Abstract

The purpose of study is to determine the type of plankton found in cultivation pond of nile tilapia infected by VNN. The method is to identify the plankton in the water pond and testing the VNN that infects nile tilapia related to plankton eaten using Polymerase Chain Reaction (PCR). VNN identification results on the nile tilapia digestive tract stomach and intestines as well as plankton found in the stomach and intestines indicate VNN positively. Plankton found in the digestive tract of fish is division of Chlorophyta, Bacillariophyta, phylum Rotifera and Crustacea. It indicates that nile tilapia infected by VNN both on the gastrointestinal tract organs, stomatch or intestines and plankton were found with positive VNN digestive duct, while plankton found in water pond is VNN negative. The VNN infection depends on the nature of specifications VNN on nile tilapia and the plankton in the nile tilapia digestive duct.

Abstract

In this study, the effect of cinnamaldehyde (CA) towards Streptococcus agalactiae on zebrafish egg were investigated. To obtain the optimal concentration, CA and S. agalactiae were tested by short-term effects assay. Fresh zebrafish egg, CA and S. agalactiae were observed in different groups, single and mix treatment. The result showed that the larval survival rate (%) of single CA concentration (2.5 and 5 µg mL-1) were not significantly different compared to the control, with the value 94 ± 2.8% and 93 ± 3.3% respectively. Meanwhile, the single S. agalactiae concentration (109 and 1010 CFU mL-1) were significantly different in comparison to the control with the value 73 ± 2.1% and 6 ± 2.5%. Mix groups, larval survival rate (%) value were significantly different with the best combination value was 17 ± 4.7% at 5 µg mL-1 of CA and 109 CFU mL-1 of S. agalactiae. Even though CA may potential for zebrafish egg shelter against S. agalactiae infection. But a mixture (CA and S. agalactiae) treatments were not recommended for this study because the low value was shown compared to the single treatments. This possibility may occur because of the debris formation of two substances which leads to the poor environment and causes to low fish larval survival rate (%) value. For further study, the water quality assay is needed.

Abstract

Indonesia is known as a country with the highest potential production of tuna, with total production more than six hundred thousand ton per year as value as 6.3 trillion IDR per year.However, Tuna fishery have several challenges, as can be identify by the declining in productivity and an average length fish had been caught, and also tuna fishing groundsaregetting farther. This apparently appears as a result of inaccurate determination of fish stock as the basic management unit of tuna fisheries, especially in yellow fin tuna (YFT). This study aims to define population stocks of YFTin South Java Sea based on genetic information. Samples were taken from three fish landing ports (MuaraBaru, Cilacap and Benoa). Restriction Fragment Length Polymorphism (RFLP) technique were applied for Cytochrome Oxidase I genes, using restriction enzymes;HaeIII, RsaI, BamHI, XhoI, and AluI. Genetic variation in populations were evaluated with POPGENE32 software. Result indicate that finer scale of stocks present YFT and should bemanaged differently for effective management purpose and to maintain sustainability of this species.

Abstract

Shrimp aquaculture wastewater after the end of the rearing period has a high total organic matter (TOM) content of up to 87.74 mg/L. If the wastewater is disposed of directly without any efforts to reduce or treatment, it will disturb public waters ecosystem because of the high organic matter. The aim of this study was to determine the rate of reduce of organic matter in pond aquaculture waste. The study was carried out in January 2021. Pond wastewater samples were taken from the Tambak at the end of the maintenance period (120 days of maintenance). The trademarks of the bacterial consortium are EM4, Bio HS, BIO Prisma, Starbio plus, BIO2000, and Biolet. The experimental research was arranged in a completely randomized design with 6 treatments and 4 replications. Each treatment was filled with 2 liters of wastewater plus 6 ml of a liquid bacterial consortium. As for the solid bacterial consortium, 1 gram/L was added. Measurements of total organic matter, temperature, pH, dissolved oxygen, salinity and carbon dioxide were carried out every 24 hours for 72 hours of observation. The results showed that during 72 hours of immersion the highest percent reduction in organic matter was in treatment E (BIO 2000) of 95% (87.74 mg/L to 4.11 mg/L). The lowest percentage decrease was in treatment D (Starbio plus) of 83.4% (87.74 mg/L to 14.54 mg/L). However, in general 6 types of bacterial consortium trademarks can degrade organic matter. The levels of temperature, pH, dissolved oxygen and salinity are in optimum conditions, while carbon dioxide exceeds the threshold caused by the high activity of overhauling organic matter by bacteria. The conclusion was obtained that the trademark of the bacterial consortium BIO2000 was the most effective in reducing organic matter for 72 hours.Keyword : Shirmp wastewater, Tambak, Bioremediation, Organic Matter, Microorganis

Abstract

The fish of the Bagridae Family in Indonesia reach until 60 species and one of them is from the genus Msytus. The method is used for fast and accurate species identification was needed. DNA barcoding was a new identification method with molecularly approach. The CO1 gene was amplified and PCR products were sequenced and analyzed using bioinformatics software. Editing of sequencing results and determining the nucleotide composition were analyzed with Mega5 software. The DNA sequence was aligned with ClustalW vers. 1.4. Sequences are compared with GenBank data using BLAST (Basic Local Aligment Search Tools) and BOLDSystems. Phylogenetic tree was made using Neighbor_Joining method. One example of the identification of fish from the confirmed Mystus genus is Mystus nigriceps being the Mystus singaringan.

Abstract

Ikan merupakan salah satu biota perairan yang digunakan sebagai bioindikator karena sangat peka terhadap perubahan lingkungan perairan. Kondisi perairan yang tidak mendukung dapat menyebabkan stress pada ikan yang salah satunya ditandai oleh perubahan kadar gula darah ikan. Tujuan penelitian ini adalah untuk menganalisis hubungan kualitas perairan Sungai Brantas dengan kadar gula darah Ikan Gambusia (Gambusia affinis). Penelitian dilaksanakan pada sepuluh lokasi sampling di sepanjang aliran Sungai Brantas dengan mengukur enam parameter kualitas air (in-situ dan ex-situ) dan mengambil sampel Ikan Gambusia untuk diukur kadar gula darahnya. Hasil penelitian menunjukkan bahwa kadar gula darah tertinggi dan melampaui batas normal ditemukan pada sampel Ikan Gambusia yang ditangkap di wilayah Mojokerto. Kondisi ini berkaitan erat dengan rendahnya kadar DO dan tingginya konsentrasi BOD, ammonia, dan fenol di lokasi tersebut.

Abstract

Waters that are presumably contaminated with heavy metals need to be observed to ensure the level of pollution to perform water restoration. This study was aimed to determine the status of water quality based on heavy metal contents. The survey method was employed in this study, and it was conducted in Pasuruan sea waters in different seasons; September 2019 represented the dry season, and April 2020 represented the rainy season. The water sampling stations were chosen by using purposive sampling in three sampling locations: the sea waters of Kraton, Lekok, and Nguling districts. The heavy metals and water quality parameters were analyzed by using Storet Index. The results indicated that the highest level of heavy metals was found in Lekok waters in September, consisting of Hg (1.22 mg/l), Cd (1.20 mg/l), and Pb (0.55 mg / l). The seawater of Nguling district had the lowest content in April, while Kraton's seawater was moderate. The water quality status based on the Storet Index suggested that the sea waters of Kraton, Lekok, and Nguling districts were classified as moderately polluted with a score of -16 to -24. The highest score was in September in the rainy season with a score of -24 and the score in April in the dry season was -16. It means that the score is increasing but still in the moderately polluted category. The measurement results of temperature, pH, dissolved oxygen, salinity, and current velocity were in optimum results for aquatic organisms in the dry season or the rainy season. Regarding the water quality status in Pasuruan sea waters, it can be concluded that the heavy metal contents exceeded the quality standard set by the government. Therefore, improved management is required to prevent the deterioration of the pollution status in Pasuruan sea waters

Abstract

Penelitian ini dilakukan dengan menggunakan sample ikan betok (Anabas testudineus Bloch 1972) yang berasal dari perairan rawa Kalimantan Selatan, dengan tujuan untuk mendeskripsikan keragaman  genetik dan aspek konservasinya dengan metode amplifikasi mtDNA. Proses amplifikasi mtDNA ikan betok terjadi di daerah D Loop.  Hasil analisis mt-DNA D Loop ikan betok menunjukkan bahwa, analisis keseimbangan populasi Hardy-Weinberg  berkisar antara 0,02 - 0,09, sedangkan haplotipe tertinggi terdapat pada rawa monoton (0,9384), kemudian tadah hujan (0,7111) dan pasang surut (0,6).  Heterozigositas ditemukan unik pada populasi rawa monoton (BAAAA) dan rawa pasang surut (BAACA) dan umum di temukan di ketiga ekosistem rawa (AAABA).  Ikan betok di bagi menjadi dua stok populasi yaitu populasi rawa monoton dan pasang surut serta stok tadah hujan.  Konsep utama dalam konservasi genetik adalah fitness population dimana populasi dipertahankan minimal 500 ekor/kawasan. Untuk meningkatkan keragaman genetik ikan betok, dilakukan dengan cara introduksi individu-individu baru yang memiliki keragaman genetik yang lebih tinggi kedalam populasi lokal, restocking dan membuat kawasan suaka yang dilindungi oleh Dinas Perikanan setempat bersama-sama dengan masyarakat di sekitar perairan rawa tersebut. The research was conducted using climbing perch samples originated from the swampy waters of the southern Borneo, and the objektive of this study to investigate the genetic diversity and the conservation aspect using mtDNA amplification method.  mtDNA amplification process occurs in the D Loop region.  The results of the analysis of D-Loop mtDNA of climbing perch showed that, the analysis of Hardy-Weinberg equilibrium population ranged from 0.02 to 0.09, while the highest haplotypes found in swamp bogs (monotonic) (0.9384) then rainfed (0.7111) and tides (0.6). Heterozygosity was found uniquely in the swamp monotonic population (BAAAA) and marsh tides (BAACA) and common in all three ecosystems found in the swampy area (AAABA) . Climbing perch stock divided into two populations monotone and tidal swamp population and rainfed stock.  The main concept of genetic conservation is the fitness population where the population is maintained at least 500 tail/region.  To increase the genetic diversity of climbing perch, can be done by the introduction of new individuals wich has a higher genetic diversity into the local population, restocking and create reserves of protected areas by the Local Fisheries Authority together with the community around the swampy waters.

Abstract

Usaha pembenihan ikan merupakan alternatif pemecahan masalah untuk mengatasi kesenjangan antara ketersediaan dan kebutuhan benih dalam budidaya ikan. Keberhasilan usaha pembesaran ikan betok dibatasi oleh tingkat kelangsungan hidup larva bagi pertumbuhan dan jenis pakan alami larva ikan di habitatnya. Tujuan penelitian ini adalah untuk mengetahui jenis dan kelimpahan pakan alami yang dikonsumsi larva ikan betok di perairan rawa monoton Danau Bangkau Kalimantan Selatan. Penelitian dilaksanakan pada bulan Desember 2011-Januari 2012. Pengambilan sampel pakan dan larva dilakukan pada dua tempat yang berbeda pada kedalaman 30 cm. Hasil penelitian menunjukkan jenis plankton yang dikonsumsi oleh larva ikan betok berubah sesuai dengan ukuran bukaan mulut ikan dan kelimpahan plankton di perairan. Jenis plakton dominan yang dimakan oleh larva betok yaitu Mougeotia sp. 265 sel/L diikuti oleh Coconeis sp. 246 sel/L, Keratella sp. 174 sel/L, Chlorococcum sp. 110 sel/L, Brachionus sp. 98 sel/L, dan Navicula sp. 47 sel/L. Jenis plakton dominan yang dimakan berubah sesuai dengan umur larva. Business seeding fish an alternative problem-solving to overcome the gap between availability and needs seeds. The success of the business of enlargement climbing fish bounded by the degree of continuity larvæ live for growth and a kind of natural fish larvae feed on their habitat. The purpose of this research was to know the type of plankton that were consumed by climbing perch fish larvae in the waters of the monotonous swamp of Danau Bangkau. The research was conducted from December, 2011- January, 2012. The sample feed and larvæ carried on two different places at the depth of 30 cm. The result showed plankton species consumed by climbing fish larvae change according to size openings mouth fish and abundance plankton in waters. A kind of plankton dominant which are eaten by the climbing larvæ  namely Mougeotia sp. 265 cells/L followed by Coconeis sp. 246 cells/L, Keratella sp. 174 cells/L, Chlorococcum sp. 110 cells/L, Brachionus sp. 98 cells/L, and Navicula sp. 47 cells/L. A kind of plankton dominant eaten changed in accordance with the age of larvæ.

Abstract

Contaminants sourced from numerous anthropogenic activities threaten a river ecosystem continuously. Anthropogenic modification alters water quality and affects aquatic biota life. Most of the pollution of the aquatic environment comes from chemical materials such as heavy metals, fertilizers, pesticides, and plastics. Heavy metals are the most dangerous pollutants as they are toxic, carcinogenic, biomagnifying, and bioaccumulative. Pollution affects the survival of aquatic biota, including macroinvertebrates. The sedentary nature of macrozoobenthos makes it to be contaminated with ease, which then affects its survival. Currently, the damaging impact of pollutants on the physiology of macrozoobenthos has been reported in several studies though it is still limited to the use of particular families. This review aims to inform the physiological impacts caused by pollutants on the macrozoobenthos community that lives along the river from upstream to downstream. Pollutants trigger disturbances in physiological functions, which are termed oxidative stress. Macrozoobenthos perform a series of mechanisms, including producing enzymes as the primary defense.
