Abstract

Brassica rapa L., a member of the mustard green family, is a high-value crop with notable economic potential and nutritional benefits in Indonesia. Within the Brassicaceae family, commonly cultivated mustard varieties, such as mustard green, white mustard, and pakcoy, exhibit distinct differences in morphology and characteristics. Pakchoy is especially prized for its health benefits, primarily attributed to its dietary fiber content, which aids digestion and binds bile acids, reducing cholesterol levels. This study evaluated the effects of various liquid organic fertilizers (LOF) and pakchoy varieties on plant growth and yield. The research utilized a factorial Complete Randomized Design (CRD) with two main factors. The first factor was three types of LOF: rice water (L1), coconut water *L2), and banana stem (L3). The second factor consisted of three pakchoy varieties: Nauli (V1), Masbro (V2), and Gardena (V3). Observed parameters included plant height, leaf count, fresh weight, root count, and root length, recorded at 7, 14, 21, and 28 days after planting (DAP). Data were analyzed using ANOVA, with significant differences further tested by LSD at a 5% significance level. The results demonstrated that the rice water LOF (L1) combination and the Nauli variety (V1) produced the best outcomes across all growth parameters. These findings highlight the effectiveness of using rice water LOF alongside the Nauli variety as an optimized approach to improving pakcoy growth, yield, and overall agricultural productivity.

Abstract

Plastic waste represents a significant global environmental challenge due to its resistance to natural degradation and its widespread pollution impact. In Indonesia, the management of household plastic waste remains inadequate, largely due to limited public awareness and engagement. To address this issue, a community-based initiative was developed to build local capacity for managing plastic waste by creatively repurposing used water gallons into functional trash bins. This approach involved direct, hands-on training and ongoing mentoring of residents in Bangi, Woromarto Village, Purwoasri District, Kediri Regency. The effectiveness of the program was evaluated through participatory observation and systematic documentation of the products created by community members. The findings revealed a significant increase in community participation in recycling activities and the strategic placement of the repurposed trash bins throughout the village, thereby improving local waste management practices. Beyond providing a practical solution for reducing plastic waste accumulation, this initiative fostered positive behavioral changes by encouraging residents to adopt more responsible and sustainable practices centered on environmental stewardship and maximizing the value of discarded materials.

Abstract

Human papillomavirus (HPV) is a virus that causes infection on the surface of the skin and has the potential to cause cervical cancer. This viral infection is characterized by the growth of warts on the skin in various areas of the body, such as the arms, legs, mouth, and genital area. Because the virus can endanger health, it is necessary to design an HPV vaccine to overcome this problem. In this study, we performed a study characterization of HPV types 16 and 18 sequences to obtain immunogenic epitopes retrieved from the National Center for Biotechnology Information (NCBI) web server. Then, epitope prediction was performed using the immune epitope database (IEDB) web server and selected to get the best vaccine candidate for HPV types 16 and 18. We recommend 16P1 as an epitope-based peptide vaccine candidate for HPV type 16 and 18P4 for type 18. Both vaccine candidates are antigenic, non-allergenic, and non-toxic. The 16P1 and 18P4 have the lowest global energy values among the other candidates. However, further research is needed to be able to develop the best vaccine (in vitro and in vivo experiments).

Abstract

Human Immunodeficiency Virus type 2 (HIV-2) has been identified to exhibit an ability to resist antiretroviral administration and many scientists has predicted increases in the pathogenicity of HIV-2. The development of a vaccine against the type 1 virus (HIV-1) infection has reached the phase 3 clinical trial stage, but currently there is no information on the development of a vaccine against HIV-2. Vaccine development to trigger an increase in the coverage of the expansion of protection can be done through B cell polytope. This study aims to provide an important preliminary for the construction of vaccine candidates by identifying the peptides that make up the B cell polytope in the HIV-2 envelope glycoprotein region. The HIV-2 sequence was obtained from the database. The study followed by 3D modelling, prediction of linear B-cell epitope mapping, antigenicity, allergenicity, peptide properties, and immune simulation was carried out via a webserver. The 3D structure of the peptide was displayed through molecular visualization software. The results showed that the 23-mer peptides E1 'HPRYTGVKNIRDITLTEPGRGSD', F1 'NFIENRKGTQHN' 12-mer, M1 'YLKDQARLNS' 10-mer, N1 'PWVNDSIQPNWNNMTWQQWELQVRD' 25-mer, and O1 'KLQNSWNMGVQTO' can be used as a candidate polytope HIV-2 vaccine because it is recognized by B cells is an antigenic peptide with stable molecule, non-allergenic. The peptides trigger proliferation and activation of B cells to produces a humoral response and work as functionally protective antibody for neutralization of HIV-2. Abstract

Stelechocarpus burahol (kepel) belongs to the Annonaceae family, and is considered to be a native species to Indonesia which is mainly distributed on the island of Java. However, the plant’s existence is currently difficult to find, so it is categorized as rare in Indonesia. A molecular approach using DNA barcoding technique is significant to assist plant identification. The gene that is widely used and proven to be accurate for Annonaceae is trnL-F. This study was aimed to evaluate the efficiency of trnL as DNA barcode for the identification of S. burahol and its relatives (Annonaceae) from Java Island. In total 10 specimens have been used in this study. Whole genome DNA was isolated by Tiangen kit and amplified by Polymerase Chain Reaction (PCR) technique using a specific primer. Results showed that trnL was easily amplified with a DNA fragment length of 500-600 bp. The trnL amplicons have successfully sequenced as indicated by the high QV20+ values. The sequence compositions were high in AT bases (63.9%). BLAST analysis of the sequences showed that S. burahol and sister species have been confirmed its identity according to the reference sequences in NCBI with query cover identities 98%-100%. This research can be concluded that trnL-F is suitable and recommended as a DNA barcode for S. burahol and its relatives. However, further research is suggested to combine analysis of both coding (rbcL, matK, etc) and non-coding (trnL) markers for better identification results.

Abstract

The Annonaceae family exhibited a high level of diversity and is spread over many regions. The designation of the names of several species of the Annonaceae family varies in each region.  Therefore, characterization is needed to confirm and classify the taxonomy position of the species. This study aims to characterize the variation and analyze the clustering pattern of several species of the Annonaceae based on vegetative morphology characters. The type of method used in this research is exploratory descriptive, with seventeen specimen species consisting of ten Annonaceae studied in this research. Characters observed include tree age, stem diameter, plant height, stem color, type of sapling, type of branching, leaf shape, leaf tip shape, leaf base shape, leaf width, leaf length, leaf thickness, petiole length and thickness, leaf hairs , color of young and mature leaves, type of leaf edge, number of leaf veins, color of young branches, leaf hairs on young branches, and number of nodes on the stem per meter. These qualitative and quantitative characters were observed using Descriptors for Cherimoya (Annona cherimola Mill.) from Bioversity International and CHERLA. Data analysis was conducted in the Paleontological Statistics program with cluster analysis method (Bray-curtis similarity index) to produce a dendrogram topology. Results showed that there were variations in vegetative morphological characteristics among the ten species examined. Dendrogram topology was divided into 3 groups relatively following the tribe division, with similarity index 0.64 to 0.96. Xylopia sp. was separated from the others in Group 1 (Xylopiae). Group 2 (Uvariae) consisted of Fissistigma sp., Desmos chinensis, Artabotrys suaveolens, Uvaria purpurea, and Desmos sp. Group 3 (Annoneae) consisted of Annona muricata, Annona montana, Stelechocarpus burahol, and Annona glabra. Vegetative morphological characters have proven to be useful and effective in differentiating species in the Annonaceae family; it is recommended for initial identification of species, especially in the Annonoideae subfamily.
