Abstract

Keinginan hidup sehat dengan mengonsumsi makanan dan minuman alami menjadi bagian gaya hidup masyarakat. Jeruk menjadi salah satu buah yang menjadi makanan fungsional untuk menjaga dan memelihara kesehatan. Tujuan penelitian ini adalah mengetahui perbedaan aktivitas antioksidan dan antimikroba pada buah Jeruk Lemon(Citrus limon) dan Jeruk Nipis(Citrus aurantiifolia). Metode penelitian adalah penelitian eksperimen laboratorium dengan analisis deskriptif. Penelitian ini dilakukan pada bulan Februari-April 2017 di Laboratorium Mikrobiologi dan Kultur Jaringan Tumbuhan Jurusan Biologi Universitas Negeri Malang. Screening fitokimia dengan metode reaksi warna, total fenol dengan metode Folin Ciocalteu, aktivitas antioksidan dengan metode DPPH dan aktivitas antimikroba dengan metode Cakram. Hasil uji screening menunjukkan adanya saponin dan alkaloid tetapi tidak terdapat flavonoid, terpenoid dan tanin. Uji total fenol didapatkan kandungan total fenol pada Jeruk Lemon(Citrus limon) 110,25 mg GAE/ 100ml sedangkan pada Jeruk Nipis(Citrus aurantiifolia) 116,5 mg GAE/ 100ml. Aktivitas antioksidan Jeruk Lemon(Citrus limon)49.593g/ml dan Jeruk Nipis(Citrus aurantiifolia)49.589g/ml. Uji aktivitas antimikroba diperoleh luas zona hambatan yang tertinggi pada konsentrasi 100% dari masing-masing buah jeruk. Kesimpulan dari penelitian ini adalah ada perbedaanaktivitas antioksidan dan antimikroba pada kedua jeruk, dimana buah jeruk lemon (C.limon) aktivitas antioksidannya lebih tinggi daripada dan Jeruk Nipis(C. aurantiifolia), sedangkan aktivitas antimikroba jeruk Nipis(C. aurantiifolia)lebih tinggi daripada jeruk lemon (C.limon).

Abstract

Paradigm change in 21st century poses a challange for higher education in teaching science as inquiry. In another side, teaching science as inquiry meet some problem because of every student in the classroom was different. This research is aimed to know effectiveness of differentiated science inquiry based on research results of gene variation analysis in genetics course to increase students cognitive learning outcome. This research used pre-experimental research design on one class that consist of 26 biology students. Statistical analysis used in this study is paired sample t-test on pre-test and post-test to measure students cognitive learning outcomes. If there are significances between the pretest and posttest, data analysis was continue to N-Gain score to determine the increasing category of students cognitive learning outcome. The average pretest score was 64,3 and posttest score was 77,2. The statistical analysis result showed there was a significant difference between students cognitive learning outcome (p<0,005), meanwhile the N-Gain score between pretest and postest is 36,2 and categorized a medium criteria. That result mean although there are difference students learning outcome before and after learning by using differentiated science inquiry model based on research result of gene variation analysis, that learning is not too recommended to increase students cognitive learning outcome.

Abstract

This study aims to reveal the anticancer mechanism of the bioactive compound of Pisangulin angulata and to assess the feasibility of that results as teaching material. This research includes descriptive explorations. In the first stage, an in silico analysis was performed by molecular docking method between physalin compounds and GLI1 protein. The second steps of this study aim to develop teaching materials based research using the Analysis, Reorganizing, Piloting Class, and Evaluating (ARPE models). Feasibility test was carried out by experts and practitioners. Think Pair Share was used in the pilot project. Student motivation and misconception were recorded using SMI and CRI instrument. This study reveals that physalin B has higher activities than controls. The type of chemical bond that is formed between GLI1 amino acids residues with physalin is hydrogen bonds and hydrophobic bonds. The visualization of the types of bonds in that molecular docking between GLI1 amino acid residues and physalin has a high degree of feasibility (89) and can be used to enrich Chemistry for Biology lectures. The visualization of these chemical bonds can increase learning motivation and can improve the understanding of the concept of chemical bonds.

Abstract

Penelitian ini bertujuan untuk menganalisis potensi senyawa repensol dalam menginhibisi replikasi virus hepatitis B (HBV) melalui pendekatan in silico dengan menggunakan perangkat komputasi dan perangkat lunak (software). Analisis dilakukan dengan metode simulasi penambatan molekular (Molecular Docking Simulation) yang menggunakan software PyRx, serta divisualisasikan dengan PyMol dan Discovery Studio. Data yang digunakan dalam penelitian yaitu struktur senyawa repensol, protein target berupa protein kapsid HBV, dan ligan referensi berupa 4-methyl heteroaryldihydropyrimidine (4-metil HAP) yang dikoleksi dari database PubChem dan PDB. Hasil docking menunjukkan bahwa ikatan yang terbentuk antara senyawa repensol dengan protein kapsid HBV memiliki nilai binding affinity lebih rendah dari ligan referensi, yaitu -7.0 kkal/mol. Visualisasi menunjukkan senyawa repensol membentuk binding site yang sesuai dengan ligan referensi dan melibatkan 13 residu asam amino dari protein kapsid HBV, yaitu PRO D:138, ILE D:139, SER D:141, PHE D:110, THR D:109, LEU D:37, THR D:33, ILE D:105, PRO D:25, TYR D;118, TRP D:102, LEU D:140, dan PHE D:23. Jenis ikatan yang terbentuk antara senyawa repensol dengan protein kapsid HBV yaitu ikatan van der Waals, hidrogen konvensional, Pi-Sigma, Pi-Pi stacked dan Pi-alkil.

Abstract

Mata kuliah evolusi merupakan salah satu mata kuliah yang dianggap sulit oleh mahasiswa, sulitnya pemahaman mahasiswa terhadap konsep evolusi disebabkan oleh beberapa masalah sehingga perlu dilakukan beberapa observasi terhadap komponen-komponen dalam proses pembelajaran Evolusi. Penelitian ini bertujuan untuk menganalisis Keterampilan Berpikir mahasiswa terhadap Mata Kuliah Evolusi di Universitas Sulawesi Barat Penelitian ini menggunakan metode survey dengan cara penyebaran angket dan pemberian tes untuk mengetahui pengetahuan mahasiswa tentang konsep evolusi. Berdasarkan survey yang dilakukan terhadap mahasiswa STKIP Yapim Maros Program Pendidikan Biologi dengan 36 orang responden menunjukkan bahwa Keterampilan Berpikir Kritis Mahasiswa STKIP Yapim Maros. Berdasarkan hasil analisis yang didapatkan, maka dapat disimpulkan bahwa keterampilan berpikir mahasiswa biologi STKIP Yapim Maros terhadap mata kuliah Evolusi masih rendah. Oleh karena itu perlu dilakukanpembenahan dalam penelitian lanjutan.Kata kunci: Berpikir Kritis, Evolusi, Pembelajaran

Abstract

Keterampilan berpikir analitis merupakan kemampuan yang harus dikuasai mahasiswa untuk menyelesaikan masalah dalam biologi. Penelitian ini merupakan penelitian deskriptif yang bertujuan untuk mendeskripsikan kemampuan berpikir analitis mahasiswa. Sejumlah 38 mahasiswa yang telah menempuh mata kuliah biologi umum menjadi subyek penelitian. Instrumen yang digunakan pada penelitian ini berupa tes tipe uraian yang mengacu pada keterampilan berpikir analisis yang terdiri dari membedakan (differentiating), mengorganisasi (organizing), dan menghubungkan (attributing) masing-masing indikator 3 soal. Berdasarkan analisis data dideskripsikan kemampuan analisis mahasiswa pada t indikator membedakan 3,01 (baik), mengorganisasikan 2,93 (baik), dan menghubungkan 2,47 (kurang baik). Indikator menghubungkan perlu mendapatkan perhatian lebih.Kata Kunci: keterampilan berpikir analitis, profil, dan biologi umum.

Abstract

This study aims to determine the various kinds of compounds from the saponin group found in local Indonesian garlic and map the biological potential of these compounds, especially the potential in the health sector. Saponins are a group of compounds characterized by the presence of foam. Saponins have various benefits both in the health, agriculture, animal husbandry, and also in the food and cosmetic industries. Fifteen compounds from the saponin group were identified from local Indonesian garlic obtained from West Papua, using Liquid Chromatography Mass Spectrometry (LCMS) and validated based on their similarity with the NCBI PubChem database. Saponin compounds in local Papuan garlic, then, their biological potential was mapped using the PASS Server and showed that there were seventeen kinds of biological potential in the saponin compound group from Indonesian local garlic. The three most dominant potencies are as anticancer, antifungal, and hypolipemic. Based on this research, it can be concluded that Indonesian local garlic has great potential to be developed and applied in the health sector, especially as an anticancer related to the saponin content in tubers.

Abstract

A high-fat-enriched diet causes an increase in the level of oxidized LDL (Ox-LDL) in the blood. The presence of Ox-LDL will activate macrophages to secrete pro-inflammatory cytokines and lead to severe inflammation. Single bulb garlic has a potential anti-inflammatory effect due to of high-fat diet. This research aimed to investigate the effect of single bulb garlic extract (SBGE) on the pro-inflammatory cytokines TNF-α (CD11b+TNF-α+) and IL-1b (CD11b+IL-1b+) in the spleen, spleen weight, and TNF-α secretion in HFD mice. Twenty-four mice were divided into six groups: normal (healthy mice); HFD (HFD mice without any treatment); HFD + Simvastatin (HFD mice receiving simvastatin); HFD + SBGE 100; HFD + SBGE 200; and HFD + SBGE 400 (HFD mice receiving 100, 200, and 400 mg/kg BW of SBGE for 4 weeks). Blood serum was collected at the end of treatment, and macrophage was isolated from the spleen. The relative number of CD11b+TNF-α+ and CD11b+IL-1b+ were examined using flow cytometry. SBGE treatment significantly (p<0.05) reduced the spleen weight and the relative number of CD11b+TNF-α+ and CD11b+IL-1b+ in the spleen of HFD mice. SBGE treatment also prevents the elevation of TNF- α levels in the blood serum. The optimal dose of SBGE to diminish the relative number of CD11b+TNF-α+, CD11b+IL-1b+ in the spleen, and TNF-α in the serum was 100 mg/kg BW.

Abstract

The objectives of this research were to examine the effect of Sechium edule ethanolic extract to repair beta pancreas cells damage and tomeasure nitrogen oxide (NO) concentration in streptozotocin-induced rats. Type I diabetic rats were obtained by intraperioneally injected therats with multiple low dose streptozotocin (MLD-STZ) for 5 days. In this research, rats were assigned into 5 treatment groups: untreated rats ingroup 1 were considered as negative control, while MLD-STZ induced type 1 diabetic rats in group 2 were considered as positive control. Inanother 3 groups, type 1 diabetic rats were orally treated with three doses of Sechium edule ethanolic extract (14, 28, 42 mg/kg bw) respectivelyfor 7 consecutive days. Repairing of beta pancreas cells were analyzed descriptively whereas concentration of NO were analyzed using one wayANOVA and continued with LSD test (α= 0.01). The result showed that the administration of Sechium edule ethanolic extract significantlydecreased NO concentration (P0.01) up to 67.9 µM and repaired beta pancreas cells damage. Dose of 42 mg/kg bw of Sechium edule ethanolicextract is the effective dose for diabetic rat therapy. Key words: Sechium edule extract, diabetic mellitus, rat, beta pancreas cells

Abstract

Cayenne pepper is consumed as a food spice because of its color, spicy taste, aroma and complete nutritional value. Cayenne pepper sold in traditional markets is not separated between whole cayenne pepper and broken cayenne pepper. This study aims to: 1) calculate the ALT of mold colonies on whole and damaged cayenne peppers, 2) identify contaminant mold species found in intact and damaged cayenne peppers. Samples of whole cayenne pepper and damaged cayenne pepper were isolated by grinding 10 grams of cayenne pepper, it was then inoculated on Potato Dextrose Agar (PDA) medium and incubated at 25oC for 7 x 24 hours. Then the ALT of mold colonies was calculated on whole and damaged cayenne pepper. Then, each isolated mold contaminating colony's morphology and microscopic characteristics were described. Finally, each contaminant mold isolate was identified. Furthermore, the study unpacked that: 1) The ALT of contaminant mold colonies on whole cayenne pepper was 1.0x103 colonies/g, while the average ALT of contaminant mold colonies on damaged cayenne pepper was 2.1x105 colonies/g, 2) There were 8 species of mold found in cayenne pepper namely Mucor racemosus Fres, Aspergillus niger van Tieghem, Penicillium griseofulvum Dierckx, Penicillium frequentans Westing, Penicillium citrinum Thom, Penicillium nalgiovense Laxa, Penicillium digitatum Sacc and Penicillium variabile Sopp.
