Abstract

Penelitian ini bertujuan melakukan isolasi dan penentuan aktivitas mikrobial senyawa bioaktif reduktiomisin dari bakteri Streptomyces sp. Ank181 daratan (terrestrial). Penelitian ini dilaksanakan di laboratorium kimia organik, Institute of Organic and Biomoleculare Chemistry, University of Goettingen, Germany. Isolat dan subkultur agar bakteri Streptomyces sp. Ank181 diperoleh dari koleksi sampel genus Streptomyces Professor Dr. H. Anke, Institute for Biotechnology and Drug Research, Kaiserslautern, Germany. Struktur reduktiomisin dalam penelitian ini dielusidasi berdasarkan data spektroskopi dan spektrometri massa. Tiga senyawa metabolit sekunder telah berhasil diisolasi dan diidentifikasi dari bakteri tanah genus Streptomyces sp. Ank181, yaitu reduktiomisin, asam 2,3-dihidroksibenzoat, dan asam indole-3-karboksilat. Hasil uji antimikroba menunjukkan bahwa reduktiomisin bersifat bioaktif terhadap bakteri, jamur dan sitotoksik terhadap Artemia salina.

Abstract

Penelitian ini bertujuan mengetahui aktivitas sulfur dan selenium nanopartikel terhadap cacing Steinernema feltiae (S. feltiae) dan perbandingan toksisitasnya terhadap sel neuroblastoma (neuro 2A). Sulfur dan selenium nanopartikel “aqueous nanoparticles” dikarakterisasimenggunakan Nano Zetasizer pada pH 7 dan suhu 25 C. Dalam penelitian ini uji nematoda dilakukan terhadap S. feltiae. Hasil penelitian menunjukkan bahwa sulfur nanopartikel mempunyai aktivitas yang sangat tinggi terhadap S. feltiae dengan lethal dose 50 (LD50) berkisar pada 6,99 μg/ml setelah 24 jam inkubasi. Sementara itu, live and dead assay dilakukan terhadap neuroblastoma sel (Neuro 2A cell lines). Hasil penelitian menujukkan bahwa ‘aqueous’ sulfur nanopartikel (NPS) menunjukkan aktivitas yang lebih baik dibandingkan dengan ‘aqueous’ selenium nanopartikel (NPSe) terhadap sel neuroblastoma (neuro 2A cell lines) dengan IC50 1 μg/ml.

Abstract

Patient satisfaction based on a patient’s opinion is the main criterion and can be one of the best ways to measure the quality of healthcare services. In this study, we determined patient satisfaction in hospital healthcare services at Zainal Abidin Hospital (RSUZA) and the socio-demographic factors associated with patient satisfaction. We conducted a cross sectional study at RSUZA. To recruit participants, purposive sampling and accidental sampling are used in this study. The binomial proportion is used to determine the total number of respondents. The questionnaire was tested for was performed. The chi-square test of independence was used to analyze the difference in each answer on each variable. The Kendall tau correlation was used to explore the correlation between social demographic variables and the total patient satisfaction value. The questionnaire used in this study is valid (p < 0.000) and reliable (Cronbach’s Alpha = 0.785). Of 904 respondents, 809 respondents (89.5%) were satisfied with the services provided by RSUZA. There was no difference on respondent in the gender category (p = 0.309), the age category (p = 0.095), and the respondent type (p = 0.377). Sociodemographic factors such as age (p < 0.000), education (p < 0.000), and occupation (p < 0.000) were significantly related to the level of patient satisfaction. We concluded that the majority of respondents are satisfied with the healthcare services provided by RSUZA. Healthcare providers need to focus on the patient’s age, educational level, and occupation in order to increase patient satisfaction, which is a benchmark for the quality of a healthcare service

Abstract

Blumea balsamifera (L.) DC. or Sembung is a flowering plant belonging to the genus Blumea of the family Asteraceae. Many pharmacological activities of this plant show potential in human therapy. In this study, an investigation was conducted on the ethanolic extract of B. balsamifera collected from a geothermal area known as Ie-Jue, in Aceh Province, Indonesia. The results showed that the ethanolic extract of B. balsamifera contained secondary metabolites of flavonoids and tannins. Chemical constituents of ethanolic extracts B. balsamifera further analysis using gas chromatography-mass spectrometry (GC-MS) show that active compounds from this plant was Proximadiol (C15H28O2) with relative area 41.76%. This research underscores the compelling potential of the Ie-Jue geothermal area as a promising reservoir of flora owing to the plant's adaptability to geothermal extremities.

Abstract

Bakông Asóe is a typical Acehnese snack loved by older people in Aceh since ancient times. This snack is made from wet tobacco which is used as chewing tobacco. The process of drying Bakông Asóê in the open triggers heavy metal contamination by air pollution from motor vehicles. The process of storing Bakông Asóê in a wet state also triggers the growth of microbes such as bacteria and fungi due to a growth medium in the form of water. The purpose of this study was to determine the content of heavy metal contamination (Pb, Cd, Se, Cu and Hg) and microbiological contamination in the form of bacteria (Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa) and fungi (Candida albicans and Aspergillus niger) in Bakông Asóê which sold on the side of the road without adequate hygiene. The research refers to a qualitative research approach with the type of research that is descriptive research. This study showed that heavy metal exposure in the form of Pb, Cu and Se in the three samples was still in the safe range for consumption. However, exposure to heavy metals in the form of Cd and Hg in the three samples exceeded the consumption standard. The results of the analysis of microbiological contamination in the three Bakông Asóê samples were proven to contain bacterial and fungal contamination, where cocci-shaped bacteria were more commonly found and fungi with the genus Aspergillus sp were also more dominant in the three Bakông Asóê samples. The conclusion of this study is positive for chemical and microbiological contamination in Bakông Asóê, which is sold in Aceh, Ule Kareng and Lambaro Markets.

Abstract

The influence of foreign ions on the response of Al3+ ISE-based polyurethane membranes from castor oil has been determined using SSM. The results showed that the presence of the foreign ions tested did not affect the sensitivity and response of Al3+ ISE. The influence of foreign ions Na+, K+, Li+, Pb2+, Ca2+, Co2+, Cu2+, Ni2+, Cr3+, and Fe3+ on the selectivity of ion-selective electrodes (Al3+ ISE) in the concentration range 1x10-9 – 1x10-4 M has been performed using separate methods. The results show that the presence of foreign ions does not affect the performance of Al3+ ISE with a Kij value < 1. The selectivity sequence of foreign ions is Fe3+ > Cr3+ > Pb2+ > Cu2+ > Ni2+ > Co2+ > Ca2+ > Li+ > Na+ > K+ respectively.

Abstract

Garlic (Allium sativum) contains organosulfur compound that plays an important role as an antibacterial and antifungal activities. Ascorbic acid or vitamine C also has been show has a good activity as an antioxidant and as an antifungal. The aims of the research is to determine the effect of the combination of petroleum ether garlic extract with vitamin C against Candida albicans. Zone of inhibition testing done by Kirby-Bauer method. The results showed that the combination of petroleum ether garlic extract with vitamin C in concentration of 50% did not show an activity significantly. Meanwhile, the activity of petroleum ether garlic extract alone at concentration of 50 and 75% showed activities towards Candida albicans with a diameter of inhibition zone are 19.46 and 27.46 mm respectively.

Abstract

This study focuses on the development of a machine learning ensemble approach for the classification of Beta-Secretase 1 (BACE1) inhibitors in Quantitative Structure-Activity Relationship (QSAR) analysis. BACE1 is an enzyme linked to the production of amyloid beta peptide, a significant component of Alzheimer's disease plaques. The discovery of effective BACE1 inhibitors is difficult, but QSAR modeling offers a cost-effective alternative by predicting the activity of compounds based on their chemical structures. This study evaluates the performance of four machine learning models (Random Forest, AdaBoost, Gradient Boosting, and Extra Trees) in predicting BACE1 inhibitor activity. Random Forest achieved the highest performance, with a training accuracy of 98.65% and a testing accuracy of 82.53%. In addition, it exhibited superior precision, recall, and F1-score. Random Forest's superior performance was a result of its ability to capture a wide variety of patterns and its randomized ensemble approach. Overall, this study demonstrates the efficacy of ensemble machine learning models, specifically Random Forest, in predicting the activity of BACE1 inhibitors. The findings contribute to ongoing efforts in Alzheimer's disease drug discovery research by providing a cost-effective and efficient strategy for screening and prioritizing potential BACE1 inhibitors.

Abstract

This research aims to synthesize silver nanoparticles (AgNPs) using an aqueous leaf extract of Calotropis gigantea obtained from the geothermal manifestation Ie Seu-Um, Aceh Besar, Aceh Province, Indonesia. The C. gigantea leaf extract was mixed with AgNO3 solutions at concentrations of 2, 5, and 9 mM, respectively. The mixture was stirred at 80 rpm by a magnetic stirrer for 48 hours in the dark. The change in solution color indicated the reduction of Ag+ to Ag0. The resulting AgNPs synthesized using C. gigantea leaf extract (AgNPs-LCg) exhibited cloudy grey, reddish dark brown, and light brown colors when synthesized with AgNO3 concentrations of 2, 5, and 9 mM, respectively. The particle sizes of AgNPs-LCg had maximum frequencies at 246.98 nm (synthesized using AgNO3 2 mM), 93.02 nm (synthesized using AgNO3 5 mM), and 171.25 nm (synthesized using AgNO3 9 mM). The zeta potential values of AgNPs-LCg using 2, 5, and 9 mM AgNO3 were -41.9, -40.1, and -31.4 mV, respectively. Based on the solution color, nanoparticle size, and stability value of AgNPs, it can be concluded that the use of AgNO3 at 5 mM is optimal for the green synthesis process of AgNPs-LCg.

Abstract

This study investigates the application of the Gradient Boosting machine learning technique to enhance the classification of Atopic Dermatitis (AD) skin disease images, reducing the potential for manual classification errors. AD, also known as eczema, is a common and chronic inflammatory skin condition characterized by pruritus (itching), erythema (redness), and often lichenification (thickening of the skin). AD affects individuals of all ages and significantly impacts their quality of life. Accurate and efficient diagnostic tools are crucial for the timely management of AD. To address this need, our research encompasses a multi-step approach involving data preprocessing, feature extraction using various color spaces and evaluating classification outcomes through Gradient Boosting. The results demonstrate an accuracy of 93.14%. This study contributes to the field of dermatology by providing a robust and reliable tool to support dermatologists in identifying AD skin disease, facilitating timely intervention and improved patient care.
