Abstract

Cantigi is an endemic plant of sub-alpine area of Mount Tangkuban Parahu in Bandung, Indonesia. Previous study showed ethanol extract of young red leaves had antioxidant activity, however no information on this activity if changed into nanoparticles. The purpose of this study was to determine the effects of gelatin and glutaraldehyde concentrations on the characteristics of Cantigi extract loaded gelatin nanoparticles and to evaluate the antioxidant activity of nanoparticles. Cantigi leaves were extracted by maceration using n-hexane, ethyl acetate, and ethanol 96%. The ethanol extract was dried, made into nanoparticles by varying gelatin (0.1; 0.2; and 0.3 g) and glutaraldehyde (0.1; 0.2; and 0.3 mL) amounts, and conducted at 500 rpm and 40 °C for 3 hours. Nanoparticles were evaluated for particle size, zeta potential, morphology, and antioxidant activity. Nanoparticles with glutaraldehyde amount variation had particle sizes (PS) of 105.9±26.2; 37.1±8.7; and 32.5±7.4 nm; polydispersity indeces (PI) of 0.508; 0.717; and 0.563; zeta potential values (ZPV) of 0.55; 0.89; and 0.78 mV; and antioxidant activities (IC50) of 56.15±0.16; 53.67±0.10; and 51.57±0.39 ppm, respectively. Then, nanoparticles with gelatin amounts variation had PS of 22.5±5.1; 37.1±8.7; and 83.3±21 nm; PI of 0.604; 0.717; 0.326; ZPV of 1.27; 0.89; 0.18 mV; and antioxidant activities of 51.58±0.19; 53.67±0.12; and 55.46±0.04 ppm, respectively. Nanoparticle morphology was spherical. Cantigi leaf extract can be made into gelatin nanoparticles; the smaller the concentration of the polymer used and higher the concentration of the glutaraldehyde, the smaller the resulted particle size and increased antioxidant activity. Antioxidant activities of nanoparticles was lower than those of the extract (IC50 16.84±0.30 ppm).

Abstract

Cassava pulp, a low cost solid byproduct of cassava starch industry, has been proposed as a high potential ethanolic fermentation substrate due to its high residual starch level, low ash content and small particle size of the lignocellulosic fibers. As the economic feasibility depends on complete degradation of the polysaccharides to fermentable glucose, the comparative hydrolytic potential of cassava pulp by six commercial enzymes were studied. Raw cassava pulp (12% w/v, particle sizeμm) hydrolyzed by both commercial pectinolytic (1) and amylolytic (2) enzymes cocktail, yielded 70.06% DE. Hydrothermal treatment of cassava pulp enhanced its susceptibility to enzymatic cleavageas compared to non-hydrothermal treatment raw cassava pulp. Hydrothermal pretreatment has shown that a glucoamylase (3) was the most effective enzyme for hydrolysis process of cassava pulp at temperature 65 °C or 95 °C for 10 min and yielded approximately 86.22% and 90.18% DE, respectively. Enzymatic pretreatment increased cassava pulp vulnerability to cellulase attacks. The optimum conditions for enzymatic pretreatment of 30% (w/v) cassava pulp by a potent cellulolytic/ hemicellulolytic enzyme (4) was achieves at 50 °C for 3, meanwhile for liquefaction and saccharification by a thermo-stable α-amylase (5) was achieved at 95 °C for 1 and a glucoamylase (3) at 50 °C for 24 hours, respectively, yielded a reducing sugar level up to 94,1% DE. The high yield of glucose indicates the potential use of enzymatic-hydrothermally treated cassava pulp as a cheap substrate for ethanol production.

Abstract

The effects of mycorrhizal endosymbiotic bacteria Bacillus subtilis B10 and composite of arbuscular mycorrhizal fungal spores in green house experiment were examined in order to evaluate their effectiveness and compatibility with oil palm seedlings in the presence of a fungal pathogen Ganoderma boninense, the most serious pathogen in oil palm (Elaeis guineensis Jacq) in Indonesia. A three factors experiment were conducted, with mycorrhizal inoculation (M0 and M1), bacterial B. subtilis B10 inoculation (B0 and B1), and G. boninense inoculation (G0 and G1) as the first, second, and third factors, respectively. The results showed that disease severity index, plant height, root dry-weight, and phosphorus uptake were affected by co-inoculation of mycorrhizal endosymbiotic bacteria B. subtilis B10 and composite of arbuscular mycorrhizal fungi. Co-inoculation of mycorrhizal endosymbiotic bacteria B. subtilis B10 and arbuscular mycorrhizal fungi did not only reduce the percentage of basal stem rot incidence, but also significantly increased plant height and phosphorus uptake by oil palm seedlings. Our results suggest that in oil palm seedlings mycorrhizal endosymbiotic bacteria B. subtilis B10 worked synergistically with arbuscular mycorrhizal fungi in increasing plant adaptation toward biotic stress of pathogen G. boninese and could be promising biocontrol agents.

Abstract

Surfactin is a lipopeptide biosurfactant that show potential biomedical application due to its activities such as antiviral, antibacterial, antifungi, anticancer, and antimycoplasma. Bacillus amyloliquefaciens MD4-12, isolated from oil-contaminated soil, produced promising yield of surfactin in McKeen medium. The production of surfactin was influenced by many fermentation process parameters such as carbon, nitrogen, minerals and also environmental conditions such as pH and agitation. Therefore, to obtain high yield of surfactin by Bacillus amyloliquefaciens MD4-12, optimization of process production was conducted in shake flask fermentation using response surface methodology. McKeen medium composition was used as basal medium.  Screening of the best carbon and nitrogen source were selected in preliminary experiments followed by selection of the influencing significant parameters on surfactin production using Plackett-Burman design. Selected parameters were optimized by central composite design and for the data analysis was used response surface methodology. The result showed that the optimum medium composition contained (g/L) 45.0 glucose, 6.33 urea, 1.0 monosodium glutamate, 1.85 MgSO4.7H2O, 0.4 KCl, 0.5 K2HPO4 and 0.5 mL trace elements. The surfactin yield at optimal condition was 1.25 g/L, increased 2.4 times compared to condition prior to optimization.
