in the fast current category. Current speed descriptively provides information on the life of H. halmahera. Primary data shows that the direction of the surface current moves to the east and north. Secondary data in vertical distribution shows weak zonal and meridional current shifts. The tidal pattern was found to be two high and low tides (semidiurnal). The distribution of surface temperature was found to be around 30-32oC. The vertical temperature profile found three layers, namely 1-13m (30oC), 65m (25oC) and 71m (20oC). The chemical environment showed all normal variables. The physical and chemical environmental conditions support H. halmahera to inhabit the sea area of Morotai Island. The environmental description has provided a brief understanding of how the ecological role of the H. halmahera species. Management and sustainability of the environment in the future due to climate change can be done with information on habitat characteristics.Keywords:coral triangleHalmaheranicheNorth Malukutropical

Abstract

The South Halmahera Islands feature a comprehensive marine coastal ecology. The ecosystems found were seagrass, mangrove and coral reef ecosystems. The components that sustain the existence of this ecosystem are employed equitably because of its geographical location on the equator. Mangrove ecosystems that provide benefits directly and indirectly to the community. The purpose of this study was to determine the condition of the mangrove ecosystem based on the hemispherical photography method and the type of zonation. Tawabi Island, Kayoa, South Halmahera Regency served as the site of the equator research. The research station is determined as many as 3 stations. Mangrove percent cover canopy using the hemispherical photography and transect quadrant for density. Field data gathering procedures with transects drawn perpendicular from the sea to the land. Zonation characteristics and thickness using zone tracking. Density mangrove using ecology analysis and zonation using canvas 9.0. The results of the research founded were Rhizophora apiculata, Rhizophora mucronata and Sonneratia alba. Mangrove percent cover canopy were good category (75-81%), where station I (75%), station II (81%) and station III (77%). Station I having 600 trees per ha, station II having 500 trees per ha, and station III having 400 trees per ha (low density category). Mangroves were found in 3 zones Sonneratia alba, Rhizophora apiculata and Rhizophora mucronata. Abstract

Mangrove, Rhizophora mucronata, grows in the intertidal area, which contains much organic matter and varying salinity. The organic matter content is influenced by the bacterial community that inhabits the ecosystem, but information regarding the bacterial community, especially in the mangrove root system, is not widely available. There are several challenges in completing this information, one of which is that the method used is still in a conventional form. Developments in environmental DNA analysis can support and complement this information. However, this method must be optimized because the organic matter content and salt variations affect the extraction results. Thus, this study aimed to determine the optimal approach for extracting bacterial DNA from mangrove sediments. The analysis used two methodologies: manual DNA extraction techniques based on buffer modification and DNA extraction kits. There were four different treatments, namely the soil DNA isolation plus kit (M1), the fecal / soil microbial quick-DNA miniprep kit (M2), glass powder with charcoal (M3), and glass powder with skimmed milk (M4). DNA samples were obtained from each method and assessed for concentration and purity using a nanodrop. In addition, the resulting DNA's quality was analyzed using 1.5% agarose. The results obtained were in the M2 treatment, which showed optimal results compared to the others. M2 uses a bead-based beating and spin column method to achieve optimal DNA concentration through high molecular weight. DNA barcoding was capable of resolving field identification challenges and providing taxonomic clarification based on DNA sequence data.   Abstract Equatorial small islands host dynamic coastal ecosystems, yet seagrass biodiversity and molecular identification remain poorly documented. Complete information on coastal ecosystems has not been provided for seagrass in the equatorial latitudes. Seagrass is dynamically distributed throughout the equatorial latitudes. The area that the equatorial latitudes also traverse is known as the indotropic region. Eastern Indonesia’s Kayoa Island is situated directly on the equator. However, seagrass biodiversity and DNA-based species confirmation in this region remain insufficiently documented. There are no published research on seagrass, particularly on biodiversity. The aim of this study is to reveal the diversity of seagrass in equatorial latitudes on the small islands. The methodology in this study was purposive sampling with a quadrant line transect was used to obtain the samples. In situ identification of seagrass species was done. Seagrass samples were taken from the leaves for DNA analysis purposes. Leaf samples were analyzed using molecular methods in the laboratory. To determine the distribution of seagrass ecology and environmental were subjected to PCA analysis. The results showed that six seagrass species were identified. It has been discovered that seagrass may develop in the tropical region under typical environmental conditions. Despite the fact that six species were discovered, diversity was limited. In this region, seagrass cover is typically high and supports a healthy species density. Species diversity was low, whereas percent cover and density were locally high. Overall, the study indicated relatively low diversity and density values. DNA barcoding has revealed differences in morphological identification.This discovery represents preliminary data that can be investigated further in subsequent studies.

Noctiluca scintillans is a mixotrophic dinoflagellate commonly associated with eutrophic coastal systems and bloom dynamics. This study aims to characterize the morphometric variability of N. scintillans in Jakarta Bay, a tropical coastal environment strongly influenced by anthropogenic nutrient enrichment, and to evaluate how cellular morphology reflects functional and ecological responses under eutrophic conditions. Plankton samples were collected in February 2024 and analyzed using high-resolution microscopy to quantify key morphometric traits, including cell diameter, biovolume, aspect ratio, surface area-to-volume (SA:V) ratio, and nucleus-to-cytoplasm (N:C) ratio. Differences between red (RNS) and green (GNS) morphotypes were assessed using non-parametric statistical analyses, and preserved measurements were corrected to estimate live cell size. A total of 121 cells were analyzed, with RNS overwhelmingly dominating the population. RNS exhibited significantly larger cell diameter and volume than GNS, while SA:V ratios differed significantly between morphotypes, indicating functional differences in size-related morphology. In contrast, aspect ratio did not differ significantly, suggesting overall conservation of cell shape across morphotypes. Estimated live cell sizes indicate that N. scintillans populations in Jakarta Bay are dominated by relatively small cells compared to those reported from temperate regions. Variability in the N:C ratio further suggests heterogeneity in cellular physiological states within the population. Overall, these findings establish a morphometric baseline for N. scintillans in Jakarta Bay and highlight the value of morphometric traits as integrative indicators of physiological variability and environmental forcing in tropical eutrophic coastal systems.
