Abstract

Free radicals are compounds that can cause various diseases and can be inhibited with antioxidant compounds. One of the antioxidant compounds that can be obtained from natural sources is the kepundung plant (Baccaurea racemosa). This research aims to determine the fraction that contains flavonoid compounds and determine the percent inhibition of the fraction resulting from gravity column chromatography from the methanol extract of kepundung leaves (Baccaurea racemosa) using the DPPH method. Extraction used the sonication method and continued with fractionation using gravity column chromatography. A UV-VIS spectrophotometer was then used to measure the antiradical activity and perform a qualitative flavonoid and antioxidant test using TLC. The study's findings demonstrated that the yield of a 96% methanol extract of kepundung leaves was 17%, and that there were 15 sets of fractions produced by gravity column chromatography. Positive results for flavonoids and antioxidants were shown by fractions B, G, H, K, L, and N. The fraction with the highest antiradical activity was fraction L with a % inhibition value of 79,94%.

Abstract

The poinsettia red leaves (Euphorbia pulcherrima Willd.) have the potential as antibacterial and antioxidant agents and are widely utilized by the community for wound healing purposes. This research aims to determine the specific and non-specific parameter values of methanol extract from poinsettia red leaves. Samples of poinsettia red leaves were processed into extracts using the maceration method with 96% methanol as the solvent. The extract was standardized by determining specific and non-specific parameters. The test result data were analyzed descriptively in both qualitative and quantitative manners. The standardization results of specific parameters showed the identity of the extract as methanol extract of poinsettia red leaves, with organoleptic properties of a thick greenish-black extract, distinctive poinsettia aroma, and bitter taste. The water-soluble compound content was 51,47% ± 0,437 and ethanol-soluble 61,22% ± 0,117. Results for non-specific parameters indicated drying loss of 12,50% ± 0,419, moisture content of 12,62% ± 0,173, total ash content of 5,25% ± 0,538, acid-insoluble ash of 0,34% ± 0,048, residual solvent 0,00% ± 0,000, total plate count was TFTC (Too Few To Count), and mold and yeast count was 1,5 x 104 colonies/g sample. This research can serve as a reference for determining specific and non-specific parameters of methanol extract from poinsettia red leaves.

Abstract

The use of analgesics for pain management has limitations related to side effects, effectiveness, and dependence that are often caused. The malacca plant (Phyllanthus emblica L.) has been used empirically on Sumbawa Island to treat throat pain. This study aims to test the analgesic activity of malacca stem bark extract (EKBM) in vivo with acetic acid induction method. Mice were randomly divided into 5 groups including negative control (K-) CMC Na 1%, positive control (K+) ibuprofen 52 mg/KgBB mice, and various doses EKBM1, EKBM2, and EKBM3 consecutively 250, 500, 750 mg/KgBB mice orally. Analgesic activity was assessed based on the number of writhes, % analgesic protection, and % analgesic effectiveness. Phytochemical screening results showed that EKBM contains flavonoids, saponins, steroids, and tannins. The mean number of writhes in groups K (-), K (+), EKBM1, EKBM2, and EKBM3 were 105,60; 75,60; 60,60; 59,40; and 40,20, respectively. The results of one way ANOVA on the number of writhes showed that there were significant differences between all groups (p < 0.05). Furthermore, LSD test was conducted which showed that there were significant differences (p < 0.05) in all EKBM groups compared to K (+) and K (-). There was no significant difference between EKBM1 and EKBM2 groups (p > 0.05). The % analgesic protection values obtained by EKBM1, EKBM2, and EKBM3 were 42,61%; 43,74%; and 61,93%, respectively, while the % analgesic effectiveness values were 150%; 154%; and 218%. Based on the analgesic activity test, EKBM at three dose variations showed analgesic activity.

Abstract

Acne vulgaris is a chronic inflammation of the skin’s pilosebaceous unit, caused by oxidative stress due to reactive oxygen species (ROS) release. Oxidative stress conditions can be treated using natural antioxidants, namely ashitaba (Angelica keiskei). In contrast to previous studies that tested column chromatography fraction of the macerated ethanol extract, this study examined the liquid-liquid partition fraction of 80% methanol sonicated extract of Ashitaba herb. This study aims to determine the antiradical activity of various fractions of Ashitaba herb extract against 1,1-diphenyl-2-picrylhidrazyl (DPPH) free radicals. 80% methanol extract of the Ashitaba herb was fractionated using chloroform, ethyl acetate, and water solvents. Each ashitaba herb fraction was identified for the content of flavonoids and phenolic compounds and the antiradical DPPH activity was tested with ascorbic acid standard using a UV-Vis spectrophotometer. The IC50 value of each fraction was statistically analyzed using One-Way ANOVA and Posthoc Tukey. The results of phytochemical identification showed that ashitaba herb fractions positively contain flavonoids and phenolics. Antiradical activity of the chloroform, ethyl acetate, and water fractions against DPPH was indicated by IC50 values of 164.22±5.68 ppm (moderate); 8.70±0.12 ppm (very strong); and 243.60±8.14 ppm (weak). Statistically, each ashitaba herb fraction differed significantly from ascorbic acid (IC50 2.37±0.05 ppm). The ethyl acetate fraction has the strongest antioxidant activity and is in the same category of antioxidant strength as the ascorbic acid standard.

Abstract

Free radicals are reactive fragments that can trigger a variety of chronic diseases. Antioxidant compounds are needed to counteract the attack of free radicals. Sumbawa oil is one of the natural traditional remedies that has the potential to be a natural antioxidant because it contains compounds such as lauric acid, palmitic acid, linoleic acid, stearic acid, and methyl palmitate. The study aimed to determine the metabolite compounds contained in the Sumbawa oil brand “Rimba” and their activity to capture the DPPH free radical. The Sumbawa oil determined their metabolite compounds using Gass Chromatography-Mass Spectrometry (GC-MS). The antioxidant activity of Sumbawa oil was analyzed using the DPPH radical capture method. The GC spectra showed the 5 compounds detected in the Sumbawa oil. In addition, the MS spectra showed these 5 compounds were delta-3-carene, trans-caryophyllene, palmitic acid, palmitoleic acid, and stearic acid. The antioxidant activity of Sumbawa oil was found to be very weak with an IC50 value of 13.49 ± 0.34 mg/mL. Therefore, Palmitoleic acid and palmitic acid were reported to have potential antioxidant activity. These compounds were lipophilic antioxidants that did not properly detect their antioxidant activity using the DPPH method. So, the antioxidant activity of Sumbawa oil was recommended to be analyzed using the β-carotene bleaching method.

Abstract

Ashitaba herb contains flavonoids, phenolics, tannins, alkaloids, and terpenoids, which are known to have antioxidant, antibacterial, and anti-inflammatory properties. Therefore, ashitaba has the potential to be developed into anti-acne preparations, one of which is cream. Creams have various advantages, namely the ability to deliver hydrophilic and hydrophobic compounds better, provide better moisture, and have emollient and occlusive properties by forming a protective layer on the skin and maintaining skin hydration longer. The purpose of this study was to determine the physical properties of methanol extract cream preparations of ashitaba herb. The research method used was laboratory experimental. Ashitaba herb was extracted using the sonication method with 80% methanol solvent to produce a solid extract. The ashitaba herb's methanol extract was combined to create a cream preparation, and its physical characteristics such as homogeneity, pH, viscosity, spreadability, adhesion, and cream type were assessed. The physical property testing revealed that the cream preparation had a unique scent, a thick consistency, and a green hue. This resulted in a homogenous oil-in-water (O/W) type cream preparation with a pH of 6.29, viscosity of 37.050 cp, spreadability of 5.23 cm, and adhesion of 4.59 seconds. Thus, it can be said that the production of the methanol extract of ashitaba herb cream satisfies the requirements and has good physical qualities.
