Abstract

This study aims to produce a valid biology module based on emotional spiritual quotient (ESQ) in an evolution topic for senior high school students grade XII. This conducted research used the Plomp development method. In this model, there were three steps established, such as; preliminary research, developing or prototype phase, and assessment phase. For implementation validity, the research subjects were a biology teacher and five biology lecturers. The instrumentation used were; interview questionnaire, self-evaluation sheet, and validity questionnaire. The obtained data analyzed by qualitative and quantitative methods. From the research, yield a new biology module with ESQ based in evolution lesson for senior high school students grade XII. Based on the expert review, obtained the score of validity 3.71 which indicated very valid criteria.

Abstract

Refill Drinking Water Depots (DAMIU) in the community are easy to find at affordable prices, which is a concern in the feasibility of refill drinking water quality. E.coli is one of the pathogenic bacteria present in drinking water that has poor quality. When it enters the body it can cause symptoms of diarrhea, fever, vomiting and others. The purpose of this study was to create a specific primer for E.coli that can be used to detect E.coli and determine the optimum temperature of primer annealing. Researchers do a new primer design because the existing design does not necessarily produce the same results due to various factors of different experimental conditions. The primer design was carried out on an in silico-based and had to meet the criteria for a good primer because it would be used in vitro. E.coli gene sequences was aligned with Shigella sp. using Pairwise Alignment. Primer candidates were analyzed using NCBI's Primer3 and Geneious Prime tools. The result is that the first primer pair is forward 5'–ATGCAGTGGTTCCTTATCTCACA-3' reverse 5'- ATCCTTAATGGCACTGCGCT-3', amplifying the amplicons along 417 bp in the yraJ gene. The second primer pair, forward 5'-CAGAACGTTTTTCATTCAGCAGG-3' reverse 5'-GCCACTACCAGATCGAGTCA-3' amplifies the 573 bp amplicon in the rne gene. The optimum annealing temperature for both pairs of primers was 59.5oC.

Abstract

Endphytic microbes are microscopic living organisms (bacteria and fungi) that live in plant tissues (xylem and phloem), leaves, roots, fruit, and stems. These microbes live symbiosis of mutual benefit. Morus macroura miq. or Andaleh is a native plant of western Sumatra that is considered rare. The purpose of this study was to isolate and identify entophytic bacteria found in young leaves and old leaves andaleh taken from the village Andaleh, Kec. Batipuah West Sumatra. The methods used to isolate endophytic bacteria from andaleh leaves are spread plate and streak plate. Identification of endophytic bacteria in macroscopic or colony morphology. A total of 12 isolates of endophytic bacteria from young andaleh leaf and 8 isolates of endophytic bacteria from andaleh leaf. Identification Microscopic using gram staining obtained bacteria 18 gram positive endophytic bacterial isolates and 2 isolate gram negative bacteria.

Abstract

Cassava (Manihot esculenta) is one of the food crops that contain high carbohydrate and is used as a raw material for various industries. Cassava also contains several types of proteins, including beta-carotene. However, some plants also contain various levels of beta-carotene. Beta-carotene is one of the groups of carotenoids that serve as precursors of vitamin A. This bioinformatics study aims to determine the similarities and differences in the enzyme encoding genes involved in carotenoid biosynthesis pathways in cassava with several plants containing beta-carotene from nucleotide sequences and encoding gene proteins carotenoids (Lcy α and Lcy β). Data on nucleotide and protein sequences were obtained through the NCBI database. Similarities and differences in nucleotide and protein sequence data are seen from the phylogenetic tree using the MEGA 7.0 program. Based on the nucleotide gene Lcy α Manihot esculenta with several plants containing beta-carotene is not related. Based on the phylogenetic tree nucleotide sequence protein Lcy α and Lcy β Manihot esculenta with several plants containing beta-carotene there were differences in similarity

Abstract

Dental and oral infection problems can be overcome using natural antimicrobial compounds from bacterial endophytic resulted from bacterial fermentation. This study aims to determine the effectiveness of extracts of Andalas endophytic bacterial fermentation products in treating oral cavity infections. Fermentation products was extracted using maceration methods. Antimicrobial activity of Andalas endophytic bacterial fermentation product was evaluated to S. mutans and C. albicans by disk diffusion method. The study showed that the optimum concentration of extract in inhibiting microbial growth that causes oral cavity infections was at 12.5%.

Abstract

Anti-Biotic resistance is a health problem globally. It is able to overcome with a new anti-microbial compound that can be produced from Andalas endophytic bacteria. This compound can be obtained through the fermentation process. In order to separate this active-compound, it is needed to use the extraction method. In this method, the solvent is functioned as the extractor. One of the solvent which is commonly used is ethanol. This research is aimed to know the effect of ethanol concentration toward antibacterial activity from extracted bacterial fermentation products of Andalas endophytic bacteria isolate JDT 1B. The fermented products are extracted by using the maceration method. The concentrations are 100%, 80%, 70%. A test of anti-microbial activity is used disk diffusion method. The extracted concentrations tested for each solvent are 50%, 25%, 12,5%, and 6.25%. Anti-bacterial activity is analyzed by using factorial design. The factorial result showed there is no significant contrast between ethanol concentration mentioned toward anti-bacterial activity from extracted bacterial fermentation products of Andalas endophytic bacteria Isolate JDT 1B. The concentration of extracted fermentation product using 70% ethanol has the same inhibition zone as control is 6,25%.

Abstract

Tuberculosis (TB) is a serious disesase in the world. According to the WHO, it is estimated more than 3 million people die every year as a result of this infectious disease. One factor that causes diffi culty handling TB chemoteraphy is not effective against the bacteria Mycobacterium tuberculosis that causes TB . Effectiveness of treatment is often hampered by the emergence of bacterial resistance against M. Tuberculosis chemotherapy agents are given. From some research found that bacterial resistance may occur in more one type of chemotherapy agent also known as multi-drug resistance (MDR). Mycobacterium tuberculosis develop resistance mechanisms that are different from other bacteria in general. In prokaryotes, resistance is generally due to the transfer of genetic, either through plasmids,transposons and other. Reference sequence beta sub unit of RNAP protein M. Tuberculosis with accession number NP_215181.1 and M. tucerculosis rpoB gene with accession number NC_000962.3 used to obtain preliminary information from the data base www.ncbi.nlm.gov and www.uniprot.org . Mutation done according to several studies literature. Analysis of the composition, profi le, location and structure of protein using www.expasy.org, TMHMM and http://bioinf.cs.ucl.ac.uk/psipred. The primer design is done with Primer Design Program. Based on the analysis of mutation in the beta subunit of RNAP protein M. Tuberculosis, codon 531 (Ser ->Leu), it is known that mutations cause changes in some properties and structure of proteins. Possible changes affecting the nature of bacterial resistance to antibiotics rifampicin. However, further analysis needs to be done with the analysis of the docking technique.

Abstract

Apis cerana, which is often known as honey bee, has spread from Afghanistan, China, Japan, to Indonesia. This distribution cannot be separated from Apis cerana's ability to survive by needing food. Pollen and nectar are the main food sources for Apis cerana in maintaining life. Apis cerana is widely cultivated by the community with its potential. This article aims to describe in more detail about the forage plants from Apis cerana and how the feeding strategies of honey bees were assembled from several studies in various regions, both in Indonesia and outside Indonesia as well as comparing these daily activities with other Apidae. The discussion is focused on the comparative analysis of the daily activities of Apis cerana with other Apidae to see the uniqueness of Apis cerana which is a factor in the number of bees being cultivated.

Abstract

This study aims to determine the effectiveness of the use of integrated blended learning on multiple intelligences (multiple intelligence) and student character education during the COVID-19 period. This type of research is a meta-analysis research. The research sample came from a search of 21 national and international journals. Search articles through databases sourced from Google Scholar, Eric, IEEE, Science Director and Wiley. The sample selection technique is purposive sampling technique. The data sampled relates to the use of Blended Learning, Multiple Intelligence and character education. Data analysis used quantitative data analysis techniques by conducting effect size and N-Gain tests using JASP software version 26. The results showed that the use of STEM integrated blended learning was effective in increasing students' multiple intelligence with an average value of 76.25 and student character education of 75 with the value of effect size (ES) is 0.44 and the N-Gain is 0.65 category medium.

Abstract

The purpose of this study was to determine the potency and optimum pH of AAS thermophilic bacteria isolates in producing bioethanol. This research was carried out from September 2021 to December 2021, at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Padang State University. This research is an experimental study consisting of 6 treatments and 3 replications with variations in incubation pH of 6.5, 7, 7.5, 8, 8.5, and 9. The data obtained were analyzed by means of variance (ANOVA) with Duncan's further test. New Multiple Range Test (DNMRT). The results showed that the isolates of thermophilic bacteria SSA had the potential to produce bioethanol with isolates SSA 8 being the producer of bioethanol with the highest concentration of 1.001%, and variations in incubation pH affected the concentration of bioethanol produced with the optimum pH of thermophilic bacteria in producing bioethanol, namely pH 8 which produced the highest concentration of bioethanol is 1.001%.Tujuan dari penelitian ini adalah untuk mengetahui potensi serta pH optimum isolat bakteri termofilik SSA dalam menghasilkan bioetanol. Penelitian ini dilaksanakan pada bulan September 2021 hingga bulan Desember 2021, di Laboratorium Mikrobiologi  Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri Padang. Penelitian ini merupakan penelitian eksperimen yang terdiri dari 6 perlakuan dan 3 ulangan dengan variasi pH inkubasi 6,5, 7, 7,5, 8, 8,5, dan 9. Data yang diperoleh dianalisis dengan sidik ragam (ANOVA) dengan uji lanjut Duncan’s New Multiple Range Test (DNMRT). Hasil penelitian menunjukkan bahwa isolat bakteri termofilik SSA memiliki potensi dalam menghasilkan bioetanol dengan isolat SSA 8 merupakan penghasil bioetanol dengan kadar tertinggi yaitu 1.001%, dan  variasi pH inkubasi berpengaruh terhadap konsentrasi bioetanol yang dihasilkan dengan pH optimum bakteri termofilik dalam menghasilkan bioetanol yaitu pH 8 yang menghasilkan konsentrasi bioetanol tertinggi yaitu 1.001%.
