Abstract

Model Pembelajaran Project Based Learning ialah Model Pembelajaran yang berpusat pada kegiatan pembuatan proyek yang dilakukan oleh peserta didik. Model Pembelajaran ini dapat membantu Peserta didik untuk lebih memahami materi yang diajarkan di kelas dengan melakukan kegiatan praktik secara langsung di kehidupan nyata. Penelitian yang dilakukan peneliti ini bertujuan agar peserta didik mampu mencapai ketuntasan dalam mata pelajaran biologi khususnya pokok bahasan pertumbuhan dan perkembangan makhluk hidup dengan menerapkan model pembelajaran PjBL. Penelitian in berjenis penelitian tindakan kelas. Penelitian ini dilakukan di SMA Negeri 1 Kabaruan. Subjek penelitian yang digunakan yaitu Peserta didik kelas XII IPA yang berjumlah 22 orang. Hasil penelitian menunjukkan presentasi ketuntasan belajar pada siklus I sebesar 5% dengan rata – rata 41 dan pada siklus II menjadi 82% dengan rata – rata 78,9. Hasil penelitian ini menunjukkan bahwa materi pembelajaran pertumbuhan dan perkembangan makhluk hidup dengan menggunakan model pembelajaran Project Based Learning dapat meningkatkan hasil belajar peserta didik dan mencapai ketuntasan di kelas XII SMA Negeri 1 Kabaruan.

Abstract

Binocular light microscopy (BLM) is an excellent match for a scanning electron microscope (SEM) and a trinocular light microscope equipped with a micrometer (TLM). The practicality, user-friendliness, and short-time analysis of BLM make this method a good choice for spicule analysis. However, its effectiveness and accuracy are yet to be confirmed. This study aimed to validate the effectiveness of BLM by comparing its usefulness to both TLM and the gold standard methods. BLM was first subjected to measuring megascleres and microscleres of 2 sponges. Then, by using the If function built-in Excell and t-test in SPSS 16.0, the compatibility of BLM was evaluated against SEM by measuring the length of spicules from 4 Sangihe sponges and their counterpart species from different locations. Furthermore, the t-test analysis was used to validate the compatibility and effectiveness of our method to the TLM by measuring the spicules of four sponges. Both the F-function and the t-test analysis proved BLM was compatible with SEM with both measurements showing a perfect match for megascleres typed spicules of 4 compared sponges. This new technique also showed a perfect match with SEM (p = 0.367, t-test) and with TLM (p = 0.963, t-test). Keywords: Spicules, sponges, SEM, Wallacea, biomaterial, sponge taxonomy

Abstract

Together with bacterial and viral infection, fungal infection represents the world's top ten killer diseases, desperately requiring new antifungal drugs. This research aimed to evaluate the antifungal activity of the sponge Styllisa flabelliformis against the pathogenic fungi Aspergillus flavus and Aspergillus parasiticus by the standard agar diffusion technique. Three concentrations (1, 10, and 10 mg/mL) for the extract and 0.5 mg/mL for ketoconazole and fluconazole were prepared and evaluated in triplicate against the tested fungi. Whereas ketoconazole poorly inhibited A. parasiticus and fluconazole weakly inhibited A. flavus, the extract of S. flabelliformis exerted antifungal activity against A. parasiticus (6.8 ± 1.8; 8.3 ± 3.2; and 9.5 ± 2.1) mm and A. flavus (6.8 ± 1.1; 11.5 ± 1.4; and 14.3 ± 1.1) mm 1, 10 and 100 mg/mL respectively. PASS analysis showed jasplakinolide as a promising antifungal agent with potential activity (Pa) of 0.736. STITCH analysis further confirmed that jasplakinolide worked by inhibiting the expression of cytoskeleton genes that prevented fungi from synthesizing chitin and inhibiting the formation of the fungi’s cell walls and hyphae, different from the ergosterol synthesis inhibition in ketoconazole and fluconazole, implying the potential of jasplakinolide as an antifungal agent.

Abstract

Kecamatan Atinggola merupakan salah satu kecamatan dalam wilayah Kabupaten Gorontalo Utara yang memiliki potensi penghasil kelapa dengan luas areal perkebunan 4,563 ha dan jumlah produksi kelapa 9,080 ton/ha/ tahun. Produk samping dari pengolahan minyak kelapa berupa ampas minyak kelapa atau blondo sekitar 60% dari total bahan baku santan yang digunakan dan hanya dimanfaatkan sebagai campuran pada bahan makanan tertentu seperti tambahan pada tumisan sayur atau sambal padahal blondo mengandung protein 24,22% yang dapat dimanfaatkan sebagai bahan baku pembuatan kecap manis. Kegiatan diseminasi dan pengolahan kecap manis berbahan baku blondo dilakukan bersama dengan mitra sasaran Kelompok Tani IKM F2-1 Dusun Harapan, Kecamatan Atinggola dengan metode sosialisasi, diseminasi dan pengolahan kecap yang melibatkan peran aktif dari peserta dan kelompok mitra dimana produk kegiatan berupa produk kecap manis berbahan baku blondo serta kegiatan ini dapat meningkatkan pengetahuan dan ketrampilan mitra dalam pengolahan kecap berbahan baku blondo

Abstract

Suatu kegiatan Pemberdayaan Kemitraan Masyarakat (PKM) telah dilaksanakan untuk memberdyakan masyarakat berwirausaha tani terpadu guna menunjang pariwisata pantai desa Marinsow Kecamatan Likupang Timur Minahasa Utara. Metode yang digunakan adalah kombinasi metode kombinasi metode Andragogik dan Pilot Project aplikasi teknlogi tepat guna usaha tani terintegrasi dengan mengoptimalkan lahan dan usaha tani Mitra berada persis di tepian jalan utama menuju kawasan wisata pantai Paal desa Marinsow. Berdasaran kegiatan sampai saat ini diperoleh hasil yaitu, (1) terfasilitasi upaya menghadirkan satu unit cultivator untuk revitalisasi pengolahan lahan pertanian, (2) terfasilitasi pengadaan mulsa plastic hitam perak (MPHP) dan peralatan pertanian lainnya untuk menunjang usaha budidaya tanaman pangan, (3) terfasiitasi pemasangan tower dan pengadaan satu unit tangki air untuk pemenuhan irigasi lahan pertanian saat musim kemarau, (4) terfasilitasi peningkatan pengetahuan dan keterampilan mitra dalam pengolahan lahan menggunakan cultivator, (5) terfasilitasi peningkatan pengetahuan dan keterampilan mitra memproduksi dan mengemas aneka produk kacang tanah menggunakan vacuum sealer dengan plastic vacuum emboose. Abstract

Desa Imana terletak di Kab.Gorontalo Utara memiliki tanaman kelapa yang sangat melimpah dan menjadi salah satu sumber penghasilan masyarakat disekitarnya. Tujuan dari kegiatan ini memberikan pelatihan kepada warga dalam pengolahan kelapa guna menghasilkan minyak kelapa secara tradisional dan berkualitas pada desa Imana kecematan Atinggola, Kab Gorontalo Utara. IKM Mikas melakukan proses pembuatan minyak kelapa secara tradisional dengan metode cara basah berbeda dari pembuatan minyak pada umumnya, adapun tahapan proses pembuatan melalui sortasi bahan baku, pengepresan santan, pemeraman santan selama 24 jam, ekstraksi santan melalui pemanasan hingga diperoleh minyak murni kelapa. Berdasarkan studi literatur minyak kelapa tradisional mengandung 5 jenis asam lemak yaitu asam laurat (44,83%), asam miristat (22,83%), asam palmitat (12,82%), asam oleat (11,68%), dan asam stearat (3,90%). Minyak kelapa yang dihasilkan IKM Mikas dari segi aroma dari sisi aroma yang jauh lebih harum dibanding dengan produk-produk pabrikan. Menurut Bapak Supanto Patingki yang merupakan Ketua IKM Mikas, setiap bulannya IKM Mikas memproduksi minyak kelapa kampung sebanyak 4 kali (sepekan sekali) Setiap produksi menghasilkan 100 botol

Abstract

Indonesia is ranked 7th out of 10 countries with the highest number of people with diabetes mellitus, which is estimated to increase every year.α -amylase is an enzyme that catalyzes the hydrolysis of α-1,4 glycosidic bonds of polysaccharides to produce dextrins, oligosaccharides, maltose and D-glucose. Apis dorsata Binghami honeycomb contains secondary metabolites that have antidiabetic potential, including inhibiting the action of the α-amylase enzyme. The purpose of this study was to determine the in vitro antidiabetic potential of Apis dorsata binghami nest extract. This study uses a descriptive research method where the research data are obtained through laboratory experiments. Honeycomb extraction using maceration method. Analysis of total flavonoid content using UV-Vis spectrophotometric method. Antidiabetic potency test using -amylase enzyme inhibition method. The results of the extraction study showed that the % yield of the extract was 11.82% with a brownish yellow color. Analysis of the total flavonoid content showed the results of 3.33 mgQE/g. The inhibitory activity of the -amylase enzyme, the IC50 value of the extract obtained was 158.48±7.42 g/mL, the acarbose value was 165.96±7.08 g/mL. These results indicate that the extract of A.dorsata Binghami nest has hyperglycemic activity by inhibiting complex carbohydrate hydrolyzing enzymes such as inhibition of the -amylase enzyme which is better than acarbose because the IC50 value of the extract is smaller. If the IC50 value is smaller then the enzyme inhibition is stronger. For further research, it is recommended to use a more specific enzyme, namely the -glucosidase enzyme.

Abstract

Indonesia is known with its high biodiversity. One of them is sea cucumber. Various species of sea cucumbers are found in Indonesian sea in Talaud Regency at Kabaruan Island. Identification of sea cucumbers to reveal the biodiversity of sea cucumbers in Kabaruan Island, which is almost unknown. This research was to identify six samples of sea cucumber (Holothuroidea) in Kabaruan Island based on morphology and DNA barcoding using the COI gene. The differences between each species can be seen from the morphological characteristics, the length and width of sea cucumbers, papillae, pattern and color. The results showed that there were 4 species from 6 samples, which is Holothuria leucospilota, Holothuria whitmaei, Eleutherozoa sp., and Holothuria atra. That six samples with the code A1, A2, B1, C1, and C2 have a homology level of 99%-100% with the target nucleotide sequence from GenBank. Meanwhile, the sample’s code A3 has a homology level of 90% with the Eleutheroza sp. so there is a possibility that sample A3 is a new taxa that has the highest similarity to the Eleutheroza sp. From the six samples, it was found that samples A2 and B1 had the highest level of similarity which is 100%. Meanwhile, the samples with the lowest percentage values are samples A3 and B1 and samples A2 and A3, with a percentage value of 82.75%. There were 4 species from six samples found in Kabaruan Island. There is a possibility that Sample A3 is a new species that is similar to Eleutheroza sp.

Abstract

Chicken meat (Gallus gallus domesticus) plays an important role in society with its role as one of the sources of fulfillment of animal protein in Indonesia. With the high consumption of chicken meat by the public, the hygiene and safety of chicken meat must be ensured so as not to incur losses to the people. This study aimed to determine the bacterial contamination based on the testing of Total Plate Counts, Coliform, Escherichia coli, and Salmonella sp. Meet the requirement of the Indonesian National Standard on chicken meat in the traditional market of Langowan Selatan District. The method used in this research is a descriptive method to analyze and describe the results of the study, the testing using several indicators to directly conclude bacterial contamination. The results of the total plate count bacterial contamination test of the colonies, TPC in sample 1 = 29,1x kol/, sample 2 = 29,4x kol/, sample 3 = 27,7x kol/. MPN of Coliform and Escherichia coli results obtained in samples S1, S2, and S3 are 11x kol/gr. The results of microbact Salmonella sp. In samples S1 are positive, and S2 and S3 are negative. Based on observations, it is suspected that contamination occurs because sellers use equipment that is less sterile and there is no cover or barrier so the chicken meat is in direct contact with the air and customer. It is concluded that based on bacterial testing indicators, chicken meat in the traditional market of Langowan Selatan District has exceeded the limit of SNI 7388 2009 and is contaminated.

Abstract

Suatu kegiatan pengabdian kepada masyarakat melalui Program Pemberdayaan Masyarakat oleh Mahasiswa (PMM) Aplikasi Teknologi Budidaya Tanaman Hortikultura di Kelurahan Kelapa Dua Bitung telah dilaksanakan dengan baik. Kegiatan PMM ini tersebut bertujuan untuk menumbuhkembangkan jiwa berwirausaha siswa Sekolah Menengah Atas Negeri 3 Bitung dengan merevitalisasi usaha budidaya tanaman hortikultura dengan mengintroduksi teknologi budidaya dengan menggunakan system Aquaponic. Secara khusus (1) menciptakan usaha pertanian tanaman pangan dengan mengoptimalkan pemanfaatan halaman sekolah. (2) menghasilkan program aksi menciptakan usaha tani dengan memanfaatkan halaman sekolah dalam upaya menanamkan jiwa berwirausaha sekaligus menjadi media pembelajaran sains bagi siswa. (3) meningkatkan pengetahuan dan keterampilan kecakapan hidup (life skill) melalui usaha budidaya aquaponik bagi anak usia sekolah di SMA Negeri 3 Bitung. Metode dan pendekatan yang diterapkan dalam pelaksanaan kegiatan PMM yaitu metode Project Based Learning berbasis wirausaha budidaya hortikultura dan ikan dalam upaya revitalisasi implementasi program P5 tema Kewirausahaan. Metode Project Based Learning pada dasarnya merupakan pembelajaran yang memberikan kesempatan kepada mitra (siswa SMA) melatih kemampuan analisis untuk menyelesaikan masalah juga kegiatan ini menjadi laboratorium dan sumber belajar bagi siswa untuk mata pelajaran IPA dan Pendidikan Ekonomi. Hasil kegiatan yaitu, terfasilitasi percontohan budidaya tanaman dengan system aquaponik di samping sebagai upaya menunjang pemenuhan kebutuhan konsumsi juga sekaligus sebagai media pembelajaran dan implementasi Projek Penguatan Profil Pelajar Pancasila tema Kewirausahaan
