Abstract

This study aims to see the effect of giving noni extract (Morinda citrifolia L.) on the quality of spermatozoa of mice (Mus musculus L.). This study was conducted using an experimental method, with a completely randomized design (RAL), 3 treatments with 5 replicates with 15 male mice divided into three treatment groups, namely the control group (P0), the treatment group with a concentration of 0.3 g/BW (P1), and the treatment group with a concentration of 0.6 g/BW (P2) which then the data were analyzed using ANOVA. This research was conducted from March 2 to April 4, 2023. Observations of the effect of giving noni extract were made by making wet preparations of mice sperm and then observing under a microscope. Based on the research conducted, it was found that noni extract had a different effect from each concentration on motility and morphology in sperm. From the research, it can be concluded that the higher the concentration of noni extract given to mice will reduce the quality of sperm in mice However, at a concentration of 0.3 g/BB, it was found that noni had a positive effect on sperm morphology and motility in mice (Mus musculus L).

Abstract

The cere fish species (Gambusia affinis) is a small fish, which is 2-3 cm. This fish is a wild fish that fills rivers, ditches and ditches. Cere fish are generally easy to adapt and reproduce. However, the water that irrigates rivers, ditches and ditches often contains community wastes, whether in the form of household waste, laundry, or even oil waste from workshops. So this study aims to obtain the LC5o-72 hour value of waste originating from river waste, laundry, and workshop oil on cere fish mortality. This research was conducted using a completely randomized design experiment. Each waste was treated in 4 treatments with different doses, namely 5 ml/L, 10 ml/L, 25 ml/L, and 50 ml/L for 72 hours with checking every 3 hours, 24 hours, 48 hours, and 72 hours. The results showed that laundry waste had a tolerance limit at a concentration of 10 ml/L and waste oil at a concentration of 25 ml/L.

Abstract

Borax, a toxic material, can disrupt the testes, hypothalamus, and spermatogenesis in the male reproductive system, leading to infertility. Apple peel contains quercetin, a substance crucial for boosting antioxidant levels and preventing free radical damage. The study aimed to assess the impact of apple peel extract on spermatozoa quality and testicular histology in borax-induced mice. The study utilized a Complete Randomized Design (CRD) with five treatment groups and five replications of each group: C- (without treatment), C+ (Borax), T1, T2, and T3, where borax was administered alongside various doses of apple peel extract: 0,2 mg, 0,4 mg, and 0,8 mg respectively. Testicular sections were collected for histopathological analysis and caudal epididymal sections for sperm quality assessment. Data were analyzed using ANOVA and DMRT tests using SPSS 23. The results showed that apple peel extract significantly increased the quality of mice spermatozoa in borax-induced mice (p<0.05). Apple peel extract to borax-induced mice also positively affected testicular diameter and weight, as well as the diameter of the seminiferous tubules and the number of Leydig cells. The optimal dose of apple peel extract in improving the quality of spermatozoa and testicular histology of mice was found to be0.4 mg / 20g BW.

Abstract

Abstrak Fertilitas merupakan hal yang sangat penting pada wanita untuk menghasilkan keturunan. Salah satu dari banyaknya faktor kesuburan wanita adalah nutrisi yang dikonsumsinya. Buah tomat (Lycopersicum esculentum Mill.) mengandung senyawa likopen yang memiliki banyak manfaat diantaranya sebagai zat antioksidan, menjaga kestabilan sel serta menurunkan resiko infertilitas. Tujuan dari penelitian ini untuk mengetahui pengaruh ekstrak tomat (Lycopersicum esculentum Mill.) terhadap fertilitas mencit betina (Mus musculus L.). Penelitian ini menggunakan metode Rancangan Acak Lengkap (RAL) yang terdiri dari tiga perlakuan dengan lima ulangan. Perlakuan kontrol (P0) yang tidak diberi esktrak tomat, perlakuan 1 (P1) dan perlakuan 2 (P2) dengan dosis ekstrak tomat 0,4 ml/hari dan 0,8 ml/hari. Pemberian ekstrak tomat, dilakukan setiap hari selama 15 hari secara oral menggunakan spuid modifikasi. Hasil penelitian menunjukkan adanya pengaruh ekstrak tomat (Lycopersicum esculentum Mill.) pada fertilitas mencit betina. Pada pemberian dosis 0,4 ml/hari menunjukkan terjadinya peningkatan kesuburan mencit pada fase estrus dibandingkan dengan kelompok perlakuan lainnya. Pemberian ekstrak tomat (Lycopersicum esculentum Mill.) dapat meningkatkan kualitas kesuburan mencit betina. Kata kunci: Fertilitas, Lycopersicum esculentum Mill., Mencit, Betina, Estrus

Abstract

Preservasi spermatozoa merupakan langkah penting dalam teknologi reproduksi ikan untuk mendukung program budi daya dan pelestarian spesies. Air kelapa muda memiliki komposisi elektrolit dan mineral yang mirip dengan cairan fisiologis, sehingga menjadi solusi pengencer alami yang potensial untuk pengawetan spermatozoa. Glukosa dalam madu berfungsi sebagai sumber energi untuk mempertahankan motilitas. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pengenceran dengan air kelapa muda dan konsentrasi madu yang berbeda terhadap motilitas dan morfologi spermatozoa ikan nila. Penelitian ini menggunakan metode eksperimental dengan Rancangan Acak Lengkap (RAL) yang terdiri dari 4 perlakuan dan 5 kali ulangan. Jenis pengencer yang digunakan terdiri dari air kelapa muda dan madu dengan berbagai konsentrasi, yaitu 0%, 3%, 6%, dan 9%. Data yang diperoleh mencakup motilitas (%) dan morfologi kemudian dianalisis secara statistik dengan One-way ANOVA dan morfologi spermatozoa ikan nila. Nilai kepercayaan data adalah 95% dengan nilai P-value <0,05. Hasil penelitian menunjukkan bahwa perlakuan pengenceran air kelapa muda dan madu dengan dosis 3% berpengaruh terhadap motilitas spermatozoa ikan nila selama 7 hari penyimpanan. Penelitian ini dapat digunakan sebagai referensi untuk pengawetan spermatozoa.

Abstract

Hypercholesterolemic, a condition characterized by excess cholesterol content, can lead to coronary heart disease, atherosclerosis, and high-risk heart tissue degeneration. Current drugs, like simvastatin and pravastatin, focus on lowering cholesterol levels without addressing tissue degeneration. This study aimed to determine the effectiveness of sidempuan salacca vinegar in improving cardiac tissue degeneration in hypercholesterolemic mice. The experimental research involved two groups, the treatment group given different doses of sidempuan salacca vinegar and the control group. Histopathological observations showed improvement in cardiac tissue degeneration in hypercholesterolemic mice after treatment with salacca vinegar. The administration of salacca vinegar could serve as a reference for developing alternative hypercholesterolemic drugs that address both cholesterol levels and heart degeneration.

Abstract

EDNRB gene is a gene located on chromosome 13q22, plays a role in the signaling pathway during the development of the ENS intestinal nerves. Mutations in the EDNRB gene in humans are associated with Hirschsprung Disease (HD). Mutations in the EDNRB gene are the second most common cause after the RET gene in cases of HD with a percentage of 5–10%. To detect mutations, PCR products are needed. Primers are one of the basic components of PCR, while optimization of annealing temperature and optimization of primer concentration are factors in the success of primer attachment to the target gene. This study aims to design EDNRB exon 4 gene primers and determine the optimum annealing temperature and primer concentration to amplify the EDNRB gene. Primer design with Geneious Prime, and good primer criteria are analyzed in silico. Optimization of annealing temperature and primer concentration is carried out with gradient PCR and variations in primer concentration. The results of this study obtained the best primers, namely the forward primer 5'- CAGTAAGTGTGGCCTGAAAG-3' and the reverse primer 5'-GTGGAACCGAAGTGACTAGA-3', which amplified the EDNRB gene exon 4. The optimum PCR conditions for amplification of exon 4 of the EDNRB gene were at an annealing temperature of 57.2 ℃ and at a primer concentration of 0.5 μM.

Abstract

Consumers have the right to correct information about the products they consume. One example of a food product that is vulnerable to counterfeiting is chicken nuggets. To overcome this problem, qPCR can be an effective solution. Previous research has designed primers targeting the chicken ND5 gene. However, the PCR conditions for this primer pair have never been optimized. This research aims to optimize qPCR conditions and carry out chicken nugget authentication trials. Genomic DNA was isolated from five weight variations of chicken meat samples as a qPCR standard curve and three chicken nugget samples. Optimizing the annealing temperature with gradient PCR gave the best results at a temperature of 57.6OC. The results of the authentication test showed that the chicken meat content in TD nuggets was 54.7%, C nuggets 43.8%, and D nuggets 37.3%. This value is still in accordance with the Indonesian National Standard (SNI).

Abstract

Teratogen berefek membahayakan terhadap perkembangan embrio. Tujuan penelitian ini untuk mengetahui pengaruh teratogen AlCl3 terhadap perkembangan embrio ayam (Gallus gallus domesticus). Metode penelitian ini dilakukan secara eksperimental dengan perlakuan induksi AlCl3 0,2 mg/L dan kontrol (tanpa perlakuan). Pemberian perlakuan diberikan kepada masing-masing 5 butir telur ayam yang kemudian diinkubasi selama 48 jam. Pengamatan preparat embrio ayam menggunakan whole mount method dengan pewarnaan metilen blue dan eosin. Hasil penelitian menunjukkan ukuran embrio pada kelompok perlakuan lebih kecil (0,6 cm) dibandingkan dengan embrio kontrol (1cm). Jumlah somit pada kelompok perlakuan sejumlah 12 pasang sedangkan pada kelompok kontrol 28 pasang somit. Ukuran kepala lebih kecil jika dibandingkan dengan kelompok kontrol. Penelitian ini menunjukkan bahwa AlCl3 dapat menyebabkan malformasi pada embrio ayam, terkait dengan ukuran embrio, jumlah somit, dan ukuran kepala.
