Abstract

Health is the most important and primary thing in human life. Industry and human lifestyle development have both positive and negative impacts. One of the adverse consequences to be aware of is the proliferation of a variety of degenerative diseases. Sidaguri leaves (Sida rhombifolia L.) grow naturally on roadsides, grasslands, forests, fields, areas exposed to bright sunlight, and slightly sheltered places. It is a plant that is widespread in tropical areas ranging from lowlands to 1,450 m above sea level throughout the world. The plant's chemical content includes alkaloids, hypoparin, flavonoids, triterpenoids, sterols, sugars, vaccissinol, vaccissinone, betaine, and phenylalanineSidaguri leaves (Sida rhombifolia L.) are medicinal plants from the Malvaceae family. Almost all parts of this plant can be used for medicinal purposes, particularly in alternative medicine. This study aimed to determine the total saponin content in a 70% ethanol extract of Sida rhombifolia L. Sidaguri leaves, extracted using the maceration method and a 70% ethanol solvent. Using vanillin, H2SO4, and diosgenin as standards, we used UV-Vis spectrophotometry to find out how much total saponin was present. We measured antioxidant activity using the DPPH free radical scavenging test. The test showed that the 70% Sidaguri ethanol extract had a total saponin content of 196.28 ± 1.60 mg DE/g. It also had an IC50 value of 208.63 ± 7.73 mg/mL against DPPH in the antioxidant activity test. This shows the potential of Sidaguri leaf extract as a natural antioxidant.

Abstract

DNA (deoxyribonucleic acid) extraction method is the process of separating DNA from the sample. In this process, the DNA obtained must be protected from contamination by RNA, carbohydrates, lipids, and proteins. Contamination of RNA, carbohydrates, lipids, and proteins can increase DNA purity. DNA purity was measured using a NanoDrop 2000 spectrophotometer measured by the absorbance ratio at 260 nm and 280 nm wavelengths. Good quality DNA will have an A260/A280 ratio of 1.7–2.0 and a concentration > 0.03 pg. This study aimed to obtain the appropriate DNA extraction method for fresh meat samples (a mixture of rat and chicken meat). This research consisted of two stages: the DNA extraction stage using the Progenus EasyFast™ Extraction Kit for Meat Products and the amplification stage using the EASYFAST™ Rat Detection Kit. This study used 16 samples of a mixture of rat meat and chicken with concentrations of rat meat: 5, 10, 15, and 20%. At the extraction stage, the incubation time was optimized for 15, 30, 45 minutes, and 1 hour. The results showed that the one hour incubation had a lowest CT value in the results of PCR amplification.

Abstract

Improving entrepreneurship skills is very important in the current state of the COVID-19 pandemic. The number of layoffs carried out by the industrial world is one of the reasons for the high number of layoffs now. Entrepreneurial skills are an important part of educating school children from the start so that after graduation they are expected to open up job opportunities for themselves and others. Vocational High School (SMK) is now one of the schools that can produce skilled and innovative personnel. One of the private vocational schools in Surakarta that is trying to do that is SMK BATIK 1. The UHAMKA FFS Service Team provides training and guidance on the pharmacy expertise program to make foot care preparations, in the form of foot sanitizer and foot lotion. Preparations made from natural ingredients lemongrass extract, ginger extract, and coffee extract. Activities include lectures and workshops. Activities provide benefits in the form of increasing the soft skills and hard skills of students in utilizing natural materials in dosage forms which will increase the selling value.

Abstract

The meat-based products are highly susceptible to counterfeiting, primarily due to high consumer demand of meat derivative products, such as meatballs. This demand creates opportunities for food fraud by specific industries, including adulterating meat with non-halal species, such as rats. This research aimed to detect rat meat contamination in meatball samples from the Indonesian local market using Real-time Polymerase Chain Reaction (RT-PCR). The RT-PCR amplification involved an initial denaturation step at 95°C for 3 minutes, followed by denaturation at 95°C for 15 seconds, and annealing/extension at 60°C for 1 minute. The rat-specific probe primer included in the kit produced an increasing curve in the External Positive Control (EPC) with a Ct value of 27.22, and no amplification occurred in the Negative Control (NTC). The analysis of 30 samples from meatball vendors yielded negative results, as there was no increase in the FAM (rat) curve, indicating that none of the meatballs were contaminated with rat DNA.
