Abstract

The cytotoxicity of ethyl acetate extract from Tampa badak (Voacanga foetida (Bl.) Rolfe) leaves against Artemia salina leach larvae was determined using the brine shrimp lethality test (BSLT) method and was evaluated against T47D breast cancer cells using MTT assay method. The result of BSLT showed a consistent result with MTT assay, which is the result obtained that ethyl acetate extract is very toxic against A. salina Leach larvae with LC50 value of 8.61 µg/mL and very cytotoxic against T47D breast cancer cells with IC50 values of 0.87; 0.66; and 0.95 µg/mL at the 24, 48 and 72 hours of incubation times, respectively. The MTT assay data were analyzed using a two-way ANOVA statistical method to see the effect of the dependent variable (concentration and time) on the independent variable (% viability). Based on the statistical test result, there is a difference in % viability between concentrations of 0.1; 1; and 10 µg/mL (p <0.05), but the length of incubation does not affect % viability (p >0.05).

Abstract

Piperine, the primary alkaloid of Piper nigrum and Piper longum, is believed to have anticancer properties. Although research on cervical cancer is still in its early stages, piperine's anticancer capabilities have been established in numerous types of cancer and have been shown to target pathways involved in the cell cycle and apoptosis. This study aims to determine how piperine administration time and concentration affect the viability of Hela cells. The MTT method was used to conduct this study. The concentration of piperine utilized was 1,000; 500; 250; 125, and 62.5 µg/mL, and it was administered for 24 and 48 hours. The IC50 values, obtained after incubation for 24 and 48 hours, were 711.21 µg/mL and 419.75 µg/mL, respectively. We may conclude that the viability of HeLa cells is influenced by piperine in a time- and concentration-dependent manner (p<0.05).

Abstract

Community health education is crucial, including education on the rational use of antibiotics, as well as halal food and medicine. Additionally, providing free medical treatment has a significant impact, encouraging the community to prioritize health and maintain it as a primary concern. This community service project was conducted in the Desa Perkampungan Adat Sijunjung. The objective of this activity is to maintain and improve the community's health status. One way to achieve this is through community service activities. The service involved educational sessions on medicines and health, as well as free medical treatment, in collaboration with the Faculty of Pharmacy and the Faculty of Medicine at Universitas Andalas. The education targeted pregnant women and the elderly, focusing on proper medication selection, rational use of antibiotics, and education on halal medicines and food. Free medical treatment involved specialist doctors and pharmacists, who also provided counseling on medications to patients. The results of the activity showed high community enthusiasm, with more than 100 prescriptions filled. The education on the rational use of medications, particularly antibiotics, is expected to increase community awareness and knowledge. This activity also highlights the importance of pharmacists in providing patient-oriented pharmaceutical services, as well as the importance of promotional and preventive efforts in maintaining public health. It can be concluded that free medical treatment is expected to encourage the community to be more health-conscious and less fearful of seeking medical care.

Abstract

Post-Immunization Adverse Events (AEFI) are common among vaccine participants. AEFI needs to be evaluated because Covid-19 vaccination is an effort to prevent Covid-19 infection which is still relatively new and people are still afraid of post-immunization adverse events occurring. In order for community participation to increase, it is deemed necessary to carry out an AEFI evaluation for both the first dose and the second dose (complete dose). This study aims to evaluate the symptoms of AEFI 1 with AEFI 2 by measuring the number of respondents who caused local and systemic reactions after COVID-19 vaccination. The retrospektive and prospective cross sectional study were used in this study with about 113 Astrazeneca vaccine participants at the UTA'45 Jakarta vaccine center and the data met the inclusion and exclusion criteria. Data was collected through questionnaires, AEFI forms and interviews based on symptoms of local and systemic reactions. Of the 113 participants, 29 were female, while 84 were male and the reactions they caused were different, such as pain, redness, swelling at the injection site, such as fever, muscle aches throughout the body, joint pain and also weakness. Comparison of symptoms of local and systemic reaction between KIPI 1and KIPI 2. In those without symptoms, there was increase number, but in those with symptoms, the number of respondens decrease, and statistically there was sig, P value <0,05.

Abstract

Hypoalbuminemia is generally identified in patients undergoing preoperative care, the recovery phase after surgery, and those in the healing process. At DR. M. Djamil General Hospital, the therapy to improve albumin levels in patients receiving intensive postoperative care involves using 20% human albumin and 25% human albumin. This study aims to evaluate and compare the effectiveness of 20% and 25% human albumin in increasing albumin levels in postoperative hypoalbuminemia patients at the institution. Data collection for this research was conducted retrospectively with a cross-sectional approach, where data were analyzed to determine how the increase in albumin levels after the administration of 20% or 25% human albumin affected clinical outcomes in patients. Findings from the proportionality testing in the subject characteristics involved in the research showed no disparity in the proportion of patient characteristics between the group receiving 20% human albumin (n=27) and the group receiving 25% human albumin (n=33) (p>0.05). Furthermore, the comparative mean analysis indicated a statistically significant difference in the levels of hypoalbuminemia before and after albumin infusion therapy in each group (p<0.05). The average increase in hypoalbuminemia in the group using 20% human albumin was recorded at 0.46 g/dL, while in the group with 25% human albumin, the increase reached 0.66 g/dL. Statistical analysis of the mean difference showed a statistically significant average increase in albumin concentration among the studied groups, with a p-value of less than 0.05. In this context, 25% of human albumin contributed to a significantly higher average increase in albumin concentration. Given the significant cost difference between 20% and 25% human albumin, it is recommended to use 20% human albumin for patients with albumin levels above 2.1 g/dL. Meanwhile, for patients with albumin levels below 2.1 g/dL, it is more appropriate to use 25% human albumin.

Abstract

Cancer is a problem in the health sector because this disease has an increasing incidence of morbidity and mortality. While the treatment still leaves some drug side effects, which cause discomfort for cancers sufferers. The arises because most cancer drugs work non-selectively. Because of some of the problems above, it is necessary to encourage the search for alternative drugs treatment of cancer. Voacanga foetida (Blume) Rolfe (Apocynaceae) is a species of plant from the genus Voacanga known as the tampa badak from West Sumatra. This plant is known to be source of indole alkaloids and also bis-indole alkaloids potential as cancer drugs. Therefore, in this study we are interested to explore the potency of this plant as natural product sources with potential cytotoxic activity. The study was started from the isolation process and continued by characterization of the isolated compounds and then testing their cytotoxic activity T47D breast cancer cell line. The isolation process of alkaloid compounds were performed by acid-base extraction, liquid-liquid fractionation, and followed by separation using column chromatography. Then, the potency of cytotoxic activity of the isolates were screened through Brine Shrimp Lethality Test (BSLT) method and evaluated by MTT assay. The structure of three isolated alkaloids (VFB-DB1.1; VFB-DB2.1; and VFB-DB2.2) were charachterized using spectroscopic analyses, including UV and FT-IR. One isolated compound (VFB-DB1.1) was also characterized using 1H NMR spectroscopy. Based on the similarity of 1H NMR spectra of isolated compound when compared with the literature, compound VFB-DB1.1 was identified as voacangine. Then, the BSLT and MTT assay result showed that VFB-DB1.1 exhibited LC50 value of 15.8 µg/mL againt Artemia salina L.and IC50 value of 8.9 µg/mL against T47D breast cancer cells, respectively.

Abstract

Coagulation disorders, one of the causes of death of severe Covid-19 patients and comorbidities that also affect coagulation is Diabetes Mellitus. In the guidelines, it is recommended to use prophylactic anticoagulants, namely Unfractionated Heparin (UFH) and Low Molecular Weight Heparin (LMWH) to overcome the problem of this coagulation disorder by paying attention to the value of coagulation parameters, namely, D-Dimer, PT, APTT and platelet values. The purpose of this study was to see the relationship between the use of prophylactic anticoagulants with a decrease / increase in the value of the coagulation parameter. The research design was retrospective pre-post test cohort study with research subjects of comorbid Covid-19 Diabetes patients treated in the ICU of RSUP Dr.M Djamil Padang. As a result, in the group of patients using UFH there was a significant decrease in D-Dimer values (p = 0.001) while the LMWH group was not significantly related (p = 0.05). For the association of anticoagulants with PT values, APTT in both groups was not significant. As for platelet values, there was a significant decrease in the UFH group. Conclusion for severe Covid-19 patients with comorbid Diabetes The use of UFH is recommended as a therapeutic option.

Abstract

AstraZeneca vaccine has become one of the efforts to overcome the COVID-19 pandemic. This study aims to determine the relationship between the AstraZeneca vaccine and antibody titers and its association with exposure to COVID-19 infection. This study used a retrospective and prospective cohort design with convenience sampling. A total of 102 respondents who had received a complete dose of the AstraZeneca vaccine and were willing to be followed up for 18 months were involved in this study. Sociodemographic data, comorbidity history, lifestyle, and antibody titers were measured at two times: before the first dose and after the second dose of vaccinations. This study found that there was no significant relationship between comorbidities and respondent behavior (smoking and alcohol consumption) with COVID-19 antibody titers. Physical activity (exercise) showed a positive relationship with COVID-19 antibody titers. Respondents who exercised regularly had higher antibody titers than those who did not. There was a positive relationship between comorbidities, smoking, and alcohol consumption with the risk of exposure to COVID-19 infection after vaccination. This study found that physical activity (exercise) can increase COVID-19 antibody titers and provide protection against COVID-19 infection after vaccination.
