Abstract

Mastitis is an inflammation of the mammary glands that can cause a sharp reduction in milk production and income for farmers. Subclinical mastitis is mastitis without clinical symptoms but can result in a decrease in the quantity and quality of milk production. One of the main pathogens that cause mastitis namely Staphyloccocus aureus. S. aureus become the biggest problem of mastitis because the cure rate is classified as very low. The risk factors for the occurrence of mastitis, both clinical and subclinical mastitis, can cause a decrease in milk production caused by several factors, including genetic, feed and environmental factors. This study aims to determine the presence of subclinical mastitis in milk of dairy goats with sufferers in Siliragung District, Banyuwangi Regency. Sample testing was carried out at the SIKIA Instrument Laboratory, Airlangga University. Isolation was carried out using MSA (Mannitol Salt Agar) media for further identification consisting of Gram staining and biochemical tests. Some of the biochemical tests used are the catalase test, the coagulase test and the VP test. The results of isolation, observation of characteristics and identification of bacteria in the research that has been carried out show that 64.51% (40/62) of the samples examined were positively identified S. aureus. Results showed that the risk factors for causing subclinical mastitis included cleaning the cage, the number of lactating goats, washing hands before milking, manure and waste management, weaning age and lactation age.

Abstract

This study aims to determine the relationship between factors and cases of repeated breeding in beef cattle in Klabang District, Bondowoso Regency. Beef cattle farming in Klabang District, Bondowoso Regency is currently experiencing problems in meeting the demand for beef due to low productivity. These problems occur due to cases of repeated breeding and decreased livestock efficiency and productivity. This case is suspected due to the lack of sanitation from farmers. This descriptive study used 69 cows with a simple random sampling method. Primary data was obtained from direct observation of livestock, followed by interviews with farmers using an in-depth study system. Meanwhile, secondary data was obtained from inseminator records which included the period of cultivation, the farmer's ability to detect the estrous cycle, and the farmer's knowledge of reproduction, housing, feeding, and copulation in beef cattle. The collected data were analyzed using descriptive statistics and then processed using SPSS 22.0. The Chi-Square test (X2) is used to determine the relationship between causative factors and repeated breeding. The results of this study indicate that the incidence of repeated breeding in Klabang District, Bondowoso Regency is 100% of the sample. All factors such as length of breeding, farmer's ability to detect estrous cycles, and farmer's knowledge about reproduction, housing, feed, age of livestock, and copulation in beef cattle correlate with the incidence of repeated breeding.

Abstract

This study aimed to evaluate the protective effects of α-tocopherol and zinc sulfate on oxidative stress parameters in ovarian granulosa cells of female Wistar rats (Rattus norvegicus) exposed to lead acetate (Pb(C₂H₃O₂)₂). A total of 25 rats were randomly divided into five groups: Control (C), lead exposure only (T0), α-tocopherol + lead (T1), zinc sulfate + lead (T2), and α-tocopherol + zinc sulfate + lead (T3). Treatments were administered orally for 21 days. Superoxide dismutase (SOD) levels were measured spectrophotometrically, and p53 expression was analyzed using immunohistochemistry. The results showed that SOD levels significantly decreased in group T0 compared to the control (p <0.05), while treatment with either α-tocopherol (T1), zinc sulfate (T2), or their combination (T3) significantly improved SOD levels compared to T0. Moreover, p53 expression was markedly elevated in T0, indicating enhanced oxidative stress and potential apoptosis, whereas all antioxidant-treated groups showed reduced p53 expression, with T3 demonstrating values comparable to the control group. In conclusion, the combination of α-tocopherol and zinc sulfate provided a synergistic antioxidant effect, effectively enhancing SOD levels and suppressing p53 expression in granulosa cells of lead-exposed rats. This suggested a promising therapeutic potential of these compounds in mitigating lead-induced ovarian toxicity.

Abstract

Goats are small ruminants that are reared by the rural community for financial income and nutrition. This study was aimed to determine the effect of vitamin C addition in tris–egg yolk extender on the lifespan of Sapera goat spermatozoa stored in 5o C. Semen was collected from 1.5 years old Sapera male goat. The ejaculates were diluted in Tris-egg yolk (T-EY) extender added with 0, 0.2, 0.3, and 0.4 g vitamin C/mL extender for T0, T1, T2, and T3 groups, respectively. The spermatozoa motility, viability, and morphological abnormality were assessed along with storage at 5° C. The result showed that spermatozoa motility was the highest (p <0.05) in T1 compared to other groups and qualified for AI use for up to 72 hours. In 24 hours storage, the spermatozoa viability was not significantly different (p >0.05) among the groups. The spermatozoa viability in the T1 group was the highest (p <0.05) among the groups along 48-96 hours of storage. The spermatozoa morphological abnormalities of the T1 group was the lowest (p <0.05) compared to other treatment groups in the range of 24-72 hour storage. In the control group (T0), the less than 5% spermatozoa morphological abnormalities (qualified for artificial insemination) were only in the 24 hours storage, while those of the T1 group were up to 72 hours. It could be concluded that the addition of 0.2 g vitamin C/100 mL T-EY extender maintained the quality of Sapera goat semen for 72 hours at 5°C.
