Abstract

Identification of alternate bioactive compounds isolated from microbial endophytes from plants may lead to better solution against antibiotic resistance. Main objective of this research is to determine the bioactivity of antibacterial compounds produced by endophytic actinomycetes, which was isolated from stem bark of Neesia altissima Bl. at Halimun Salak Mount. Detection of bioactive compounds was assayed using Thin Layer Chromatography (TLC) by employing bioautography. Bioactivity was assayed using crude extract against five microorganisms using agar diffusion methods. Tetracycline was used as a positive control. The result of antibacterial activity from endophytic actinomycetes isolate revealed that the bioactive compounds were effective in inhibiting the growth of bacteria Bacillus cereus (1.25 mm), Salmonella typhimurium (5.75 mm), and Shigella flexneri (4.37 mm). Bioactivity of ethyl acetate extract from bioactive compounds of endophytic actinomycetes has an antibacterial activity against bacteria Bacillus cereus (13.00 mm) and Staphylococcus aureus (15.90 mm), at a concentration of 250,000 ppm. Each of TLC fraction was able to inhibit growth of B. cereus at a minimum concentration of 10,000 ppm. In conclusion, the bioactive compounds produced by endophytic actinomycetes can have inhibitory effect towards B. cereus. The category of that compounds are narrow spectrum. The bioactive compound isolated from endophytic actinomycetes may have other implications such as anticancer, antifungal, and antioxidant.

Abstract

The oral cavity as a passageway for various types of substances can be a nesting place for various types of microbes that can cause microbial infections, including candidiasis. Inggu leaf extract and extract of the endophytic bacteria Pseudomonas azotoformans in previous studies have shown the resulting antimicrobial activity. The aim of the study was to screen for antimicrobial bioactive compounds from inggu leaf extract and endophytic bacteria as nutraceutical oral preparations. A mouthwash formulation was made with the addition of crude extract of inggu leaves and pure extract of the endophytic bacteria Pseudomonas azotoformans as antimicrobial agents. The method used is laboratory experimental. The formulation was made by adding the crude extract of inggu leaves and pure extract of the endophytic bacteria Pseudomonas azotoformans and divided into 3 formulations A, B, and C with a ratio of 1:1, 1:2, and 2:1 respectively. The results showed that each mouthwash preparation had an inhibition zone on preparations A, B and C, which were 8.55 mm; 8.55mm; and 9.53mm. The normality test result for each preparation was 0.05. The homogeneity test results obtained a significance value of 0.074 (p > 0.05). The results of the one way ANOVA test found a significance value of 0.000 (p <0.05), there was a significant difference in the variation in the concentration of the mouthwash formula in inhibiting the growth of the Candida albicans fungus. The inhibition zone formed indicated the presence of antimicrobial activity from the mouthwash and all formulas were included in the moderate criteria according to the Davis & Stout method. All preparations are homogeneous because there is no precipitate, have the dominant smell and color of inggu leaves, and have a sweet taste that is slightly mixed with bitter and spicy from inggu leaves.

Abstract

The infection disease is still important problem in Indonesia, especially in eastern part of Indonesia. The problem of the bacterial resistance has resulted in increased morbidity, mortality, and costs of healthcare. This situation is therefore drives many researches on discovery for new antibiotics and novel bioactive compounds to combat multidrug resistant microbes, such as Mycobacterium tuberculosis, Staphylococcus spp., and Streptococcus spp.  The aim of this study is to know the development of the endophytic microorganisms. The endophytic bacteria provide vast potential in producing various novel natural products, including secondary metabolites similar to their hosts. The plant is one of host for the endophytic microorganisms reside. In Indonesia, that endophytic is used medicinally for treatment as well as from the host. The bioactive compound of endophytic microorganisms are used medicinally for treatment antibacterial, antiviral, anticancer, antioxidants, antidiabetic, and immunosuppressive. These showed that endophytic bacteria are promising source of new antibiotics against susceptible and resistant forms of various infectious microbes.

Abstract

Permintaan cabai di Indonesia terus meningkat setiap tahunnya namun hal ini berbanding terbalik dengan produktivitas tanaman cabai yang semakin menurun setiap tahun. Salah satu penyebab utama terjadinya penurunan produktivitas ini adalah adanya hama yang menjangkiti tanaman cabai sehingga cabai menjadi tidak layak untuk dikonsumsi. Biopestisida merupakan langkah alternatif dalam membasmi hama tanaman tanpa merusak lingkungan dan tidak meracuni tanaman. Penggunaan limbah sebagai agen biopestisida selain menguntungkan bagi tanaman juga bagi lingkungan karena dapat menjadi solusi dalam pemanfaatan limbah. Salah satu limbah yang masih belum banyak dimanfaatkan adalah cangkang telur. Tujuan penelitian untuk mengetahui aktivitas antifungi dari air rendaman membran cangkang telur sebagai biopestisida pada tanaman cabai kecil. Metode yang digunakan dalam penelitian ini adalah metode eksperimen berbasis laboratorium secara in vitro dengan Rancangan Acak Lengkap (RAL). Metode difusi kertas cakram atau Metode disc diffusion digunakan untuk melihat aktivitas antifungi. Data dianalisis dengan menggunakan aplikasi Statistical Product abd Service Solution (SPSS) for windows versi 24 dengan beberapa uji diantara uji Analysis of Variance (ANOVA) dan uji Mann Whitney. Penelitian ini memilih dua isolat fungus yang diisolasi dari tanaman cabai berpenyakit, yang diberi kode Fungus A dan Fungus B. Hasil penelitian menunjukkan bahwa air rendaman membran cangkang telur memiliki aktivitas antifungi dengan rata-rata zona hambat yang terbentuk berkategori sedang untuk Fungus A dan berkategori kuat untuk Fungus B. Uji statistik menunjukkan tidak adanya pengaruh lamanya proses perendaman membran cangkang telur terhadap aktivitas antifungi.

Abstract

This activity is a collaboration between the community service team of the University of Indraprasta Graduate School (Unindra) and the Centre for the Development of Educational Competence, Education and Research (P2KPTK2) in West Jakarta. Pkm activities were carried out online through zoom meetings on 1-5 August 2022 and expected to provide insight to low-level elementary school teachers in the work area of P2KPTK2 West Jakarta in terms of the use of innovative learning models in thematic learning in schools. The methods used in this activity are lectures, questions and answers and practice making educational games based on the Internet. After the community service, there is an increase in the value of the post-test results or a change between the lowest, highest, and rare values in general. At the lowest level there is a decrease of 10 points while at the highest there is no increase. The ratio increased by 32.97 points. Generally speaking, the rate of improvement in competence after the community service was 27.45%. The school is expected to provide guidance on the importance of this activity for teachers to improve competence. In addition, teachers should also be aware of the importance of improving themselves through such training to adapt to the demands of the implementation of education in the era of technology as it is now.

Abstract

Mollusca merupakan hewan yang memiliki sifat tripoblastik slomata. Mollusca termasuk ke dalam jenis invertebrata yang memiliki tubuh lunak multiseluler namun biasanya memiliki cangkang yang kuat sebagai pelindung yang terbuat dari kalsium karbonat. Filum Mollusca juga memiliki sifat yang merugikan karena dapat merusak tanaman dan terlibat dalam daur hidup cacing parasit yang akan menyebabkan penyakit pada tanaman dan hewan ternak. Tujuan penelitian ini untuk mengetahui dan menganalisis keanekaragaman filum Mollusca di lahan persawahan Desa Pari Pandeglang, Banten. Penelitian ini dilakukan pada bulan April 2023 di wilayah lahan persawahan Desa Pari Pandeglang, Banten. Penelitian ini menggunakan metode kualitatif deskriptif dengan cara pengoleksian sampel pada titik yang sudah ditentukan pada garis transek. Hasil penelitian didapatkan dua kelas dari fillum Mollusca yaitu Gastropoda dan Bivalvia. Pada kelas Gastropoda ditemukan tiga famili dengan masing-masing famili terdiri satu spesies yaitu Ampullariidae (Pomacea canaliculata), Viviparidae (Filopaludina javanica), dan Polygyridae (Mesodon thyroidus). Sementara Bivalvia ditemukan sebanyak satu famili dan satu spesies yakni Unionidae (Pilsbryoconcha exilis). Indeks keanekaragaman jenis di lahan persawahan Desa Pari Pandeglang, Banten yaitu 1,09 dengan kategori sedang, indeks kemerataan 0,78 dengan kategori stabil, indeks kekayaan jenis 0,55 dengan kategori rendah, indeks dominasi 0,42 dengan kategori sedang. Keanekaragaman filum Mollusca di lahan persawahan Desa Pari Pandeglang, Banten dapat dikategorikan stabil.

Abstract

Keanekaragaman hayati mengacu pada kekayaan hidup di bumi dengan berbagai tumbuhan, hewan, dan mikroorganisme genetika yang dikandung dan ekosistem dimana melangsungkan kehidupan. Tujuan dari penelitian ini untuk mengetahui biodiversitas tingkat spesies pada burung di Agrowisata Cilangkap. Penelitian ini dilakukan di Agrowisata Cilangkap Jakarta Timur. Teknik penelitian yang dipakai dalam penelitian ini yaitu Encounter rates (tingkat pertemuan) dan dilakukan analisis data mengenai indeks keanekaragaman jenis, kemerataan jenis, kekayaan jenis, dan dominansi spesies. Diperoleh sebanyak 579 individu dari 21 spesies dan 14 famili burung yang terdapat di Agrowisata Cilangkap Jakarta Timur. Berdasarkan hasil analisis tersebut, maka dapat disimpulkan tingkat keanekaragaman spesies burung yang terdapat di Agrowisata Cilangkap Jakarta Timur termasuk dalam kategori sedang.

Abstract

Tujuan penelitian adalah untuk mengetahui pengaruh langsung kecerdasan adversitas dan rasa ingin tahu terhadap penguasaan konsep IPA siswa, pengaruh langsung kecerdasan adversitas terhadap rasa ingin tahu siswa dan pengaruh tidak langsung kecerdasan adversitas melalui rasa ingin tahu terhadap penguasaan konsep IPA siswa SMPN di Jakarta Barat. Metode penelitian yang digunakan adalah survei dengan sampel sebanyak 86 siswa. Analisis hipotesis statistik yang digunakan dalam penelitian ini dengan menggunakan analisis jalur (path analysis). Hasil penelitian menunjukkan bahwa terdapat pengaruh langsung yang signifikan kecerdasan adversitas terhadap penguasaan konsep IPA dengan thitung = 20,103 > ttabel = 1,989, terdapat pengaruh langsung yang signifikan rasa ingin tahu terhadap penguasaan konsep IPA dengan thitung = 9,411 > ttabel = 1,989, terdapat pengaruh langsung yang signifikan kecerdasan adversitas terhadap rasa ingin tahu dengan nilai thitung = 8,988 > ttabel = 1,989 dan terdapat pengaruh tidak langsung yang signifikan kecerdasan adversitas melalui rasa ingin tahu terhadap penguasaan konsep IPA dengan thitung = 3,858 > ttabel = 1,989. Kesimpulan dari penelitian menunjukkan bahwa terdapat pengaruh langsung yang signifikan kecerdasan adversitas dan rasa ingin tahu terhadap penguasaan konsep IPA, kecerdasan adversitas terhadap rasa ingin tahu dan terdapat pengaruh tidak langsung yang signifikan kecerdasan adversitas melalui rasa ingin tahu terhadap penguasaan konsep IPA.

Abstract

Futoamide, Gentialutine, Gentiabetine, 1-[(2E,4E)-2,4-dec- adienoyl] pyrrolidine, Lycopodine, Dihydro-lycopodine are bioactive compounds that can be obtained Pseudomonas azotoformans UICC B-91. Previous research results indicate that P. azotoformans extract- ed with dichloromethane, chloroform, and ethyl acetate have antican- didal activity against Candida albicans ATCC 10231. C. albicans is considered to become an opportunistic pathogen and well-known as the main cause of candidiasis. This study aims to determine the mech- anism of inhibition of bioactive compounds from P. azotoformans UICC B-91 on the growth of C. albicans using the molecular docking method. Docking was carried out using the targeted (Lanosterol 14 alpha demethylase and Glucan endo-1,3-beta-D-glucosidase) dock- ing method with an exhausted parameter of 50. The size of the grid- box was adjusted to the position of the amino acid residues based on predictions of binding sites using PrankWeb. The docking results were obtained in the form of binding affinity resulting from the inter- action of the compound with the protein. Results showed that the three sample compounds had the potential to form strong and stable bonds with both protein targets with only two ligands show a binding energy value of less than -7 kcal/mol. In addition, the speed and stability of the bond between the sample and the target protein cannot exceed control ligands, thus it can be predicted the structure of ligand. Fu- toamide, 1-[(2E,4E)-2,4-decadienoyl] pyrrolidine, Lycopodine, and Dihydrolycopodine had binding potential with Lanosterol 14 alpha demethylase. For Glucan endo-1,3-beta-D-glucosidase, only Fu- toamide had the potential to form stable and strong bonds that similar to the control. It can be concluded that the futoamide, one of ligand from P. azotoformans compounds has the potential as a multitarget inhibitor of the two C. albicans proteins, because it has various affini- ties and interaction stability for Lanosterol 14 alpha demethylase and Glucan endo-1,3-beta-D-glucosidase.

Abstract

The low level of knowledge about the appropriate use of antibiotics causes the practice of irrational antibiotic use to increase. The aim of this community service is to provide education about understanding antibiotics and how to use them appropriately and rationally. The community service method is by providing education through group discussions, workshops and seminars to health workers, posyandu cadre mothers and the community around Ratujaya Village, Depok. The level of knowledge about antibiotic use was measured using a questionnaire, pretest and posttest. The results achieved by this community service after providing education on understanding antibiotics and their use to the community show that people who have good and sufficient knowledge about antibiotics are 88.57%, meaning that more people understand about antibiotics than those who don't. It can be concluded that the education on the use of antibiotics provided to the public can sufficiently increase public knowledge regarding antibiotics.
