Abstract

Physiological quality of seeds could be seen from viability and vigor. The used of matriconditioning treatment using biological agent and bokashi fertilizer could be the way to enhance physiological quality of seeds. The objective of experiment was to determind the most effective dosage interaction of biological agent and bokashi fertilizer to improve physiological quality of seeds.This research was held at Ciparanje Experimental Farm and Laboratory of Seed Technology Faculty Agriculture Universitas Padjadjaran.from November 2016 until February 2017.  The experiment’s method design using randomized block design factorial pattern with Duncan’s Multiple Range Test (DMRT) at 5 %. The Cultivar that used for this research was Grobogan with Bokashi that tested at 0 ton/ha, 16 ton/ha, 32 ton/ha, and 48 ton/ha respectively combined with matriconditioning that used biological agent and control without biological agent. The biological agent that used for this treatment were Trichoderma spp., Azotobacter spp., Trichoderma spp + Azotobacter spp., with three replication.  The result of the experiment showed that interaction of biological agent and bokashi fertilizer affected the weight of 100 seeds and vigor index. The treatment that used Trichoderma spp. + Azotobacter spp. and bokashi fertilizer 48 ton/ha has showed that this dosage was the best for weight of 100 seeds parameter. Thus the treatment with trichoderma and azotobacter without bokashi fertilizer showed that this treatment affected the best for vigor index.Keyword: Soybean seeds, physiological quality, Trichoderma spp., Azotobacter spp., Bokashi.

Abstract

The use of high-yielding strawberry varieties such as Sweet Charlie needs to be followed by proper propagation methods to produce large amounts of strawberry seeds quickly and free from disease. In vitro propagation techniques can be done through the formation of a callus. This study aims to determine the effect of the combination of NAA and CaP treatment on the in vitro induction of the Sweet Charlie strawberry variety leaf callus. This experiment was arranged using a completely randomized design (CRD) which consisted of 9 treatments, namely concentrations of NAA (0 mg L-1, 5 mg L-1, 10 mg L-1) and CaP (0 mgL-1, 1 mgL-1, 2mg L-1) which was repeated three times. The results showed that using NAA 5 mgL-1 was the best treatment to support callus induction of the Sweet Charlie strawberry variety in vitro. The supporting with the parameter percentage of callus formation reaching 100%, callus appearance time at 13.5 days after planting (DAP), callus diameter of 9.00 mm, callus weight of 0.52 g, and the percentage of crumbly textured callus was 50%.

Abstract

Temu putih (Curcuma zedoaria) merupakan tanaman herbal yang memiliki potensi untuk mengobati berbagai penyakit seperti kanker dan tumor. Saat ini hampir 94% bahan baku obat tradisional berasal dari dalam negeri, namun penyediaan bahan baku industri obat tradisional masih terbatas. Teknik kultur jaringan dapat digunakan sebagai alternatif untuk produksi bibit dalam waktu yang singkat dengan jumlah yang banyak. Thidiazuron memiliki potensi memacu pembelahan sel secara cepat pada sel yang bersifat meristematik sehingga dapat meningkatkan multiplikasi tunas. Penggunakan TDZ dengan konsentrasi yang tinggi dapat menyebabkan tunas kerdil dan penghambatan pada pembentukan akar. Tujuan penelitian ini ialah untuk mengetahui pengaruh serta memperoleh konsentrasi terbaik TDZ terhadap proliferasi tunas temu putih. Penelitian dilaksanakan di Laboratorium Kultur Jaringan, Fakultas Pertanian, Universitas Padjadjaran, yang dirancang dengan Rancangan Acak Lengkap yang terdiri dari 5 perlakuan diantaranya Kontrol, 0 ppm; 0.3 ppm; 0.6 ppm dan 0.9 ppm TDZ yang diulang sebanyak tiga kali. Hasil penelitian menunjukkan Pemberian zat pengatur tumbuh thidiazuron berpengaruh pada terhadap jumlah tunas, jumlah akar, jumlah daun dan tinggi planlet temu putih. Pemberian 0.6 ppm TDZ menujukkan pengaruh yang baik terhadap multiplikasi tunas temu putih.

Abstract

Seed propagation of strawberry by in vitro culture is needed to obtain high quality seeds of strawberry in large quantities. In in vitro culture, a large number of shoots is expected can be produced at the multiplication stage. Cytokinins type of PGR can be added to culture media to produce a large number of shoot. The focus of this research was to compare the effect of BAP and Kinetin affected Sweet Charlie strawberry multiplication. The experiment was located at the Seed Technology Tissue Culture Laboratory, Faculty of Agriculture, Padjadjaran University. The experimental design was a Completely Randomized Design (CRD) with two treatments of different types of cytokinin, namely Benzyl Amino Purine (BAP) and Kinetin at 1 mg L-1 on Murashige and Skoog (MS) basic media, each treatment was repeated four times. This study showed that BAP had a better effect than Kinetin on the number of shoots, number of leaves, and wet weight of strawberry Sweet Charlie cultivar. The addition of BAP was recommended over Kinetin to obtain the highest number of shoots, the highest number of leaves, and the highest wet weight of culture on strawberry Sweet Charlie cultivar.

Abstract

Growth regulator is one of the important factors in the success of plant propagation by tissue culture. 6-Benzylaminopurine (BAP) is one of the most widely used cytokinin because it is more stable, less expensive and more effective. Thidiazuron (TDZ) can increase the action of other cytokinins, either exogenous cytokinin or endogenous cytokinin. 1-Naphthaleneacetic acid (NAA) belongs to the more stable auxin group, because it is not easily decomposed and oxidized by enzymes released by plants. This study aimed to obtain the best types and concentrations of BAP, TDZ, and NAA for the in vitro multiplication of strawberry explant. The experiment was conducted at the Seed Technology and Tissue Culture Laboratory, Faculty of Agriculture, Universitas Padjadjaran, from April to July 2022. The explants used were axillary shoots of strawberry var. Sweet Charlie is derived from plantlets that were propagated in vitro. The experimental design used was a completely randomized design with six treatments and four replications. The media used were Murashige dan Skoog (MS) with the addition of plant growth regulators (PGRs) in the form of BAP (0 ppm and 0,5 ppm), TDZ (0 ppm and 0,1 ppm), and NAA (0 ppm and 0,1 ppm). The application of different types and concentrations of BAP, TDZ, and NAA resulted in different effects of in vitro shoot multiplication of strawberry explant var. Sweet Charlie. Concentration of 0.1 ppm TDZ produced the highest number of shoots (5.38).

Abstract

The superior seeds are indicated by clean, good germination rate, pest and disease-free. One of the problems in providing superior tomato seeds is the presence of pulp attached to the tomato seeds, leading to less clean of seed. To remove the muscus on tomato seeds, the seeds have to be extracted. Other than seeds extraction, seeds exposure also could optimize seeds growth by increasing the metabolism of the tomato. The aim of this research is to determine the best extraction technique and magnetic field exposure for obtaining high tomato seeds germination. The complete random design used in this research consist of two factors, seeds extraction and magnetic field with three time replications. The result showed that seeds extraction for 24 hours is the best treatment for improving the maximum germination percentage, growth potential, growth rate, speed growth, uniformity index than others. Magnetic exposure at 6mT is the best treatment to improve seeds maximum growth and seeds speed growth.

Abstract

 The addition of cytokinin to the multiplication of white turmeric (Curcuma zedoaria) seedlings in vitro proved effective in shoot multiplication, but the addition of high cytokinin concentrations could cause stunted shoots and stunted growth. The addition of the hormone gibberellins (GA­3­) is often used in tissue culture for shoot elongation, so increasing the viability of plantlets. The experiment aims to determine the effect of GA3  and obtain the best concentrations of GA3 consisting of 0.50, 1, and 1.5 ppm GA3 on optimizing the growth of zedoary plantlets. The experimental parameters included the number of shoots, roots, leaves, plantlet height, and chlorophyll content observed at 6 WAP after subculturing. The results showed that giving 1 ppm GA3 had the best effect on optimizing plantlets' growth, namely the growth component of the number of shoots and plantlets' height.

Abstract

One of the internal factors that play an important role in seed quality is the level of the phytohormone auxin. Efforts to increase auxin in seeds include developing new varieties with increased indigenous auxin, such as those found in mutant tomatoes from Micro-Tom, namely iaa9-3 and iaa9-5. This research was conducted to determine the germination response of iaa9-3 and iaa9-5 mutant tomato seeds at different levels of the fruit maturation stage. The research was carried out at the Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University, and the Seed Testing Laboratory of the Center for Standard Testing of Vegetable Plant Instruments. The response design used was a completely randomized factorial design consisting of two factors and repeated three times. The first factor was the iaa9-3, iaa9-5, and Wild-Type Micro-Tom (WT-MT) mutants as controls. The second factor is the fruit maturation stage, which consists of Breaker, Pink, and Red. The research results showed that there was an interaction between genotype and fruit maturation stage on the parameters of germination, growth simultaneity, and hypocotyl length.  The harvest stage for tomatoes to produce normal strong seed germination was the pink stage in all tomato genotypes tested. The best germination rate was shown by WTMT seeds at the pink stage, genotype iaa9-3 at the red stage, and genotype iaa9-5 at the red or pink stage. The effect of fruit maturity stage on the synchronization of sprout growth was relatively not significantly different in mutant tomatoes, but it had an effect in WTMT tomatoes, namely the best pink stage.

Abstract

This study aims to determine the best interaction of potassium (K) fertilizer dose and retardant concentration on the growth, yield, and seed quality of G0 potato grown in the medium land of Jantinangor. The experiment was conducted at Screen House Station Ciparanje, Jatinangor, Faculty of Agriculture Universitas Padjadjaran from November 2022  until January 2023 at an elevation of ± 685 meters above sea level. The experimental design used was the Factorial Randomized Block Design (RBD). The first factor was the K fertilizer dose including 50%, 100%, and 150%. The second was the retardant concentration including without retardant, 100 ppm paclobutrazol, 100 ppm prohexadione-Ca, and 150 ppm prohexadione-Ca. The experimental results showed that there was an interaction effect between the K dose and retardant concentration. The interaction effect of 150% K and 100 ppm prohexadione-Ca showed the highest percentage of L class seed tuber and the lowest tuber weight loss. Independently, 150% K improved growth rate, number of tubers (6.67 knol/plant), and weight of tubers (73.33 g/plant). Independently, 100 ppm paclobutrazol concentration increased the chlorophyll content index. 150 ppm prohexadione-Ca concentration suppressed leaf area and plant height but increased stomatal conductance, fast emergence shoot time, and shoot length. 100 ppm prohexadione-Ca concentration produced the highest number of tubers and the weight of tubers, i.e., 6.78 knol/plant and 74.33 g/plant, respectively.

Abstract

Di Indonesia sebagian besar budidaya tomat dilakukan di dataran tinggi yang memiliki suhu rendah, namun pertumbuhan di sektor ekonomi yang cepat diikuti dengan perkebangan infrastruktur seperti bangunan dan jalan berdampak negatif terhadap pertumbuhan dan perkembangan tanaman. Pada tomat, perubahan temperatur yang melebihi suhu optimum menyebabkan cekaman panas. Cekaman panas akan menurunkan hasil dan kualitas tomat menyebabkan penurunan kualitas polen sehingga mempengaruhi keberhasilan proses penyerbukan dan pembuahan yang merupakan tahap awal proses pembentukan buah. Saat ini telah dilepas varietas baru tomat yang dapat tumbuh di daerah dataran rendah yang bersuhu tinggi yaitu varietas oval dan mutiara. Kedua varietas ini masih belum diketahui oleh masyarakat sekitar sehigga diperlukan kegiatan dalam rangka sosialisasi dan diseminasi tomat varietas baru beserta memperkenalkan teknik cara memproduksi benih tomat unggul. Kegiatan ini dilakukan di Desa Cieles Kecamatan Jatinangor Kabupaten Sumedang dan merupakan salah satu lokasi percobaan Universitas Padjadjaran dengan ketinggian 756 m di atas permukaan laut. Tujuan kegiatan ini adalah untuk melakukan sosialisasi produksi dan pengolahan benih tomat unggul yaitu varietas opal dan mutiara. Metode yang dilakukan pada kegiatan diantaranya kegiatan penyuluhan dan praktek secara langsung serta pendampingan.  Hasil kegiatan yang sudah dilakukan 1) pembuatan demplot percontohan pertanaman tomat yang ditanam oleh warga, (2) Praktek pengolahn benih tomat, (3) hasil survey informasi tentang tomat di masyarakat dan ketertarikannya. Setelah kegiatan berlangsung maka banyak warga yang mulai membudidayakan tanaman tomat varietas unggul ini.
