Abstract

Aglaia elliptica Blume is one of plants that contain cyclopenta[b] tetrahydrobenzofuran compound with strong cytotoxic effect on various types of cancer cell lines. The objective of this study is to determine the inhibition effect of ethanol extract of A. elliptica leaves on mammary tumor growth in Sprague Dawley female rats induced by 7.12-dimethylbenz[a]anthracene (DMBA). The study was conducted with induction of 50 female rats with DMBA at a dose of 20 mg/kg orally for 11 times. Twenty-five female rats suffering mammary tumor were divided into 5 groups. Three groups were treated with ethanol extract at a dose of 50, 100 and 200 mg/200 g BW one day after the tumor appeared for 30 days, 1 group as a negative control and 1 group as a positive control (doxorubicin 2 μg/200 g BW). The results showed that the induction of DMBA resulting tumor incidence by 74% and tumor multiplicity of 2 nodules/rat. Histopathological analysis of mammary tumor, suggested that the carcinogenesis has reached the level of ductal carcinoma invasive (DCIV). The administration of ethanol extract at dose of 50, 100 and 200 mg/200 g BW suppressed the growth of mammary tumor volume by 30 %, 33.5% and 37.4%, respectively.

Abstract

The evaluation of toxicity of a mixed herbal extract containing mahkota dewa fruits (Phaleria macrocarpa Scheff. Boerl) and pegagan leaves (Centella asiatica L. Urban) on Wistar-strain rats had been carried out by the determination of the urea and creatinine content in urine and plasma after feeding. Oral doses of 100 mg, 500 mg, and 2500 mg of the mixed extract/kg body Weight were administered for 16 consecutive weeks to three groups of rats. Each treated group consisted of 15 males and 15 females, and the control group was represented by 10 males and 10 females. Samples of urine and plasma of the treated groups were taken at the time right before treatment (Week zero) and at Sm, 16th, 18th Week, while those of the control were taken at zero Week, 8th, and 16th Week, respectively. The result showed that theurea and creatinine contents among the treated and control groups were not signihcantly different. It could be concluded that oral administration ofthe mixed extract by a dose up to 2500 mg/kg body weight for 16 Weeks did not influence the urea and creatinine contents both in urine and plasma of the treated animals. Based on this result, it could be assumed that the use of the mixed extract is safe.

Abstract

Obesity or overweight is defi ned as the accumulation of abnormal or excessive fat in adipose tissue whichcan damage health.Several plants have been used empirically as antiobesity.This study aims to obtain candidate antiobesity compounds that act as pancreatic lipase enzyme inhibitors, fi nd some amino acids that involved on the active site of the receptors and modelling the interactions of these active compounds at antiobesity receptors.The study start with internal validation of the receptors used (4DOQ), then carried out the docking process for the native ligand, comparison compound (Orlistat) and each test compound with the same receptor. The softwares used are PLANTS, YASARA, ChemSketch, and Pymol. The results of the 4DOQ receptor validation have an RMSD value of 1.7097 Ǻ. The results of the docking score for orlistat (-65.3258), and 4 active compounds namely chlorogenic acid (-69,3009), 5-O-caff eoylquinat acid (-70.6911), 4,5-O-dicaff eoilkuinic acid (-85 , 3375), 5-Op-coumaroilkuinic acid (-67,1886). Amino acids that play a role in the affi nity on the active side of the ligand bond with the pancreatic lipase receptor are THR21 (Threonine), ASN84 (Asparagine), GLY19 (Glycine), GLN156 (Glutamine), ALA85 (Alanine), ILE63 (Isoleucine), ARG62 (Arginine) . The conclusion of this study there are 4 chemical compounds in green coff ee beans that are active as antiobesity with the mechanism of action of the pancreatic lipase enzyme inhibitor, namely chlorogenic acid, 5-O-caff eoylquinic acid, 4,5-O-diphoeilquinic acid, 5-op-coumaroilquinic acid.

Abstract

8-Hydroxy 9,12-octadecadienoic acid (8-HODE) is an oxidized aliphatic fatty acid belongs to the group of oxylipins, which shows bioactivity. It was reported previously that Curvularia lunata BioMCC FE-00283, an endophytic fungi from Cibotium barometz be able to produce 8-HODE by submerged fermentation. Glucose, monosodium glutamate, corn oil are media components which showed significant effect toward HODE production by C. lunata BioMCC FE-00283. In this study we reported the enhancement of 8-HODE production by adding potato extract to previouse medium. These four medium components, which were potato extract, glucose, monosodium glutamate and corn oil, were optimized for 8-HODE production by Response Surface Metodology (RSM). A full factorial Central Composite Design (CCD) was used to explain the interaction effect between media components. Maximum HODE production predicted by the quadratic model was 16.46 mg/L where media composition consits of 36.50% potato extract, 3.60 g/L glucose, 15.92 g/L monosodium glutamate and 0.96 ml/L corn oil. Experimentally verification using these medium composition production of 8-HODE reached 16.189 ± 0.526 mg/L. This optimization,which was added potato extract as variabel in medium optimization was enhanced 3 times fold of 8-HODE concentration compare to the 3 variabel optimization.

Abstract

The influence of temperature and time on ripening of Noni fruits (Morinda citrifolia L.) for the production of Noni juice was determined using mature, but unripe Noni fruits. The design of this experiment was a split-plot. The ripening times used as the main plot were 1, 2 or 3 months, and the ripening temperatures used as the sub-plot were 30°C, 40°C, and 50°C. The results showed that the difference of Noni juice properties at 50°C as ripening temperature during 1, 2 or 3 month were not significant, as were those at 40°C during 2 and 3 months. The concentration of antraquinon increases with the ripening time of Noni fruits.

Abstract

Hydroxyl octadecadienoic acid (HODE) is an oxidized aliphatic fatty acid from the oxylipins group, which shows bioactivity and can be produced by Curvularia lunata BioMCC FE-00283 by submerged fermentation. Glucose, monosodium glutamate, and corn oil were media components that showed significant effect toward HODE production by C. lunata BioMCC FE-00283. Composition optimization of these three medium components was carried out by Response surface methodology (RSM). A full factorial central composite design (CCD) was used to explain the interaction effect between media components. Maximum HODE production as predicted by the quadratic model have chromatogram area of 4540000, which was verified experimentally to be 1091338 ± 152489. Eventhough experimental veriiication showed lower result compared to the model prediction, however it was higher compared to the HODE production using basal media with unoptimized composition of carbon, nitrogen, and inducer which only have chromatogram area of 295045 ± 71200. The verification also showed that optimization increased HODE production by 3.7 fold compared to that produced by the unoptimized media.

Abstract

Drying process of Mengkudu (Morinda citrifolia L.) juice by spray drying method had been carried out. Two kinds of fillers were used in this process, i.e. 5%, 15%, 20%, and 25% of lactose monohydrate or 15% and 20% of maltodextrin, both in w/w %. The result obtained showed, that the dried powder containing 15 % of maltodextrin was better (dry fine and brown) than those with other concentration of fillers.

Abstract

Plasmid transformation is the introduction and incorporation of exogenous plasmid into cells or protoplast. The purpose of this research is transformation of pTRLI plasmid into protoplasts of Aspergillus terreus and obtain stable transformants. The research was initiated by isolation of pTRLI plasmid and determining its purity and concentration by nanodrop machine. Furthermore, protoplasts of A. terreus were isolated enzymatically by addition of chitinase, cellulase, and maserozyme enzymes. pTRLI plasmid was transformed into protoplasts of A. terreus by addition of calcium chloride and polyethyleneglycol (PEG) solutions. These transformants were grown in Czapek-Dox agar medium containing pyrithiamine 1 mg/L and the number of transformants/µg of pTRLI plasmid was calculated. The number of transformants were produced ranging from 12 to 19 transformants/µg of pTRLI plasmid. The success of the transformation was indicated by ptrA gene in transformants that could be amplified by PCR of 801 base pairs in size. It was concluded that PEG solution could be used to transform pTRLI plasmid into protoplasts of A. terreus and transformants are stable up to live generations by growing the transformants in Czapek-Dox agar medium containing pyrithiamine 1 mg/L.

Abstract

Tuberculosis is an infectious disease caused by Mycobacterium tuberculosis. Nicotinic acid derivatives such as isoniazid have the strongest anti-tuberculosis properties. For pharmacokinetics studying of isoniazid (INH), a method is needed to determine the levels of INH in plasma. Objective: The aim of this research is to partial validate of high performance liquid chromatography (HPLC) for analysis of INH in rat plasma. Method: For the preliminary study, rat plasma was used. The HPLC system used is a stationary phase C18 with length 250mm and temperature of 30°C, mobile phase hexane sulphonate acid 20mM pH 2.47–methanol (65:35). The analytical parameters in partial validated were linearity, lower limit of quantification (LLOQ), precision, accuracy, and recovery. Results: The results of linearity test of INH showed r value of 0.9996. LLOQ of this method was 0.1258μg/mL. The resulting accuracy and precision value met FDA requirements with a percent recovery ranging from 96.57–107.99%. Conclusion: The HPLC system was a valid method for analysis of INH in rat plasma.

Abstract

Pharmacy as the main type of distribution facility is still in great demand by investors. However, Jaminan Kesehatan Nasional (JKN) has the effect of decreasing turnover in some conventional pharmacies so that many pharmacies are closed. A pharmacy with a modern concept is expected to be an exit strategy of that problem. The purpose of this research is to find out and describe the concept of modern pharmacy according to the perceptions of respondents in DKI Jakarta province and its marketing strategy using the marketing mix concept. This research is a descriptive exploratory research by interviewing 100 respondents who have visited the pharmacy using a questionnaire. Data from interviews were analyzed using statistical software. The results showed that 98% of respondents agreed with the existence of a modern pharmacy concept. Modern pharmacy concepts that can be accepted by respondents are pharmacies that sell complete products, prices compete with surrounding pharmacies, locations close to housing or offices, friendly employees, fast and easy service processes, frequent promotions and social services in the community and a waiting room spacious and comfortable. The right marketing strategy for modern pharmacies is to prioritize excellent service and carry out vigorous promotions on online media and collaborate with private offices, government and universities in DKI Jakarta province.
