Abstract

In human, coronary heart disease causes by severe pathological atherosclerosis. In this study, we established animal model to study atherosclerosis caused by hyperlipidemia. This study therefore was undertaken to define the effect of increasing atherosclerosis risk factor, include body weight as well as age, cholesterol concentration and dietary fat in rabbit chow, and time of treatment. Male New Zealand White rabbits were divided into 4 groups; Group I and III were consisted of 2 months rabbit were fed with standard rabbit chow. To introduce atherosclerosis, the chow for Group II was contained 0.25% cholesterol and 5% palm oil; whereas the chow for group IV was contained 0.5% cholesterol and 5% coconut oil to induce higher atherosclerotic lesion. Results showed that group II and IV developed hyperlipidemia. However, aortic cholesterol concentration in those groups did not different significantly (P > 0.05). We suggest that low carbohydrate composition in diet, 50% lower compared to the previous researches, was able to increase high-density lipoprotein (HDL) concentration. This study demonstrated the complex interactions between low carbohydrate diet and cholesterol metabolism and the dramatic effects of reducing atherosclerosis risk factor; however, even though hyperlipidemic condition was achieved, total plasma cholesterol HDL ratio was maintained low. Abstract

Cancer is one of the leading causes of death in the world. Excess endogenous estrogen is a risk factor for breast cancer. Red betel leaf herbal plants have been used as an alternative cervical and colon cancer treatment. This study aimed to obtain the active compounds that play a role in ERα receptor inhibitors in silico and to determine the anti-breast cancer cytotoxic activity of the extract and fraction of red betel leaf against MCF-7 cells. We use in silico research method using the YASARA Structure software with anti-breast cancer receptors, namely 3ERT, and in vitro using the MTT test on the anti-breast cancer cytotoxic activity of MCF-7 cells. There are 38 compounds that were obtained from the research. The results of the in silico test showed that the bioactive compound that played a role in inhibiting the ERα was 2-(4-Hydro xyphenyl)-2-phenyl-N(3,3-diphenylpropyl)-acetamide (44578655), a compound from water fraction, with an inhibition constant of 2.82 × 10-8 µM and Gibbs free energy of –10.2880 Kcal/mol. In vitro results showed that the best cell growth inhibition value was obtained from the n-hexane fraction at a concentration of 500 ppm of 73.42%. The conclusions of this study indicate that the bioactive compound of red betel leaf is water fraction is the best fraction inhibition. However, the hexane fraction proved to have cytotoxic activity against breast cancer MCF-7 cells.

Abstract

Tofu is one of the main human foodstuffs and has become an alternative substitute for animal protein. Tofu contains relatively high protein, and active compounds, but the waste produced during the tofu processing process is very abundant, namely 3.5 million tons per year in Indonesia. The production of active compounds from tofu waste can use bromelain enzymes. The bromelain enzyme in this study was produced from the fermentation of pineapple leaf waste using Pediococcus pentosaceus E7. The production of active compounds from tofu waste has not been widely reported, so this study aimed to produce active compounds from tofu waste. This study began with bromelain production, measurement of bromelain activity, bromelain precipitation, peptide production from tofu waste, Liquid Chromatography-Mass Spectrometry (LC-MS) analysis, and antibacterial testing of active compounds derived from tofu waste degradation. The results showed that the peptide content of tofu liquid waste degraded using the bromelain enzyme was 41.01±0.06 mg/mL. The LCMS results showed that 18 bioactive compounds were contained in the degradation products of tofu liquid waste. The active compounds derived from the degradation of tofu waste are capable of inhibiting the growth of Salmonella Typhimurium ATCC 14028, yielding a clear zone index of 2.53±0.12.
