Abstract

AIglaia elliptica Blume is one of plant species from the genus of Aglaia, belonging to the family of Meliaceae. This group of plants is known to have powerful cytotoxic effect. This research was conducted to isolate and characterize major compound(s) from ethanol extract of Aglaia elliptical leaf that can be used as marker compound(s) for extract standardization. Approximately 1000 g of Aglaia elliptica Blume leaf dry powder was extracted with ethanol using maceration method. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Six sub-fractions were obtained from the Vacuum Liquid Chromatography (VLC) process of ethyl acetate fraction. Based on the HPLC profile and separation process using column chromatography and preparative HPLC, a major compound (with the weight of 36.80 mg) was successfully isolated from sub-fraction H. Analyses on its UV absorbance pattern, mass spectra, and proton NMR identified the isolated compound as a compound with molecular weight of 300, C18H24N2O2, known as odorin. Cytotoxicity test showed that odorin has inhibition effect against MCP-7 cell line with IC5O of 153.32 ppm.

Abstract

Sambiloto (Andrographis paniculata Nees) is one of a medicinal plants containing andrographolide as its primary bioactive component, which is able to inhibit breast cancer cell proliferation. Andrographolide is a diterpene lacton that is sparingly soluble in water. The aim of this study is to improve the solubility of andrographolide in A. paniculata herbs ethyl acetate fraction by microencapsulation method prepared using spray drying. The ethyl acetate fraction of A.paniculata herbs was microencapsulated using PVP K30 and HPMC as the coating polymer in different ratios of ethyl acetat fraction and polymer (1:5; 1:7,5 and 1:10). Microspheres containing ethyl acetate fraction of A.paniculata herbs were evaluated their saturation solubility and in vitro dissolution in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium. The result showed that the microencapsulation could increase the saturation solubility and dissolution rate of andrographolide as compared to the ethyl acetate fraction and andrographolide standard. Compared to the solubility of andrographolide from A. paniculata ethyl acetate fraction, the solubility of andrographolide on ethyl acetate fraction microspheres in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium were increased approximately 23.42-39.34, 19.92-34.83 and 24.56-38.25 folds respectively. Moreover, compared to the solubility of andrographolide standard, the solubility of andrographolide on ethyl acetate fraction microspheres in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium were increased approximately 16.99-28.53, 19.76-34.54 and 17.26-26.87 folds respectively.

Abstract

Good Pharmaceutical Distribution Practice is called as CDOB in Indonesia is a guidelines to ensure pharmaceutical product quality throughout the distribution chain..This guideline consist of nine aspects, and pharmaceutical wholesalers (PBF) are required to implement the principle in the guideline as part of compliance to the pharmaceutical wholesaler regulation. The purpose of this research is to assess the extent of the implementation of Good Pharmaceutical Distribution Practices in the pharmaceutical daily operation.This research is descriptive with random sampling technique to determine which pharmaceutical wholesalers to be selected. The samples were 41 pharmaceutical wholesalers in Surabaya and they were given questionnaire they need to answer.. The data were collected in the period of August until September 2015, where the CDOB certifi cation was not mandatory yet. According to data collected, results show that 40 out of 41 (97.56%) PBF do not possessed CDOB certifi cate. From 41 PBFs, 18 (43.90%) PBF has met ≥80% CDOB, 16 (39.02%) PBF has met ≥65 - <80% CDOB, 6 (14.63%) PBF met ≥ 50 - <65% CDOB and 1 (2.44%) PBF met <50% CDOB. There was 1 (2.44%) PBF never conduct a study of the quality management, 4 (9.76%) offi cial pharmacist of respective PBF had never attended CDOB’s training and 5 (12.20%) PBF all their personnel have not been trained CDOB, 5 (12.20%) PBF never check the registration number, there are 2 (4.88%) PBF rarely keep the appropriate medication storage temperature indicated on the drug packaging.Overall, it may be said that not all the PBF in Surabaya implemented 9 aspects of CDOB. This could aff ect directly or indirectly to the quality of pharmaceutical products they distributed. CDOB certifi cation however, just became mandatory in November 2017.

Abstract

Cytotoxicity tests on ethanol extract of Aglaia elliptica Blume leaves and its fractions on cervical cancer cell line (HeLa) were carried out. Dried powder of Aglaia elliptica leaves was extracted using ethanol andthen fractionated by n-hexane, ethyl acetate, and buthanol solvents. The cytotoxicity tests using MTT method (prolipheration inhibiton test) on HeLa cell lines were conducted for ethanol extract, n-hexana, ethyl acetate, and buthanol lractions.The results showed that the ethanol extract of Aglaia elliptica and its fractions have inhibition effect against HeLa cell line with IC50 of 282.44 ppm, 177.64 ppm, 16.03 ppm, 211.77 ppm, 181.33 ppm for ethanol extract, n-hexane, ethyl acetate, buthanol, and Water fractions respectively.

Abstract

Fenugreek seed (Trigonella foenum-graecum L.) is one of Indonesian medicinal plant with many known activities such as antidiabetic, antihypercholesterolemia, phytoestrogen and anticancer. Fenugreek seed contains steroidal sapogenines (i.e. diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside), alkaloid trigonellin and flavonoid (i.e vitexin, orientin). This experiment was carried out to investigate the cytotoxic activity and its potency to induce apoptotic on breast cancer cell line, MCF-7. Cytotoxic activity was carried out by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium) method. Fenugreek seed was prepared by extraction (methanolic and ethanolic extract) and fractionation using solvents with different polarity (hexane, ethylacetic, buthanolic and water fractions). The potency of apoptotic induction assay was carried out using flowcytometric method staining with propidium iodide (20μg/mL) on 100 ppm sample. Apoptotic potency was evaluated based on % population of cells in subG0/G1 phase. Results showed that ethylacetic fraction has the lowest IC50 (41.81 ppm) and significantly (p<0.05) increase % population of cells in subG0/G1 phase (20.87%) on 100 ppm concentration. It is concluded that fenugreek seed posses cytotoxic activity and potency of apoptotic induction on MCF-7 cells.

Abstract

Bioanalysis method is needed to pharmacokinetic study of INH as antituberculosis. The main problem is INH structure is similar to that of acetyl isoniazid (AcINH) as its metabolite. Therefore, a selective separation method is needed to separate the INH from its metabolite and matrix. The aim of this study was to test the selectivity of separation method of INH and AcINH in human plasma in-vitro by high performance liquid chromatography (HPLC). The HPLC system used a reversed phase with UV detector and hexane sulphonate as an ion counter. The optimum conditions was obtained by using C18 as stationary phase, hexane sulphonate (pH 2.47)-methanol (65:35) as mobile phase, with flow rate of 1mL/min, and UV detector at wavelength of 265nm. The selectivity of method separation was indicated by a resolution value of ≥ 1.5. The tailing factor for INH and AcINH were 1.297, dan 1.912, respectively. The k values ware ​​less than 10 and N values ​​were greater than 5000 indicate good separation efficiency. The developing of HPLC was a selective for separating of INH and AcINH in human plasma in-vitro.

Abstract

Andrographolide is a diterpene lacton and sparingly soluble in water. The aim of this study is to prepare microspheres of A . paniculata ethylacetate fraction by microencapsulation using spray drying method. The ethyl acetate fraction of A. paniculata herbs was microencapsulated using PVP K30 and HPMC as the coating polymer in different ratios of 1:5; 1:7,5 and 1:10. Microspheres containing the ethyl acetate fraction of A. paniculata herbs were characterized in term of recovery factor, morphology, particle size distribution, water content and entrapment efficiency. The results showed that the A. paniculata microspheres have the recovery factor of 19,50% to 31,64%, spherical shape varies from smooth surface to irregular shape, particles size of 1,44% to 19,23 µm, moisture content of 5,15% to 13,44%, and entrapment echiency between 94,83 to 112,15%.

Abstract

Screening for citotoxicity has been conducted on 36 endophytic fungi isolated from medicinal plants in East Lombok. Isolates were cultivated in 100 mL potato dextrose yeast extract medium in 250 mL Erlenmeyer, incubated at 28°C and shaked in 150 rpm for 10 days. Broth was extracted with buthanol, ethyl acetate and dichloromethane. Extracts were concentrated under vacuum concentrator and assayed for citotoxicity against Artemia salina using the BSLT method. Screening using 100 mg/L of crude extract showed that 9 extracts were active out of 108 extracts assayed. Further screening based on LC50 showed that the ethyl acetate extract from fungus ENLT 74.3d.2 was the most active with LC 50=30.21 mg/L. Fungus was isolated from Cibotium barometz and described as Curvularia lunata. The most active extract was fractionated by column silica gel 60 and eluted using hexane, ethyl acetate, and methanol. These fractions showed that the ethyl acetate fraction was the most active fraction (LC50 = 4.69 mg/L). Purification was conducted by HPLC using column C18 and eluted by gradient system of acetonitril water 15% to 100% in 25 minutes. The active compound retention time was 14-16 min, λmax = 233 nm, and at 5 mg/L could inhibit 28% MCF-7 cell proliferation.

Abstract

Trypsin is an enzyme which is important for research as well as for the pharmaceutical industry, medicine and health, which is especially developed as a raw material for digestive enzymes. This study aimed to characterize PST from Lactobacillus plantarum FNCC 0270 that have not been studied as a producer of trypsin. Fermentation for producing PST by L . plantarum FNCC 0270 was performed in the LKB fermentor with working volume of 3.5 liters, at temperature of 37 oC, pH value of 8, agitation rate of 77 rpm and aeration of 0.5 vvm. The media composition were 3.64% baker’s yeast, 1.21% glucose, 0.13% skim milk. The fementation resulted in enzyme activities of 0.00129 U/mL and protein content of 0.49 mg/mL. Purification of the PST conducted by ultrafiltration Hollow Fiber Catridge 5 kD, presipitated with ammonium sulfate saturated (30-70%), 30 kD membrane polyesthersulfone, ion exchange column chromatography Q-XL and HiTrap affinity column chromatography. All of system increased purity of the crude PST. SDS-PAGE of PST indicated the molecular weights of 47.35; 38.42;21.40 and 12.96 kD. The PST was stable at pH value of 8 and temperature range of 25-35 oC. It is proved that the same half-life was 693.2 minutes. Km and Vmax values of the PST were 0.231 mM and 0.001 U/mL, respectively using N-α-benzoyl-DL-arginine-p-nitroanilide as the substrate. PST was inhibited by EDTA, Ca2+, Zn2+, Mg2+, Mn2+ and specific substrates. Based on the result of characterization of PST from L. plantarum FNCC 0270 can be concluded that the trypsin–like protease (PST) was trypsin.

Abstract

Aglaia elliptica Blume is one of plants that contain cyclopenta[b] tetrahydrobenzofuran compound with strong cytotoxic effect on various types of cancer cell lines. The objective of this study is to determine the inhibition effect of ethanol extract of A. elliptica leaves on mammary tumor growth in Sprague Dawley female rats induced by 7.12-dimethylbenz[a]anthracene (DMBA). The study was conducted with induction of 50 female rats with DMBA at a dose of 20 mg/kg orally for 11 times. Twenty-five female rats suffering mammary tumor were divided into 5 groups. Three groups were treated with ethanol extract at a dose of 50, 100 and 200 mg/200 g BW one day after the tumor appeared for 30 days, 1 group as a negative control and 1 group as a positive control (doxorubicin 2 μg/200 g BW). The results showed that the induction of DMBA resulting tumor incidence by 74% and tumor multiplicity of 2 nodules/rat. Histopathological analysis of mammary tumor, suggested that the carcinogenesis has reached the level of ductal carcinoma invasive (DCIV). The administration of ethanol extract at dose of 50, 100 and 200 mg/200 g BW suppressed the growth of mammary tumor volume by 30 %, 33.5% and 37.4%, respectively.
