Abstract

Penelitian ini bertujuan untuk mengidentifikasi dan mengetahui kebutuhan LKPD berbasis pendekatan inkuiri terhadap pengetahuan peserta didik pada materi Animalia. Metode yang digunakan yaitu penelitian deskriptif menggunakan pendekatan kuantitatif dan kualitatif. Sampel pada penelitian ini yaitu satu orang guru biologi yang mengajar kelas X dan 33 orang peserta didik di SMAN 1 VII Koto Sungai Sarik. Hasil penelitian yaitu (1) Pada materi Animalia, guru sudah menggunakan LKPD tetapi belum maksimal, (2) Materi Animalia sebagai materi yang memiliki materi yang kompleks untuk dipelajari lebih lanjut, (3) Peserta didik memiliki pengetahuan terhadap materi Animalia, dan (4) Pengetahuan peserta didik dalam mempelajari materi Animalia semakin bertambah sesuai dengan kemampuan peserta didik. Kesimpulan penelitian ini yaitu LKPD berbasis pendekatan inkuiri pada materi Animalia dibutuhkan untuk memaksimalkan proses pembelajaran serta dapat mengasah keterampilan ilmiah peserta didik.

Abstract

Bat Cave is a new tourist attraction located in Indarung Village, Lubuk Kilangan District, Padang City. Changes in habitat will affect the fauna in it, especially the herpetofauna, because herpetofauna is very sensitive to environmental changes. Herpetofauna research in this area has never been done. The research was conducted in December 2022, the observation method used was the transect method combined with the VES (Visual Encounter Survey). The purpose of this study was to determine the diversity of herpetofauna species in the Bat Cave area. Based on the research, nine types of herpetofauna were found, consisting of six types of amphibians and three types of reptiles. Total number of individuals obtained was 57 individuals. The most common type found was Limnonectes blythii, namely 23 individuals. In this study, one individual was found endemic to West Sumatra, namely Ichthyophis elongatus. Based on the research conducted, the diversity of herpetofauna species in the Bat Cave area is low.

Abstract

Changing the function of habitat into a tourism area will have an impact on the diversity of Anuran species. This condition is currently happening in the Talago Nagari Paninjauan Waterfall Area, Tanjung Raya District, Agam Regency, West Sumatra. This study is therefore important for revealing the diversity of Anuran species before the impact to this habitat increase in the future. This study used the VES (Visual Encounter Survey) sampling technique, and information on various localities and the microhabitat of each species was recorded. A total 48 individuals, with 12 species belonging to six families of Anuran was collected. Species diversity in the Talago Waterfall area shows a medium level of diversity, with an index of 2.2653, a medium species richness index of 2.8414. In this area, no dominant species were found with a low dominance index (0.1207). If the level of tourism activity in the Talago Waterfall area increases, it will have an effect in the future by causing habitat disruption and the presence of Anura; thus, related parties must be able to take appropriate action on tourism activities in the area.

Abstract

Rice snails are one of the pests that have the potential to cause high losses to rice plants. Rice snail mucus contains glycosaminoglycans, proteoglycans, glycoprotein enzymes, copper peptides, metal ions, collagen, elastin, glycolic acid, allantoin, antimicrobial peptides and vitamin C. Vitamin C or ascorbic acid is one of the vitamins needed by the body that functions as a help process in the body's metabolism. To determine vitamin C levels can be done using the UV-VIS spectrophotometry method with a wavelength of 260 nm. This study aims to determine vitamin C levels from rice snail mucus concentration of 100% and rice snail mucus concentration of 50%. This study is descriptive quantitative, analyzing vitamin C levels obtained from a spectrophotometer can be seen from the absorbance of rice snail mucus. Based on the results of research obtained from the analysis of vitamin C levels of rice snail mucus, the concentration of 100% is 2,372. While vitamin C levels in rice snail mucus concentration of 50% were obtained at 0.911. So it can be concluded that there are vitamin C levels in rice snail mucus with a concentration of 100% greater absorbance than 50% rice snail mucus.

Abstract

Sumatera Barat memiliki tingkat keanekaragaman hayati yang cukup tinggi di Indonesia. Keanekaragaman hayati perlu dijaga dengan meningkatkan literasi sains peserta didik. Literasi sains berfungsi dalam meningkatkan kemampuan peserta didik dalam memahami dan mengkomunikasikan sains. Penelitian ini bertujuan untuk mengembangkan booklet. Penelitian ini merupakan penelitian pengembangan menggunakan model pengembangan 4-D. Penelitian ini dilaksanakan dengan 3 yaitu define, design, dan develop. Subjek penelitian ini 2 dosen ahli, 1 guru mata pelajaran biologi dan 27 peserta didik kelas X IPA SMA 1 Pertiwi. Objek penelitian ini adalah booklet keanekaragaman herpetofauna di Sumatera Barat. Lembar penilaian yang digunakan adalah lembar validitas, dan lembar praktikalitas, dengan teknik analisis data yang digunakan adalah statistik deskriptif. Hasil uji validasi yakni 92,35% kriteria sangat valid. Hasil uji praktikalitas oleh guru 90,13% kriteria sangat praktis dan hasil uji praktikalitas oleh peserta didik 90,06% kriteria sangat praktis. Maka disimpulkan booklet keanekaragaman herpetofauna di Sumatera Barat sangat valid dan sangat praktis.

Abstract

Sungai Batang Gadis merupakan sungai yang sangat penting sebagai penyedia air untuk mendukung kelangsungan hidup dan kegiatan perekonomian utama masyarakat, yaitu pertanian. Disepanjang pinggiran sungai Batang Gadis banyak ditemukan daerah pertanian masyarakat yang memanfaatkan irigasi dari aliran sungai Batang Gadis. Tujuan dari penelitian ini adalah untuk menentukan tingkat pencemaran pestisida di Sungai Batang Gadis Mandailing Natal. Metode : Metode survey digunakan dalam penelitian ini dengan mengambil dua titik sampling yaitu Muara Mais dan Tamiang. Penentuan titik sampling menggunakan Purposive sampling method. Pengambilan sampel air, tanah dan ikan yang diambil dianalisis menggunakan Gas Kromatografi. Hasil : Hasil penelitian menunjukkan residu pestisida hanya ditemukan pada stasiun II (Muara Mais). Residu pestisida yang ditemukan pada air, sedimen tanah dan ikan dari golongan organofosfat yang terdeteksi adalah diazinon, malation, dan klorfirifos sedangkan dari golongan organoklorin aldrin, dieldrin dan endosulfan. Selain residu pestisida juga ditemukan residu pupuk yaitu fosfat, amonia, nitrat, nitrit dan sulfida. Kesimpulan : Residu pestisida pada Sungai Batang Gadis yang ditemukan hannya pada titik lokasi yang ditemukan daerah pertanian disepanjang pinggiran sungai tersebut. Kadar residu pestisida dan juga residu pupuk yang ditemukan masih berada di bawah Batas Maksimal Residu (BMR) pestisida. Konsentrasi residu pestisida organofosfat tersebut yang terbesar terdapat pada tanah, lalu ikan dan terakhir air, sedangkan organoklorin terbesar pada ikan, air kemudian tanah.

Abstract

Peroksidase (POD) merupakan enzim antioksidan yang memiliki beragam fungsi dalam siklus hidup tanaman, salah satunya adalah sebagai pertahanan dalam melawan ROS dengan mengkatalisis konversi H2O2 menjadi udara dan O2 . Kemampuan aktifitas enzim POD dalam mengatur kandungan H2O2 memungkinkan enzim tersebut dapat mempertahankan tanaman dari cekaman. Metode yang dapat digunakan untuk amplifikasi gen POD salah satunya yaitu kuantitatif reverse transcription- PCR (qRT-PCR). Metode ini memerlukan beberapa komponen penting salah satunya yaitu primer ( forward dan reverse ). Primer yang digunakan dalam amplifikasi gen harus spesifik terhadap gen target sehingga dapat mengenali dan menempel pada gen target yang diinginkan. Penelitian ini bertujuan untuk mendesain primer yang sesuai untuk amplifikasi gen POD menggunakan teknik qRT-PCR. Primer dirancang menggunakan perangkat PrimerQuest. Primer yang telah dirancang kemudian dianalisis untuk spesifikasinya dengan geneious prime . Kemudian spesifikasi primer di cek menggugakan primer BLAST. Hasil desain primer dengan kriteria terbaik untuk amplifikasi gen POD yaitu Forward POD 5'-AAATGCGTCGATCTACTGTACCT-3' dan Reverse POD 5'-GTGTTGAAAATGGCAATAAACCGG-3'

Abstract

Rice is the main food source for Indonesians. The demand for rice to meet people's needs increases every year due to population growth and efforts to improve nutrition by the community. One of the problems that can affect rice production is drought. Drought is an environmental condition when plants do not get enough water to grow and develop optimally, which can cause a decrease in rice production. To find out how rice growth responds to drought, a study was carried out by giving drought simulation treatments using polyethylene glycol (PEG) on several rice varieties. This study used a completely randomized design that was arranged in a factorial manner with two factors. The first factor was the rice varieties (Harum, Situbagendit, Rosna) the second factor was 0% and 20% PEG concentration. The data obtained were then analyzed statistically using a two-way ANOVA test, and if the results were significantly different, then proceed with Duncan's test at the 5% level.The results showed that the drought simulation treatment had a negative effect on rice growth. Drought simulation treatment using 20% PEG resulted in a decrease in Kadar air relatif (KAR), root length, plant height, and root dry weight of rice. The highest decrease in KAR was found in the sensitive rice variety (Rosna), which was 43.42%. The highest average root length (7.99 cm) was on the sitabaendit variety, and the lowest (5.61) was on the rosna variety. The highest average crown height (17.32 cm) and the lowest (6.61) were on the rosna variety.

Abstract

This study aims to see the effect of giving noni extract (Morinda citrifolia L.) on the quality of spermatozoa of mice (Mus musculus L.). This study was conducted using an experimental method, with a completely randomized design (RAL), 3 treatments with 5 replicates with 15 male mice divided into three treatment groups, namely the control group (P0), the treatment group with a concentration of 0.3 g/BW (P1), and the treatment group with a concentration of 0.6 g/BW (P2) which then the data were analyzed using ANOVA. This research was conducted from March 2 to April 4, 2023. Observations of the effect of giving noni extract were made by making wet preparations of mice sperm and then observing under a microscope. Based on the research conducted, it was found that noni extract had a different effect from each concentration on motility and morphology in sperm. From the research, it can be concluded that the higher the concentration of noni extract given to mice will reduce the quality of sperm in mice However, at a concentration of 0.3 g/BB, it was found that noni had a positive effect on sperm morphology and motility in mice (Mus musculus L).

Abstract

Sweet potato varieties (Ipomoea batatas) Pucuk Hitam Panyalaian and Madu Pucuk Hitam Panyalaian have the potential to be a superior commodity in West Sumatra, but the information of genetic diversity is very limited. Data on the diversity of sweet potato germplasm is beneficial in the selection of plants to obtain superior cultivars in plant breeding. Aim: The aim of the study was to find specific primers for the study of sweet potato genetic markers using Ipomoea batatas chloroplast genes matK. Methods: DNA extraction from sweet potato young leaves; designing forward and reverse primers Ipomoea batatas chloroplast genes matK; PCR; sequencing; bioinformatics analysis and species identification by comparing the NCBI database. Results: Plant DNA barcoding PCR using designed primers matK were successfully resulting single DNA band in different amplicon size in some samples. This indicating that the designed primers used were able to distinguish variation in one species.
