Abstract

Grouper (Epinephelus sp.) is a superior Indonesian fishery commodity and has high economic value, has a high price and is an export commodity. Currently, grouper cultivation has developed, to meet the need for seeds, there is a need for a grouper hatchery business whose technology can be applied. Grouper seeds that are in demand, especially for the export market, are cantang grouper, which is a hybrid of female tiger grouper (Epinephelus fuscoguttatus) and male kertang grouper (Epinephelus lanceolatus). The aim of this research was to evaluate the reproductive performance and production performance of hybrid cantang grouper at the hatchery scale. The method used was active participation by directly following activities in the field and obtaining primary data (observation, interviews) and secondary data (literature study) at the Brackish Water Aquaculture Fisheries Center Situbondo, East Java. The stages of cantang grouper hatchery activities include container preparation, natural food culture, parent rearing, parent selection, parent spawning, egg handling, larva and fry care, water quality management, pest and disease control, as well as harvesting, packaging and transportation. The Hatching Rate (HR) value during the research obtained from stocking 150,000 eggs was 115,200 eggs or 76.8%. The Survival Rate (SR) value during the research obtained from stocking 115,200 fish with a total of 16,271 fish harvested or 14.1%.

Abstract

This study aims to identify the fucoidan content extracted from Sargassum sp. collected from Sundak Beach, Gunung Kidul. Fucoidan is a sulfated polysaccharide compound with various biological activities, including anticoagulant, antitumor, and anti-inflammatory activities. Sargassum sp. was chosen as the source of fucoidan due to its abundance in coastal waters and its pharmacological potential. The extraction method was carried out using a hot water extraction technique with acid treatment, followed by a purification process using dialysis. The extracted fucoidan content was then analyzed using spectrophotometry and Thin Layer Chromatography (TLC) to determine its concentration and purity. Fourier Transform Infrared (FTIR) spectroscopy was used to identify the functional groups of the compound. The results showed that fucoidan was successfully extracted from Sargassum sp. with a significant yield. The main component of the extracted fucoidan from Sargassum sp. in this study was fucose, as indicated by the TLC results. The Rf value of the fucoidan hydrolysate from Sargassum sp. and the standard fucoidan, which was 0.51, matched that of fucose, indicating that both samples contained fucose. This study concludes that Sargassum sp. from Sundak Beach, Gunung Kidul, is a potential source for high-quality fucoidan production.

Abstract

Research on the use of black cumin (Nigella sativa) as an immunostimulant in aquaculture has shown interesting potential. In this review, we explore the potential of black cumin as an immunostimulant in aquatic animals. Based on a recent literature review, we highlight the positive effects of black cumin extract in enhancing immune responses in aquatic organisms. The active compounds in black cumin, such as thymoquinone and α-hederin, are thought to have the ability to stimulate the immune system, increase cytokine production, and increase phagocytic activity. Implications of using black cumin in aquaculture include the potential to improve the health and productivity of aquatic animals. However, challenges such as dose optimization, effectiveness in various aquatic species, and environmental risk management need to be addressed. Suggestions for further research include more in-depth studies of the mechanism of action, clinical trials on various aquatic species, as well as environmental impact assessments. With a careful and coordinated approach, the use of black cumin as an immunostimulant has the potential to increase sustainability and productivity in aquaculture practices.

Abstract

The aquaculture sector is growing rapidly, but it faces various challenges, particularly concerning the health of fish and other aquatic organisms.This study aims to evaluate the potential of mangosteen peel extract as an antimicrobial agent against the bacterium Aeromonas hydrophila. This bacterium is known as a pathogen in fish and can cause significant diseases in aquaculture. Mangosteen peel extract was chosen because it contains active compounds such as xanthones, which have been proven to have antimicrobial properties. The initial method involves extraction with methanol, followed by filtration and separation using an evaporator. The method used to determine the content of mangosteen peel extract is FTIR analysis. Next, the antibacterial activity of the crude mangosteen peel extract is tested. The antioxidant testing is based on the agar diffusion method. Aeromonas hydrophila bacteria are cultured at a tested LD50 density of 0.05 ml on a TSA plate medium, spread evenly across the entire agar surface with a glass stick. Paper discs with a diameter of 6 mm, previously dipped in crude mangosteen peel extract, are placed on different TSA media. The test is repeated three times (triplicate) to obtain the average inhibition zone (clear zone), then incubated at 29°C, and observations are made of the inhibition zone's development at the 24th hour. The inhibition zone of mangosteen peel extract (Garcinia mangostana) against Aeromonas hydrophila bacteria, measuring 0.2±0.0 cm (20 mm), falls into the category of very strong. This indicates that mangosteen peel has the potential to be used as an alternative immunostimulant in place of antibiotics for the control of Motile Aeromonas Septicemia.

Abstract

Desa Rempek adalah sebuah desa yang terletak di Kecamatan Gangga, Kabupaten Lombok Utara, Provinsi Nusa Tenggara Barat. Salah satu mata pencaharian masyarakat desa Rempek adalah pada bidang perikanan, yaitu budidaya udang vannamei menggunakan kolam bundar yang dilakukan oleh Kelompok “Lempenge” sejak tahun 2020. Namun keterbatasan pengetahuan mengenai teknik budidaya udang seperti konstruksi tambak yang benar, pengelolaan kualitas air, pengelolaan pakan, dan berbagai permasalahan teknis lainnya dalam budidaya udang vannamei, khususnya pengelolaan pakan, masih menjadi kendala utama bagi organisasi budidaya perikanan. Kegiatan pengabdian ini bertujuan untuk meningkatkan pemahaman dan pendampingan terkait manajemen pakan dan suplementasi vitamin kompleks sehingga kualitas dan hasil produksi (kuantitas) udang vannamei dapat maksimal. Metode kegiatan pengabdian ini meliputi survei lapangan, pembuatan materi sosialisasi dan bimbingan teknis kegiatan, pembentukan model teknologi, pendampingan pelaksanaan teknis, dan evaluasi. Dari kegiatan pengabdian ini didapatkan bahwa manajemen pakan yang baik ditambah dengan suplementasi vitamin kompleks dapat meningkatkan pertumbuhan dan produksi udang vannamei yang dibudidaya dengan kolam bundar. Berdasarkan kegiatan pengabdian ini dapat disimpulkan bahwa dengan adanya pengabdian ini dapat meningkatkan pemahaman para pelaku budidaya dalam manajemen pemberian pakan yang tersuplementasi vitamin kompleks.

Abstract

Cromileptes altivelis humpback grouper is a commodity with high economic value. Humpback grouper is Indonesia's main export commodity. The high value of sales and production of humpback grouper is constrained by the many disease attacks in the humpback grouper aquaculture industry. An alternative solution that has been continuously researched for several years to tackle humpback grouper disease is the use of immunostimulants. The use of immunostimulants orally can increase non-specific immunity of humpback grouper to ward off disease attacks and is environmentally friendly. Immunostimulants such as Lactococcus lactis HNL12, Lactococcus lactis HNL12 combined with Schizochytrium limacinum (algae flour), Peptidoglycan, Phycocyanin (Spirulina platensis), Bacterium, Vitamin C, Chromium Yeast, Commercial Yeast can enhance the non-specific immune system in humpback grouper (Cromileptes altivelis). These immunostimulants increase non-specific immunity such as Respiratory burst Activity (RB), superoxide dismutase (SOD), alkaline phosphatase (AKP), lysozyme (LZM), Phagocytosis Activity, Phagocytosis Index, Total leukocytes, hemoglobin, hematocrit, phagocytic activity, lysozyme activity, serum protein, albumin and globulin, total erythrocytes, haemoglobin, hematocrit. These immunostimulants can also prevent attacks of Vibriosis disease caused by the bacteria Vibrio harveyi, Vibrio algynoliticus and Vibrio parahaemolyticus as well as Viral Necrosis Virus (VNN) disease by increasing the survival rate compared to fish that are not given immunostimulants.Keywords: cromileptes altivelis, immunostimulant, non-specific immunity, fish disease

Abstract

Motile aeromonas septicemia (MAS) adalah salah satu penyakit yang sering menyebabkan kematian pada ikan, dan disebabkan oleh bakteri Aeromonas hydrophila. Salah satu cara untuk menanggulangi penyakit bakteri yang menyerang ikan adalah dengan menggunakan imunostimulan. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian fucoidan secara oral terhadap perkembangan gejala penyakit ikan lele yang diinfeksi A. hydrophila.  Penggunaan metode asam digunakan untuk mengekstraksi fucoidan dari Sargassum sp. Penelitian ini melibatkan empat perlakuan dan tiga ulangan. Perlakuan P1, P2, P3, dan P4, masing-masing meliputi kontrol, 2, 4, dan 6 g fucoidan per kilogram pakan. Pakan diberikan kepada ikan dalam jumlah 5% dari berat kering pakan terhadap biomassa ikan. Setelah itu, dilakukan uji tantang dengan memberikan dosis 0,1 mL per ikan dari A. hydrophila dengan kepadatan 2,77 x 107 sel per ml secara intraperitonial satu minggu setelah pemberian fucoidan secara oral. Hasil penelitian menunjukkan bahwa pemberian fucoidan secara oral dengan dosis 2, 4, dan 6 g fucoidan per kilogram pakan tidak menghasilkan peningkatan yang signifikan dalam sintasan (P>0,05) dan tidak mampu menunda MTD pada ikan yang terinfeksi A. hydrophila KP1 jika infeksi dilakukan di bawah LD50. Perlu dilakukan penelitian lebih lanjut dengan menambahkan dosis fucoidan pada pakan ikan lele untuk mendapatkan hasil yang lebih baik.Motile aeromonas septicemia (MAS) is a disease that often causes death in fish, and is caused by Aeromonas hydrophila. One of ways to overcome bacterial diseases that attack fish is to use immunostimulants. The aim of this study was to determine the effect of oral administration of fucoidan on the development of disease symptoms in catfish infected with A. hydrophila. The acid method was used to extract fucoidan from Sargassum sp. This experiment involved four treatments and three replications. Treatments P1, P2, P3, and P4, respectively, included control, 2, 4, and 6 g of fucoidan per kilogram of feed. Feed was given to fish in an amount of 5% of the dry weight of the feed to fish biomass. After that, a challenge test was carried out by giving a dose of 0.1 mL per fish of A. hydrophila with a density of 2.77 x 107 cells per mL intraperitoneally one week after oral administration of fucoidan. The results showed that oral administration of fucoidan at doses of 2, 4, and 6 g of fucoidan per kilogram of feed did not result in a significant increase in survival (P>0.05) and was unable to delay MTD in fish infected with A. hydrophila KP1 if infection carried out below LD50. Further study needs to be done by adding a dose of fucoidan to catfish feed to get better results.

Abstract

Sekotong adalah salah satu daerah dengan potensi besar dalam budidaya kepiting bakau. Namun, dalam praktiknya, para pembudidaya sering menghadapi kendala dalam pemanfaatan sumber daya pakan yang efektif dan berkelanjutan. Oleh karena itu, program sosialisasi teknik pembuatan tepung ikan di Desa Sekotong bertujuan untuk memberdayakan para pembudidaya kepiting bakau melalui pengenalan metode efisien dalam pengelolaan pakan, serta peningkatan pengetahuan terkait pemanfaatan sumber daya ikan lokal, khususnya ikan rucah. Metode yang digunakan dalam kegiatan ini adalah survei kegiatan pendahuluan, pelatihan, workshop, demonstrasi praktis, monitoring dan evaluasi. Kegiatan diikuti oleh beberapa stakeholder: pembudidaya kepiting bakau, dosen dan mahasiswa Unram, dosen dan mahasiswa Unair, serta remaja setempat. Hasil dari kegiatan ini adalah masyarakat pembudidaya kepiting bakau dapat memahami, menjelaskan, dan membuat tepung ikan rucah secara mandiri. Sosialisasi teknik pembuatan tepung ikan di Desa Sekotong, Lombok Barat, telah memberikan dampak positif dalam meningkatkan pengetahuan dan keterampilan para pembudidaya kepiting bakau. Untuk menjaga keberlanjutan program, disarankan adanya pengembangan lebih lanjut terhadap jejaring kerjasama antarpetani, serta pendampingan lanjutan dalam menghadapi perubahan lingkungan dan pasar.

Abstract

Pembangunan sektor perikanan memiliki peran krusial dalam memperkuat ekonomi lokal dan meningkatkan ketahanan pangan di banyak wilayah, termasuk Desa Sekotong. Budidaya kepiting Bakau (Scylla serrata) telah menjadi salah satu kegiatan utama yang berpotensi meningkatkan pendapatan masyarakat di Desa Sekotong. Pemberdayaan masyarakat melalui pelatihan dan sosialisasi telah terbukti efektif dalam meningkatkan pengetahuan dan keterampilan dalam bidang budidaya perairan. Namun, sejauh ini, program pemberdayaan masyarakat yang dilakukan sebelumnya masih terfokus pada pemberian pakan alami dan teknologi budidaya lainnya, belum menyentuh aspek pembuatan pakan berbasis tepung ikan rucah. Dalam konteks ini, kegiatan pengabdian ini bertujuan untuk memberikan pemahaman dan keterampilan kepada pembudidaya kepiting Bakau mengenai pembuatan pakan berbasis ikan rucah serta pemanfaatannya dalam budidaya kepiting. Metode yang digunakan adalah melalui serangkaian pelatihan, workshop, demonstrasi praktis, dan monitoring serta evaluasi. Hasilnya menunjukkan peningkatan signifikan dalam pengetahuan dan keterampilan peserta terkait dengan pembuatan pakan kepiting berbasis ikan rucah. Program ini memberikan manfaat yang nyata bagi masyarakat Desa Sekotong dalam meningkatkan pendapatan mereka melalui peningkatan produksi dan kualitas budidaya kepiting Bakau. Selain itu, melalui keterlibatan mahasiswa dan dosen dari perguruan tinggi, program ini juga memberikan kontribusi dalam pengembangan ilmu pengetahuan dan teknologi di bidang budidaya perairan. Kesimpulannya, kegiatan pengabdian ini berhasil memberikan dampak positif bagi pembudidaya kepiting Bakau di Desa Sekotong. Evaluasi berkala dan kerjasama antara berbagai pihak diharapkan dapat memastikan berlanjutnya keberhasilan program ini dalam jangka panjang.

Abstract

Abstract: Sargassum sp. seaweed is included in the category of brown macro algae that has not been cultivated and has not been optimally utilized. Sargassum sp. has economic value because it contains alginate and iodine which are utilized in the food, pharmaceutical, cosmetic, and textile industries. Sargassum sp. has a high carbohydrate content that can be converted into bioethanol and bioenergy. So that this potential can be developed to improve the community's economy and increase efforts to protect the environment from pollution from the use of fossil fuels. Sargassum sp. seaweed can be found in the waters of Gerupuk, Sengkol Village, Pujut District. Gerupuk is a hamlet that is included in the Mandalika Creative Economic Zone, among the community's livelihood activities is seaweed cultivation. Seaweed farmers in Gerupuk only cultivate K. alvarezii seaweed. They only know Sargassum sp. as a seaweed that grows wild and has no economic value. Sargassum sp. in Gerupuk is not utilized, so it needs to be educated on its benefits and utilization. This community service activity will be carried out in Gerupuk Hamlet, Sengkol Village, Pujut District, Central Lombok Regency. The output targets of this community service activity are appropriate technology for the cultivation of Sargassum sp. for bioethanol production, publications at national community service seminars and articles published in national community service journals indexed by sinta. The stages of implementing this activity are coordination of activities, counseling, demonstration of Sargassum sp. seaweed cultivation, mentoring, evaluation of activities, and collaboration with the Lombok Marine Aquaculture Center (BPBL). The expected results of this activity are that seaweed farmers in Gerupuk can cultivate Sargassum sp. to improve the household economy and the community economy.
