Abstract

Rendang is one of the popular foods in Indonesia made from beef as the main ingredient, which has great potential in tourism development. As a country that has a Muslim majority population, this is very influential for the halal tourism industry. Halal tourism is the parent of tourism in accordance with Islamic principles. The issue of halal food is a sensitive issue for the community. The rise of mixing of non-halal ingredients such as pork in processed food products has worried the public, especially for adherents of the Islamic religion. However, this can be detected molecularly using the Polymerase Chain Reaction (PCR) technique which will amplify the DNA of the target gene of the species to be identified. Each gene has a primer with a different concentration and annealing temperature, so that prior to PCR with research samples, it is necessary to optimize the primer concentration and annealing temperature in order to obtain the appropriate PCR composition and conditions so as to obtain optimal PCR results. In this study, optimization of bovine-specific BOS primers that amplify the ND5 gene in mitochondrial DNA was carried out, namely ND5 (NADH dehydrogenase subunit 5). The results showed that the optimum annealing temperature was 48.2ºC, the optimum primary concentration was 0.5 μM.

Abstract

Indonesia memiliki varietas jeruk lokal yang tidak kalah dengan jeruk impor, terutama dalam hal kesegaran buah. Namun, masyarakat lebih tertarik dengan warna kulit jeruk sehingga masyarakat lebih memilih jeruk impor ketimbang jeruk lokal, khususnya di Sumatera. Oleh karena itu, perlu dilakukan upaya pelestarian dan peningkatan karakteristik jeruk ini untuk meningkatkan kualitasnya melalui pemuliaan tanaman. Penelitian ini bertujuan untuk mengoptimalkan metode isolasi DNA sampel buah jeruk dengan Chelex-TE dan untuk mengetahui profil genetik jeruk lokal Sumatera dan jeruk impor menggunakan genotipe RAPD. Sampel yang digunakan adalah beberapa jeruk lokal di Sumatera (Citrus Siam Gunung Omeh, Citrus Madu, Citrus Keprok Maga, Citrus Keprok Brastepu dan Citrus Pasaman) dan Citrus impor (Citrus Sunkist, Citrus Clemengold, Citrus Murkot dan Citrus Wokam). DNA diisolasi menggunakan metode Chelex-TE 10% yang dioptimalkan untuk beberapa parameter seperti ukuran butir, kulit buah dan daun. RAPD dilakukan dengan menggunakan 10 primer RAPD. Hasil penelitian menunjukkan bahwa metode isolasi Chelex Chelex-TE 10% yang optimal adalah ukuran sampel 1 butir. Amplifikasi jeruk lokal Sumatera dan jeruk impor menggunakan 10 primer tunggal menghasilkan pita polimorfik. Nilai kesamaan jaccard menunjukkan bahwa kelima sampel jeruk tersebut merupakan varietas lokal Sumatera yang diamati memiliki variasi genetik yang tinggi dibandingkan dengan keempat jeruk impor tersebut. Abstract

Endophytic bacteria have been extensively studied as a source of new antimicrobial compounds. Research into new antimicrobials is important as an alternative to the treatment of resistant microbes. One potential source of antimicrobial compounds is the Taxus (Taxus sumatrana). This study aimed to investigate the antimicrobial activity produced by the endophytic bacteria of the Taxus. Endophytic bacteria were isolated from the roots of Taxsus plants growing in Mt. Singalang, X Koto District, West Sumatra. Bacteria were isolated by inoculating pieces of Taxus root tissue into a PDA medium. Endophytic bacteria were identified macroscopically and microscopically. Antimicrobial activity was tested with the test microbe using the spot test diffusion method. As a result of the research, 23 isolates of endophytic Taxus bacteria were obtained, of which 21 isolates were gram-positive and 2 isolates were gram-negative bacteria. The results of the antimicrobial activity test yielded only 2 isolates of endophytic bacteria with antibacterial activity.

Abstract

Advances in the field of biotechnology have led to the use of enzymes in industry increasingly. The high public awareness of environmental pollution makes enzyme technology as an alternative to replace various chemical processes. Enzymes can be used in various industrial sectors such as textiles, food, detergents, paper and cosmetics, as well as biofuels. One of the enzymes that is often used is the enzyme xylanase. Thermophilic microorganisms that can produce thermostable enzymes are thermophilic bacteria. One of the important factors affecting the success of the manufacturing process of the production of the xylanase enzyme is the concentration of the starter. The production of the thermophilic bacterial xylanase enzyme SSA 4 produced the highest specific enzyme activity at a concentration of 4% which was 0.137 U/mg. This shows that low starter concentrations can overwhelm microbial growth well. The increase in the concentration of the starter causes the enzyme activity to decrease, this happens.

Abstract

This study aims to determine the Knowledge and Skills of Science Teachers MGMP IPA North Region Solok Regency in Planning, Implementing and Evaluating 21st Century Learning This research is a type of descriptive research. The population in this study came from the North Region Science MGMP of Solok Regency. The sample of this study came from SMPN 1 X Koto Diatas, SMPN 1 Singkarak and SMPN 1 Junjung. The sample technique is purposive sampling. Data collection techniques are tests, observations, questionnaires, interviews and documentation. Data analysis in the form of quantitative and qualitative descriptive analysis. The results of the study can be concluded that the knowledge and skills of MGMP IPA science teachers in the northern region of Solok district are very good in planning, implementing and evaluating students' 21st century learning. This can be seen from the 21st century learning analysis value of SMP 1 X Koto Diatas of 78.15 good category, SMPN 2 of 78.56 good category Kubung and SMPN 1 X Koto Singkarak of 89.46 excellent category. So, teachers are guided to be able to apply 21st century learning, so as to improve the quality of Indonesian education

Abstract

Biofuel is an alternative energy to replace fossil fuels. The most popular form of biofuel today is bioethanol. Biofuels are considered as a suitable alternative to fossil fuels because they are more environmentally friendly. Production of bioethanol utilizing thermophilic microorganisms is more profitable because thermophilic microbes generally have the characteristics of being able to use a variety of substrates, low contamination, and resistance to high temperatures. The use of microbial consortia tends to give better results than the use of monoculture isolates to optimize bioethanol yields. The aim of this study was to determine the compatibility and ability of the best thermophilic bacterial triculture consortium from the Mudiak Sapan hot springs in producing biofuel. This research is a descriptive research, to test isolates of a consortium of thermophilic bacteria that produce bioethanol. The results of this study showed that consortium isolates tended to give better results than monoculture isolates. The best thermophilic bacterial triculture consortium from the Mudiak Sapan hot springs in producing biofuels namely MS 12, 17, 18 produced a bioethanol content of 0.863%.

Abstract

This study aims to produce learning videos for Bacteria and Protists using Sparkol Videoscribe software for class X IPA students that are valid, practical, and effective. This research method is research and development (research and development) using the Plomp development model. This development stage consists of three stages, namely the preliminary research phase, the development or prototyping phase, and the assessment phase. The results of this study indicate that the developed learning videos have a validity value of 85.63% with a very valid category based on expert judgment. The practicality assessment by the teacher was 89.13% in the very practical category and the large group evaluation assessment by students obtained a practicality value of 87.69% in the very practical category. The results of the effectiveness test showed that the class that used learning videos had higher average competency values of knowledge, attitudes, and skills, compared to classes that did not use learning videos. It can be concluded that the learning videos that have been developed are in the category of very valid, very practical, and very effective for increasing students' learning competence

Abstract

Technological developments demand that education is directed at learning that equips students with 21st century skills. One of them is critical thinking skills. Students' critical thinking skills can be developed through the use of appropriate learning models and supported by prior knowledge that students already have. However, the use of learning models applied in schools has not optimally developed students' critical thinking skills and tests have never been carried out to determine students' prior knowledge. One of the learning models that can develop critical thinking skills is the problem-based learning model. The purpose of this study was to determine the effect of problem-based learning models and prior knowledge on the critical thinking skills of class VIII students of SMP Negeri 1 Lengayang. This type of research is a quasi-experimental with a population of class VIII students at SMP Negeri 1 Lengayang in the odd semester of the 2022/2023 academic year. Taking two sample classes using purposive sampling technique. The design of this research is Randomized Control-Group Posttest Only Design. The instrument used in this study was a written test. The data analysis technique for testing the hypothesis is the independent sample t test and the two-way ANOVA test. The results of this study prove that: The problem-based learning model influences the critical thinking skills of class VIII students of SMP Negeri 1 Lengayang; the problem-based learning model and high prior knowledge affect the critical thinking skills of class VIII students of SMP Negeri 1 Lengayang; The problem-based learning model and low prior knowledge affect the critical thinking skills of class VIII students of SMP Negeri 1 Lengayang; The problem-based learning model has no interaction with prior knowledge in influencing the critical thinking skills of class VIII students of SMP Negeri 1 Lengayang

Abstract

Rice is the main food source for Indonesians. The demand for rice to meet people's needs increases every year due to population growth and efforts to improve nutrition by the community. One of the problems that can affect rice production is drought. Drought is an environmental condition when plants do not get enough water to grow and develop optimally, which can cause a decrease in rice production. To find out how rice growth responds to drought, a study was carried out by giving drought simulation treatments using polyethylene glycol (PEG) on several rice varieties. This study used a completely randomized design that was arranged in a factorial manner with two factors. The first factor was the rice varieties (Harum, Situbagendit, Rosna) the second factor was 0% and 20% PEG concentration. The data obtained were then analyzed statistically using a two-way ANOVA test, and if the results were significantly different, then proceed with Duncan's test at the 5% level.The results showed that the drought simulation treatment had a negative effect on rice growth. Drought simulation treatment using 20% PEG resulted in a decrease in Kadar air relatif (KAR), root length, plant height, and root dry weight of rice. The highest decrease in KAR was found in the sensitive rice variety (Rosna), which was 43.42%. The highest average root length (7.99 cm) was on the sitabaendit variety, and the lowest (5.61) was on the rosna variety. The highest average crown height (17.32 cm) and the lowest (6.61) were on the rosna variety.

Abstract

Refill Drinking Water Depots (DAMIU) in the community are easy to find at affordable prices, which is a concern in the feasibility of refill drinking water quality. E.coli is one of the pathogenic bacteria present in drinking water that has poor quality. When it enters the body it can cause symptoms of diarrhea, fever, vomiting and others. The purpose of this study was to create a specific primer for E.coli that can be used to detect E.coli and determine the optimum temperature of primer annealing. Researchers do a new primer design because the existing design does not necessarily produce the same results due to various factors of different experimental conditions. The primer design was carried out on an in silico-based and had to meet the criteria for a good primer because it would be used in vitro. E.coli gene sequences was aligned with Shigella sp. using Pairwise Alignment. Primer candidates were analyzed using NCBI's Primer3 and Geneious Prime tools. The result is that the first primer pair is forward 5'–ATGCAGTGGTTCCTTATCTCACA-3' reverse 5'- ATCCTTAATGGCACTGCGCT-3', amplifying the amplicons along 417 bp in the yraJ gene. The second primer pair, forward 5'-CAGAACGTTTTTCATTCAGCAGG-3' reverse 5'-GCCACTACCAGATCGAGTCA-3' amplifies the 573 bp amplicon in the rne gene. The optimum annealing temperature for both pairs of primers was 59.5oC.
