Abstract

Cynometra ramifolia Linn is known as Sala plant. This plant, especially in Solo City categorized as rare plant and traditionaly used to cure uric acid, diabetes, hypertension, and others pain. It is interesting to know the other activities by done the test for human cancer cell-lines. This research objective to identify the cytotoxicity effects of ethanol extract from C.ramiflora the stem bark and leaves against T47D, HeLa and WiDr cancer cell lines. The cytotoxicity test of ethanol extract from C.ramiflora the stem bark and leaves determined by MTT assay by calculating the level of IC50  which was based on the percentage of the cell death following the 24 hours incubation with the extract. The results  showed that ethanol extract of stem bark has cytotoxic effect to HeLa, T47D and WiDr cell-lines with the IC50  of 1000; 0,90 and 6,29 g/mL respectively. The leave cytotoxic effect to HeLa, T47D and WiDr cell-lines with the IC50 of 1,92;  6,37 and 0,41 g/mL respectively. This research indicated that the ethanol extract isolated from C. Ramiflora leaves a selective cytotoxicity effect to WiDr cell line.

Abstract

Mangrove plants as traditional medicine have long been used by the society for the therapy of anticancer diseases. So far, the potency of mangrove plants as anticancer has not been studied intensively. The methanol extract of R. mucronata stem bark has cytotoxic activity on myeloma cells. This study aims to determine the isolates of the ethyl acetate fraction of the methanol extract of the stem bark of R. mucronata and R. apiculata, and to examine the potency of cytotoxic activity against MCF-7 and T47D cells. The powder of the stem bark of R. mucronata and R. apiculata were extracted with methanol by maceration, and then the methanol extract was fractionated consecutively using n-hexane, ethyl acetate, and methanol. Ethyl acetate fraction was then purified by column chromatography with a diameter of 2 cm and a height of 30 cm with a static phase in the form of silica gel 60 (0.063-0.2 mm) Merck 7734 and eluent n-hexane:ethyl acetate (6: 4, 5: 5, 6: 4, 7: 3, 8: 2, 9: 1) and n-hexane: methanol (9,5: 0,5) 10 ml each. The eluates that produce the same number and pattern of stains were combined into one isolate. Then the eluate was evaporated at room temperature. Then the purification results of the extract R. mucronata were isolated using preparative TLC with 0.25 mm GF254 silica gel as static phase and n-hexane mobile phase: ethyl acetate (6: 4), while the apiculata extract with n-hexane mobile phase: ethyl acetate (5: 5). The results of the stain separation of the compound were scraped off and separated from the static phase using a solvent. The purity of isolates compound was examined using TLC in the static phase of silica gel GF254 0.25 mm. The qualitative test results of the ethyl acetate fraction of R. apiculata stem bark with an NMR spectrophotometer showed a composition of cis-3- (3,5-dihydroxyphenyl) acrylic acid, while the ethyl acetate fraction of R. mucronata stem bark contained n-hexan-3-ol compounds. The cytotoxicity test of ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol extract was carried out on MCF-7 and T47D cells using the MTT method. The results proved that the ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol fraction from the ethanol extract of R. mucronata and R. apiculata stem bark, based on cytotoxic activity, were inactive against MCF-7 and T47D cells.

Abstract

Kulit kacang tanah mengandung luteolin yang berpotensi sebagai antiinflamasi secara in vitro dan in vivo. Penelitian ini bertujuan untuk mengetahui efek antiinflamasi ekstrak etanol kulit kacang tanah (Arachis hypogaea L.) pada tikus jantan galur Wistar yang diinduksi karagenin Penelitian menggunakan rancangan acak lengkap pola searah. Dua puluh lima ekor tikus putih jantan galur Wistar dibagi menjadi 5 kelompok. Kontrol negatif menggunakan CMC-Na 0,5% 2,5 mL/200 gBB, kontrol positif menggunakan Na-diklofenak 6,75 mg/kgBB. Ekstrak etanol kulit kacang tanah yang diujikan yaitu dosis 50, 100 dan 200 mg/kgBB. Larutan uji diberikan secara peroral dengan volume pemberian 2,5 mL/200 gBB 1 jam sebelum kaki hewan uji diradangkan dengan karagenin 1% secara subplantar. Pengukuran volume kaki tikus dilakukan tiap 0,5 jam selama 6 jam. Data volume udem dihitung nilai AUC (Area Under the Curve) rata-rata volume udem terhadap waktu dan persen daya antiinflamasi. Data dianalisis dengan anava satu jalan dan dilanjutkan uji LSD (Least Significant Difference) dengan taraf kepercayaan 95%.  Hasil penelitian menunjukkan bahwa ekstrak etanol etanol kulit kacang tanah (Arachis hypogae L.) dengan dosis 50, 100 dan 200 mg/kgBB mempunyai persen daya antiinflamasi berturut-turut sebesar 16,33%; 26,39% dan 31,70%, sedangkan Na-diklofenak 6,75 mg/kgBB sebesar 32,24%.Kata kunci: kulit kacang tanah (Arachis hypogaea L.), ekstrak etanol, antiinflamasi

Abstract

Previous research has shown that some compounds in leaves and seeds of sugar apple have a cytotoxic activity. The aim of this research was to determine the cytotoxicity of semipolar fraction of ethanolic extract from sugar apple stem bark (Annona squamosa L.) on T47D cancer cells. The semipolar fraction of ethanolic extract from sugar apple stem bark was collected by fractionation using Vacuum Liquid Chromatography (VLC) with hexane:ethyl acetic (9:1, 8:2, 7:3, and  6:4)  as  mobile  phase.  Cytotoxicity from the fractions of five different concentration namely; 25, 50, 100, 150, and 250, µg/mL was measured by MTT assay. The potency of the cytotoxicity was defined by the ability of the fraction to inhibit the growth of T47D cells indicated by the value of IC50. Qualitative analysis of contained compounds in the fraction was done by Thin Layer Chromatography (TLC) method using silica gel F 254 as a stationary phase and hexane:ethyl acetic (7:3) as a mobile phase. UV 254 and 366 nm lamp also Dragendorff, citroboric, and FeCl3 spray reagents were used to visualize the spots of the secondary metabolites. The result proved that the semipolar fraction of ethanolic extract from sugar apple stem bark showed potential cytotoxicity on T47D cancer cells with IC50value of 70,77 µg/mL. Qualitative analysis showed that the fraction contained flavonoids and alkaloids which is presumably responsible for its cytotoxic activity.

Abstract

Shorea accuminatissima, which is known locally as Meranti kuning, is one species of Dipterocarpaceae family. This plant contains compounds that are expected to have antioxidant activity. This research was carried out to examine the radical scavenger activity of polar fraction of methanolic extract from the tree bark of S. accuminatissima by using DPPH (2,2-difenil- 1-pikrilhidrazil) method with vitamin C as comparator. Powder of tree bark of S. accuminatissima was macerated in methanol. The methanolic extract was then evaporated to obtained dry extract. Extract with concentration of 200, 100, 50, 25 and 10 µg/mL were used to test its radical scavenger activity. The result showed that polar fraction of methanolic extract and vitamin has IC value of 319.83 and 3.72 µg/mL, respectively.

Abstract

The issue of animal feed and the efficiency of the goat breeding production process, is still a big problem for conventional goat breeders. Therefore, the use of fermented feed technology to save time providing feed, as well as the use of a communal cage system for efficiency and effectiveness in the management of goat farms, needs to be trained and mastered by goat breeders. The purpose of this IbM is to develop the capability of the goat business and increase the yields of goat farmers in the village of Sindon, kec. Ngemplak kab. Boyolali Some solutions offered to help partner problems are (1) introduction and training to goat breeders in Sindon village, Ngemplak district, Boyolali Regency regarding the use of fermented feed technology for integrated management of goat livestock, (2) training and providing technology of leaf counting machines for the production process of fermented feed goat livestock, (3) training and assistance for themanagement of goats by utilizing a communal cage system to increase the efficiency andeffectiveness of the production process. The expected outcome of this IbM activity is to increase the ability of the goat business, increase income and the effectiveness of the goat farming process in Sindon village, Ngemplak district, Boyolali district.AbstrakPermasalahan pakan ternak dan efisiensi proses produksi peternakan kambing, masih menjadi persoalan besar bagi para peternak kambing konvensional. Oleh karenanya, pemanfaatan teknologi pakan fermentasi untuk menghemat waktu penyediaan pakan, serta pemanfaatan sistem kandang komunal untuk efisiensi dan efektifitas dalam pengelolaan peternakan kambing sangat perlu dilatihkan dan dikuasai oleh peternak kambing. Tujuan IbM ini adalah pengembangan kemampuan usaha ternak kambing dan peningkatan hasil budidaya pada peternak kambing di desa Sindon, kec. Ngemplak kab. Boyolali. Beberapa solusi yang ditawarkan untuk membantu permasalahan mitra adalah (1) pengenalan dan pelatihan pada peternak kambing desa Sindon kecamatan Ngemplak Kabupaten Boyolali tentang pemanfaatan teknologi pakan fermentasi untuk pengelolaan ternak kambing terpadu, (2) pelatihan dan pemberian teknologi mesin pencacah dedaunan untuk proses produksi pakan fermentasi ternak kambing, (3) pelatihan dan pendampingan untuk pengelolaan ternak kambing dengan pemanfaatan sistem kandang komunal untuk peningkatan efisiensi dan efektifitas proses produksi. Luaran yang diharapkan dari kegiatan IbM ini adalah peningkatan kemampuan usaha ternak kambing, peningkatan pendapatan serta efektifitas proses budidaya ternak kambing di desa Sindon kecamatan Ngemplak kabupaten Boyolali.

Abstract

Indonesian original medicine requires new development, one of the important stages is by testing which can be used as a parameter to determine standards for certain extracts. Capidada leaves (Sonneratia alba J. Smith) and ketapang leaves (Terminalia cattapa L.) are a type of mangrove that have various activities such as anti-bacterial, anti-inflammatory, antioxidant, as well as the benefits of capidada leaves and ketapang leaves. Therefore, it is necessary to standardize extracts to ensure quality that matches the quality. This research tries to test the water content, ash content, and drying shrinkage levels, which later can be used as a reference for the determination of extract quality standards. The extract was obtained through maceration using 96% ethanol. Test results obtained on capidada leaves are water content (10,38 ±0,52%v/b), total ash content (17,97±1,75%), drying shrinkage levels (10,35±0,35%), and the test results obtained for the ketapang leaves are water content (7,08±0,38%), total ash content (10,51±0,43%) and drying shrinkage levels (13,67±1,58%). To find out all aspects that are not directly related to pharmacological activities but affect the aspects of quality, safety, the stability of extracts and preparations produced, this research studied the ash content, water content, and drying shrinkage levels on ethanol extracts of mangrove capidada leaves (Sonneratia alba) J. Smith.) And ketapang (Terminalia cattapa L.)

Abstract

Artocarpus had been used as traditional medicine for cancer, cirrhosis, malaria, hypertension, and diabetes. This research aimed to evaluate the cytotoxic activity of methanol leaf extract of Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi against MCF-7 breast cancer cell. The leaf extracts of Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi were obtained by the method of maceration with methanol. Phytochemical screening wasperformed by Thin Layer Chromatography with the use of silica GF-254 for stationary phase and ethyl acetate: n-hexane (2:8) for mobile phase. The cytotoxic test was conducted by MTT with diverse concentrations of extract, namely 500, 250, 125, 62.5, and 31.25 μg/mL. The results of cytotoxic test were measured by ELISA reader. Based on phytochemical screening, tannin, and alkaloids were found in three extracts. Cytotoxic test demonstrated that those extracts had low cytotoxic activity against MCF-7 breast cancer cells, with IC50 of 120.85; 339.46 and 530.88 μg/mL, respectively, for Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi.

Abstract

One of the disease that have a high contribution to the death of women in the world is breast cancer. It leads to the research and exploration to find its cure. This study is conducted to know the anti-cancer activity onethanol extract, nonpolar, semipolar, and polar fractions of lesser yam (Dioscorea esculenta L.) herbs. It also aims to find out the content of compound on semipolar fraction Dioscorea esculenta L. herbs. The extraction was carried out using maceration technique with 96% ethanol as solvent. Then, the fractination was done using vacuum liquid chromatography method with n-hexane and acetone as mobile phase with the ratio of 9:1, 8:2, 7:3, 6:4, and 5:5. After that, the phytochemical test was undertaken using the thin layer chromatographywith n-hexane and acetone as mobile phase with proportion 7:3. Meanwhile, the cytotoxic test was conducted using MTT assay method with IC50calculation was counted using linear regression between the log concentration and the percentage of living cells. The semipolar fraction of Dioscorea esculenta L. herbs has IC50value of 632,42 µg/mL, whereas the ethanol extract, nonpolar and polar fractions have the IC50 value 1000 µg/mL. Furthermore, the compound of the semipolar fraction of Dioscorea esculenta L. herbs are saponins, terpenoids, alkaloids, and phenolics.

Abstract

Mareme (Glochidion arborescens Blume.) merupakan tumbuhan liar yang banyak tumbuh di pekarangan. Daun Mareme dapat digunakan sebagai obat tradisional dan diduga mengandung senyawa aktif antioksidan golongan flavonoid dan fenol. FT-IR (Fourier Transform Infrared Spectroscopy) merupakan metode yang digunakan untuk identifikasi gugus fungsi pada suatu sampel dan salah satu metode yang sering digunakan untuk identifikasi senyawa metabolit sekunder tanaman obat. Penelitian ini bertujuan untuk identifikasi senyawa metabolit sekunder yang terkandung dalam isolat daun Mareme. Serbuk daun Mareme diekstraksi menggunakan metode maserasi, ekstrak kental diuji skrining fitokimia dan difraksinasi menggunakan Kromatografi Cair Vakum serta diisolasi menggunakan kromatotron. Isolat daun Mareme diidentifikasi menggunakan FT-IR. Ekstrak kental daun Mareme diperoleh sebanyak 95,52 gram dan mengandung senyawa metabolit sekunder flavonoid, saponin, steroid, terpenoid, tannin dan polifenolat. Fraksi non polar diperoleh sebanyak 3,75 gram, fraksi semi polar sebanyak 3,90 gram dan fraksi polar sebanyak 3,25 gram, serta diperoleh isolat sebanyak 850 mg. Isolat daun maeme memiliki gugus fungsi -OH, - CH alifatik, -C=O ester, dan -C-O, dari hasil analisis FT-IR isolat daun Mareme mengandung senyawa flavonoid.
