Abstract

Ikan tuna merupakan komoditas penting Indonesia dengan nilai produksi yang meningkat setiap tahun. Pengolahan ikan tuna menghasilkan limbah 50-70%. Jeroan merupakan limbah yang dapat dimanfaatkan untuk menghasilkan biomaterial baru, contohnya heparin. Tujuan penelitian adalah untuk mendapatkan komponen heparin dari beberapa bagian jeroan ikan tuna, yaitu hati, lambung, usus dan sekum pilorus. Ekstraksi heparin dilakukan dengan metode enzimatis menggunakan enzim papain. Ekstrak kasar heparin dipurifikasi lebih lanjut dengan fraksinasi aseton. Gugus fungsi heparin murni dianalisis menggunakan FTIR spektrofotometer dan kandungan heparin ditentukan menggunakan sulfate GAG assay. Hasil penelitian menunjukkan bahwa usus dan sekum pilorus menghasilkan rendemen ekstrak sebesar 0,66±0,06% dengan kandungan heparin 43%. Heparin murni dari jeroan ikan tuna memiliki gugus fungsi penciri senyawa heparin, yaitu karboksil, asetil, hidroksil, siklus ester, dan atom N tersulfasi. Penelitian ini menunjukkan bahwa jeroan ikan tuna dapat menjadi alternatif sumber heparin.

Abstract

Fungi endofit laut merupakan fungi yang berasosiasi dengan jaringan organisme laut, yaitu rumput laut, koral, lamun, bunga karang maupun sedimen di lingkungan laut. Fungi dapat memproduksi metabolit sekunder berupa senyawa enzim ekstraseluler, di antaranya selulase, amilase, pektinase, kitosanase, lipase, maupun protease. Penelitian ini bertujuan menentukan potensi isolat fungi endofit berdasarkan aktivitas kitosanolitik, selulolitik, dan proteolitik untuk mendegradasi kitosan. Isolat fungi endofit laut diisolasi dari jaringan rumput laut, lamun, dan mangrove dari perairan Sukabumi dan Kepulauan Buton, Indonesia. Fungi sebanyak 20 isolat dilakukan penapisan aktivitas enzimatik (kitosanolitik, selulolitik, dan proteolitik) dengan metode difusi agar untuk mengetahui diameter zona bening yang dihasilkan. Proses hidrolisis dilakukan dengan menginokulasi isolat ke media hidrolisis yang berisi koloidal kitosan selama 7 hari. Hasil penelitian menunjukkan 12 isolat memiliki aktivitas kitosanolitik, 8 isolat selulolitik, dan 10 isolat proteolitik. Lima isolat memiliki aktivitas terhadap semua aktivitas enzimatik yang diuji. Dua isolat dengan kode KVA dan BM48A memiliki diamater zona bening kitosanolitik tertinggi. Kedua isolat tersebut diidentifikasi melalui DNA Barcoding sebagai spesies dengan galur Trichoderma harzianum KTR3 dan Aspergillus sydowii KTR50. Rendemen hidrolisat kitosan yang dihidrolisis menggunakan isolat T. harzianum KTR3 dan A. sydowii KTR50 7,44% dan 6,74%. Nilai viskositas yang dihidrolisis menggunakan isolat T. harzianum KTR3 dan A. sydowii KTR50 berturut-turut 21,10 cP dan 9,26 cP serta nilai bobot molekul 9,06 kDa dan 4,47 kDa. Aktivitas enzimatik yang dihasilkan oleh isolat fungi endofit laut memiliki kemampuan dalam mendegradasi kitosan, hal ini dibuktikan dengan terjadinya penurunan nilai viskositas dan bobot molekul kitosan.

Abstract

Coastal plant locally known as terong pungo (Solanum sp.) was used by Aceh community as traditional medicine for toothache. The objectives of this study were to isolate endophytic fungi from the leaves of terong pungo and determine the antibacterial activity of the fungi against bacteria causing dental caries and infection. Eight endophytic fungi were isolated. Antagonism test among the isolates was performed to select the most dominant fungus. TP6 was the selected fungus based on the antagonism assay. Crude extracts of the fungus were macerated from the culture broth using ethyl acetate. The strongest antibacterial activity of the extracts was obtained when the fungus was at 12 days of cultivation. The fungal crude extract was strongly active against Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa with inhibition zone diameter of 20, 21 and 23 mm, respectively.

Abstract

Multidrug-resistant (MDR) bacterial infections have become a serious problem for the last several decades. To solve this issue, the finding of new antibiotic candidates is an urgency. Natural pigment is known to have biological activity against pathogenic bacteria. Coastal fungi are an unexplored source of natural pigment to fight MDR bacteria. This research was aimed to isolate coastal endophytic fungi from smooth ant plant (Hydnophytum formicarum), to screen endophytic fungi that produce red pigment, to extract the red pigment, to determine antibacterial activity of the red pigment, and to identify the coastal endophytic fungi producing the red pigment. In this study, seven fungi were isolated as endophytic fungi from H. formicarum. There were three isolates that produced extracellular pigment, i.e., RS 1A produced red pigment, RS 3 produced black pigment, and RS 6A produced yellow pigment. Our study focused on the red pigment which was produced by endophytic fungus strain RS 1A. The yield of red pigment was 8.8657% (w/w). This study showed that red pigment had antibacterial activity against Escherichia coli, Acinetobacter baumannii, and Proteus mirabilis strain MDR. Based on molecular and morphological identification, the endophytic fungus strain RS 1A was identified as Aspergillus versicolor.
