Abstract

Efforts to inhibit the transmission of COVID-19 are continuing to this day. One of them is using hand-washing soap to prevent the transmission of this virus. The Faculty of Pharmacy, Airlangga University, seeks to implement SDGs-3 through community service activities to increase community empowerment, especially for the younger generation. The utilization of natural ingredients containing many active compounds has yet to be discovered. The solution is to provide information on the source of the active ingredients for hand washing soap, for example, the Rosella flower, explaining the mechanism of soap in inhibiting the spread of viruses and how to make hand washing soap. The activity was carried out in Gresik with counseling and training methods. The outcome was an increase in participants' understanding, as indicated by an average post-test score of >65, with an increase in score of >10% compared to the pretest. In addition, one product of Rosela flower hand washing soap was also obtained. From this activity, it was concluded that there was an increase in understanding of 28%. It is hoped that this increase in understanding and skills for the younger generation can mobilize the surrounding community so that prevention of the transmission of COVID-19 is more massive and conditions of good health and well-being in society will be achieved.

Abstract

This service activity is motivated by efforts to maintain health after the Covid-19 pandemic. The community carries out preventive efforts by providing stocks of their medicines at home. The culture of storing medicines in the household requires proper drug management to prevent drug damage due to improper storage, drug use errors, and environmental pollution due to errors in handling damaged or expired drugs. The pharmacist's role is vital in providing drug management education to the community. Another preventive effort undertaken by the community is to maintain a healthy body by applying a diet where fast food currently contains a lot of Food Additives (BTP), which can threaten long-term public health. For this reason, people need to get an education directly, so they are wise in using BTP. The solution is to educate the public regarding managing household medicines and healthy food for parents and guardians of RA Al Manshur Play Group (KB) students in Bojonegoro. Activities are packaged in counseling methods and training/practice activities. The expected outcome of the activity is an increase in the understanding of guardian parents, as indicated by an average post-test score of >65 with an increase in score of >10% compared to the pretest. By increasing the understanding of parents and teachers about managing medicines and healthy food in the family, it is hoped that conditions of good health and well-being in society will be achieved.

Abstract

In this study, a methylation reaction was carried out for the p-coumaric acid compound using potassium carbonate as a base and dimethyl carbonate as a methylation agent with acetone as a solvent. The methylation reaction was carried out using microwave irradiation from microwave domestic with a power of 200 watts. The purity test was carried out on a TLC plate using three different eluents and produced a single stain that was different from the initial compound. The phenol functional group tested with FeCl3 reagent showed a positive purple color. Compound identification was done using an FT-IR spectrophotometer and a 1H-NMR spectrometer. In the synthesized FT-IR spectrum, there was an absorption of the ester group at wave numbers 1671 cm-1 (C=O group) and 1243 cm-1 (C-O group), there was a band at 3378 cm-1 indicating the presence of the phenol group. There were differences in fingerprint results in the FT-IR spectrum of the p-coumaric acid compound and the results of the methylation reaction. The 1H-NMR spectrum confirmed the presence of three methyl ester protons of the carboxylic acid and the phenolic OH. This indicated that the synthesis produced a compound, namely methyl (E)-3-(4-hydroxyphenyl) acrylate. Abstract

Pulmonary fibrosis was one of the conditions that occurred in Post-COVID Syndrome patients. Development of specific treatment as an agent to treat pulmonary fibrosis was still ongoing. Ferulic acid was a compound that had a potential lung antifibrotic agent through the inhibition of TGF-β1-mediated signaling. Molecular docking studies using the AutoDock Tools program version 1.5.7 were conducted on ferulic acid and its analogs to determine the activity of compounds as antifibrotic. Furthermore, the interaction of the compounds with the receptor was visualized using the Discovery Studio 2021 program. The results showed that ferulic acid and its analogs had lower free energy than Pirfenidone, which was a standard lung antifibrotic drug. 4-Benzoyloxy-3-methoxycinnamic acid was the compound that had the greatest activity. The group that contributed to the ligand-receptor interaction was the aromatic group with π interaction. Keywords:  Molecular Docking, AutoDock, Antifibrotic, TGF-β1, Ferulic Acid

Abstract

Previous studies had shown that ethyl p-methoxycinnamate (EPMS) had various bioactivities, such as analgesic and anti-inflammatory through COX inhibition. Due to its unique structure, this compound was modified into a selective COX-2 inhibitor. In this research, EPMS was isolated from Kaempferia galanga L. and used as a starting material to be reacted with hydrazine hydrate to obtain 3-(4-methoxyfenil)propanehydrazide. This synthesis was carried out using conventional heating and microwave irradiation. The microwave irradiation was chosen because of its potential to reduce reaction time and increase yield. Next, the crude product was purified using column chromatography to produce a pure product, where characterization and structural analyses of the compound were carried out using the TLC test, melting point test, IR, 1H-NMR, and UV spectroscopy. The results showed that the product produced was 3-(4-methoxyphenyl)propanehydrazide with a yield of 0,76% by conventional thermal heating and 7,90% by microwave irradiation. Keywords:  EPMC, Hydrazine Hydrate, 3-(4-Metoxyphenyl)propanehydrazide, Conventional Heating, Microwave Irradiation

Abstract

Introduction: In emergency conditions such as the COVID-19 pandemic, many drugs treating SARS-CoV-2 are currently being developed through the concept of repurposing the existing drugs. This study aims to evaluate the physicochemical, pharmacokinetic and toxicity (ADMET) profiles of 15 drugs that have been used clinically for other viral diseases, and inspect their activity as antivirals against SARS-CoV-2. Methods: The physicochemical properties were obtained by using the ChemDraw and the ADMET were predicted using pkCSM on line tools. The selected drugs based on Lipinski's Rules were docked into main protease of SARS-CoV-2 (PDB. 6LU7) using Molegro program. Results: Six drugs complied with Lipinski's Rules and showed good ADMET profile except for their hepatotoxicity, but favipiravir and oseltamivir were predicted to be non-hepatotoxic. Oseltamivir also showed high binding affinity with free energy score below -100 kcal/mol. Conclusions: Oseltamivir is a potential antivirus for COVID-19 based on physicochemical, ADMET profile, and in silico activity against protease of SARS-CoV-2.

Abstract

Background: Cancer is one of the top causes of death worldwide. A wide range of illnesses known as cancer can start in almost any organ or tissue in the body when abnormal cells multiply uncontrollably. Cancer patients have higher levels of the Topo IIα protein in their cells, this protein has been proposed as a relevant target for anticancer treatment development. Objective: This study aims to predict the anticancer activity of pinostrobin and 5-O-acylpinostrobin derivatives against topoisomerase IIα by docking molecular and QSAR study. Methods: In silico analysis was performed using the structure of the topoisomerase IIα (PDB: 5GWK)) as templates. Molecular docking analysis was performed with AutoDock Vina. Result: All 5-O-acyl pinostrobin derivatives, showed lower ΔG values than the parent pinostrobin. The 5-O-acetyl pinostrobin compound showed the highest score, namely -9.14 kcal/mol. 5-O-acetyl pinostrobin is predicted as the most powerful inhibitor that can cause inhibition of topoisomerase IIα. Conclution: The results of the best QSAR equation obtained can be used as a reference for predicting the activity of the new pinostrobin derivatives to be synthesized by inserting the electronic (Etot) parameter values of the compounds into the equation.
