Abstract

This study aimed to analyze the microbial contamination of chicken meat and its pH value from traditional markets in Banyuwangi, East Java to ensure public health. A total of 30 raw chicken meat samples from three traditional markets were used in this study and examined for the presence of Escherichia coli and Salmonella spp. E. coli and Salmonella spp. were identified using Gram staining, culturing, and biochemical tests. The Most Probable Number (MPN) method was used for coliform and E. coli. Measuring the pH value of chicken meat was carried out after purchasing the meat and 24 hours after storage at cold temperatures. The results showed coliform contamination in all samples with an average of >1100 CFU/G with E. coli contamination of 20% (6/30), exceeding the maximum recommended limit. Furthermore, the identification of Salmonella spp. in all samples showed negative results. Chicken meat pH value decreased with the length of postmortem time; immediately after purchase (0 hours), the pH value was 5.78 ± 0.26, and after 24 hours of storage, it raised 6.02 ± 0.28. In conclusion, there was high coliform contamination out of 30 raw chicken meat samples taken from some traditional markets in Banyuwangi. Furthermore, E. coli contamination was as much as 20%, and Salmonella spp. was negative.

Abstract

Probiotics are live microbes that are used as supplementary feed that work to increase the good bacteria that live in the digestive tract by suppressing pathogenic microorganisms in the intestine so that good bacteria can grow can optimize the feed absorption process. This study aims to determine the effect of adding probiotics as feed additive Against the height and width of the duodenal villi of the laying chicken in early production. Probiotics can nourish the digestive tract and increase nutrient digestibility so that nutrient intake is met for livestock. Bacillus subtilis and Bacillus coagulans as a prophylaxis against bacterial diseases by balancing intestinal microbes, increasing digestibility, and secreting protease, lipase, and amylase enzymes. The purpose of this study was to determine the effect of probiotics Bacillus subtilis and Bacillus coagulans on duodenal histomorphology. Twenty-four layers were randomly divided into 4 groups. The control group was not given probiotics, groups P1, P2, and P3 were given probiotics of 2 ml, 4 ml, and 6 ml. Probiotics were given for 14 days. One-way ANOVA showed a significant difference in each group (P<0.05) and continued with the DUNCAN test. The average length and width of the duodenal villi in each treatment showed that the control group was significantly different from the P1, P2, and P3 groups. The P3 group had the highest average length of duodenal villi. The increase in villi height and villi width is due to the increase in short-chain fatty acids induced by probiotics. An increase in the height and width of villi indicates the absorption capacity of the small intestine, the work of digestive enzymes on the intestinal mucosa is more maximal in the intestine with a wider morphological size, so that absorption and growth are better. The conclusion from the research that has been done is that the use of Bacillus subtilis and Bacillus coagulans as probiotics has an effect on the layer duodenal villi which is characterized by an increase in the average length of the duodenal villi according to the increase in the probiotic dose.

Abstract

This study detected the presence of Brucella sp. antibodies in Sapera goats in Siliragung sub-district, Banyuwangi District using serological tests. The screening test used in this study was the Rose Bengal Test (RBT). Brucellosis causes goats to experience abortion and disruptive of reproduction system. The method used in this research is purposive sampling method using the criteria of sapera goats that have never been vaccinated. Using the blood serum of Sapera goats taken at the Siliragung subdistrict farm, Banyuwangi. The results of the study did not show any samples that were considered positive for Brucellosis in the Rose Bengal Test. In this study, no Brucella sp. antibodies were found in Sapera goats in Siliragung sub-district, Banyuwangi District.

Abstract

Laying breeds of chickens or commonly called layers are a breed of chicken that has potential value as an egg producer. The length of the layer production period is 80 – 90 weeks. During layer production, layers require high energy to produce, while the level of energy in the ration determines layer consumption. Various efforts have been made to increase the efficiency of using chicken feed in tropical areas, one of which is the use of additional feed additives. Feed additives that can be used are probiotics which come from microorganisms such as Lactobacillus acidophilus, Lactobacillus plantarum and Lactococcus lactis. This research aims to determine the effect and potential of adding a combination of probiotics Lactobacillus acidophilus, Lactobacillus plantarum, and Lactococcus lactis in increasing the quantity and quality of layer eggs during the production period. This research is experimental using a Completely Randomized Design (CRD). This study used a layer population of 20 individuals from the Isa Brown strain aged 40 weeks which were divided into four groups consisting of P1, P2, P3 and P4 with each group consisting of 5 individuals. The data that was collected was analyzed using Analysis of Variance (ANOVA) using SPSS version 24.0 software, then continued with the Duncan test on the data which showed significant differences (P<0.05). Based on the results of the study, the addition of the probiotics Lactobacillus acidophillus, Lactobacillus plantarum, and Lactococcus lactis in drinking water at doses of 2 mL/L, 4 mL/L, and 6 mL/L cannot increase the quantity of layer eggs, but can improve the external quality of layer eggs (egg weight, egg length, egg diameter, shell weight, shell thickness), and can improve the internal quality of layer eggs (yolk score, egg yolk index, egg white index, and haugh unit).

Abstract

This research was conducted to identify the occurrence of Anaplasma sp. infection in pigs (Sus scrofa) at Morgan Farm, Pahandut Subdistrict, Palangka Raya City. Palangka Raya City is one of the cities that has great potential to develop pig livestock, but the shortcomings of the farms in Palangka Raya City still apply traditional housing with poor cage sanitation. Samples in this study were obtained from Morgan Farm with a total of 68 pig blood samples, then examined at the Animal Health and Veterinary Public Health Laboratory of Palangka Raya City, then starting with the preparation of PBS solution and making blood review preparations using 10% Giemsa staining. The blood obtained was then reviewed using object glass, absolute methanol, and stained with Giemsa 10%. Identification was carried out using a trinocular microscope with a magnification of 400x and immersion oil was dripped on the object glass. The results of this research showed that 29 pig blood samples were positive out of 68 pig samples, with an occurrence rate of 42.6%. The morphology of Anaplasma sp. found is that there is a round, purplish-blue dot shape at the edge of red blood cells. From these results it can be concluded that the infection of Anaplasma sp. in pigs (Sus scrofa) at Morgan Farm, Pahandut District, Palangka Raya City is 42.6%, which means that the infection is very high for Palangka Raya City.

Abstract

This study is to evaluate the quality of milk generated by Ettawa crossbreed goats in Sempu District, Banyuwangi. A total of 30 raw milk samples were collected from Ettawa crossbreed goats, aged 3 to 4 years, across 6 different farms. The organoleptic testing method was employed to evaluate the physical properties of milk samples. Furthermore, tests for specific gravity and pH were conducted. Subsequently, the Total Plate Count (TPC) and Most Probable Number (MPN) methods were utilized to evaluate bacterial presence. The organoleptic evaluations validate that the goat's milk samples adhere to the Indonesian National Standard (SNI) 01-3141-2011 and the Thai Agricultural Standard (TAS) 6006-2008. The mean specific gravity of goat's milk is 1.0316 g/mL, whereas the average pH value is 6.45. The findings indicate that the specific gravity meets the criteria set out by the TAS 6006-2008. The pH value of 6.45 is below the threshold established by TAS. The mean TPC result complies with the Standard Limit SNI, which is under 1x10⁶ CFU/mL. Coliform levels in the milk samples adhere to SNI requirements (below 1 x 10² CFU/mL). The statistics indicate that the milk from the Ettawa crossbreed in Sempu District, Banyuwangi, satisfies the minimal standards set by TAS (2008) and SNI 7388-2009. Despite coliform levels conforming to the average standard, this examination indicated the presence of coliforms in multiple farms. Therefore, it is essential to provide heat treatment or pasteurization to the milk before its ingestion by the general populace.

Abstract

The Asian vine snake (Ahaetulla prasina) is an arboreal snake that can inhabit various types of trees in both lowland and highland areas. Based on its habitat, the Asian vine snake has a high potential to be infected by parasites. One of the parasitic diseases that can infect snakes is blood protozoa, which have the potential to be zoonotic. It can be transmitted between animals and humans through food contamination due to environmental pollution by snake feces which contain infective stage of protozoa. Clinical symptoms of this infection include lethargy, dehydration, and anorexia, which are common signs of parasitism. Case reports related to blood protozoa in snakes in the Indonesia are still very limited and have never been reported in Asian vine snake, even though many people keep Asian vine snake as exotic pets.

Abstract

Scabies is an infectious disease which is caused by Sarcoptes scabiei mites and considered as an emerging/re-emerging parasitic disease. Recent study using the Polymerase Chain Reaction (PCR) method has a higher sensitivity for detecting mites than using the skin scraping method. This study aims to detect the Second Internal Transcribed Spacer (ITS-2) encoding gene of S. scabiei. ITS-2 is a highly conserved area and always used to identify and differentiate host species infected by S. scabiei. S. scabiei isolates were obtained from skin scrapings from stray cat suspected of being infected with Scabies from Pasar Induk Bondowoso, Pasar Tamanan and Pasar Pujer. Detection on the presence of S. scabiei was carried out microscopically and then confirmed by molecular examination using the PCR method and visualized by electrophoresis on 2% agarose gel. The Polymerase Chain Reaction (PCR) results showed DNA bands at 304 bp Based on the result it can be concluded that samples are positively infected by Scabies can be detected by the ITS-2 encoding gene of Sarcoptes scabiei.

Abstract

Luka eksisi adalah luka yang disebabkan akibat terpotongnya jaringan oleh benda yang tajam (Partogi, 2008). Kitosan mampu sebagai zat dalam penyembuhan luka salah satunya dalam proses angiogenesis. Tujuan penelitian ini yaitu untuk mengetahui efektivitas salep kitosan dari limbah kulit udang terhadap angiogenesis dalam penyembuhan luka eksisi pada tikus putih jantan. Sebanyak dua puluh ekor tikus putih (Rattus norvegicus) jantan dibagi ke dalam lima kelompok perlakuan yaitu K+ luka eksisi dengan pemberian povidone iodine, K- luka eksisi dengan pemberian salep tanpa kitosan, P1 luka eksisi dengan pemberian salep kitosan 1,5%, P2 luka eksisi dengan pemberian salep 2,5% dan P3 luka eksisi dengan pemberian salep kitosan. Terapi diberikan sehari sekali selama tujuh hari. Hasil data rata-rata jumlah pembuluh darah baru dianalisis dengan uji Anova yang menunjukkan adanya perbedaan yang signifikan (p<0,05) dan dilanjutkan dengan uji LSD (Least Significant Differences). Kelompok P1 dan P2 tidak memiliki perbedaan yang nyata, tetapi berbeda nyata dengan kelompok K+, K- dan P3. Kesimpulan dari penelitian ini kitosan efektif dalam meningkatkan jumlah pembuluh darah baru pada proses penyembuhan luka eksisi.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui tingkat cemaran bakteri Salmonella sp. pada susu segar dari peternakan sapi perah di kecamatan licin kabupaten banyuwangi. Sampel susu segar di ambil pada april 2021 sebanyak 16 sampel dari KPSP Ijen makmur di kecamatan Licin Kabupaten Banyuwangi. Sampel diambil sebanyak 25 ml dari masing-masing peternak. Hasil yang di dapat setelah melakukan isolasi dan identifikasi menggunakan media Salmonella Shigella Agar (SSA), dan dilanjutkan uji identifikasi menggunakan TSIA dan Urea didapatkan  dari ke 16 sampel semua dinyatakn negatif bakteri Salmonella sp.. Hal ini dapat terjadi karena para peternak sapi perah di Kecamatan licin sudah mulai menyadari pentingnya kebersihan kandang dan sanitasi kandang yang baik sehingga tingkat cemaran bakteri Salmonella sp. dapat dicegah.
