Abstract

Kapulaga fruit, temu putri rhizome and sengugu bark have been reported as antibacterials, suggesting potency to be developed as antibiofilm agents. This research has investigated the inhibition activity on biofilm formation of Methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli. Following petroleum ether extraction, the remaining biomass was macerated in ethanol 70 %. After solvent evaporation, the ethanol extract was partitioned to yield hexane, ethyl acetate and water fractions. The inhibition activities of ethanol extracts, hexane, ethyl acetate and water fractions of samples were carried out in vitro on microplates flexible U-bottom 96 wells using crystal violet staining and were recorded at λ 595nm. Phytochemical identification were performed by thin layer chromatography. Results showed that extract or fraction samples which have the highest biofilm inhibition activity toward MRSA were hexane fraction of kapulaga IC50 0.23±0.01 mg/mL, temu putri IC50 0.45±0.03 mg/mL and the ethyl acetate fraction of senggugu IC50 0.35±0.022 mg/mL. Biofilm inhibition activity toward E. coli, were shown by the ethyl acetate fractions of kapulaga IC50 0.32±0.17 mg/mL, temu putri IC50 0.46±0.03 mg/mL and senggugu IC50 0.39±0.02 mg/mL.

Abstract

Pada penelitian ini, kelopak rosela dikeringkan kemudian dimaserasi dengan etanol 70% - HCl (99:1) kemudian difraksinasi berturut-turut menggunakan n-heksana, etil asetat, n-butanol dan air. Semua fraksi diuji aktivitas antibakteri terhadap S. aureus dan E.coli dengan menggunakan metode difusi, aktivitas antibiofilm dan aktivitas antikuorum sensing terhadap Pseudomonas aeruginosa. Fraksi etil asetat adalah fraksi yang paling aktif terhadap S. aureus menghasilkan daerah hambatan pertumbuhan pada konsentrasi 10% dan 20% sebesar 15,89 ± 0,37 mm dan 15,93 ± 0,72 mm, sedangkan terhadap E. coli sebesar 17,25 ± 0,86 mm dan 17,35 ± 0,48 mm pada konsentrasi 10% dan 20%. Pada uji aktivitas antibiofilm didapat % penghambatan pada S. aureus sebesar 62,05% ± 17,83 dan pada E.coli sebesar 11,11% ± 23,13, uji aktivitas antikuorum sensing memberikan daerah hambatan sebesar 19,23 ± 1,52 mm dan 20,89 ± 2,35 mm pada konsentrasi 10% dan 20%. Fraksi etil asetat dilakukan isolasi terhadap satu senyawa aktif dengan menggunakan metode VLC dan KLT preparatif kemudian diidentifikasi strukturnya dengan menggunakan spektrofotometer 1H-NMR, 13C-NMR, COSY-NMR, HMBC dan LCMS. Abstract

Myrmecodia pendens Merr & Perry (Rubiaceae) is widely used in Papua as traditional medicine against various diseases. Preliminary study revealed that ethyl acetate fraction has potency to increase mice Balb/c lymphocyte proliferation in vitro. This study aimed to investigate the active compound responsible for the activity. Dried powders of plant tubers were macerated in ethanol 95% followed by solvent evaporation. The crude extract was further fractionated by liquid-liquid partition with n-hexane, ethyl acetate and water, successively. The ethyl acetate fraction was subjected to a silica gel column and separated by vacuum. The subfraction yields were tested on mice Balb/c lymphocytes to observe the influence on lymphocyte proliferation by MTT assay. Total phenolic content of subfractions were determined by Folin-Ciocalteau method. Chemical characteristics of active compound was identified by GC-MS, UV-Vis spectroscopy, IR and H-NMR. The result showed that total phenolic content has no significant correlation to the lymphocyte proliferation activity. Subfraction 7 yielded from ethyl acetate-methanol (7:3v/v) contained a major constituent (78% by GCMS, 83.75% by Densitometry). GCMS and UV spectra suggested a phenolic substructure. IR and H-NMR suggested that the phenolic was glycosilated and had aliphatic moiety.

Abstract

Curcumin is a lipophilic compound which suffers from the poor bioavailability after oral administration. Increasing its dissolution rate can be a successful strategy to improve the bioavailability. Along with the formulation developments, a rapid and simple analytical method to determine curcumin concentrations in the dissolution medium is required. The aim of this study was to develop and to validate an analytical method based on thin layer chromatography (TLC) to determine curcumin concentrations in a dissolution medium containing 0.5% w/v sodium lauryl sulfate (SLS) and 20 mM sodium phosphate buffer. A polyvinylpyrrolidone K30 based solid dispersion of Curcuma longa extract and its corresponding physical mixture were dissolved in a medium containing 0.5% w/v SLS and 20 mM sodium phosphate buffer (pH 6.0). Dissolution samples were spotted on a normal TLC plate and eluents of various compositions were evaluated. The retardation factor (Rf), resolution (Rs), and asymmetry factor (As0.05) of the optimized method were determined. Using the optimized eluent, proper separation of curcumin peak was achieved with an Rf of 0.50, Rs of 2.62 and As 0.05of 0.87. Linearity (5-30 μg/mL) was demonstrated by r value of 0.9965. The TLC method provided precision with RSD ≤3.50 and accuracy with recovery value of 94-105%.

Abstract

Harmsiopanax aculeatus, Harms, also known as kapur, have been traditionally used as medicinal plants to treat malaria in Maluku, Indonesia, although scientific evidence of the antimalarial active compounds of this plant has not been studied. This study was conducted to evaluate the anti-plasmodial and cytotoxicity activity of the active fraction of H. aculeatus leaves. The fractions tested were FG2, FG3 and FG4 obtained by separation of methanol extract using Vacuum Liquid Chromatography (VLC) by gradually using n-hexane, chloroform, ethyl acetate and methanol solvents. Anti-plasmodial activity in vitro was evaluated in Plasmodium falciparum strain FCR3 which is chloroquine resistant using microscopic method. The activity of cytotoxicity against vero cells was evaluated using MTT colorimetric method. The anti-plasmodial activity and cytotoxicity were expressed by IC50, inhibitory concentration of 50% Plasmodium or cells growth after incubation with the fraction tested, which calculated using probit regression analysis with SPSS.  The results showed that the fraction of FG2, FG3 and FG4 had a promising anti-plasmodial activity with IC50 value of 7.48; 8.24 and 7.75 μg/mL, respectively. The cytotoxicity activity of the three fractions on vero cell were 5814.43; 7780.48 and 1022.44 μg/mL, respectively. Moreover, the selectivity index (IC50 Vero cells/IC50 Plasmodium) of those were 777.33,944.23 and 131.93, respectively indicated that the three fractions were active, non-toxic and selective against P. falciparum. Further study will be conducted to isolate the active compounds of the fraction.

Abstract

Apoteker yang mempunyai pengetahuan yang baik dan persepsi yang positif terhadap kualitas fitofarmaka diharapkan akan merekomendasikan produk fitofarmaka kekonsumen di apotek. Penelitian bertujuan untuk mengetahui gambaran pengetahuan danpersepsi apoteker terhadap kualitas fitofarmaka, serta pengaruhnya terhadap niat merekomendasikan produk fitofarmaka. Penelitianmerupakan penelitian korelasional dengan instrumen berupa kuesioner. Responden adalah apoteker yang bekerja di apotek yang menjual fitofarmaka di Yogyakarta. Kuesioner terdiri dari 4 bagian yaitu karakteristik responden, pengetahuan fitofarmaka, persepsikualitas fitofarmaka, dan niat merekomendasikan produk fitofarmaka. Persepsi kualitas dan niat merekomendasikan diukur denganskala Likert. Teknik pengambilan sampel dilakukan secara acak sederhana. Data dianalisis dengan analisis deskriptif dan regresi liniersederhana. Hasil penelitian menunjukkan bahwa pengetahuan apoteker di apotek kota Yogyakarta secara rata-rata adalah cukup (nilai mean 6,67). Persepsi apoteker tentang kualitas fitofarmaka adalah baik (positif) berturut-turut dari nilai dimensi tertinggi adalah keamanan, efikasi, ketersediaan, akseptabilitas, merek, dan harga. Pengetahuan apoteker tentang fitofarmaka memberikan pengaruh positif dan signifikan terhadap persepsi kualitas fitofarmaka. Pengetahuan dan persepsi apoteker tentang kualitas fitofarmaka masing–masing memberikan pengaruh positif dan signifikan terhadap niat merekomendasikan produk fitofarmaka. Kata kunci: fitofarmaka, pengetahuan, persepsi kualitas

Abstract

Health service facilities which are considered to have an excellency will also have a greater opportunity tobe selected by the customer. The formation of positive perceptions from customer that is expected to bringsatisfaction will provide a positive impact on efforts to increase profit and service in Hospital Pharmacy Unit. Thisstudy was aimed to measure the services quality in Outpatients Pharmacy Unit at Regional General Hospital Slemanwhich were reviewed from input, process, and outcome variables.The input variables were assessed through observation of the pharmacy services supporting resources,interviews with the insurer Pharmacists unit responsible for outpatient and job satisfaction questionnaires ofOutpatient Pharmacy Unit employees. The variable process was assessed through the results of drug use indicators.The variable outcome was assessed through value gap customer satisfaction questionnaires based on SERVQUAL methods. Subjects in the study covered the pharmacy services supporting resources (9 persons of employee), prescription drugs, and customers of Outpatient Pharmacy Unit at General Hospital Sleman (257 prescription drugs and customers). Input variables data were analyzed by descriptive analysis. Process variables data were checkedfor completeness from its data fields, tabulated and analyzed using a triangulation method. Outcome variables datawere analyzed by gap analysis.Results showed that the outcome of service did not match the quality services target of Pharmacy Unit at General Regional Hospital Sleman. The gap values of all dimensions service quality were negative, outpatient customers satisfaction index score was 74.41%. Some of the resources input variables need improvement and have not fulfilledthe Law on Hospitals, Government Regulation on Pharmacy Practice, Standard for Pharmacy Services in Hospital, aswell as there were still some obstacle on process variables that affect the pharmacyservices. Key words: pharmacy services quality, the variable input, process, outcome,Outpatient Pharmacy Unit GeneralHospital Sleman.

Abstract

Sterculia quadrifida, R.Br. is one of the plants that grows on the mainland of Timor, East Nusa Tenggra. Sterculia quadrifida, R.Br. commonly known as Peanut tree in English and faloak in Indonesian has various pharmacological activities and has been widely used in traditional medicine. This review article aimed to provide a comprehensive overview of the potential of the faloak plant as an herbal medicine by looking at various aspects such as ethnobotany, phytochemicals, pharmacology and toxicity. Literature searches are carried out in scientific databases that are accepted worldwide such as ScienceDirect, Scopus, PubMed, Springer Link, Wiley Online Library and advanced searches on Google scholars, books, abstracts, theses, scientific reports, and several non-impact and non-indexed journals with the search keywords "Sterculia quadrifida" or "Peanut tree" or "Red fruit kurajong" or "Faloak". From the various literature that has been collected, faloak has been shown to have pharmacological activities such as anticancer, antioxidant, antifungal, immunomodulatory, antiviral, antibacterial, antidiabetic, and antipyretic. The existence of this activity cannot be separated from the chemical compounds contained therein such as flavonoids, alkaloids, terpenoids, phenols, tannins and saponins. Faloak also has low toxicity with LD50> 5000 mg/kg body weight rats.

Abstract

Saponin merupakan salah satu jenis metabolit sekunder dari tanaman yang memiliki aktivitas antibakteri namun aktivitas antibiofilmnya terhadap C. albicans ATCC 10231 belum pernah dilaporkan. Penemuan kandidat antibiofilm baru terhadap biofilm C. albicans menjadi tantangan yang harus di atasi dalam mencegah infeksi yang berhubungan dengan biofilm. Penelitian ini bertujuan untuk mengetahui efektivitas saponin dalam menghambat dan mendegradasi C. albicans ATCC 10231. Pengujian planktonik, penghambatan biofilm dan pengujian degradasi biofilm ditentukan dengan menggunakan metode microtiter broth. efektivitas antibiofilm saponin terhadap biofilm dianalisis dengan menghitung minimum biofilm inhibitor konsentrasi (MBIC50) dan nilai minimum biofilm eradication concentration (MBEC50). Senyawa saponin 1 % memberikan aktivitas penghambatan pada fase planktonik C. albicans sebesar 79,48 % ± 0,01, fase pertengahan biofilm sebesar 56,00 %  ± 0,02, fase pematangan 35,55 ± 0,03 Hasilnya juga memberikan bukti aktivitas saponin dapat mendegradasi 50 % biofilm C. albicans sebesar 51,03 ± 0,01. Oleh karena itu, senyawa saponin potensial untuk dikembangkan sebagai kandidat obat-obat antibiofilm baru terhadap biofilm C. albicans.

Abstract

Daun kersen (Muntingia calabura L.) diketahui mengandung beberapa senyawa yang memiliki aktivitas sebagai antibakteri terhadap Staphylococcus aureus, salah satunya adalah flavonoid. Flavonoid bekerja sebagai antibakteri dengan beberapa mekanisme aksi, diantaranya menghambat sintesis asam nukleat, menghambat fungsi membran sitoplasma dan menghambat metabolisme energi dari bakteri. Hasil penelitian ini diharapkan dapat mengetahui kadar bunuh minimum (KBM) ekstrak dan fraksi daun kersen terhadap S. aureus, untuk mengetahui aktivitas antibakteri yang paling besar dari ekstrak etanol dan masing-masing fraksi serta korelasi antara aktivitas antibakteri dengan kandungan flavonoid total. Serbuk sampel kering dimaserasi dengan etanol 96%. Ekstrak etanol dipartisi dengan fraksinasi cair-cair secara berturut-turut menggunakan heksan, etil asetat, dan air. Ekstrak etanol dan masing-masing fraksi kemudian diuji aktivitas antibakterinya terhadap S. aureus dengan metode dilusi menggunakan media Mueller Hinton Broth (MHB) pada konsentrasi 20 mg/mL, 15 mg/mL, 10 mg/mL, dan 5 mg/mL. Sebagai kontrol pembanding digunakan antibiotik Seftazidim, pelarut DMSO, dan media MHB. Kuersetin digunakan sebagai standar pembanding dalam mengukur kandungan flavonoid total menggunakan Spektrofotometer UV-Vis. Analisis hasil dilakukan dengan membandingkan kadar flavonoid total dari ekstrak etanol dan masing-masing fraksi dengan KBM. Hasil penelitian menunjukkan bahwa fraksi etil asetat merupakan fraksi yang paling aktif dengan KBM 0,312 mg/mL, sekaligus sebagai fraksi yang memiliki kadar flavonoid total paling besar yaitu sebesar 5,624 % QE. Analisis korelasi Pearson menunjukkan aktivitas antibakteri dari ekstrak etanol dan masing-masing fraksi, 93% dipengaruhi oleh kandungan flavonoid total.
