Abstract

One of the crucial stages in the ramie plant (Boehmeria nivea L. Gaud) tissue culture is the acclimatization phase, which transitions between the in vitro and ex-vitro environments. Determining the appropriate age of plantlets and providing nutrients in the substrate play significant roles in the success of acclimatization. This research is conducted to determine the most suitable age of plantlets for acclimatization and assess the impact of providing nutrients through Trichoderma spp. application on the growth of ramie seedlings during the acclimatization phase. The experiment was conducted between January and March 2023 at the Screenhouse Bale Tatanen, Faculty of Agriculture, Universitas Padjadjaran, Jatinangor, Sumedang. The experimental design used was a Randomized Complete Block Design (RCBD) with two factors: plantlet age with three levels (4, 8, and 12 weeks old) and Trichoderma spp. fertilizer application with two levels (without and with Trichoderma) with 4 replications. The results showed that the 4-week-old plantlets exhibited the best height, root number, root length, and leaf number growth. Additionally, the application of Trichoderma spp. increased the stem diameter of the ramie seedlings during the acclimatization phase.

Abstract

The objective of the study was to evaluate the seed vigor of two soybean cultivars under submergence stress conditions. This research was carried out from January to March 2024 at the Seed Technology Laboratory, Faculty of Agriculture, Universitas Padjadjaran. The first test to compare the two cultivars used the F test as variance analysis, and the differences between the two conditions of water level were measured using the t-test at the 5% significant level. In each treatment, it was replicated 4 times and duplicated. Furthermore, the vigor characters were measured using a plastic rolled paper test (PRP) test for 40 seeds in each test. The results of this study indicate that submergence stress conditions can give a real affection to germination percentage, vigor index, root length, hypocotyl, and epicotyl length. The higher germination percentage and vigor index in submergence stress were obtained in the Demas 1 cultivar, 80% and 7,22 respectively. Besides, the hypocotyl and epicotyl length in submergence stress showed higher compare to Dega 1, 11.58 cm and 2.54 cm, respectively. Even though, the Dega 1 cultivar showed the best response in root length under submergence stress conditions with a length of 6 cm. These results indicated that the different response cultivars on Submergence stress may be caused by the genetic and internal condition of the seed.

Abstract

White turmeric is a medicinal plant widely used to treat various diseases. The limited availability of white turmeric seedlings is influenced by the scarcity of quality seeds due to the long dormancy of the rhizomes and the high incidence of pathogen attacks. Tissue culture techniques are one alternative to address the problem of white turmeric seedling availability. This study aims to determine the best concentrations of meta-topolin and kinetin for in vitro shoot multiplication of white turmeric. The research was conducted from January to June 2022 at the tissue culture laboratory, Faculty of Agriculture, Universitas Padjadjaran. This study used a completely randomized design consisting of 7 treatments: control (0 mg L-1), meta-topolin (1 mg L-1; 2 mg L-1; 3 mg L-1), kinetin (1 mg L-1; 2 mg L-1; 3 mg L-1). Analysis of variance (ANOVA) and Duncan’s multiple range test were used to analyze the statistical effects of meta-topolin and kinetin application. Applying 1 mg L-1 meta-topolin yielded the best results for shoot number, indicating that 1 mg L-1 meta-topolin is a potential cytokinin for the propagation of Curcuma zedoaria Rosc.

Abstract

This research aimed to study the addition of ameliorant material in the soil to increase fertility rates, especially for potato plants grown at the height of 750 m above sea level. The experiments were conducted on medium plains 750 m above sea level in the Jatinangor area, Indonesia, with land type Incepticol. The composition of ameliorant consists of Cattle manure, biochar from coconut shells, and dolomite. The seeds of the potatoes used are relatively good varietal when cultivated in a medium plain. The design of the environment used is the group’s random draft repeated three times, the treatment design consists of K0: control; K1: Cattle manure 10 Mg ha-1; K2: Cattle manure 20 Mg ha-1; K3: Cattle manure 8 Mg ha-1, dolomite 1 Mg ha-1, 1 Mg ha-1 biochar; K4: Cattle manure 16 Mg ha-1, dolomite 2 Mg ha-1, 2-Mg biochar ha-1, K5: Cattle manure 8 Mg ha-1, dolomite 2 Mg ha-1; K6: Cattle manure 16 Mg ha-1, dolomite 4 Mg ha-1; K7: Cattle manure 8 Mg ha-1, biochar 2 Mg ha-1; K8: Cattle manure 16 Mg ha-1, biochar 4 Mg ha-1. Results showed that the composition and dose of ameliorant could increase the value of soil pH but have not improved the C/N ratio and absorption of phosphorus plants. The best result was obtained from the composition treatment of 16 Mg ha-1 Cattle manure, 2 Mg ha-1 dolomite, and 2 Mg ha-1 biochar. Soil pH is capable of increased by 16.83%, and the size of the potato bulbs of Class L (450-550 mm) amounted to 35, 11%, size M (350-450 mm) by 25.89%, and size S (200-350 mm) amounting to 34.37% when compared with controls.

Abstract

Various intensifications of potato cultivation, such as the provision of paclobutrazol and nitrogen (N) fertilizer, are thought to impact the quality of the seeds produced. This study aims to evaluate the effects of different N fertilizer doses and paclobutrazol concentrations applied in the preharvest period on the sprouting performance of G0 potato tuber seeds after storage. Nine treatment combinations, each repeated three times, were tested, using 50%, 100%, and 150% of the recommended N dose and paclobutrazol concentrations of 50, 100, and 150 part per million (ppm), applied at 30 and 45 days after planting, respectively. The interaction effect between N fertilizer and paclobutrazol concentration was not significantly affected on all observed variables. Preharvest application of 100% N fertilizer produced the largest seedlings, indicated by the highest shoot length at 56 and 74 days after storage. Preharvest application of 150 ppm paclobutrazol produced the highest shoot length, shoot emergence rate, and seedling dry weight than other treatments. The present study implied the importance of preharvest N and paclobutrazol for improving the sprouting performance of G0 potato seed tuber.
