Abstract

Latar Belakang: Farmasis remaja memiliki peran dalam pembangunan kesehatan. Salah satu kegiatan yang dilakukan adalah penerapan swamedikasi. Tujuan: Pentingnya peran farmasis remaja dalam swamedikasi mendorong dilakukannya kegiatan pengabdian dengan tema pengenalan peran farmasis remaja dalam swamedikasi. Metode: Peserta diberikan pretest dan selanjutnya diberi intervensi berupa materi umum terkait swamedikasi, diskusi dan praktek swamedikasi bersama tutor. Media yang digunakan berupa materi dalam bentuk power point, modul dan alat peraga. Kegiatan diakhiri dengan melakukan posttest pada peserta yang sama. Hasil: Hasil pretest menunjukan bahwa hanya 14 peserta (20,29%) yang memiliki pengetahuan baik, sedangkan 50 peserta (72,46%) berpengetahuan cukup dan 5 peserta (7,25%) memiliki pengetahuan kurang. Nilai rata-rata hasil pretest adalah 57,10. Setelah diberi intervensi, hasil posttest menunjukan bahwa 66 peserta (95,65%) memiliki pengetahuan baik, sedangkan hanya sejumlah 3 peserta (4,35%) yang masih memiliki pengetahuan cukup. Nilai rata-rata posttes adalah 85,60. Kesimpulan: Adanya peningkatan nilai ini menunjukkan bahwa pemberian intervensi dalam program pengenalan farmasis remaja dalam swamedikasi yang diberikan dapat meningkatkan pengetahuan siswa SMK Pelita Bangsa Yogyakarta tentang swamedikasi. Perlu dilakukan penelitian sejenis dengan jangka waktu antara pretest dengan posttest lebih dari 1 bulan.

Abstract

Morinda citrifolia L. fruits are rich in polysaccharides of which are non toxic and possess hospes-mediated-antitumor potential. In Indonesia, noni fruit has been used empirically to enhance immune system. Previous research revealed that noni polysaccharide-rich fraction showed macrophage activity enhance-ment. This research aimed to optimize the method of noni-polysaccharides-rich fractionation using four different methods for polysaccharide isolation (method I, II, III, IV) and to evaluate the immunostimulatory effects of the isolated polysaccharides on macrophages and lymphocytes activities. Immunostimulatory effects were examined on male Balb/c mice in vitro. The polysaccharides were evaluated by phenol sulfuric acid test. Qualitative methods were used to detect the phytochemical characteristic. Total phenolic content was determined by using Folin-Ciocalteau method. The study showed different methods resulted in different yields, phytochemical characteristics and immunostimulatory activities of polysaccharides-rich fraction. The phytochemical test from method I, III and IVb revealed the presence of alkaloid. Method II resulted fractions with the presence of saponin. There were no protein and phenolic substances detected from all methods. The highest result of polysaccharide content was found in method IVb (32.58%). Method III showed the highest activity on phagocytic activity of the macrophage, while method IVb exhibited the highest stimulation on lymphocyte proliferation.Key words: Isolation method, polysaccharide, Morinda citrifolia L. immunostimulatory effect

Abstract

Biofilms associated with human infection have high levels of pathogenicity due to their resistance to antibiotics. The discovery of an active antibiofilm agent against polymicrobial biofilms is a necessary consequence for coping with biofilm-related infections. Thymol and Eugenol are essential oils that have potential as antibacterial and antifungal. This study aimed to determine the effectiveness of thymol and eugenol inhibits C. albicans, P. Aeruginosa, E. coli S. aureus and polymicrobial biofilm. Biofilm formation inhibition assay and biofilm degradation assay of thymol and eugenol were determined using microtiter broth method.The antibiofilm efficacy of thymol and eugenol towards polymicrobial biofilms were analyzed by calculating minimum biofilm inhibitor concentration (MBIC50) and minimum biofilm eradication concentration (MBEC50) values. The data were analyzed using Statistical Package for the Social Sciences (SPSS) with 95% confidence level. Thymol and eugenol showed inhibitory activity against the formation of mono and polymicrobial biofilms of the microbial tested.The result also demonstrated an evidence of activity of thymol and eugenol in breaking down mono and polymicrobial biofilm. Therefore, thymol and eugenol serves as a potential source for new antibiofilm drugs towards polymicrobial biofilm.

Abstract

This study was aimed to isolate and toidentify structures of the sponge secondary metabolites as well as to test their cytotoxic activity on mouse lymphoma cell line L5178Y.The sponge extract was fractionated by liquid-liquid partition followed with a vacuum liquid chromatography method. Structure elucidation was performed on the basis of extensive spectroscopic analysis involving one and twodimensional NMR spectroscopy as well as mass spectrometry. Cytotoxicity was tested on mouse lymphoma cell line L5178Y by using the microculture tetrazolium (MTT) assay.This study found a mixture of 5-pentadecyl-1H-pyrrole-2-carbaldehyde and (6’E)-5-(6’pentadecenyl)-1H-pyrrole-2-carbaldehyde as major constituents of the sponge extract. Those compounds were expected to be theactive constituent to show growth inhibition of mouse lymphoma cell line (L5178Y) in vitro.Key words : Mycale phyllophila, cytotoxic agent, NMR spectroscopy.

Abstract

Dental  plaque  is  a  mouth  cavity  health  problem  related  to  microbial biofilm,  where  Streptococcus  mutans is  predominant.  Adding  of  essential  oils blend  in  mouthwash  has  been  reported  to  increase  the  dental  plaque  inhibitory activity.  The  essential  oils  of  clove  leaves  (Syzygium  aromaticum (L.)  Merr.  & Perry) and cinnamon cortex (Cinnamomum burmanniNees ex Bl.), are known as potential  antibacterial  and  antibiofilm  towards  S.  mutans.  This  research  aims were  to  reveal  the  influence  of  blending  the  clove  leaves  and  cinnamon  cortex essential  oils  in  antibacterial  and  anti  biofilm  activity  against  S.  mutans and  to find out the optimum composition. Antibacterial assay was performed in nutrient broth  media,  on  microplate  flat-bottom  polystyrene  96  wells.  Biofilm  formation inhibition  and  degradation  assays  were  done  in  BHI  +  2  %  sucrose  on microplate flexible U-bottom PVC 96 wells. Crystal violet1 % was used to stain the biofilm and Optical Density(OD) was measured at λ 595 nm. Simplex Lattice Design formula  was  used  to  calculate  the  blend  optimum  composition.  TLCbioautography and GC-MS assays were done to revealthe active substances. As conclusion,  it  was  proven  that  blending  the  clove  leaves  and  cinnamon  cortex essential  oils  increased  the  antibacterial  and  biofilm  degradation  potency,  but reduced  the  biofilm  formation  inhibitory effect against  S.  mutans. The  optimum composition  of  the  essential  oils  blend  was  27:73  (% v/v).  From  our  results  we suggest  that  the  clove  leaves  and  cinnamon  cortex  essential  oil  blend  used  in this study be developed as anti dental plaque.Key words : clove leaves, cinnamon cortex, essential oils blend, Streptococcus mutans

Abstract

Quorum sensing adalah suatu bentuk komunikasi bakteri yang membantu mengatur perilaku koloni bakteri. Quorum sensing merupakan mekanisme komunikasi berdasarkan ekspresi gen dan populasi bakteri yang mempengaruhi perkembangan biofilm, pompa effluks, produksi toksin, dan faktor virulen lainnya. Quorum sensing inhibitor mengurangi patogenisitas organisme, mengurangi sifat virulen organisme, dan membantu sistem imun untuk membersihkan infeksi bakteri. Quorum sensing inhibitor dapat dikombinasi dengan antibiotik untuk membersihkan patogen yang persisten. Minyak atsiri kulit batang Krangean (Litsea cubeba) diketahui dapat menghambat pertumbuhan bakteri dan pembentukan biofilm Streptococcus mutans. Karena quorum sensing berperan dalam pembentukan biofilm maka dilakukan penelitian aktivitas daya antiquorum sensing kulit batang  Krangean terhadap Pseudomonas aeruginosa. Penentuan kadar hambat minimal dari ekstrak n-heksan, etil asetat, dan metanol dilakukan dengan metode mikrodilusi. Dari hasil penelitian, didapatkan hasil bahwa ekstrak etil asetat merupakan ekstrak yang paling aktif terhadap Pseudomonas aeruginosa dengan kadar hambat minimal 8µg/µl. Ekstrak aktif  etil asetat kemudian diuji daya antiquorum sensing dengan metode sumuran. Hasilnya menunjukkan bahwa ekstrak aktif etil asetat dengan loading sampel 25 mg per sumuran, memiliki aktivitas antibakteri dan hambatan produksi pioverdin. Senyawa aktif diidentifikasi dengan KLT kemudian dilakukan uji bioautografi.  Active compound group was unable to be determined by TLC then bioautography assay. Golongan senyawa yang terdapat di dalam ekstrak etil asetat yaitu alkaloid dan fenolik.

Abstract

Kanker adalah penyakit mematikan dan angka kejadiannya di Indonesia sangat tinggi. Data di Departemen Kesehatan didapati angka 1,8 per 100 ribu penduduk. Ironisnya saat ini belum ditemukan antikanker yang efektif dan aman. Minyak kayu manisyang diperoleh dari tanaman kayu manis (Cinnamomum burmannii) diduga memiliki aktivitas antikanker. Penelitian sebelumnya  bertujuan untuk isolasi, standarisasi minyak kayu manis sudah dilakukan. Saat ini penelitian dilanjutkan terhadap uji aktifitas serta keamanan minyak kayu manis untuk  mendapatkan fitofarmaka antikanker. Pengujian aktivitas antikanker  dengan menguji kemampuan penghambatan sel kanker terhadap sel line kanker WiDr (kanker kolon). Keamanan minyak kayu manisditentukan dengan melakukan uji toksisitas sub kronis terhadap mencit putih jantan.Hasil penelitian ini menunjukkan bahwa minyak kayu manis memiliki aktifitas sitotoksik terhadap kultur sel WiDr dengan IC50=13,70µg/ml.  Pemberian minyak kayu manis kepada hewan uji Tikus selama 1 bulan tidak memberikan perubahan biokimia darah BUN kreatinin, SGPT dan SGOT, serta hematologi Darah. Hasil histopatologi terhadap hepar dan ginjal juga tidak memberikan kerusakan sel kecuali pada dosis 0,04ml/200 g BB. Kata kunci:Minyak kayu manis, antikanker, WiDr, toksisitas

Abstract

Mesoyi oil distilled from the bark of Cryptocarya massoy have been used for traditional treatment of various ethnicities. However, its safety use has not been widely studied. This study aimed to investigate the cytotoxicity of mesoyi oil on vero cells in vitro. The Inhibitory concentration 50% (IC50) of mesoyi oil is evaluated using MTT assay. The results indicated that the mesoyi oil exhibited cytotoxic effects on vero cells with IC50 value of 97.4 μg//mL. This call for further studies to evaluate the cytotoxic activity in vivo.
