Abstract

BACKGROUNDMany tumors express on their receptor tyrosine kinases vascular endothelialgrowth factor activity associated with angiogenesis. Inhibition ofangiogenesis through reduction of tyrosine kinase activity is a promisingstrategy for cancer therapy. The present study aimed to determine themechanism and potency of ethyl p-methoxycinnamate (EPMC) isolatedfrom Kaempferia galanga as angiogenesis inhibitor.METHODSA laboratory experimental study was conducted using chorio-allantoicmembranes (CAMs) of nine-day old chicken eggs induced by 60ng basicfibroblast growth factor (bFGF). Ethyl p-methoxycinnamate (EPMC) potencywas determined at dosages of 30, 60, 90 and 120 μg and compared withcelecoxib 60 μg as reference drug and one negative bFGF-induced controlgroup. Neovascularization and endothelial cell count in CAM blood vesselswere evaluated. To predict the antiangiogenic mechanism of EPMC, adocking study was performed with the Molegro Virtual Docker program ontyrosine kinase as receptor (PDB 1XKK).RESULTSAngiogenesis stimulation by bFGF was prevented significantly (p<0.05)by EPMC at dosages of 30, 60, 90 and 120 μg and this activity was dosedependent. Molecular docking showed interaction between EPMC functionalgroups and tyrosine kinase amino acids at Met766, Met793, Thr854, Thr790,Gln791 and Ala743. There was an association between EPMCantiangiogenic activity and docking study results.CONCLUSIONSEthyl p-methoxycinnamate is a potential new angiogenesis inhibitor throughinteraction with tyrosine kinase. EPMC could be a promising therapeuticagent for treatment of angiogenesis-related diseases.

Abstract

Background: The difference in the nature of these aromatic amine substituents, i.e. methyl and chloro will affect the N atom of aniline as a nucleophile to attack the carbonyl C atom in the p-methoxcycinnamoyl chloride in the synthesis two amides derivate of p-methoxycinnamic acid, namely N-(p-methylphenyl)-p-methoxycinnamide and N-(p-chlorophenyl)-p-methoxycinnamide. Aim: to obtain the N-(p-methylphenyl)-p-methoxycinnamide and the N-(p-chlorophenyl)-p-metoxicinamide compound from p-methoxycinamic acid using the microwave irradiation method as source of energy. Beside that, it also to determine the effect of the presence of methyl and chloro substituents in para position of aromatic amines in the yields of reactions. Method: The reactions were carried out by microwave irradiation at three powers, i.e 120 watts, 200 watts, 280 watts. After separation and purification steps, the products were identified by spectrometric methods. Result: At power of 200 watts for reaction time of 7.5 minutes, the yield of N-(p-methylphenyl)-p-methoxycinamide is larger than N-(p-chlorophenyl)-p-methoxycinnamamide. The percentage of the product synthesis of N-(p-methylphenyl)-p-methoxycinamide was 51.84% and the percentage of N-(p-chlorophenyl)-p-methoxycinnamamide was to 47.20%. Conclusion: The effect of substituent methyl is increase the percentage yield of N-(p-methylphenyl)-p-methoxycinamide compound than that substituent chloro of N-(p-chlorophenyl)-p-methoxycinamide compound under the same reaction conditions. Based on the identification of the structure of the synthesized compound using a UV spectrophotometer, infrared spectrophotomers and 1H-NMR spectrometer it can be concluded that the synthesized compounds are N-(p-methylphenyl)-p-methoxycinnamide and N- (p-chlorophenyl)-p-methoxycinnamide.

Abstract

Ferulic acid (FA) has been reported to have antiplatelet activity through indirectly inhibiting P2Y12 receptor. In order to increase the activity of FA, by improving its lipophilicity, so that it is easier to enter the cell, synthesis methyl ferulate was conducted through Fisher esterification. Ultrasonic waves were utilized as source of energy emitted through water as the medium at two various of temperature, i.e., 55 °C and 65 °C. The purposes of this study are to produce methyl ferulate and to determine the reaction constant rate (k) and its energy activation (Ea), at temperature of 55 °C and 65 °C. Moreover, the biological activity as antiplatelet was investigated at dose 20 mg/kg BW. The antiplatelet assay was conducted by clotting time and bleeding time methods. The results were analyzed by one way ANOVA program (P<0.05). The yield of methyl ferulate are 50.3% and 67.1% at 55 °C and 65 °C, respectively. The k value at 55°C is 4x10-5 cons-1min-1, while that of at 65 °C is 9x10-5 cons-1min-1. The clotting time and bleeding time of methyl ferulate obtained were 265 sec and 175 sec, respectively. The antiplatelet activity of methyl ferulate is better than ferulic acid.

Abstract

Abstract

The halal aspect is an important factor that must be considered by Muslims in choosing food. However, the low awareness and knowledge related to halal rules and thoyyib on food had caused various serious problems. To overcome this, several government regulations have been issued, but some people still find it difficult to understand the concept of halal products. In this regard, the Faculty of Pharmacy, Airlangga University through community service conducted socialization in two schools in Rungkut Surabaya district to increase community empowerment about halal lifestyle and the critical point of food prohibition at the young generation. The method used is an online pre-test to determine the initial understanding of the participants, education in the form of counseling and discussion, and evaluation of increased understanding through pre and post-test. As partners are students, teachers, and canteen managers in Madrasah Aliyah and Madrasah Tsanawiyah school grade. Based on the analysis conducted, the results show a strong correlation between halal certification, halal awareness, and halal composition. Socialization can increase the knowledge of students and employees in those schools about the halal lifestyle and the critical point of food prohibition. Halal certification on food products affects buying interest but halal awareness and knowledge about the composition of halal food is still low in its influence on buying interest

Abstract

The development of anticancer drugs from ethyl p-methoxycinnamate (EPMC) derivatives has been done to compounds high activity in inducing cancer cells apoptosis and minimal side effects. p-Methoxycinnamoyl hydrazide derivates, modified from EPMC structure, were docked into the ligand-binding pocket of Check point kinase 1 enzymes (2YWP) and the aromatase enzyme (3S7S) using software Molegro Virtual Docker (MVD) Ver.5.5. We compared the Rerank score of native ligand with p-Methoxycinnamoyl hydrazide derivates. Rerank scores of compounds 4b and 4c (-99.98 Kcal/mol and -99.80 Kcal/mol) were lower than the native ligand A42 in inhibiting the enzyme checkpoint kinase 1. Rerank values of p-Methoxycinnamoyl hydrazide derivate compounds were greater than the native ligand EXM in inhibiting the enzyme aromatase. p-Methoxycinnamoyl hydrazide derivate compounds, especially compounds 4b and 4c, had anticancer mechanism by inhibiting the checkpoint kinase 1 enzyme pathway and showed no activity in inhibiting the aromatase enzyme.
