Abstract

Alzheimer’s disease is a progressive neurologic dementia disorder and the fifth leading cause of death among elderly individuals. Its pathogenesis is closely linked to diabetes-induced brain insulin resistance, which accelerates beta-amyloid accumulation, exacerbating disease severity. This condition significantly impacts quality of life and healthcare costs, necessitating effective treatment strategies. Medicinal plants offer a promising avenue, with bibliometric analysis identifying trending active compounds for potential development as therapies for diabetic Alzheimer’s. This study analyzed 200 articles from the Scopus database, employing Scopus and VOSviewer 1.6.20 to visualize bibliometric trends by publication, country/territory, subject area, and keyword co-occurrence. Results indicate a recent decline in research activity, with China leading in publications across pharmacology, medicine, and biochemistry. Notably, flavonoids like “quercetin,” “kaempferol,” and “gallic acid” emerged as prominent topics. Keywords: Alzheimer’s diabetes, Bibliometrics, Herbal medicine, Extracts

Abstract

The rise in antimicrobial resistance has prompted the search for potent antimicrobial agents, with plants from unique environments, such as geothermal areas, offering potential due to their diverse phytochemical profiles. This study evaluated the antiplanktonic and antibiofilm activity of M. edule fractions from the geothermal area, Jaboi, Sabang. Crude extracts of M. edule were sequentially fractionated using hexane, ethyl acetate, and ethanol. The resulting fractions were further analyzed for their phytochemical content and antiplanktonic and antibiofilm activities. The ethyl acetate fraction demonstrated superior antiplanktonic and antibiofilm activities compared to other fractions, inhibiting 50% of Pseudomonas aeruginosa planktonic and biofilm formation at a concentration of 10 mg/mL. The most potent fraction exhibited the phenolic content, 672.84 mg GAE/g extract, surpassing the other fractions. The GC-MS analysis shows the presence of pyrogallol, hexadecanoic acid, cedran-diol, and sitosterol. These findings highlight the potential of the ethyl acetate fraction of M. edule as a source of bioactive compounds with promising antiplanktonic and antibiofilm properties, laying the groundwork for future research into its therapeutic applications against biofilm-associated infections.

Abstract

Nontuberculous mycobacteria (NTM) are environmental microorganisms, also known as opportunistic pathogens, found in patients with pulmonary tuberculosis. The emergence of antibiotic resistance is increased by prolonged antibiotic treatment for NTM infections. Therefore, alternative sources of new antibiotics are essential for the treatment of NTM infections. A marine actinobacterium, Streptomyces sp. BTA 1-131, isolated from a marine sponge, Melophlus sarasinorum, has been reported as a potential source of antibacterials and anticancer agents. The present study aimed to investigate the potential of Streptomyces sp. BTA 1-131 against two NTMs: Mycobacterium smegmatis and Mycobacterium fortuitum. Streptomyces sp. BTA 1-131 was fermented in three cultivation media (SYP, ISP2, and YS), and the secondary metabolites were extracted using methanol. The bioactivity screening showed inhibition of all methanolic extracts against the growth of M. smegmatis and M. fortuitum. The methanolic extract, which could inhibit both mycobacteria, was a crude extract derived from SYP liquid medium fermentation. The isolated compounds in this study were preliminarily identified using thin-layer chromatography (TLC). The TLC results showed different potential compounds in the crude extracts of Streptomyces sp. BTA 1-131 also highlighted the impact of the fermentation medium on the production of metabolites from Streptomyces. This study also added knowledge about the importance of the Indonesian marine actinobacterium Streptomyces sp. BTA 1-131 as a promising producer of anti-NTM compounds.

Abstract

The leaves of the H. aculeatus plant are known as a traditional antimalarial medicinal plant in Maluku. Several studies have reported on the leaf activity of this plant, but studies on in vitro and in vivo antiplasmodial activity and the compounds in the active fraction (F7) of H.aculeatus leaf have not been reported. This study aims to determine the antiplasmodial activity of the F7 of H. aculeatus leaves and to analyze the compounds contained therein. In vitro, antiplasmodial activity was tested on Plasmodium falciparum strain FCR3 using a microscopic method. A cytotoxicity test was performed on Vero cells using an MTT assay. In vivo, the antiplasmodial activity of F7 was carried out using the 4-day suppressive test method by treating Swiss mice infected with P. berghei. Analysis of the compounds in an F7 was done using spray reagent and UV Vis DAD and HPLC DAD-MS with a UV detector. The results showed that the F7 was very active in vitro (IC50 of 0.7 µg.ml-1) and in vivo (ED50 of 2.49 mg.kg BW-1.d-1) also selectively (SI of 8159.94) inhibited the growth of Plasmodium. The F7 contains a group of essential oils, triterpenoids, phenolic compounds, and flavonoids after being analyzed using spray reagents. Based on the analysis results, five compounds were identified: fomoxanthone, cyclopentene, microspherone, indole 3-carbaldehyde, and naamine. In addition, seven compounds were not identified.

Abstract

The P. major plant has long been utilized in traditional herbal medicine and is widely accessible in Indonesia and globally. Extensive research has demonstrated its efficacy in wound healing through both clinical and preclinical studies. Gel formulations are recognized for establishing a conducive moist environment essential for wound healing, thus rendering them suitable for topical wound treatment. This study assesses the stability of HPMC gel base, carbomer 940, and their combination as carriers for the ethanol extract of P. major leaves. Stability testing involved a freeze-thaw cycle test at temperatures of 4°C and 40°C for three cycles. The findings indicate no significant alteration in the organoleptic properties and pH of the gel formulations before and after stability testing. However, t-test analysis reveals a noteworthy decrease in viscosity for carbomer and its combination, albeit not for HPMC alone. In conclusion, HPMC exhibits superior stability as a gel base compared to Carbomer 940 and their combination.

Abstract

The traditional use of faloak (Sterculia quadrifida R. Br.) stem bark as an effective treatment for various diseases has led to its widespread cultivation and collection. Therefore, this study aims to determine the correlation between the intensity and color variation of faloak stem bark with its antioxidant activity and phytochemical content using chemometrics analysis. The study procedures were carried out by collecting samples from different locations in East Nusa Tenggara, Indonesia. Stem bark intensity and color variations were then associated with chemical profiles of extract produced. Chemical profiles analyzed were Total Phenolic Content (TPC), Total Flavonoid Content (TFC), and antioxidant activity as measured using the DPPH, b-carotene bleaching, and CUPRAC methods. Subsequently, the data were subjected to multivariate statistical analyses utilizing Principal Component Analysis (PCA) and Cluster Analysis (CA). The results showed that color variations on faloak stem bark had no significant impact on cluster formation. CA and PCA showed grouping according to gray value representing color intensity. Meanwhile, PCA revealed significant correlations between gray value of faloak stem bark extract and TPC, antioxidant activity, and extract yield, with TFC having no association. Based on these results, color detection system could be developed to facilitate S. quadrafida bark collection as herbal medicine raw materials in the field.
