Abstract

Kuantitas dan kualitas hasil ekstraksi DNA dari jaringan FFPE sangat tergantung dari proses awal yang harus dilakukan pada sampel sebelum masuk kedalam tahapan ekstraksi DNA. Proses awal yang sangat menentukan keberhasilan ekstraksi DNA ini adalah proses deparafinisasi, yaitu proses penghilangan parafin jaringan dari parafin. Penelitian ini bertujuan untuk membandingkan kualitas DNA yang diambil dari sampel arsip FFPE (Formalin-fixed paraffin-embedded tissue) dengan metode deparafinisasi menggunakan xylene dan mineral oil di RSUP Dr. Mohammad Hoesin Palembang. Sebanyak 16 sampel arsip FFPE diambil dari Laboratorium Patologi Anatomi RSUP dr. Mohammad Hoesin Palembang. Seluruh sampel kemudian  dibagi menjadi 2 bagian sama rata kemudian dilakukan proses deparafinisasi menggunakan xylene dan mineral oil. DNA yang telah diisolasi akan diuji kuantitasnya menggunakan spektrofotometer dengan panjang gelombang 260nm.  Uji kualitas akan dilakukan dengan menghitung OD Ratio 260nm:280nm. Selain itu DNA juga akan diamplifikasi lalu dilakukan visualisasi dengan sinar UV. Konsentrasi yang didapatkan kemudian diuji dengan uji Wilcoxon dan menunjukkan adanya hubungan signifikan antara metode deparafinisasi dan konsentrasi yang didapat (P=0.036). Primer beta-actin (524bp)tidak ditemukan potongan gen yang diinginkan setelah elektroforesis, kemudian diuji dengan primer Mitin (142bp) menunjukan potongan gen yang diinginkan pada saat elektroforesis dan visualisasi UV.Terdapat hubungan yang signifikan antara metode deparafinisasi FFPE dengan kuantitas dan kualitas DNA yang didapat.

Abstract

Karsinoma ovarium tipe epitel merupakan 75% dari semua kasus tumor ovarium. Di dunia kanker ovarium merupakan keganasan ginekologi peringkat ketiga dari keganasan saluran genital pada wanita. Tumor marker keganasan ovarium tipe epitel adalah CA-125 yang ditemukan meningkat pada hampir semua kasus tumor ovarium (80%). Diduga terdapat perbedaan peningkatan kadar CA-125 pada berbagai subtipe epitel tumor ganas ovarium yaitu serosum (37,5%), endometrioid (50%), musinosum (62,5%), undifferentiated (84,3%) dan clear cell (100%). Tujuan penelitian ini untuk mengetahui hubungan kadar CA-125 dengan subtipe tumor ganas epitel ovarium pada penderita yang dirawat di RSUP Mohammad Hoesin Palembang. Penelitian ini menggunakan studi observasional analitik dengan desain cross sectional. Populasi penelitian adalah semua penderita subtipe epitel tumor ganas ovarium yang melakukan pemeriksaan kadar CA-125 di Bagian Patologi Anatomi RSMH Palembang sejak Januari 2013 sampai September 2016. Dari 61 sampel berbagai subtipe epitel tumor ganas ovarium yang didominasi subtipe serosum (32,8%), diikuti subtipe musinosum (26,2%), disusul oleh subtipe endometrioid (19.7%), clear cell (14,8%), borderline musinosum (4,9%) dan subitpe brenner (1.6%). Kadar CA-125 meningkat dijumpai delapan kali lipat (88,5%) lebih banyak dibanding subjek dengan kadar normal (11,5%). Hasil uji one-way ANOVA menunjukkan tidak terdapat perbedaan antara kadar CA-125 dengan masing-masing subtipe epitel tumor ganas ovarium. Hasil uji statistik Chi-square menunjukkan bahwa tidak terdapat hubungan yang bermakna antara kadar CA-125 dengan subtipe tumor ganas epitel ovarium. Tidak terdapat hubungan yang bermakna antara kadar CA-125 dengan subtipe tumor ganas epitel ovarium di RSMH  Palembang.

Abstract

COVID-19 secara dominan mempengaruhi jenis kelamin pria dan usia lanjut dengan komorbiditas, terutama penyakit paru-paru, jantung, arteri hipertensi (AH) dan diabetes mellitus (DM). Penderita DM sangat riskan jika terinveksi COVID-19. Pada kegiatan pengabdian pada masyarakat kali ini dilakukan  penyuluhan dan pelatihan untuk optimalisasi Pencegahan Covid-19 Pada Penderita Diabetes Melitus Melalui Edukasi dan Pelatihan Formulasi Minuman Kesehatan Kunyit, Susu, Kayu Manis (Kusuka) Untuk Meningkatkan Sistem Imun. Metode pelaksanaan kegiatan meliputi Metode pelaksanaan pada kegiatan kali ini mencakup tahapan persiapan, pelaksanaan kegiatan dan dan evaluasi pelaksanaan kegiatan. Berdasarkan Hasil pelaksanaan kegiatan menunjukkan bahwa persiapan kegiatan berjalan dengan baik. Diskusi yang dinamis antara pemateri dan warga ketika pelaksanaan kegitan menunjukk antusiasme warga yang cukup tinggi serta hasil evaluasi yang diberikan pada warga melalui wawancara menunjukka bahwa warga dapat memahami mengenai pencegahan Covid-19 bagi Penderita Diabetes Melitus dan terampil dalam membuat minuman kesehatan untuk meningkatkan system Imun. Sehingga dapat disimpulkan bahwa melalui kegiatan sosialisasi dan pelatihan dapat meningkatkan wawasan dan pengetahuan warga, serta keterampilan warga dalam formualsi minuman kesehatan dari susu dan rempah untuk meningkatkan sistem imun.

Abstract

Background: The stem cell transplantation successful influenced by the quality of the umbilical cord blood which includes the number of CD34 + and Total Nucleated Cells (TNC). Objectives: The study aims to determine the correlation between umbilical cord length and it’s diameter, with the number of CD34 + and TNC cells as indicators of the quality of cord blood storage feasibility. Several other factors as maternal age, gestation period, and infant birth weight also examined. Methods: Thirty four of umbilical cords from the delivered woman in Dr. Mohammad Hoesin Hospital (RSMH) Palembang were collected from May to June 2018. The length and diameter of the cord were immediately measured after delivery. The evaluated cells were counted with a flow cytometer at Klaster Stem Cell and Tissue Engineering Research Centre (SCTE) IMERI Faculty of Medicine University of Indonesia (FKUI). Results: Spearman correlation test show that there was no correlation between the length and diameter of the umbilical cord, maternal age, gestational period and infant weight, with the number of CD34 + and TNC cells in the cord (p> 0.05). However, the profile analysis indicated that the longer and larger the diameter of the umbilical cord, the higher concentration of the CD34+ and TNC cells. Conclusion: This study suggested that the younger maternal age, older gestational age, and higher infant birth weight, also normal hemoglobin level, tend to increase the number of CD34+ and TNC cells in the cord blood.

Abstract

Background : The estimated inci­dence of Acute Kidney Injury (AKI) is two to three cases per 1,000 patients. Seven percent of hospitalized patients and about two-thirds of patients in intensive care units develop acute kidney injury and the mortal­ity rates range between 25 and 80 percent. Disruption in epithelial brush border of proximal tubular cell causes N-Acetyl-β-D-Glucosaminidase (NAG) to be released to the urine and the amount of enzyme could be directly correlated with tubular disruption. Objective : The aims of this research is to determine the sensitivity and specificity of NAG urine examination as an early biomarker for acute kidney injury. Methods : There’s 66 subjects who met the inclusion criteria. All the subject were checked for the NAG urine level with Cloud Clone kit and creatinine serum were also checked 48 hours after admission. Results : The results showed in the cut-off point of 7.98 Ng/mL, urine NAG has a sensitivity of 68.6% and specificity 77.4%, positive predictive value 77.42%, negative predictive value 68.57 % and accuracy of 72.73% Conclusions : The result shows that urine NAG examination is more sensitive and specific as an early biomarker for Acute Kidney Injury compared to creatinine serum. Abstract

A B S T R A C TIntroduction. Preeclampsia is one of the leading causes of maternal and perinatalmorbidity and mortality and is still a disease of theory. Klotho is a new gene, inseveral biological processes in the pathophysiology of preeclampsia that play a rolein regulating endothelial nitric oxide production, angiogenesis, production ofantioxidant enzymes and protection against endothelial dysfunction. The Klotho G-395A genotype AA promoter polymorphism is the cause of hypertension. This studyaims to determine the relationship of the Klotho G-395A promoter polymorphism tothe incidence of preeclampsia. Methods. This study is an analytical study with acase-control design. The research was conducted at Pembina community healthcentre of Palembang and the public hospital of Prabumulih in February - July 2020and involving 50 case group and 50 control group. To determine the genotype andallotype of the Klotho G-395A gene promoter polymorphism, using polymerase chainreaction examination. Result. The results showed that the risk factors for maternalage and maternal gestational age had a significant relationship with the incidence ofpreeclampsia (p-value 0.015; p-value 0.000). There was a significant relationshipbetween the Klotho G-395A genotype GA + AA promoter polymorphism and theincidence of preeclampsia (p-value 0.024; OR = 2.571; 95% CI = 1.122-5.895), whileallotypes in the study sample also had a significant relationship with the incidenceof preeclampsia. preeclampsia (p-value 0.025; OR = 1.978; 95% CI = 1.087-3.599).Conclusion: There is a significant relationship between the Klotho G-395A genepromoter polymorphism and the incidence of preeclampsia.

Abstract

Background. Endometriosis is a chronic gynecological disorder that generally affects women of reproductive age which is characterized by implantation of endometrial tissue, both glands and stroma outside the uterus. Research shows that the initial onset of endometriosis patients begins in adolescence. The high prevalence with atypical symptoms leads to long term delay in establishing the appropriate diagnosis. Further developments in the field of early endometriosis screening will greatly assist clinicians, especially general practitioners in areas with limited health facilities, in early detection of endometriosis. Methods. Using the electronic databases, comprehensive literature searches were conducted with the specific keywords. Full-text manuscripts published were reviewed for relevancy and importantly, reference lists were cross-checked for additional relevant studies. Results. Dysmenorrhea, chronic pelvic pain, dyspareunia, dysuria, dyschezia, and glycoprotein were found dominantly in all analyzed literature. The occurrence of endometriosis affects the functioning of women in multiple aspects of life. Endometriosis places a significant burden on teens and adult women, their families, and society as a whole. Conclusion. Combination of structured past history taking, and several biomarker level might be useful as a screening tool that easy to use and cost effective, as early diagnosis is expected to prevent further progression of endometriosis especially in adolescence.

Abstract

Neutrophil extracellular traps (NETs) are immune components found in a variety of pathological states. It has been shown to have either beneficial or harmful implications, depending on how it is controlled and has been particularly observed in three major scenarios: infection, autoimmune disease, and cancer. In this article, we compiled some of the roles of NETs in pathological conditions, as well as the benefits of targeting them for improved patient outcomes. The role of NETs were primarily positive in infectious disease, whether caused by bacteria, virus, or fungal infection. In non-infectious inflammatory scenarios, on the other hand, it's the complete opposite, with the effects being mainly deleterious and even worse than the original disease states. Targeting NETs directly or indirectly may help to prevent complications and improve patient outcomes. A plethora of compounds, including immunomodulators, anti-thrombosis, nicotinamide adenine dinucleotide phosphate (NADPH)/reactive oxygen species (ROS) inhibitors, nuclease, and other compounds, may be used to accomplish the therapeutic goals.Keywords: autoimmune disease, cancer, DNase, infection, neutrophil extracellular traps
