Abstract

Utilization of marginal lands such as tidal land should be done to increase rice production as a solution of the decreasing productive land. However, rice cultivation in such land will be affected by low soil fertility, soil acidity and Fe toxicity. Fe-tolerant varieties are needed to overcome those problems. The objective of the research was to analyze the growth and production of Fe tolerant rice lines in tidal soil type C, Banyuasin, South Sumatera. Screening for Fe-tolerant character of 54 rice lines from the F8 RIL population derived from a cross between rice var. IR64 and var. Hawara Bunar using hydroponic technique under 1000 ppm Fe resulted 25 Fe-tolerant lines. The field test of 25 putative tolerant rice lines, at the tidal land in Banyuurip, Banyuasin, showed that the most lines grew better than that of var. Hawara Bunar parent. Several lines produced yield higher than that of var. Hawara Bunar parent. The field experiment resulted two rice lines, which were IRH108 and IRH195 that potential for further studies.

Abstract

Aquilaria crassna is one of the Aquilaria species that could produce agarwood. agarwood production of A. crassna can be induced by microfungi as an inoculant. Ecological interaction between the host plant, wounding and inoculant in the formation of agarwood has not yet clearly. other factors like the plant ages, plant species, environ- ment also played important roles in the formation of agarwood. This research aimed to determine the influence of the combination between planting media and Acremonium sp. and Fusarium sp. to agarwood quality of Aquilaria crassna. The experiment was consisted of two factors, which were five planting media and two inoculants fungi i.e. Acremonium sp. and Fusarium sp. The observed parameters were the percentage of senescence leaves, color of wood, level of fragrant and terpenoid content. The best agarwood fragrant was produced by the seedlings that were treated with either combination of husk charcoal media enriched with NPK fertilizer and Acremonium sp. or the combina- tion between husk charcoal media enriched with Hoagland modified solution and Fusarium sp.. The darkest color of wood was produced by the seedlings that were treated with combination of husk charcoal media enriched with NPK fertilizer and Acremonium sp., as well as for the same media with Fusarium sp.. during the experiment terpenoid could not be detected from the treated seedlings. Acremonium sp. caused more leave senescence than that of Fusarium sp..

Abstract

High methane oxidation activity of local isolated methanotrophic bacteria have a potent as methane gases reducing agent while combined with nitrogen fixing bacteria as paddy biofertilizer. The aim of the research was to evaluate the effectiveness of the bacteria as methane gases reducing agent and biofertilizer in paddy plantation. The research was arranged in a completely randomized design consisted of fertilizer types and watering system treatments with four replicates. The research showed that paddy shoot length was not affected by the treatment. On the other hand, both plant freshand dry weight, as well as the number of productive tiller were affected by interaction of fertilizer types and watering system. Fertilizer types affected grain per panicle and methane flux after fertilization. In the end of paddy vegetative stage, bacterial fertilizers were capable to reduce methane emission in different rate. The different result in methane flux was likely due to the interaction between soil local microorganisms and soil chemical component.

Abstract

Acidity, phosphorus deficiency, and droughtstress are major problems in Indonesia’s Ultisol rice farming.Development of rice lines tolerant to those stresses isexpected to be able to reduce the consumption of Pfertilizer. The objectives of the research were to evaluategenetic constitutive of rice lines (BC2F6 population) derivedfrom Situ Bagendit x Kasalath and Situ Bagendit x NIL-C433crossings, and to evaluate responses of those lines toYoshida nutrient solution under P deficiency and Al stresscondition. The research was conducted at Molecular BiologyLaboratory and Greenhouse of ICABIOGRAD, fromNovember 2011 to May 2013. The result of foregroundanalysis showed that Pup1 locus has been integrated intothe genome of BC2F6 rice lines, eventhough some lines (SK5,SK6, SK7, SK8, SK9, SK10, SK19, and SK20) showedincomplete integration. Background analysis indicated thatmajority (95.7%) of the Situ Bagendit background has beenrecovered in BC2F6 rice lines. Al stress evaluation showed SNlines were more tolerant to P deficiency and Al stress thanthat of SK lines. Pup1 locus showed good expression underlow P and no Al stress. Based on genome proportion andYoshida nutrient solution experiments, a total of three lines,namely SK13, SN2, and SN9, have potential goodcharacteristics. Molecular analysis within a marker-assistedbackcrossing (MAB) experiment should be carried out ateach generation of lines for gaining fully gene segment inadvanced generations.

Abstract

The quality of physic nut (Jatropha curcas) seeds should be maintained during storage, either as seeds for seedlings or oil production to be used for biodiesel. The effects of water activity and duration of storage on the quality, i.e. fungal population, lipid, fatty acid and free fatty acid contents, and viability of physic nut seeds were investigated. The results showed that the moisture content of seeds and total fungal population decreased at low water activities, and increased at high water activities (aw). At aw 0.64, at the beginning of storage and after 20 weeks of storage, total fungal populations were 5.4 x 103 and 1.8 x 102 cfu g-1 dry basis (db), respectively. At aw 0.93, at the beginning of storage and after 20 weeks of storage, total fungal populations were 5.0 x 103 and 3.3 x 106 cfu g-1 db, respectively. At the beginning of storage fungi infecting seeds were field fungi, i.e. Cladosporium sp., Colletotrichum sp., Fusarium semitectum, and F. verticillioides. The population of field fungi decreased with the increase of storage duration. They were replaced by postharvest fungi, i.e. Aspergillus restrictus, A. penicillioides, Eurotium chevalieri, E. rubrum, Penicillium citrinum, P. implicatum and P. oxalicum. Lipid content and viability of seeds decreased with the increase of water activities and seed moisture contents, while free fatty acid contents increased with the increase of water activities and seed moisture contents during storage. Fatty acids of lipid were dominated by unsaturated fatty acids, i.e. oleic and linoleic acids. Physic nut seeds could be stored at aw 0.64-0.75 up to eight weeks when the seeds will be used for seedling, or up to 16 weeks when they will be used for producing oil.

Abstract

AbstrakKetidaksesuaian sistem klasifikasi Alpinia berdasarkan ciri morfologi dengan ciri molekuler menyebabkan sistem klasifikasi pada marga ini masih bermasalah. Data tambahan dari anatomi Alpinia diperlukan untuk mendukung sistem klasifikasi pada marga ini. Sayatan paradermal adaksial dan abaksial dari daun tujuh jenis Alpinia diamati. Sampel diambil dari koleksi Kebun Raya Bogor (KRB) yang mewakili jenis A. galanga, A. mutica, A. padacanca, A. macrocrista, A. malaccensis, A. pusilla, dan A. ligulata. Sebelas ciri Alpinia diamati seperti; ukuran stomata, jumlah stomata, indeks stomata, kerapatan stomata, tipe stomata, ukuran sel tetangga, jumlah sel tetangga, bentuk sel tetangga, ukuran epidermis, jumlah epidermis, dan bentuk epidermis. Hasil pengamatan menunjukkan bahwa beberapa ciri dapat digunakan untuk membedakan jenis Alpinia, seperti bentuk dan ukuran epidermis; ukuran, kerapatan dan indeks stomata; serta ukuran sel tetangga. Sementara ciri lainnya, seperti tipe stomata dan bentuk sel tetangga pada semua jenis Alpinia memiliki bentuk yang sama sehingga tidak dapat digunakan untuk membedakan jenis Alpinia.Abstract The incompatibility of Alpinia classification system based on morphological and molecular character causes classification system of this genus is still problematic. Additional data from Alpinia anatomy are needed to support the classification system in this genus. Adaxial and abaxial sides paradermal incisions of seven Alpinia species were observed. The samples were taken from the Bogor Botanical Gardens (KRB) collection representing A. galanga, A. mutica, A. padacanca, A. macrocrista, A. malaccensis, A. pusilla, and A. ligulata. Eleven Alpinia characteristics were observed; stomatal size, number of stomata, stomatal index, stomatal density, stomatal type, neighboring cells size, number of neighboring cells, neighboring cell shape, epidermal cells size, number of epidermal cells, and epidermal cells shape. The results showed that several characteristics can be used to distinguish Alpinia species, such as the shape and size of epidermal cells; the size, density and index of stomata; and the size of neighboring cells. Several other characteristics, such as the type of stomata and shape of neighboring cells in all species of Alpinia have the same shape so the characters cannot be used to distinguish Alpinia species.

Abstract

Program on the ability of agriculture to provide rice as a staple food tends to decrease with the reduced land area used for rice cultivation. improvement rice production should be directed to use  marginal land  as dry and acid land, where poisoning aluminium was seen as a constrain factor. Development of Aluminium (Al) tolerant variety is needed to increase rice production. This technology was environmentally friendly, cheaper and also easier for farmers adopted. The rice lines with broad genetics background which tolerant to Al toxicity has been produced by breeding program. The diversity of Indonesian local rice germplasm is a basic foundation on development of Al toxicity tolerant rice varieties. The purpose of this research was to analyze the genotype variety using 384 SNPs markers on selected Al tolerant rice lines based on bioinformatics approach. Based on the 384 SNPs markers, the total 26 selected  genotypes were clustered into 5 groups. Each group was having varied Relative Root Length (RRL), which was an Al tolerant parameter of phenotype. These groups were indicated for having different genes or QTL that were related to the Al tolerant character. The association analysis obtained 9 significant SNPs markers that can be applied as the molecular markers for assisting selection in breeding  program.

Abstract

Typhonium flagelliforme atau yang dikenal dengan keladi tikus merupakan tumbuhan perdu yang banyak tumbuh di Indonesia. Tumbuhan ini mengandung lektin pada umbinya yang memiliki aktivitas antikanker. Namun hingga saat ini, tidak ada informasi ilmiah yang mengkaji mengenai lektin yang terdapat di umbi T. flagelliforme yang tumbuh diberbagai wilayah Indonesia. Tujuan dari penelitian ini adalah untuk mengidentifikasi dan mengkarakterisasi lektin umbi dari tujuh aksesi T. flagelliforme di Indonesia. Umbi yang digunakan dalam penelitian berumur 6 bulan tanam. Ekstrak protein umbi yang didapat dianalisis kandungan protein, toksisitas, dan aktivitas hemaglutinasinya. Aksesi yang memiliki aktivitas hemaglutinasi tertinggi, lektinnya dipurifikasi menggunakan kolom DEAE-Sepharose dan CM monolitic. Hasil menunjukkan bahwa protein umbi dari setiap aksesi memiliki toksisitas dan aktivitas hemaglutinasi. Aksesi Solok memiliki total protein dan aktivitas hemaglutinasi tertinggi dibanding aksesi lainnya. Aksesi ini memiliki kandungan lektin dengan bobot molekul 12,67 kDa. Lektin ini stabil pada temperatur 20 hingga 40 °C dan pada pH 5,0 hingga 7,2. Informasi karaktersitik ini dapat membantu dalam pengembangan obat antikanker berbasis lektin.

Abstract

The Durian Tengkurak (Durio tanjungpurensis Navia) is one of the endangered exotic species in the Malvaceae family. The species is important for conservation of the germplasm and is considered a potential genetic resource for the development of durian in the future. The objective of this research project was to assess the molecular diversity of D. tanjungpurensis in West Kalimantan, based on Inter Simple Sequence Repeat (ISSR) markers. We applied ten ISSR primers to reveal the genetic diversity of 60 individuals from six natural endemic D. tanjungpurensis populations. The genetic diversity parameters were estimated based on binary data about PCR products (present or absent bands). The results showed that the mean number of observed alleles, the mean number of effective alleles, the genetic diversity, the Shannon’s Information Index score, the number of polymorphic loci, and the percentage of polymorphic loci were 1.53, 1.29, 0.17, 0.26, 77.83, and 52.59, respectively. An analysis of molecular variance (AMOVA) showed that the genetic diversity within a population (65%) was higher than that found between the populations (35%). UPGMA clustering and principal coordinate analysis, based on the DICE similarity matrix, were used to classify the populations into three groups: 1) Hutan Rejunak and Tembaga, 2) Bukit Merindang, and 3) Hutan Rawak, Bukit Sagu 1, and Bukit Sagu 2. Further analysis of the population structure using STRUCTURE software was used to classify all the individuals into two major categories, thus uniting Groups 2 and 3 as one major category. In conclusion, a high level of genetic diversity in the Durian Tengkurak was revealed utilizing the ISSR markers employed in the study.

 AbstrakPenggunaan herbisida parakuat untuk mengendalikan pertumbuhan gulma telah menjadi metode yang paling umum. Namun, penggunaan herbisida juga dapat menyebabkan kematian sel tanaman kentang yang sedang dibudidayakan, karena herbisida parakuat dapat menginduksi pembentukan senyawa Reactive Oxygen Species (ROS) yang dapat merusak sel tanaman. Ekspresi berlebih dari gen MmCu/Zn-SOD diharapkan dapat memberikan ketahanan tanaman terhadap cekaman herbisida parakuat. Penelitian ini bertujuan untuk menganalisis ekspresi gen MmCu/Zn-SOD pada kentang transgenik IPB CP3 kultivar dan ketahanannya terhadap cekaman herbisida parakuat. Perlakuan herbisida parakuat dengan konsentrasi 75 µM pada tanaman transgenik (P3SOD6, P3SOD9, P3SOD17) dan non-transgenik (NT) secara in vitro menunjukkan bahwa semua tanaman transgenik lebih tahan terhadap cekaman parakuat dibandingkan tanaman non-transgenik. Klon P3SOD9 paling tahan terhadap herbisida parakuat, dengan perbandingan 10 tanaman hidup dari 12 tanaman yang diuji. Ekspresi gen MmCu/Zn-SOD pada galur transgenik lebih tinggi dibandingkan galur non-transgenik. Ekspresi relatif gen MmCu/Zn-SOD pada P3SOD9, P3SOD6, dan P3SOD17 secara berturut-turut lebih tinggi 233,22; 127,62; dan 3,18 kali dibandingkan ekspresi gen tersebut pada NT. Semakin tinggi ekspresi gen MmCu/ZnSOD, semakin tinggi ketahanan terhadap cekaman herbisida parakuat.AbstractThe use of paraquat herbicide to control weed growth has become the most common method. However, using the herbicide can also cause cell death in potato plants that are being cultivated because it can induce the formation of Reactive Oxygen Species (ROS) compounds that can damage plant cells. Overexpression of the MmCu/Zn-SOD gene is expected to provide resistance to paraquat herbicide stress. The study aimed to analyze the expression of the MmCu/Zn-SOD gene in transgenic potato cultivar IPB CP3 and its resistance to paraquat herbicide stress. In vitro treatment of transgenic (P3SOD6, P3SOD9, P3SOD17) and non-transgenic (NT) potato with 75 µM paraquat herbicide showed that all transgenic plants were more resistant to paraquat stress than that non-transgenic plants. The P3SOD9 clone was the most resistant to paraquat herbicide, with a ratio of 10 surviving plants of the 12 tested plants. The expression of MmCu/Zn-SOD in transgenic lines was higher than that in non-transgenic lines. The relative expressions of MmCu/Zn-SOD gene in P3SOD9, P3SOD6, and P3SOD17 were 233.22; 127.62; and 3.18 times higher than that of NT, respectively. The expression level of MmCu/ZnSOD correlates with the level of resistance to paraquat herbicide stress.
