Abstract

Cases of bacterial resistance to antibiotics are discussed with serious problems in the world of health. New antimicrobial compounds are needed which are more effective in treating infectious diseases. Isolate B.J.T.A-6 is an endophytic bacteria from Andalas plants (Morus macroura Miq.) which is known to be able to produce antimicrobial active compounds. Antimicrobial compounds can be produced by growing them on fermented media. The purpose of this study was to optimize the medium fermentation of Andalas endophytic bacteria of B.J.T.A-6  isolates in producing antimicrobial compounds. While medium fermentation is Nutrient Broth (NB), Muller Hinton (MH), and Luria Bertani Broth (LB). Antimicrobial activity tests were carried out by means of diffusion. The parameters used were the diameter of the inhibition zone against Staphylococcus aureus around the disc paper. The profile of medium fermentation optimization was analyzed statistically. The results showed that B.J.T.A-6  isolates could inhibit the highest growth of S. aureus by using LB fermentation medium.

Abstract

This study aims to determine variations in the expression of ZDS and LCYb enzymes encoding several types of yellow tubers. Cassava used, namely Carvita 25, Nangka and Mentega 2 (yellow bulb) and Menti (white bulb) were used as controls. The target enzyme coding genes (ZDS and LCYb) in cassava were identified in an online database, Phytozome using the BLAST method. Design and primary analysis for the target genes was carried out using online software, OligoAnalyzer and PCR. The expression of the target enzyme coding gene was analyzed using the Reverse Transcription PCR (RT-PCR) method. The BLAST results in the cassava genome in the Phytozome database showed that the enzymes ZDS and LCYb in cassava were encoded by two genes. The primary optimization results showed the primary annealing temperature for the LCYb enzyme coding gene was 55 ° C. The ZDS primers were not amplified after several replications, so that only the LCYb enzyme coding gene was selected for qualitative gene expression analysis using RT-PCR. A positive control in the analysis of gene expression is the housekeeping gene, PP2A. Electrophoresis results of PCR products (RT-PCR) showed negative results (no DNA bands were detected) in all samples from both yellow and white tuber samples and its housekeeping gene. This is possible from the low quality of RNA used in cDNA synthesis.

Abstract

Trichoderma spp adalah jamur non patogen yang berasosiasi dengan rhizosfer dan daun tanaman. Jamur ini juga mampu mengendalikan penyakit dan menghasilkan zat pengatur tumbuh, sehingga potensial  digunakan dalam pertaniankhususnya dalam budidaya pertanian organik.  Penelitian  kemampuan Trichoderma spp dalam meningkatkan pertumbuhan benih padi masih terbatas. Penelitian ini, melaporkan kemampuan Trichoderma setempat untuk meningkatkan poertumbuhan kecambah  benih padi. Hasil penelitian menunjukkan bahwa isolat Trichoderma mampu meningkatkan daya kecambah dan vigor benih padi. Hasil penelitian membuktikan bahwa isolat Trichoderma indegenous memiliki potensi untuk digunakan sebagai pengatur pertumbuhan benih padi. Hasil penelitian meningkatkan pemahaman lebih lanjut tentang peranan jamur dalam meningkatkan pertumbuhan padi

Abstract

The number of death couse of infection continues to increase. Treatment of infections is increasingly difficult due to increased resistance of germs to existing antimicrobial compounds. A new source of antimicrobial substances is needed. One source of antimicrobial substances is endophytic bacteria that live in the stems of Andalas plants. The aim of this study was to compare the diversity of endophytic bacteria present in old and young Andalas plant stems. The method used to isolate endophytic bacteria from the stem of the Andalas plant is the streak plate method. Bacterial identification is done macroscopically and microscopically. The results of macroscopic observation showed that there are differences in the number of isolates found in old and young Andalas plant stems. 11 isolates of endophytic bacteria from Andalas plant stems, each of which consists of 9 endophytic bacteria from old Andalas stems and two isolates from young Andalas stems. A microscopic identification was found that of the 11 isolates isolated, eight isolates were gram-positive and three isolates were gram-negative bacteria

Abstract

DNA bands formed from the results of electrophoresis with Polyacrilamide gel are considered as 1 character representing 1 allele. PCR products produce multiple bands (multy bands), which indicates that there are multi alleles in the sample. Electrophoresis is a chemical analysis method based on the movement of charged protein molecules in the electric field. Separation is carried out based on differences in the size of the molecular weight and the electric charge contained by the macro-molecule. In addition, the effect of gel concentratio n, buffer and electrophoresis time also has a role in the results of electrophoresis. This study was conducted to compare the best separation time for acrilamide gel electrophoresis with the results of cassava DNA amplification. The materials used in this study are two cassava varieties, namely: Adira IV 1, Adira IV 2, Adira IV 3, Carvita 25 1, Carvita 25 2, and Carvita 25 3. Using electrophoresis by poly-acrilamide gel with two different time effects: 1 hour 30 minutes and 3 hours. The results of electrophoresis with 3 hours gave better results of DNA visualization compared to 1 hour 30 minutes.

Abstract

Dinding sel jamur Trichoderma tersusun atas senyawa kitin yang sangat kokoh dan resisten terhadap aktivitas enzim. Teknik dan metode spesifik diperlukan untuk mendegradasi dinding sel jamur. Penelitian ini bertujuan untuk menganalisis metode isolasi DNA terbaik dalam melisiskan dinding sel jamur dengan membandingkan beberapa metode isolasi. Metode isolasi yang digunakan adalah DNeasy Plant Mini Kit, kombinasi DNeasy Plant Mini Kit dan pemanasan, serta kombinasi DNeasy Plant Mini Kit dan cara fisik (menggerus sampel dengan menggunakan mortar dan pestle dalam nitrogen cair). Penelitian ini dilakukan pada bulan Maret sampai September 2018 yang bertempat di Laboratorium Mikrobiologi, Laboratorium Bioteknologi dan Genetika, serta Laboratorium Penelitian Terpadu, FMIPA, UNP. Data dianalisis secara kualitatif dengan elektroforesis gel agarose dan secara kuantitatif dengan menghitung nilai kemurnian dan konsentrasi DNA menggunakan Nanodrop. Hasil penelitian menunjukkan bahwa metode kombinasi DNeasy Plant Mini Kit dengan cara fisik memberikan hasil isolasi DNA dengan konsentrasi yang tertinggi yaitu 2,4 μg/ml dengan nilai kemurnian 1,857. Pada metode DNeasy Plant Mini Kit dan metode kombinasi DNeasy Plant Mini Kit dengan pemanasan, konsentrasi DNA yang diperoleh berturut-turut 1,211 µg/ml dan 0,933 µg/ml dengan nilai kemurnian 1,728 dan 1,708. Hasil uji elektroforesis menunjukkan adanya pita DNA berupa garis tipis pada sampel yang diisolasi dengan kombinasi metode kit dan cara fisik, sedangkan pada kedua sampel lainnya tidak ditemukan adanya pita DNA. Dapat disimpulkan metode isolasi DNA yang dapat dijadikan rujukan untuk mendegradasi dinding sel jamur Trichoderma adalah metode kombinasi kit dengan nitrogen cair dengan catatan dibutuhkan penambahan kadar isolat yang digunakan.Kata Kunci: Isolasi DNA, Dinding sel, Trichoderma

Abstract

The purpose of this study was to determine the potency and optimum pH of AAS thermophilic bacteria isolates in producing bioethanol. This research was carried out from September 2021 to December 2021, at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Padang State University. This research is an experimental study consisting of 6 treatments and 3 replications with variations in incubation pH of 6.5, 7, 7.5, 8, 8.5, and 9. The data obtained were analyzed by means of variance (ANOVA) with Duncan's further test. New Multiple Range Test (DNMRT). The results showed that the isolates of thermophilic bacteria SSA had the potential to produce bioethanol with isolates SSA 8 being the producer of bioethanol with the highest concentration of 1.001%, and variations in incubation pH affected the concentration of bioethanol produced with the optimum pH of thermophilic bacteria in producing bioethanol, namely pH 8 which produced the highest concentration of bioethanol is 1.001%.Tujuan dari penelitian ini adalah untuk mengetahui potensi serta pH optimum isolat bakteri termofilik SSA dalam menghasilkan bioetanol. Penelitian ini dilaksanakan pada bulan September 2021 hingga bulan Desember 2021, di Laboratorium Mikrobiologi  Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri Padang. Penelitian ini merupakan penelitian eksperimen yang terdiri dari 6 perlakuan dan 3 ulangan dengan variasi pH inkubasi 6,5, 7, 7,5, 8, 8,5, dan 9. Data yang diperoleh dianalisis dengan sidik ragam (ANOVA) dengan uji lanjut Duncan’s New Multiple Range Test (DNMRT). Hasil penelitian menunjukkan bahwa isolat bakteri termofilik SSA memiliki potensi dalam menghasilkan bioetanol dengan isolat SSA 8 merupakan penghasil bioetanol dengan kadar tertinggi yaitu 1.001%, dan  variasi pH inkubasi berpengaruh terhadap konsentrasi bioetanol yang dihasilkan dengan pH optimum bakteri termofilik dalam menghasilkan bioetanol yaitu pH 8 yang menghasilkan konsentrasi bioetanol tertinggi yaitu 1.001%.

Abstract

Advances in the field of biotechnology have led to the use of enzymes in industry increasingly. The high public awareness of environmental pollution makes enzyme technology as an alternative to replace various chemical processes. Enzymes can be used in various industrial sectors such as textiles, food, detergents, paper and cosmetics, as well as biofuels. One of the enzymes that is often used is the enzyme xylanase. Thermophilic microorganisms that can produce thermostable enzymes are thermophilic bacteria. One of the important factors affecting the success of the manufacturing process of the production of the xylanase enzyme is the concentration of the starter. The production of the thermophilic bacterial xylanase enzyme SSA 4 produced the highest specific enzyme activity at a concentration of 4% which was 0.137 U/mg. This shows that low starter concentrations can overwhelm microbial growth well. The increase in the concentration of the starter causes the enzyme activity to decrease, this happens.

Abstract

Rice is the main food source for Indonesians. The demand for rice to meet people's needs increases every year due to population growth and efforts to improve nutrition by the community. One of the problems that can affect rice production is drought. Drought is an environmental condition when plants do not get enough water to grow and develop optimally, which can cause a decrease in rice production. To find out how rice growth responds to drought, a study was carried out by giving drought simulation treatments using polyethylene glycol (PEG) on several rice varieties. This study used a completely randomized design that was arranged in a factorial manner with two factors. The first factor was the rice varieties (Harum, Situbagendit, Rosna) the second factor was 0% and 20% PEG concentration. The data obtained were then analyzed statistically using a two-way ANOVA test, and if the results were significantly different, then proceed with Duncan's test at the 5% level.The results showed that the drought simulation treatment had a negative effect on rice growth. Drought simulation treatment using 20% PEG resulted in a decrease in Kadar air relatif (KAR), root length, plant height, and root dry weight of rice. The highest decrease in KAR was found in the sensitive rice variety (Rosna), which was 43.42%. The highest average root length (7.99 cm) was on the sitabaendit variety, and the lowest (5.61) was on the rosna variety. The highest average crown height (17.32 cm) and the lowest (6.61) were on the rosna variety.

Abstract

Rice has become food for most of the world's population. Indonesia is the third largest producer in the world. However, in fact rice production in Indonesia has decreased by 0.43% compared to 2020. One of the factors that can cause this decline is drought. Because rice is a semi-aquatic plant that grows normally in flooded conditions, it makes drought stress very threatening. Drought stress that occurs in plants causes plants to experience oxidative stress due to excessive accumulation of ROS. PEG is a compound that is widely used to provide drought conditions in plants. Previous research has classified several varieties of rice plants based on their level of resistance to drought. However, it is not yet known how the morphological response will be in different periods of drought stress and rewatering treatment. This research was conducted by giving treatment in the form of control (Yoshida nutrient culture solution) and drought stress (Yoshida + PEG-6000 20% solution) repeated 3 times. The observed parameters were RWC which were analyzed using standard errors and morphological images of roots and leaves. The results showed that the RWC obtained during the stress period from the third to the fifth day, Harum had the highest value according to its class as tolerant rice. After rewatering Rosna has a better recovery ability. In addition, root morphology shows differences in the form of root length, small root diameter, inhibition of adventive root growth. On the leaves include a decrease in leaf area, leaf curl up, and leaf yellowing.
